Spontaneously hypertensive rats have more orexin neurons in the hypothalamus and enhanced orexinergic input and orexin 2 receptor-associated nitric oxide signalling in the rostral ventrolateral medulla.

What is the central question of this study? Our previous study demonstrates that elevated orexin 2 receptor (OX2R) activity within the rostral ventrolateral medulla (RVLM) contributes to hypertension in spontaneously hypertensive rats (SHRs), and a lower OX2R protein level was detected in their RVLM. The present study aims to explore the mechanisms underlying elevated orexinergic activity in the RVLM of SHRs, compared with their normotensive counterparts, Wistar-Kyoto rats. What is the main finding and its importance? Increased orexinergic input into the RVLM and enhanced OX2R responsiveness i

Orexin-A Mitigates Traumatic Brain Injury by Inhibiting Neuronal Ferroptosis Through Orexin Receptor 1/Nuclear Factor Erythroid 2-Related Factor 2 Signaling Pathway.

Traumatic brain injury (TBI) contributes to neurological disability, in part through neuronal ferroptosis. Although orexin-A (OXA) attenuates TBI-induced ferroptosis and functional impairment, the underlying mechanism remains unclear. We investigated whether OXA exerts neuroprotection via the orexin receptor 1 (OX1R)/nuclear factor erythroid 2-related factor 2 (Nrf2) signaling pathway through in vitro and in vivo experiments. Rat TBI models were induced by modified Feeney's method alongside Erastin-treated PC12 ferroptosis models. OX1R (intracerebroventricular) and Nrf2 (intraperitoneal) inhib

Dysregulation of Orexin-A and Implications for Renal Function Assessment in Chronic Kidney Disease.

BACKGROUND: Chronic kidney disease (CKD) is a long-standing kidney condition in which the kidneys are gradually damaged. It usually has multiple symptoms and is often accompanied by complications, such as hormonal imbalances, high blood pressure, anemia, and sleep disorders. Orexin, occasionally mentioned as hypocretin, is a neuropeptide that is mostly generated by orexin-containing neurons in the hypothalamus. Their receptors are found in a variety of tissues, such as the brain and kidney. This study aims to evaluate the impact of renal function on orexin-A levels in CKD. MATERIALS AND METHO

The orexin system as a pharmacological target in inflammation: peripheral mechanisms and safety implications.

The orexin (hypocretin) system, composed of the neuropeptides orexin A and orexin B and their receptors OX1R and OX2R, is primarily known for its role in sleep-wake regulation and energy homeostasis. Increasing evidence indicates that orexin signaling also participates in the regulation of inflammatory processes. This review examines the pharmacological relevance of the orexin system in immune modulation, with particular emphasis on potential peripheral mechanisms that may represent novel therapeutic targets. Current evidence suggests that orexins may influence inflammatory responses through b…

Structural insights into the binding mode of the hypnotic drug vornorexant to orexin receptors.

Insomnia is a widespread sleep disorder that significantly impairs quality of life and imposes a societal burden. Although benzodiazepines and Z-drugs are commonly used for treating insomnia, these drugs often have side effects, such as excessive muscle relaxation and dependency. The orexin signaling pathway has emerged as a promising therapeutic target for insomnia, with dual orexin receptor antagonists (DORAs) offering an alternative approach to treatment. To reduce the potential of these drugs for next-day residual effects, we developed vornorexant, a novel DORA with a high receptor affinit

Reward prediction is encoded by orexin neuron activity during motivated behavior.

Orexin neurons regulate physiological functions, such as energy homeostasis, wakefulness, and motivated behaviors. However, studies linking orexin neuron activity to behavior via selectively activating/inactivating these inputs in a temporally controlled manner in rats are scarce. Here, we examined the role that orexin neurons play in motivated behavior in transgenic rats using cell type-specific fiber photometry and optogenetic manipulation. Using chemogenetics, we found that motivation for a reward increased when orexin neurons were activated. Furthermore, during motivated behavior, orexin n

Research Progress and Future Perspectives of Orexin Receptor Antagonists in Insomnia Therapy.

Insomnia is a common sleep disorder that significantly impairs quality of life and increases the risk of various chronic diseases. Although conventional sedative-hypnotic agents are effective, their clinical use is limited by tolerance, dependence, and residual next-day effects. The orexin system plays a critical role in sleep-wake regulation through orexin receptor 1 (OX1R) and orexin receptor 2 (OX2R), with OX2R serving as the primary mediator of wakefulness. Based on this mechanism, orexin receptor antagonists (ORAs) have been developed to promote sleep by suppressing wake-promoting signali

Orexin-A and Circadian Disruption in Alzheimer's Disease: Implications for Amyloid-Beta Pathology.

Alzheimer's disease (AD) is characterized by cognitive decline, circadian rhythm disruptions, and accumulation of Aβ plaques. Orexin-A, a neuropeptide involved in regulating sleep and circadian rhythms, has been implicated in these processes, although its specific role in modulating β-amyloid (Aβ) aggregation remains unclear. This study investigates how orexin-A influences Aβ aggregation and its impact on cognitive and circadian dysfunctions in AD mice subjected to acute sleep deprivation (ASD). Behavioural assessments showed significant cognitive deficits following ASD, including impaired rec

Differential modulation of contextual fear conditioning by orexin receptors in the dorsal hippocampus.

The dorsal hippocampus (DH) encodes contextual memories essential for adaptive responses to environmental threats. The lateral hypothalamus (LH), a primary source of orexinergic neurons in the brain, is also involved in the modulation of defensive behaviors. Although recent research links the orexinergic system to emotional processes, its role in the DH during the organization and expression of learned fear responses remains poorly understood. This study investigates the role of orexin receptor subtypes in DH during contextual fear conditioning. Anatomical tracing techniques revealed a direct

Associations of Cerebrospinal Fluid Orexin-A, Alzheimer Disease Biomarkers, and Cognitive Performance.

OBJECTIVE: Cerebrospinal fluid (CSF) orexin-A has been suggested to be a biomarker of Alzheimer disease (AD). In both cognitively unimpaired healthy older adults and individuals with symptomatic AD, CSF orexin-A is positively associated with CSF Aβ42, p-tau181, and total tau (t-tau) concentrations. However, a recent systematic review and meta-analysis did not support differences in orexin-A between AD and controls. In this study, we tested the association between CSF orexin-A concentrations, AD biomarkers, and cognitive performance in older adults with and without symptomatic AD. METHODS: Two

Cerebrospinal-fluid Orexin-A levels in different neurocognitive disorders: a comparison study.

In the present study, we investigated the differences in cerebrospinal fluid (CSF) orexin-A levels among patients with different neurocognitive disorders, such as mild or moderate to severe Alzheimer's disease (AD; mAD, msAD, respectively), behavioral variants of frontotemporal dementia (bv-FTD), non-fluent primary aphasia (NFPA), and idiopathic normal pressure hydrocephalus (iNPH). A total of 214 participants were evaluated (mAD, 45; msAD, 31; bv-FTD, 12; NFPA, 22; iNPH, 13; non-demented elderly controls, 91). The highest CSF orexin-A levels were found in iNPH patients (263.31 ± 56.89 pg/mL).

Orexin Receptor Antagonism: Normalizing Sleep Architecture in Old Age and Disease.

Sleep is essential for human well-being, yet the quality and quantity of sleep reduce as age advances. Older persons (>65 years old) are more at risk of disorders accompanied and/or exacerbated by poor sleep. Furthermore, evidence supports a bidirectional relationship between disrupted sleep and Alzheimer's disease (AD) or related dementias. Orexin/hypocretin neuropeptides stabilize wakefulness, and several orexin receptor antagonists (ORAs) are approved for the treatment of insomnia in adults. Dysregulation of the orexin system occurs in aging and AD, positioning ORAs as advantageous for thes

Hypothalamic orexin prevents non-alcoholic steatohepatitis and hepatocellular carcinoma in obesity.

Non-alcoholic steatohepatitis (NASH) occasionally occurs under obesity; however, factors modulating the natural history of fatty liver disease remain unknown. Since hypothalamic orexin that regulates physical activity and autonomic balance prevents obesity, we investigate its role in NASH development. Male orexin-deficient mice fed a high-fat diet (HFD) show severe obesity and progression of NASH with fibrosis in the liver. Hepatic fibrosis also develops in ovariectomized orexin-deficient females fed an HFD but not ovariectomized wild-type controls. Moreover, long-term HFD feeding causes hepat

Animal models of narcolepsy and the hypocretin/orexin system: Past, present, and future.

Animal models have advanced not only our understanding of the etiology and phenotype of the sleep disorder narcolepsy but have also informed sleep/wake regulation more generally. The identification of an inheritable narcolepsy phenotype in dogs in the 1970s allowed the establishment of a breeding colony at Stanford University, resulting in studies that provided the first insights into the genetics and neurotransmitter systems that underlie cataplexy and rapid-eye movement sleep atonia. Although the discovery of the hypocretin/orexin neuropeptides in 1998 initially seemed unrelated to sleep/wak

Reduced Plasma Orexin-A Concentrations are Associated with Cognitive Deficits in Anorexia Nervosa.

Orexins/hypocretins are neuropeptides implicated in numerous processes, including food intake and cognition. The role of these peptides in the psychopathology of anorexia nervosa (AN) remains poorly understood. The aim of the current study was to evaluate the associations between plasma orexin-A (OXA) concentrations and neuropsychological functioning in adult women with AN, and a matched control group. Fasting plasma OXA concentrations were taken in 51 females with AN and in 51 matched healthy controls. Set-shifting was assessed using the Wisconsin Card Sorting Test (WCST), whereas decision ma…

The relationship between orexin levels and blood pressure changes in patients with narcolepsy.

STUDY OBJECTIVE: Narcolepsy type 1 (NT1) is caused by a deficiency or absence of the neurotransmitter orexin. NT1 is also associated with a reduced nocturnal "dipping" of blood pressure (BP). The study objective was to analyze whether nocturnal BP values differed in patients depleted of orexin, versus those in whom production was preserved. METHODS: We performed a retrospective analysis of the polysomnographic recordings, orexin levels, and BP values of patients with NT1. Data was collected from a total of 21 patients, divided into two groups as follows: those with a complete depletion of ore

Orexin Decreases Aromatase Gene Expression in The Hypothalamus of Androgenized Female Rats.

BACKGROUND: Orexin is a hypothalamic orexigenic neuropeptide, which third cerebral injection of it mainly exerts inhibitory effects on reproductive functions. It increases significantly the Aromatase (Cyp19) gene expression in the hypothalamus of male rats. Aromatase is an enzyme which converts androgens to estradiol in the hypothalamus of rats. Prenatal or neonatal exposure of females to testosterone masculinizes the pattern of Cyp19 mRNA levels in adulthood. In the present study the effects of central injections of orexin-A on hypothalamic Cyp19 gene expression of adult female rats were inve
