The role of cardiac energy metabolism in cardiac hypertrophy and failure.

In mammalian heart, incessant production of cellular energy is vital for maintaining continuous mechanical pumping function providing the body for oxygen and nutrients. To ensure this essential function, cardiac muscle adapt to increased energy demand or compromised energy supply by reprogramming the network of genes whose products are necessary to match the production of energy to consumption. Failure in this regulation leads to severe cardiac dysfunction and has been associated with cardiac pathogenesis including cardiac hypertrophy, failure and diabetes. Metabolic adaptations are induced by

A cold-inducible coactivator of nuclear receptors linked to adaptive thermogenesis.

Adaptive thermogenesis is an important component of energy homeostasis and a metabolic defense against obesity. We have cloned a novel transcriptional coactivator of nuclear receptors, termed PGC-1, from a brown fat cDNA library. PGC-1 mRNA expression is dramatically elevated upon cold exposure of mice in both brown fat and skeletal muscle, key thermogenic tissues. PGC-1 greatly increases the transcriptional activity of PPARgamma and the thyroid hormone receptor on the uncoupling protein (UCP-1) promoter. Ectopic expression of PGC-1 in white adipose cells activates expression of UCP-1 and key

Nanocomposite Scintillators Loaded With Hafnium Oxide and Phosphorescent Host and Guest for Gamma Spectroscopy.

Gamma sensitive plastic scintillators are of critical importance in the fields of nuclear nonproliferation, medical imaging, and high energy physics. However, there is often a trade-off between high light yield and high loading of high-Z components, both of which play an essential role in gamma ray detection. This work takes advantage of triplet exciton harvesting to increase gamma light yield by utilizing 1,3-di(9H-carbazol-9-yl)benzene and 9,9-dimethyl-9H-fluorene as triplet hosts to facilitate Dexter energy transfer to bis[2-(4,6-difluorophenyl)pyridinato-C2,N](picolinato)iridium(III) (FIrP

LXR stimulates a metabolic switch and reveals cholesterol homeostasis as a statin target in Tasmanian devil facial tumor disease.

Devil facial tumor disease (DFTD) and its lack of available therapies are propelling the Tasmanian devil population toward extinction. This study demonstrates that cholesterol homeostasis and carbohydrate energy metabolism sustain the proliferation of DFTD cells in a cell-type-dependent manner. In addition, we show that the liver-X nuclear receptor-β (LXRβ), a major cholesterol cellular sensor, and its natural ligand 24S-hydroxycholesterol promote the proliferation of DFTD cells via a metabolic switch toward aerobic glycolysis. As a proof of concept of the role of cholesterol homeostasis on DF

Accurate diabatization based on combined-hyperbolic-inverse-power-representation: 1,2 2A' states of BeH2.

A diabatic potential energy matrix (DPEM) for the two lowest states of BeH2+ has been constructed using the combined-hyperbolic-inverse-power-representation (CHIPR) method. By imposing symmetry constraints on the coefficients of polynomials, the complete nuclear permutation inversion symmetry is correctly preserved in the CHIPR functional form. The symmetrized CHIPR functional form is then used in the diabatization by ansatz procedure. The ab initio energies are reproduced with satisfactory accuracy. In addition, the CHIPR-based DPEM also reproduces the local topology of a conical intersection

Synthesis, Absorption and Emission Behaviour, Solvation, AIM, Topology, and Docking Studies on 4-(((perfluorophenyl) methylene)amino)-N-(thiazol-2-yl) Benzene Sulfonamide.

This work aims to synthesize 4-(((perfluorophenyl) methylene)amino)-N-(thiazol-2-yl) benzene sulfonamide (PFTH) and characterize it using various instrumental techniques such as UV-visible (UV-vis), fluorescence, Fourier-transform-infrared (FTIR), Raman, and nuclear magnetic resonance (NMR) spectroscopy analyses, respectively. The density functional theory (DFT) calculations have been performed using correlation-consistent polarized valence double-zeta (cc-pVDZ) basis sets and Becke, 3-parameter Lee-Yang-Parr (B3LYP) functionals, respectively. In this study, we used Gauge-Including Atomic Orbi

The rat cytochrome c oxidase subunit IV gene family: tissue-specific and hormonal differences in subunit IV and cytochrome c mRNA expression.

We have isolated three members of the rat cytochrome c oxidase subunit IV gene family: one functional gene and two processed pseudogenes. The pseudogenes appear to represent the only other closely related sequences in this family. The functional gene encodes an isoform which is expressed in all tissues examined and has features characteristic of 'housekeeping' genes. These include multiple transcription start sites mapped to within an approximately 50 bp region and a GC-rich promoter lacking typical CCAAT or TATAA sequences. Although the subunit IV gene is expressed at its highest levels in ca

The epidemiology of leukemias (past, present, future).

There are three periods of the history of the epidemiology of leukemias. The first period is characterized by 3 features: the description of the occupational leukemias (radiations, benzene) with more recently the war leukemias (Hiroshima), the leukemia induced by chemotherapy; the description of the leukemias of animals induced by viruses (chicken, mice and more over cats); and the concept of the geographical hematology. The second period may be called "Burkitt period" so important are the consequences of the discoveries made in Uganda. The third period is Japanese and American with the leukem

Relationship between cerebral interstitial levels of amino acids and phosphorylation potential during secondary energy failure in hypoxic-ischemic newborn piglets.

The aim of this study was to determine the validity of the hypothesis that excitatory amino acids are related to phosphorylation potential during primary and secondary cerebral energy failure observed in asphyxiated infants. We report here the results of experiments using newborn piglets subjected to severe transient cerebral hypoxia-ischemia followed by resuscitation. We examined cerebral energy metabolism by phosphorus nuclear magnetic resonance spectroscopy and changes in levels of amino acid neurotransmitters in the cortex by microdialysis before, during, and up to 24 h after the hypoxic-i

Observation of Long-Range Collective Flow in O+O and Ne+Ne Collisions and Implications for Nuclear Structure Studies.

The long-range collective flow of particles produced in oxygen-oxygen (O+O) and neon-neon (Ne+Ne) collisions is measured with the CMS detector at the CERN LHC. The data samples were collected at a center-of-mass energy per nucleon pair of 5.36 TeV, with integrated luminosities of 7  nb^{-1} and 0.8  nb^{-1} for O+O and Ne+Ne collisions, respectively. Two- and four-particle azimuthal correlations are measured over nearly five units of pseudorapidity. Significant elliptic (v_{2}) and triangular (v_{3}) flow harmonics are observed in both systems. The ratios of v_{n} coefficients between Ne+Ne an

Crossing boundaries, forging unity: nuclear medicine and science diplomacy in Cold War Europe.

This article explores the emergence of nuclear medicine as a clinical research field in post-war Europe, focusing on the shaping of its disciplinary boundaries in the context of geopolitical divisions. It examines how this speciality was negotiated and established, highlighting the role of international exchanges involving researchers, radioisotopes and technologies. By bringing together physicists, radiologists and internists, nuclear medicine gained momentum in the 1950s, leading to the formation of first dedicated scientific societies, conferences and journals. CAR: Discovery and development by the pharmacogenetics laboratory at NIEHS, NIH.

Our studies on the molecular mechanisms of phenobarbital induction of hepatic drug metabolism began in 1995 at the Pharmacogenetics Laboratory, National Institute of Environmental Health Sciences, NIH. Almost all pharmacology textbooks at the time described the phenomenon, but the mechanisms remained unknown. Within a few years, we revealed that the nuclear receptor CAR, constitutive active/androstane receptor, is the key molecule in these mechanisms. Since then, CAR research has branched into various directions. CAR is involved in controlling energy metabolism and has been shown to be an esse

Constraints on Solar Reflected Dark Matter from XENON1T and XENONnT Data.

We report on a search for sub-GeV dark matter upscattered via the solar reflection mechanism in the heavy mediator scenario. Under the standard halo model, keV-MeV dark matter produces nuclear recoils with energies below the detection threshold of liquid xenon time projection chambers. We enhance sensitivity to low-mass dark matter (DM) by considering dark matter-electron scattering, employing dedicated event selections to reduce the detection threshold and exploiting the additional kinetic energy imparted to the dark matter particle by solar upscattering. Using XENON1T ionization-only and XEN

Dataset for predicting single-spot proton ranges in proton therapy of prostate cancer.

The number of radiotherapy patients treated with protons has increased from less than 60,000 in 2007 to more than 220,000 in 2019. However, the considerable uncertainty in the positioning of the Bragg peak deeper in the patient raised new challenges in the proton therapy of prostate cancer (PCPT). Here, we describe and share a dataset where 43 single-spot anterior beams with defined proton energies were delivered to a prostate phantom with an inserted endorectal balloon (ERB) filled either with water only or with a silicon-water mixture. The nuclear reactions between the protons and the silico

Pancreatic involvement in co-infection visceral leishmaniasis and HIV: histological and ultrastructural aspects.

The involvement of the gastrointestinal tract in the co-infection of HIV and Leishmania is rarely reported. We report the case of an HIV-infected adult man co-infected with a disseminated form of leishmaniasis involving the liver, lymph nodes, spleen and, as a feature reported for the first time in the English literature, the pancreas. Light microscopy showed amastigote forms of Leishmania in pancreatic macrophages and immunohistochemical staining revealed antigens for Leishmania and also for HIV p24. Microscopic and ultrastructural analysis revealed severe acinar atrophy, decreased zymogen gr

A key role of nuclear factor Y in the refeeding response of fatty acid synthase in adipocytes.

Fatty acid synthase (Fasn) is a key component of energy metabolism that is dynamically induced by food intake. Although extensive studies have revealed a number of transcription factors involved in the fasting/refeeding transition of Fasn expression in hepatocytes, much less evidence is available for adipocytes. Using the in vivo Ad-luc analytical system, we identified the inverted CCAAT element (ICE) around -100 nucleotides in the Fasn promoter as a critical cis-element for the refeeding response in adipocytes. Electrophoretic mobility shift assays and chromatin immunoprecipitation show that

Mitochondrial-nuclear crosstalk, haplotype and copy number variation distinct in muscle fiber type, mitochondrial respiratory and metabolic enzyme activities.

Genes expressed in mitochondria work in concert with those expressed in the nucleus to mediate oxidative phosphorylation (OXPHOS), a process that is relevant for muscle metabolism and meat quality. Mitochondrial genome activity can be efficiently studied and compared in Duroc and Pietrain pigs, which harbor different mitochondrial haplotypes and distinct muscle fiber types, mitochondrial respiratory activities, and fat content. Pietrain pigs homozygous-positive for malignant hyperthermia susceptibility (PiPP) carried only haplotype 8 and showed the lowest absolute mtDNA copy number accompanied

Oxidation State Dependence of the Actinide M4/5-Edge XANES of Actinyl (An = U, Np, Pu) Systems.

Restricted Active Space simulations including spin-orbit coupling are used to predict the An M4/5-edge XANES spectra of uranyl, neptunyl and plutonyl ions in the +5 and +6 oxidation states. The simulations reproduce the characteristic red-shift in XANES upon lowering the actinide oxidation state. This shift arises from relative changes in the ground- and core-excited state (CES) energies between An(VI) and An(V), which can be interpreted in terms of electron affinities. QTAIM analysis links the electron affinity of An(VI) states to actinide electron localization and to the more familiar concep

Ultrafast solvent-to-solute proton transfer mediated by intermolecular coherent vibrations.

Ultrafast photoinduced excited-state proton transfer (ESPT) plays a crucial role in protecting biomolecules and functional materials from photodamage. However, the influence of solute-solvent interactions on ESPT dynamics remains under active investigation. Here, we present an ultrafast spectroscopic study of ESPT in the photobase 2-(2´-pyridyl)benzimidazole (PBI) in methanol. Ultrafast absorption spectroscopy, supported by quantum chemical calculations, reveals three distinct kinetic steps: (1) a 2.2 ps solvent-to-solute proton transfer, (2) subsequent nonradiative relaxation to the ground st
