The rhizosphere metabolic circular economy (RMCE) governs the exchange, reuse, and reconfiguration of metabolites at the root-soil interface. Building on traditional binary plant-microbe models, this perspective advances an expanded RMCE framework by integrating three additional interdependent dimensions that explain how rhizosphere metabolic processes persist over time, organize across space, and recover under environmental stress. First, the live-dead plant-microbe continuum enables metabolic persistence by incorporating microbial necromass and root residues into long-term carbon sequestrati

In May 2023, characteristic symptoms of fire blight, suspected to be caused by Erwinia amylovora, were observed infected wild Cotoneaster plants in the secondary forest along the lower reaches of the Jilgilang River, Xinjiang, China. Symptomatic tissues were collected, and bacterial isolation and purification were performed. The bacterium was identified through morphological characterization of the colonies, pathogenicity assays, tobacco hypersensitivity tests, and specific polymerase chain reaction (PCR) amplification, followed by sequence analysis. The isolated bacterium exhibited morphologi

Systemic induction of pathogenesis-related (PR) proteins in tobacco, which occurs during the hypersensitive response to tobacco mosaic virus (TMV), may be caused by a minimum 10-fold systemic increase in endogenous levels of salicylic acid (SA). This rise in SA parallels PR-1 protein induction and occurs in TMV-resistant Xanthi-nc tobacco carrying the N gene, but not in TMV-susceptible (nn) tobacco. By feeding SA to excised leaves of Xanthi-nc (NN) tobacco, we have shown that the observed increase in endogenous SA levels is sufficient for the systemic induction of PR-1 proteins. TMV infection

Phytate (salts of myo-inositol-1,2,3,4,5,6-hexakisphosphate) constitutes a large portion of the organic phosphorus in most soils, but its strong interactions with soil minerals and organic matter limit its availability to plants. Phytate can be used by plants only after it is desorbed from the soil matrix, with the inorganic P being released by phytases via cleavage of its phosphomonoester bonds. While plant phytases function primarily in its internal phytate remobilization, the role of microbial phytases in facilitating phytate-P utilization by plants remains poorly understood. Ilex paraguariensis is a native plant from Southern America, where it is used as a beverage. In traditional medicine, it is used to treat many diseases including inflammation. However, we do not yet know precisely how this effect occurs. We therefore evaluated its anti-inflammatory effect in a murine model of pleurisy. The standardized CE, BF and ARF fractions, Caf, Rut and CGA were able to reduce leukocyte migration, exudate concentration, MPO and ADA activities and NOx levels. Moreover, I. paraguariensis also inhibited the release of Th1/Th17 pro-inflammatory cytokines, while increasing IL-1

The relationship between carboxylate release and the ability of plants to access phosphorus from AlPO4 and to detoxify aluminium was studied by comparing species with a low and high rate of carboxylate release, Triticum aestivum (wheat) and Lupinus albus (white lupin), respectively. Species were supplied with P at 10, 20, 40 or 100 mg P kg-1 sand in the form of sparingly soluble AlPO4 or soluble KH2PO4; control plants did not receive any P. Triticum aestivum was significantly better than L. albus at accessing P from AlPO4, despite accumulating fewer carboxylates in its rhizosphere. Rhizosphere

Low light (LL) is a major constraint on the productivity of intercropped legumes and dense planting crops, and also affects the root-associated microbial communities. However, how LL reshapes the root-microbe interactions and whether the root microbiota can mitigate LL-induced damage in legumes remain unclear. Here, a meta-analysis based on field observations revealed that negative effects predominated (>70%) in intercropped and dense planting legume systems, with the light intensity emerging as the primary determinant of yield variation. Using soybean as a model, we found that LL suppressed p

INTRODUCTION: Ambrosia trifida, a harmful invasive plant, poses significant ecological and economic threats and is expected to spread further under future warming and nitrogen deposition scenarios. According to plant-soil feedback and enhanced mutualist hypothesis, invasive plants may gain a competitive edge by recruiting specific microorganisms. However, little is known about the composition and functional potential of its rhizosphere microbiome.
METHODS: In this study, we combined metagenomics with widely targeted metabolomics to investigate the interactions between root exudates and soil m

Enhanced rock weathering (ERW) has been proposed as a measure to enhance the carbon (C)-sequestration potential and fertility of soils. The effects of this practice on the soil phosphorus (P) pools and the general mechanisms affecting microbial P cycling, as well as plant P uptake are not well understood. Here, the impact of ERW on soil P availability and microbial P cycling functional groups and root P-acquisition traits were explored through a 2-year wollastonite field addition experiment in a tropical rubber plantation. The results show that ERW significantly increased soil microbial carbon

The plant phloem is a long-distance conduit for the transport of assimilates but also of mobile developmental and stress signals. These signals can be sugars, metabolites, amino acids, peptides, proteins, microRNA, or mRNA. Yet small lipophilic molecules such as oxylipins and, more recently, phospholipids have emerged as possible long-distance signals as well. Analysis of phloem (phospho)lipids, however, requires enrichment, purification, and sensitive analysis. This chapter describes the EDTA-facilitated approach of phloem exudate collection, phase partitioning against chloroform-methanol for

The plant root tip is highly sensitive to environmental cues and exerts sensory, chemical and movement responses. However, the microbial diversification at the root tip, and the nature of root-selection for microbes, are poorly understood. This study investigated root exudation and microbiome colonisation in the chickpea (Cicer arietinum L.) root tip and root base, in diverse soils. For the endosphere microbiome, deterministic effects were more important than stochastic processes in shaping the root tip microbiome, indicated by the stable endophyte composition independent of the soil microbiot

Antibiotics are frequently used in the United States as feed efficiency promoters and medicines for livestock that is destined for human consumption. These antibiotics are released into the environment through the runoff and wastewater streams from animal feedlots and land applications of manure. The exposure of microorganisms to these antibiotics has reportedly resulted in the development of resistant species of microorganisms, which in turn can lead to human health hazards. Phytoremediation of these antibiotics can be a useful tool for countering this problem. Aquatic plants, Myriophyllum aq

Rosmarinus officinalis, also named rosemary, is a native plant from the Mediterranean region that is useful for the treatment of inflammatory diseases. Studies using experimental models and/or in vitro tests have shown the important biological effects of rosemary. In this context, the mechanism of the anti-inflammatory activity of rosemary must be investigated to support the discovery of new substances with anti-inflammatory effects. The aim of the present study was to investigate the anti-inflammatory effects of crude extract oil free obtained from the leaves of rosemary in an animal model of

Soilborne fungal pathogens must locate host roots, stably attach to their surface and assemble a persistent mycelial network before invading the plant, yet how host perception is coupled to these cellular outputs remains poorly understood. Here, we characterize HAM-7, a GPI-anchored cell wall protein of Fusarium oxysporum and orthologue of the Neurospora crassa cell wall integrity sensor, and show that it is required for two mechanistically distinct processes essential for root colonization. Targeted deletion of ham-7 strongly impaired stable mycelial retention on tomato roots while leaving ge

Plant genetic diversity can affect ecosystem functioning by enhancing productivity, litter decomposition and resistance to natural enemies. However, the mechanisms underlying these effects remain poorly understood. We hypothesized that genetic diversity may influence ecosystem processes by eliciting functional plasticity among individuals encountering kin or genetically diverse neighbourhoods. We used soil conditioned by groups of closely related (siblings) and diverse genotypes of Deschampsia cespitosa - a species known to exhibit kin recognition via root exudation - to investigate the conseq

We have been using mutagenesis to determine how biocontrol bacteria such as Enterobacter cloacae 501R3 deal with complex nutritional environments found in association with plants. E. cloacae C10, a mutant of 501R3 with a transposon insertion in degS, was diminished in growth on synthetic cucumber root exudate (SRE), colonization of cucumber seed and roots, and control of damping-off of cucumber caused by Pythium ultimum. DegS, a periplasmic serine protease in the closely related bacterium Escherichia coli K12, is required for the RpoE-mediated stress response. C10 containing wild-type degS fro

Common reed (Phragmites australis) is a phytoremediator tolerant to heavy metals. In this study, we found that 70% of the cadmium (Cd) found in the stem of common reed exists in a soluble form, with more than half of the soluble Cd in the 10- to 50-kDa fraction. Based on an enzyme degradation assay, the major component of the Cd-associated molecule is assumed to be an amylopectin-like α-glucan. This molecule may associate with Cd via the carboxyl group, rather than the thiol group. The conditions required for the disengagement of Cd from the 10- to 50-kDa fraction indicated that disulfide bond

Constructed wetlands (CWs) depend on plant-microbe associations for pollutant transformation, yet the processes linking macrophyte identity to rhizosphere microbiome assembly remain insufficiently resolved. We compared bacterial communities associated with C. articulatus (Ca) and T. geniculata (Tg) roots with an unplanted treatment (Up) in pilot-scale CWs on the Colombian Caribbean coast. Bacterial communities were characterized by 16S rRNA gene amplicon sequencing. Co-occurrence networks, taxon-environment correlations, and the phylogenetic bin-based null model iCAMP were used to assess commu
