Quinovic acid glycosides from Mitragyna stipulosa--first examples of natural inhibitors of snake venom phosphodiesterase I.

Phytochemical investigations on the non-alkaloidal extracts of Mitragyna stipulosa bark has led to the isolation of a series of triterpenoids mainly consisting of quinovic acid ([structure: see text]) and its glycoside derivatives [structure: see text] and [structure: see text]. The other constituents isolated include alpha-amyrin, 3beta-acetyl ursolic acid and a mixture of oleanolic and ursolic acid and beta-sitosterol glucopyranoside. Their structures were identified by spectral and chemical studies and compounds [structure: see text] and [structure: see text] were, respectively, identified

Anti-inflammatory and antioxidant activities and constituents of Platostoma africanum P. Beauv.

Hexane and dichloromethane extracts of Platostoma africanum P. Beauv., a plant popularly employed in Nigeria in the treatment of rheumatic s … investigation of the extracts afforded eight acidic pentacyclic triterpenes, namely, ursolic acid, oleanolic acid, epimaslinic acid, maslinic acid, corosolic acid, hyptadienic, euscaphic acid and tomentic acid, and a mixture of beta-sitosterol and stigmasterol. These constituents are reported for the first time in the genus.

Overexpression of BcERF3 increases the biosynthesis of saikosaponins in Bupleurum chinense.

Chaihu, the dried roots of some species of Bupleurum L., is a famous Chinese herbal medicine for treatment of liver- and cold-related diseases, in which saikosaponins (SSs) are the major active compounds. Many of the genetic components upstream of SS biosynthetic pathways have been characterized; however, the regulatory mechanisms remain elusive. In this study we identified the APETALA2/Ethylene Responsive Factor family transcription factor gene BcERF3 from B. chinense. The expression of BcERF3 was induced in methyl-jasmonate-treated adventitious root of B. chinense; it was also expressed at h

Optimized plant compound with potent anti-biofilm activity across gram-negative species.

Many human diseases, including cystic fibrosis lung infections, are caused or exacerbated by bacterial biofilms. Specialized modes of motility, including swarming and twitching, allow gram-negative bacteria to spread across surfaces and form biofilms. Compounds that inhibit these motilities could slow the spread of biofilms, thereby allowing antibiotics to work better. We previously demonstrated that a set of plant-derived triterpenes, including oleanolic acid and ursolic acid, inhibit formation of Escherichia coli and Pseudomonas aeruginosa biofilms, and alter expression of genes involved in …

Two new hepaprotective saponins from Semen celosiae.

Two new oleanolic acid saponins, namely celosin C (1) and celosin D (2), were isolated from the ethanol extract of Semen celosiae. Their structures were identified as celosin C (1) and celosin D (2) by spectroscopic and chemical analyses. The hepatoprotective activity of 1 and 2 with oral dose 1.0, 2.0 and 4.0 mg/kg, respectively, were investigated by carbon tetrachloride (CCl(4))-induced hepatotoxicity in mice. The results indicate that they have significant hepatoprotective effects (p<0.01).

Synthesis of Quillaic Acid through Sustainable C-H Bond Activations.

To meet the demand for quillaic acid, a multigram synthesis of quillaic acid was accomplished in 14 steps, starting from oleanolic acid, leading to an overall yield of 3.4%. Key features include C-H activation at C-16 and C-23. Through Pd-catalyzed C-H acetoxylation, the oxidation at C-23 was observed as the major product, as opposed to at C-24. A copper-mediated C-H hydroxylation using O2 successfully afforded the single isomer, 16β-ol triterpenoid, followed by configuration inversion to the desired 16α-ol compound. In summary, with steps optimized and conducted on a multigram scale, quillaic…

Bioactive-Guided Isolation and Optimization of Luffa Acutangula Nanoemulsion for In vitro, In vivo, and In silico Cholinesterase Inhibition for Alzheimer's Disease Management.

Alzheimer's disease (AD) progressively impairs memory and cognition. Luffa acutangula (LA), rich in triterpenoids, fatty acids, and iridoid glycosides, exerts anti-Alzheimer's effects by inhibiting acetylcholinesterase (AChE). This study aims to isolate bioactive compounds from LA fruits, formulate and optimize a nanoemulsion to improve brain targeting, and assess its anti-Alzheimer efficacy through in vitro, in vivo, and in silico approaches. LA was extracted using a 1:1 hydro-ethanol mixture and subsequently underwent chromatographic isolation to yield four major constituents: oleanolic acid…

Evaluation of different ecological regions for cultivation of best quality Bupleurum: a case study from Shanxi, China.

Bupleurum, a plant of the genus Bupleurum L. in the family Umbelliferae, is prevalent and extensively applied in traditional medicine systems across East and Southeast Asian countries for the treatment of colds, malaria, hepatitis and other diseases. In the current Chinese herbal medicine market, only the dried roots of two species, Bupleurum chinense DC. and Bupleurum scorzonerifolium Willd., are authentic herbs, and their cultivars dominate as the commercial source, contributing to about 80% of the market share. Shanxi Province, known as the suitable habitat for Bupleurum in China, has a div

In Vitro Anti-Toxoplasma Activity of Extracts Obtained from Tabebuia rosea and Tabebuia chrysantha: The Role of β-Amyrin.

Toxoplasmosis is a parasitic disease caused by the protozoan Toxoplasma gondii that is highly prevalent worldwide. Although the infection is asymptomatic in immunocompetent individuals, it severely affects immunocompromised individuals, causing conditions such as encephalitis, myocarditis, or pneumonitis. The limited therapeutic efficacy of drugs currently used to treat toxoplasmosis has prompted the search for new therapeutic alternatives. The aim of this study was to determine the anti-Toxoplasma activity of extracts obtained from two species of the genus Tabebuia. Twenty-six extracts, 12 ob

Synthesis and anti-influenza activity of a series of triterpene furoyloxy-acylates.

A series of lupane, oleanane and ursane furoyloxy-triterpenoids were synthesised via acylation with 2-furoyl chloride and subsequently tested for anti-influenza activity against the pandemic influenza virus A/Puerto Rico/8/1934 (H1N1). Among them, 3β-furoyloxy-oleanolic and 3β-furoyloxy-ursolic acids were the most potent with IC50 values 17 and 1 µM with selectivity indexes of 32 and 26, respectively. The molecular structure of compound 11 was determined by X-ray diffraction analysis.

Glutaminase inhibitory activities of pentacyclic triterpenes isolated from Thymus vulgaris L.

Glutaminase is an important target that is often over expressed in neurodegenerative and lifestyle related diseases but few effective inhibitors of this enzyme have yet reached clinical trials. Ursolic acid (1), betulinic acid (2) and oleanolic acid (3), three pentacyclic triterpene acids, have been isolated from the leaves of Thymus vulgaris L. Enzyme inhibition experiments demonstrated their inhibitory effects against glutaminase activity. Compound 2 significantly inhibited the glutaminase activity with IC50 of 0.31 mM, stronger than the positive control 6-diazo-5-oxo-L-norleucine (DON) with…
