Evidence for Ca(2+)-permeable AMPA receptors in the olfactory bulb.

alpha-Amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid (AMPA) receptors (AMPARs), a subtype of glutamate receptor, contribute to olfactory processing in the olfactory bulb (OB). These ion channels consist of various combinations of the subunits GluR1-GluR4, which bestow certain properties. For example, AMPARs that lack GluR2 are highly permeable to Ca(2+) and generate inwardly rectifying currents. Because increased intracellular Ca(2+) could trigger a host of Ca(2+)-dependent odor-encoding processes, we used whole cell recording as well as histological and immunocytochemical (ICC) techniques

Expression pattern of alpha CaMKII in the mouse main olfactory bulb.

Multifunctional Ca(2+)/calmodulin-dependent protein kinase II (CaMKII) is highly enriched at synapses and has been implicated in regulating the formation and function of several sensory systems, including the visual and the somatosensory systems. Although there is evidence for CaMKII expression in the olfactory system, the cellular localization of CaMKII has not been well studied and its function remains unknown. In this study, we examined the normal expression patterns of the predominant alpha CaMKII in the mouse main olfactory bulb. We showed that alpha CaMKII expression levels were high in

Optical imaging of odor preference memory in the rat olfactory bulb.

Early olfactory preference learning in rat pups occurs when novel odors are paired with reinforcing tactile stimulation that activate the noradrenergic locus coeruleus. Pairing of odor and a noradrenergic agonist in the olfactory bulb is both necessary and sufficient for odor preference learning. This suggests the memory change occurs in the olfactory bulb. Previous electrophysiological experiments demonstrated that odor preference training induces an increase in the field excitatory postsynaptic potential to olfactory nerve input and an alteration, after training, in glomerular [14C]2- deoxyg

Long-lasting depolarizations in mitral cells of the rat olfactory bulb.

We investigated the mechanisms of long-lasting depolarizing potentials (LLDs) generated in mitral cells with whole-cell patch recordings in the rat olfactory bulb slice. LLDs occur spontaneously and are evoked by either orthodromic stimulation of the olfactory nerve or antidromic stimulation of mitral and tufted (M/T) cells. LLDs are followed by a long refractory period, limiting LLD generation to approximately 1 Hz. LLD production does not appear to involve either intrinsic voltage-activated or metabotropic mechanisms. The initiation of LLDs requires activation of non-NMDA but not NMDA recept

Sensory deafferentation and olfactory bulb morphology in the zebrafish and related species.

The zebrafish, Danio rerio, has become an important model species for examining olfactory system structure and function, yet little is known about developmental changes in olfactory bulb morphology from embryo to adult. The present study examined both normal growth and the effects of deafferentation on the bulb from hatching to adulthood. In young animals, the bulb is small relative to body size and has a higher percentage of its volume occupied by incoming olfactory nerve fibers. Young animals are also more affected by sensory deafferentation. Olfactory rosette removal resulted in more than 5

Dendrodendritic recurrent excitation in mitral cells of the rat olfactory bulb.

Most neuronal interactions within the olfactory bulb network are mediated by dendrodendritic synapses. Dendritic transmitter release potentially could affect the parent dendrite as well as local neuronal elements that have receptors for the released transmitter. Here we report that under conditions that facilitate N-methyl-D-aspartate (NMDA) receptor activity (reduced GABAA inhibition and extracellular Mg2+), a single action potential evoked by brief intracellular current pulses in mitral cells is followed by a prolonged depolarization, which is blocked by an NMDA receptor antagonist. This dep

Distribution and characteristics of cholecystokinin-like immunoreactivity in the olfactory bulb of the cat.

The distribution and characteristics of cholecystokinin (CCK)-like immunoreactive (LIR) nerve fibers was examined in the cat olfactory bulb using immunohistochemistry. CCK-LIR cell bodies were not found; fine varicose CCK-LIR fibers were observed in the most layers. In the main olfactory bulb, no staining was seen in the olfactory nerve layer and white matter. The directions of CCK-LIR fibers in the glomerular, external plexiform and mitral cell layers were generally perpendicular to the bulbar surface; those in the deep granule cell layer were parallel; and those in the internal plexiform and

Glutamatergic deafferentation of olfactory bulb modulates the expression of mGluR1a mRNA.

Glutamate (Glu) released by olfactory nerve axons acts on postsynaptic ionotropic and metabotropic glutamate receptors expressed by principal neurones and interneurones of the olfactory bulb (OB). Using ZnSO4 lesioning of the rat olfactory mucosa and semiquantitative RT-PCR, we examined the effect of removal of the glutamatergic input to the OB on the expression of mGluR1a, mGluR1b and GluR1 mRNAs. Two days after lesioning, mGluR1a mRNA levels in OB increased by 45%. At this time, the expression of tyrosine hydroxylase (TH) mRNA, which is strictly dependent on olfactory nerve input, was still

Excitatory synapses in the glomerular triad of frog olfactory bulb in vitro.

Whole-cell patch clamp recording techniques were applied to periglomerular (PG) cells in slices of the frog olfactory bulb (OB) to study the properties of the excitatory synapses in the triad formed by the olfactory nerve (ON) and the dendrites of mitral/tufted (MT) cells and PG cells. The postsynaptic response evoked by ON stimulation was glutamatergic and could be dissected into NMDA and non-NMDA components of equivalent amplitudes. The dendro-dendritic synapse between MT and PG cells could be activated following antidromic stimulation of the lateral and medial olfactory tract (LOT and MOT).

Organization of inhibition in the rat olfactory bulb external plexiform layer.

1. Intracellular recordings were made from the output neurons (mitral and tufted cells) of the rat olfactory bulb during electrical orthodromic stimulation of the olfactory nerve layer (ONL) and antidromic stimulation of the lateral olfactory tract and posterior piriform cortex (pPC) to test for physiological differences among the neuron types. Many of these neurons were identified by intracellular injections of biocytin, and others were identified by their pattern of antidromic activation. 2. Both marked and unmarked mitral cells showed large inhibitory postsynaptic potentials (IPSPs) in resp

[GABA immunoreactivity in the main olfactory bulb of the frog Rana temporaria].

Immunoreactivity for gamma-aminobutyric acid (GABA) was localized at the light microscopic level in the main olfactory bulb (MOB) of the frog, Rana temporaria. By means of free-floating peroxidase-antiperoxidase immunocytochemical technique, GABA was found in a large number of neurons in the granular cell layer, in a few small somata in the mitral cell layer and in two different types of cell somata in the glomerular layer. Individual GABA-immunopositive cells were found in the olfactory nerve layer. GABA immunostaining was also localized in cell processes and fiber fragments. There were many

Development of the nerve fiber layer in the olfactory bulb of mouse embryos.

The purpose of this study was to provide an ultrastructural description of the development of the olfactory nerve fiber layer (ONL) in the olfactory bulbs of mouse embryos. The developmental age of each embryo was determined by means of the Theiler staging system. During stages 17 and 18, the olfactory axons grew through the mesenchyme toward the cerebral vesicle; they were accompanied by a group of electron-dense cells which were referred to as peripheral glial progenitor cells (GPCs) because they ensheathed the adjacent axons. In Theiler stage 19-21 embryos, the axons and the peripheral GPCs

Electrophysiological mapping of the accessory olfactory bulb of the rabbit (Oryctolagus cuniculus).

Field potentials elicited by electrical stimulation of the vomeronasal nerve were measured in the accessory olfactory bulb of the rabbit. Maps were made of the distribution of surface field potentials and of the corresponding depth profiles. The surface maps followed closely the contours of the accessory olfactory bulb: at the frontal border the field potential tended to zero and at the center of the structure the field potential attained a maximum. Depth profiles of the field potentials through the accessory olfactory bulb presented a surface-negative wave and, in depth, a positive wave. The

An intracellular analysis of dendrodendritic inhibition in the turtle in vitro olfactory bulb.

1. Intracellular recordings from an in vitro preparation of turtle olfactory bulb were used to determine the pathway responsible for producing synaptic inhibition of mitral cells.2. Inhibitory post-synaptic potentials (i.p.s.p.s) could be elicited in mitral cells by both olfactory nerve (orthodromic) and mitral cell axon (antidromic) stimulation and by suprathreshold depolarizing current pulses injected intracellularly through the recording micro-electrode. Reversing the chloride gradient by either intracellular injection of chloride or lowering the external chloride concentration reversed the

[The effect of temperature changes on evoked potentials of the fish olfactory bulb].

Perches were kept at 10 degrees C and fixed in a chamber through which water was perfused. During the experiment the water temperature was changed. The potentials induced by electric stimulation of the olfactory nerve were recorded from the dorsal surface of the olfactory bulb. The oscillations of the evoked potentials separated into several components when the temperature was lowered. Six components were found in the evoked response of the olfactory bulb at 8 degrees C.
