Effects of oleuropein on pentylenetetrazol-induced seizures in mice: involvement of opioidergic and nitrergic systems.

Oleuropein, a well-known olive polyphenol, has been shown to mediate neuroprotection in Alzheimer's disease and cerebral ischemia. We investigated the effects of oleuropein on pentylenetetrazole (PTZ)-induced seizures in male NMRI mice, with diazepam as the standard drug. We also examined the possible involvement of opioidergic/nitrergic pathways in the probable effects of oleuropein. Intraperitoneal (i.p.) administration of different doses of oleuropein (10, 20 and 30 mg/kg) significantly increased the seizure threshold 60 min prior to induction of seizure, in a dose-dependent manner. Adminis

The Enteric Nervous System: An Integrative Hub From Development to Systemic Physiology.

The enteric nervous system (ENS) constitutes the largest and most functionally autonomous subdivision of the peripheral nervous system in vertebrates. It orchestrates stereotyped motor programs, secretomotor reflexes, and neuroimmune responses without central input. This review synthesizes evidence from developmental biology, mucosal immunology, and comparative physiology to advance a framework of enteric allostatic plasticity: the proposition that ENS functional resilience arises not from hard-wired reflex arcs but from a conserved capacity for transcriptional and morphological remodeling ins

Antibiotic-induced dysbiosis of the microbiota impairs gut neuromuscular function in juvenile mice.

BACKGROUND AND PURPOSE: Gut microbiota is essential for the development of the gastrointestinal system, including the enteric nervous system (ENS). Perturbations of gut microbiota in early life have the potential to alter neurodevelopment leading to functional bowel disorders later in life. We examined the hypothesis that gut dysbiosis impairs the structural and functional integrity of the ENS, leading to gut dysmotility in juvenile mice. EXPERIMENTAL APPROACH: To induce gut dysbiosis, broad-spectrum antibiotics were administered by gavage to juvenile (3weeks old) male C57Bl/6 mice for 14 day

G protein-coupled estrogen receptor is involved in modulating colonic motor function via nitric oxide release in C57BL/6 female mice.

BACKGROUND: Estrogen may regulate gastrointestinal motor functions, but the mechanism(s) is not totally understood. Here, we investigated whether G protein-coupled estrogen receptor (GPER/GPR30) was involved in regulating colonic motor functions and explored the underlying physiological mechanisms. METHODS: Adult female C57BL/6 mice were used. The expression and localization of GPER were examined by RT-PCR, western blot, and immuno-labeling. The role of GPER in modulating colonic motor functions was assessed by the bead propulsion test in vivo and organ bath experiments in vitro. KEY RESULTS

Nitrergic neuro-muscular transmission is up-regulated in patients with diverticulosis.

BACKGROUND: Neuro-transmission impairment could be associated to motility changes observed in patients with diverticular disease. Therefore, the objective was to characterize the inhibitory neuro-muscular transmission and gene expression changes of the enteric inhibitory pathways in patients with diverticulosis (DS). METHODS: Circular muscle strips from sigmoid colon of patients with DS and controls were studied using the organ bath technique to evaluate spontaneous contractility and enteric motor neurons stimulated by electrical field and qRT-PCR to assess the expression of nNOS, iNOS, P2Y1

In vitro motility disorders associated with displaced abomasum in dairy cows.

The objective of this study was to investigate in vitro abomasal motility in dairy cows diagnosed with displaced abomasum. Longitudinal muscle myenteric plexus preparations originating from the abomasal antrum of control cows, and cows diagnosed with left displaced abomasum (LDA), right displaced abomasum (RDA) or abomasal volvulus (AV) were used. In control preparations electrical field stimulation evoked an immediate cholinergic contractile response exceeding amplitude of basal contractions by 60%. In contrast, contractile activity was significantly inhibited during electrical stimulation in

Small Intestine Neuromuscular Dysfunctions and Neurogliopathy in a Mouse Model of High-Fat Diet-Induced Obesity: Involvement of Toll-Like Receptor 4.

Obesity is associated with enteric dysfunctions, including gut dysmotility and neurodegeneration, which may involve Toll-like receptor 4 (TLR4) signaling. To investigate this relationship, we examined the impact of TLR4 deficiency on the enteric nervous system (ENS) of the small intestine in a mouse model of high-fat diet (HFD)-induced obesity. Male TLR4-/- and wild-type (WT) C57BL/6J mice were fed either a standard diet (SD; 18% kcal fat) or an HFD (60% kcal fat) for 8 weeks. ENS alterations were evaluated using real-time qPCR and confocal immunofluorescence microscopy on longitudinal muscle-

Otilonium Bromide treatment prevents nitrergic functional and morphological changes caused by chronic stress in the distal colon of a rat IBS model.

Irritable bowel syndrome (IBS) is a highly prevalent gastrointestinal disorder characterized by periods of remission and exacerbation. Among the risk factors to develop IBS, psychosocial stress is widely acknowledged. The water avoidance stress repeatedly applied (rWAS) is considered effective to study IBS etio-pathogenesis. Otilonium bromide (OB), a drug with multiple mechanisms of action, is largely used to treat IBS patients. Orally administered, it concentrates in the large bowel and significantly ameliorates the IBS symptomatology. Presently, we tested whether rWAS rats developed neuro-mu

Ineffective esophageal motility and the vagus: current challenges and future prospects.

Ineffective esophageal motility (IEM) is characterized by low to very low amplitude propulsive contractions in the distal esophagus, hence primarily affecting the smooth muscle part of the esophagus. IEM is often found in patients with dysphagia or heartburn and is commonly associated with gastroesophageal reflux disease. IEM is assumed to be associated with ineffective bolus transport; however, this can be verified using impedance measurements or evaluation of a barium coated marshmallow swallow. Furthermore, water swallows may not assess accurately the motor capabilities of the esophagus, si

Glucagon-like Peptide-2 Depresses Ileal Contractility in Preparations from Mice through Opposite Modulatory Effects on Nitrergic and Cholinergic Neurotransmission.

Glucagon-like peptide-2 (GLP-2) has been reported to influence gastrointestinal motor responses, exerting a modulatory role on enteric neurotransmission. To our knowledge, no data on GLP-2 effects on the motility of the isolated ileum are available; therefore, we investigated whether GLP-2 affects the contractile activity of mouse ileal preparations and the neurotransmitters engaged. Ileal preparations showed tetrodotoxin (TTX)- and atropine-insensitive spontaneous contractile activity, which was unaffected by the nitric oxide synthesis inhibitor, L-NNA. GLP-2 depressed the spontaneous contrac

Sacral nerve stimulation increases gastric accommodation in rats: a spinal afferent and vagal efferent pathway.

Impaired gastric accommodation (GA) has been frequently reported in various gastrointestinal diseases. No standard treatment strategy is available for treating impaired GA. We explored the possible effect of sacral nerve stimulation (SNS) on GA and discovered a spinal afferent and vagal efferent mechanism in rats. Sprague-Dawley rats (450-500 g) with a chronically implanted gastric cannula and ECG electrodes were studied in a series of sessions to study: 1) the effects of SNS with different parameters on gastric tone, compliance, and accommodation using a barostat device; two sets of parameter

Nitric oxide and glutamate are contributors of anti-seizure activity of rubidium chloride: A comparison with lithium.

The neuro-protective effects of rubidium and lithium as alkali metals have been reported for different central nervous system dysfunctions including mania and depression. The aim of this study was evaluating as well as comparing the effects of rubidium chloride (RbCl) and lithium chloride (LiCl) on different seizures paradigms in mice and determining the involvement of NMDA receptors and nitrergic pathway. To assess the seizures threshold, animals received intravenous pentylenetetrazole (PTZ, 0.5%; 1 mL/min). Male NMRI mice (6-8 weeks) received intraperitoneal (i.p.) injections of different do

Escitalopram and Clonazepam Affect Gasotransmitter-related Enzyme (nNOS, HO-2, CBS) Gene Expression in the Rat Brain.

OBJECTIVES: Gasotransmitters, nitric oxide, carbon monoxide, and hydrogen sulfide, play vital roles in neuroplasticity, neurogenesis, and neuromodulation, as well as in regulating oxidative stress, neuroinflammation, and neurotoxicity, all of which are implicated in depressive disorders. This study aimed to investigate whether escitalopram, a selective serotonin reuptake inhibitor, and clonazepam, an anxiolytic Gamma-aminobutyric acid type A receptor agonist, influence the gene expression of enzymes involved in gasotransmitter synthesis in the rat brain. METHODS: Male Sprague-Dawley rats rece

Unique distribution of interstitial cells of Cajal in the feline pylorus.

The feline gastrointestinal (GI) tract is an important model for GI physiology but no immunohistochemical assessment of interstitial cells of Cajal (ICC) has been performed because of the lack of suitable antibodies. The aim of the present study was to investigate the various types of ICC and associated nerve structures in the pyloric sphincter region, by using immunohistochemistry and electron microscopy to complement functional studies. In the sphincter, ICC associated with Auerbach's plexus (ICC-AP) were markedly decreased within a region of 6-8 mm in length, thereby forming an interruption

Mucosal glucagon-like peptide 1 (GLP-1) responses are mediated by calcitonin gene-related peptide (CGRP) in the mouse colon and both peptide responses are area-specific.

BACKGROUND: Glucagon-like peptide (GLP)-1 is an incretin hormone and its mimetics are proven antidiabetic and antiobesity drugs. GLP-1 exerts antimotility and mucosal proliferative activities but its epithelial ion transport effects are uncharacterized and these may contribute to the gastrointestinal (GI) disturbance, i.e., diarrhea experienced with some GLP-1 mimetics. Our aim was to establish GLP-1 agonist mechanisms and identify potential mucosal mediator(s) in the colonic tissue from C57BL/6J mice. METHODS: A tissue survey of GLP-1 responses (using exendin 4, Ex4) and α-calcitonin gene-re

Activation of the dopamine D2 receptor enhances colonic motility via disinhibition of somatostatinergic signalling in rodents.

Dopamine (DA) plays an important role in regulating gastrointestinal (GI) motility; however, the specific contribution of colonic dopamine D2 receptor (D2 receptor) remains incompletely understood. This study investigated the role of D2 receptor in colonic motility using pharmacological approaches in D2 receptor-deficient (D2R-/-) mice and a 6-hydroxydopamine (6-OHDA) rats model of Parkinson's disease (PD). In wild-type mice, low-dose DA and D2 receptor agonist quinpirole enhanced colonic spontaneous contractility. This pro-contractile effect was completely abolished by tetrodotoxin, indicatin
