Development of NADPH-diaphorase expression in chemosensory systems of the opossum, Monodelphis domestica.

Using NADPH-diaphorase histochemistry, the present study describes development of olfactory and vomeronasal systems in postnatal opossums, Monodelphis domestica. NADPH-dependent staining is absent at and around the time of birth. By 2 weeks of age and through adulthood, intense staining is seen along the luminal surface of the olfactory epithelium (OE) and of the vomeronasal sensory epithelium (VNE), as well as in Bowman's glands of the OE. Staining of the adult VNE is not homogeneous; it is restricted to the superficial 2/3 of the epithelium. At 2 weeks of age, staining in the brain is seen o

Cross-adaptation to odor stimulation of olfactory receptor cells in the box turtle, Terrapene carolina.

Electrical recording from small twigs of olfactory nerve and electro-olfactogram (EOG) from olfactory epithelium in a turtle shows that olfactory receptors in the nose are responsive to various odors. I have used the effects of cross-adaptation to odor stimulation on the olfactory receptors to investigate the stimulus-specific components of these responses and to provide information about the responsiveness of cells. The results of the cross-adaptation experiments strongly support the hypothesis that different categories of receptor cells exist in the olfactory epithelium.

[Nature of the impulse activity in the anterior portions of the fish olfactory bulb].

The impulse activity of anterior portions of the fish olfactory bulb depends on coming of signals from the receptors as shown by the fact that action of novokain on the receptors entirely suppressed the activity. A direct application of CoCl2 solution (1--1.5 mM) had no effect on either spontaneous or evoked by adequate stimulation impulse activity in these areas which indicates its presynaptic, i.e. the receptor nature. The surface structures of the bulb anterior portions did not respond to electric stimulation of the olfactory tract while the electric stimulation of these structures evoked a

Electrophysiological evidence for a topographical projection of the nasal mucosa onto the olfactory bulb of the frog.

Three olfactory nerve branches respectively subserving either a medial, an intermediate, or a lateral region of the dorsal olfactory receptor sheet of the bullfrog Rana catesbeiana were electrically stimulated with bipolar platinum hook electrodes. Extracellular single unit responses from 93 second-order cells in different regions of the olfactory bulb were recorded with metal-filled glass micropipets. The excitatory responsiveness of each unit to the stimulation of each of the three nerve branches (response profile) was determined. Some units were sensitive to stimulation of each of the three

Olfactory Development, Part 2: Neuroanatomic Maturation and Dysgeneses.

Olfactory axons project from nasal epithelium to the primitive telencephalon before olfactory bulbs form. Olfactory bulb neurons do not differentiate in situ but arrive via the rostral migratory stream. Synaptic glomeruli and concentric laminar architecture are unlike other cortices. Fetal olfactory maturation of neuronal differentiation, synaptogenesis, and myelination remains incomplete at term and have a protracted course of postnatal development. The olfactory ventricular recess involutes postnatally but dilates in congenital hydrocephalus. Olfactory bulb, tract and epithelium are reposito

[Effect of chronic anosmia on morphofunctional parameters of the gustatory system primary centers of the carp Cyprinus carpio].

Morphofunctional state of neurons in primary centers of olfactory, external and intraoral gustatory systems in carp yearlings was studied at different time after bilateral anosming, with use of methods of morphometrical, biochemical, and histochemical analyses. The olfactory deafferentation produces centripetal translocation of olfactory bulbs and development of their degenerative changes. Chronic elimination of olfactory reception in carps leads to activation of neurons in the facial lobe of medulla--the primary center of the external gustatory system. The neuronal complexes of the vagus lobe

Axonal redirection at the dorsoventral intraretinal boundary.

Cobaltous-lysine applied to the goldfish optic nerve backfilled retinal ganglion cells and their axons. Confined to the ventronasal and ventrotemporal retina was a small population of retinal ganglion cells whose axons traveled dorsally and parallel to the retinal margin. On reaching the boundary between dorsal and ventral retina, the axons arched, joined radially oriented bundles of axons, and traveled toward the optic disk. Control studies showed that the axons came from retinal ganglion cells rather than from retinopetal cells. The somatic area of retinal ganglion cells (RGCs) with circumfe

Sense of Smell: Structural, Functional, Mechanistic Advancements and Challenges in Human Olfactory Research.

Olfaction, the sense of smell detects and discriminate odors as well as social cues which influence our innate responses. The olfactory system in human beings is found to be weak as compared to other animals; however, it seems to be very precise. It can detect and discriminate millions of chemical moieties (odorants) even in minuscule quantities. The process initiates with the binding of odorants to specialized olfactory receptors, encoded by a large family of Olfactory Receptor (OR) genes belonging to the G-protein-coupled receptor superfamily. Stimulation of ORs converts the chemical informa

Neurotrophins are not required for normal embryonic development of olfactory neurons.

Neurons of the vertebrate olfactory epithelium (OE) regenerate continuously throughout life. The capacity of these neurons to regenerate and make new and precise synaptic connections in the olfactory bulb provides a useful model to study factors that may control or mediate neuronal regeneration. Expression and in vitro studies have suggested potential roles for the neurotrophins in the olfactory system. To directly examine whether neurotrophins are required for olfactory neuron development, we characterized in vivo the role of the neurotrophins in the primary olfactory system. For this, we gen

Promoting central nervous system regeneration: lessons from cranial nerve I.

The olfactory nerve differs from cranial nerves III-XII in that it contains a specialised type of glial cell, called 'olfactory ensheathing cell' (OEC), rather than Schwann cells. In addition, functional neurogenesis persists postnatally in the olfactory system, i.e. the primary olfactory pathway continuously rebuilds itself throughout adult life. The presence of OECs in the olfactory nerve is thought to be critical to this continuous growth process. Because of this intrinsic capacity for self-repair, the mammalian olfactory system has proved as a useful model in neuroregeneration studies. In

Mechanism of copper nanoparticle toxicity in rainbow trout olfactory mucosa.

Chemosensory perception is crucial for fish reproduction and survival. Direct contact of olfactory neuroepithelium to the surrounding environment makes it vulnerable to contaminants in aquatic ecosystems. Copper nanoparticles (CuNPs), which are increasingly used in commercial and domestic applications due their exceptional properties, can impair fish olfactory function. However, the molecular events underlying olfactory toxicity of CuNPs are largely unexplored. Our results suggested that CuNPs were bioavailable to olfactory mucosal cells. Using RNA-seq, we compared the effect of CuNPs and copp

[Neural circuit analysis using isolated whole brain preparation].

Many electrophysiological studies have been conducted on the 'limbic system', which is known to play a crucial role in mnemonic, emotional, and olfactory functions. Because of the difficulty in accessing such deep brain structures under in vivo conditions, in vitro brain slice preparations are often used. Recertly, an isolated whole brain preparation in which multi-synaptic circuits and the intracellular activity they generate are well preserved has also become available. In this paper, we described about the studies of the olfactory circuits by using this unique preparation. This experimental

Afferent and efferent connections of the nucleus sphericus in the snake Thamnophis sirtalis: convergence of olfactory and vomeronasal information in the lateral cortex and the amygdala.

This paper is an account of the afferent and efferent projections of the nucleus sphericus (NS), which is the major secondary vomeronasal structure in the brain of the snake Thamnophis sirtalis. There are four major efferent pathways from the NS: 1) a bilateral projection that courses, surrounding the accessory olfactory tract, and innervates several amygdaloid nuclei (nucleus of the accessory olfactory tract, dorsolateral amygdala, external amygdala, and ventral anterior amygdala), the rostral parts of the dorsal and lateral cortices, and the accessory olfactory bulb; 2) a bilateral projectio

Spatiotemporal patterns of expression of extracellular matrix molecules in the developing and adult rat olfactory system.

Using immunocytochemical methods, we have examined extensively the spatial and temporal patterns of expression of three extracellular matrix molecules-laminin, fibronectin, and type IV collagen-in the embryonic, postnatal (days 2 and 11) and adult rat olfactory system. The study started at embryonic day 14 when olfactory fibres and their associated migrating cells course through the nasal mesenchyme. From embryonic day 14 to the adult, a sheet-like pattern of labelling for laminin, fibronectin and type IV collagen was observed along the basal surface of the olfactory epithelium and around the

Olfactory-induced locomotion in lampreys.

The olfactory system allows animals to navigate in their environment to feed, mate, and escape predators. It is well established that odorant exposure or electrical stimulation of the olfactory system induces stereotyped motor responses in fishes. However, the neural circuitry responsible for the olfactomotor transformations is only beginning to be unraveled. A neural substrate eliciting motor responses to olfactory inputs was identified in the lamprey, a basal vertebrate used extensively to examine the neural mechanisms underlying sensorimotor transformations. Two pathways were discovered fro

The larval Drosophila mushroom body balances lateralized sensing and interhemispheric integration.

Bilaterian animals must integrate sensory input from both sides of their bodies to make coherent perceptual decisions. In Drosophila larvae, olfactory receptor neurons target the ipsilateral brain hemisphere. Using calcium imaging, unilateral sensory perturbations, connectome analysis, and optogenetics, we identified the mushroom body (MB) as a key substrate for interhemispheric integration. Kenyon cell odor responses were almost entirely ipsilateral, indicating minimal coupling between the two MBs at the input level. In contrast, modulatory MB input neurons (MBINs) responded symmetrically to

Primary and secondary olfactory centres in human ontogeny.

The olfactory centres are the evolutionary oldest and most conservative area of the telencephalon. Olfactory deficiencies are involved in a large spectrum of neurologic disorders and neurodegenerative diseases. The growing interest in human olfaction has been also been driven by COVID-19-induced transitional anosmia. Nevertheless, recent data on the human olfactory centres concerning normal histology and morphogenesis are rare. Published data in the field are mainly restricted to classic studies with non-uniform nomenclature and varied definitions of certain olfactory areas. While the olfactor

Olfactory ensheathing cells: Unique glial cells promising for treatments of spinal cord injury.

Spinal cord injury (SCI) is generally the consequence of physical damage, which may result in devastating consequences such as paraplegia or paralysis. Some certain candidates for SCI repair are olfactory ensheathing cells (OECs), which are unique glial cells located in the transition region of the peripheral nervous system and central nervous system and perform neuron regeneration in the olfactory system throughout life. Culture studies have clarified many properties of OECs, but their mechanisms of actions are not fully understood. Successful results achieved in animal models showcased that

What are Probable Olfactory Markers of Alcohol Dependence?

As the olfactory system is closely connected with the brain areas responsible for the most crucial alterations in psychiatric populations, especially cognitive and emotional impairments, the study of olfactory processing may be a relevant approach to identify specific markers of alcohol dependence. The aim of this study was to propose the probable olfactory markers for alcohol dependence through a study of the olfactory parameters that involve the central olfactory pathway. We recruited the same 41 alcohol-dependent patients in an early (day 8) and late (day 67) stage of abstinence and 41 cont

Age-related changes in the olfactory system of dogs.

Age-related changes in the olfactory system were examined in 22 dogs ranging in age from 10 to 19 years old. Atrophic changes with degeneration were observed in the olfactory epithelium of dogs older than 14 years; the changes were prominent in the dogs over the age of 17 years. Immunohistochemistry using an anti-carnosine antibody, which is a marker for the olfactory cells, demonstrated a decrease in the number of olfactory cells. Electron microscopy also showed a decrease in the number of cilia of olfactory cells and microvilli of supporting cells. Atrophic changes with the features of regen
