Advancements in research on the immune-inflammatory mechanisms mediated by NLRP3 inflammasome in ischemic stroke and the regulatory role of natural plant products.

Ischemic stroke (IS) is a major cause of mortality and disability among adults. Recanalization of blood vessels to facilitate timely reperfusion is the primary clinical approach; however, reperfusion itself may trigger cerebral ischemia-reperfusion injury. Emerging evidence strongly implicates the NLRP3 inflammasome as a potential therapeutic target, playing a key role in cerebral ischemia and reperfusion injury. The aberrant expression and function of NLRP3 inflammasome-mediated inflammation in cerebral ischemia have garnered considerable attention as a recent research focus. Accordingly, thi

Induction of tomato plant biochemical immune responses by the synthesized zinc oxide nanoparticles against wilt-induced Fusarium oxysporum.

The current study used zinc oxide nanoparticles (ZnO-NPs) to protect the tomato plant against Fusarium wilt. Gamma rays were used to synthesize ZnO-NPs, and the designed ZnO-NPs were characterized using high-resolution transmission electron microscopy (HRTEM), scanning electron microscope (SEM), dynamic light scattering (DLS), energy-dispersive X-ray spectroscopy (EDX), and ultraviolet-visible (UV-Vis.) spectroscopy. We found that the 20 kGy dose is the most effective for ZnO-NPs synthesis, with the highest O.D. = 1.65 (diluted 3 times) at 400 nm. The scale of ZnO-NPs ranged from 10.45 to 75.2

Elucidating the mechanism of action of copper heptagluconate on the plant immune system against Pseudomonas syringae in tomato (Solanum lycopersicum L).

BACKGROUND: Phytopathogenic problems caused by the bacterial pathogen Pseudomonas syringae in tomato are becoming more serious due to the emergence of strains resistant to classical pesticides. This has led to research into new formulations with lower environmental problems. One of the most promising alternatives to the use of classical pesticides is the induction of natural plant defences. New formulations based on Cu complexed with heptagluconic acid induce plant innate defences and could be an alternative to classical treatments based on inorganic Cu against bacterial speck. To study the ef

Double Plant Homeodomain Fingers 2 (DPF2) Promotes the Immune Escape of Influenza Virus by Suppressing Beta Interferon Production.

The high mutation rates of the influenza virus genome facilitate the generation of viral escape mutants, rendering vaccines and drugs against influenza virus-encoded targets potentially ineffective. Therefore, we identified host cell determinants dispensable for the host but crucial for virus replication, with the goal of preventing viral escape and finding effective antivirals. To identify these host factors, we screened 2,732 human genes using RNA interference and focused on one of the identified host factors, the double plant homeodomain fingers 2 (DPF2/REQ) gene, for this study. We found t

Plant-based vaccine: mice immunized with chloroplast-derived anthrax protective antigen survive anthrax lethal toxin challenge.

The currently available human vaccine for anthrax, derived from the culture supernatant of Bacillus anthracis, contains the protective antigen (PA) and traces of the lethal and edema factors, which may contribute to adverse side effects associated with this vaccine. Therefore, an effective expression system that can provide a clean, safe, and efficacious vaccine is required. In an effort to produce anthrax vaccine in large quantities and free of extraneous bacterial contaminants, PA was expressed in transgenic tobacco chloroplasts by inserting the pagA gene into the chloroplast genome. Aphid Probing and Phloem Feeding Trigger Distinct Plant Immune Programs, Identifying a Broad‑Spectrum Trypsin Protease Inhibitor.

Aphid feeding begins with brief exploratory probing and transitions to prolonged phloem ingestion, yet whether these distinct phases trigger different plant immune programs remains unknown. We show that probing and phloem feeding are associated with distinct plant immune cascades. Under probing-enriched conditions, we observed selective activation of jasmonate (JA) signaling, accompanied by significant ROS bursts and vascular callose deposition, whereas phloem feeding elicits only marginal salicylic acid accumulation. Probing-enriched conditions imposes severe fitness penalties on aphids, a ph

Medicinal cannabis plant extract (NTI164) modifies epigenetic, ribosomal, and immune pathways in paediatric acute-onset neuropsychiatric syndrome.

Paediatric acute-onset neuropsychiatric syndrome (PANS) is a syndrome of infection-provoked abrupt-onset obsessive-compulsive disorder (OCD) or eating restriction. Based on the hypothesis that PANS is an epigenetic disorder of immune and brain function, a full-spectrum medicinal cannabinoid-rich low-THC cannabis (NTI164) was selected for its known epigenetic and immunomodulatory properties. This open-label trial of 14 children with chronic-relapsing PANS (mean age 12·1 years; range 4-17; 71 % male) investigated the safety and efficacy of 20 mg/kg/day NTI164 over 12 weeks. Clinical outcomes wer

Two plant immune inducers achieve comparable field control of the rice stem borer through distinct physiological pathways.

BACKGROUND: Plant immune inducers are promising tools for sustainable pest management because they influence both target pests and natural enemies. Previous studies showed that ATL (3% amino-oligosaccharide +3% Alternaria tenuissima activating protein) attracts the egg parasitoid Trichogramma japonicum, whereas AOS repels the parasitoid, although neither inducer directly affects adult behaviour of the rice stem borer (RSB, C. suppressalis). Whether these contrasting ecological effects influence field efficacy and larval physiology has remained unclear. RESULTS: Field experiments demonstrated

Multi-Stimuli-Responsive Hydrogen-Bonded Organic Frameworks Nanocarriers Enable Targeted Fungicide Release and Plant Immune Regulation.

Precision plant protection requires carriers that couple crystalline order with context-responsive release. Hydrogen-bonded organic frameworks (HOFs) offer ordered, metal-free porosity and programmable host-guest interactions to realize this goal. We therefore built a multi-stimuli-responsive system (PYR@PFC-1-F@HACC) using the fluorinated HOF PFC-1-F as a pesticide carrier and label-free in planta fluorescent marker. Pyraclostrobin (PYR) was encapsulated in PFC-1-F with ∼30 wt.% loading, and the resulting composite was surface-coated with chitosan quaternary ammonium salt (HACC). The HACC she

Harnessing plant chloroplasts for oral delivery of a multi-epitope HPV vaccine: toward cost-effective systemic and mucosal immunization.

Human papillomavirus (HPV) is a major causative agent of cervical and other mucosal cancers. However, the distribution and accessibility of current prophylactic vaccines remain limited, especially in low- and middle-income countries (LMICs), due to high production costs, cold-chain dependency, and limited induction of mucosal immune responses. To addressing these challenges, we designed a rationally constructed multi-epitope HPV vaccine (HPV_MEV) incorporating conserved cytotoxic T lymphocyte (CTL), helper T lymphocyte (HTL), and B-cell epitopes from diverse high- and low-risk HPV genotypes. A plant-derived VLP influenza vaccine elicits a balanced immune response even in very old mice with co-morbidities.

BACKGROUND: The elderly are at high risk from influenza, in part because immunity wanes with age and through the accumulation of comorbidities. A novel plant-derived virus-like-particle (VLP) vaccine bearing influenza hemagglutinin can induce a balanced humoral and cellular response in old mice (16-18 months) while split virion vaccines elicit mostly antibodies. Because mice also collect comorbidities and lose immune competence as they age, we wished to determine how the plant-derived VLP vaccine would perform in animals approaching the end of their life-span. MATERIALS AND METHODS: Old (24-2

Comparison of homologous and heterologous prime-boost immunizations combining MVA-vectored and plant-derived VP2 as a strategy against IBDV.

Different immunogens such as subunit, DNA or live viral-vectored vaccines against Infectious Bursal Disease virus (IBDV) have been evaluated in the last years. However, the heterologous prime-boost approach using recombinant modified vaccinia Ankara virus (rMVA), which has shown promising results in both mammals and chickens, has not been tried against this pathogen yet. IBD is a highly contagious and immunosuppressive disease of poultry that affects mainly young chicks. It is caused by IBDV, a double-stranded RNA virus carrying its main antigenic epitopes on the capsid protein VP2. Our object

Plant-derived H7 VLP vaccine elicits protective immune response against H7N9 influenza virus in mice and ferrets.

In March 2013, the Chinese Centre for Disease Control and Prevention confirmed the first reported case of human infection with an avian influenza A H7N9 virus. Infection with this virus often caused severe pneumonia and acute respiratory distress syndrome resulting in a case fatality rate >35%. The risk of pandemic highlighted, once again, the need for a more rapid and scalable vaccine response capability. Here, we describe the rapid (19 days) development of a plant-derived VLP vaccine based on the hemagglutinin sequence of influenza H7N9 A/Hangzhou/1/2013. The immunogenicity of the H7 VLP vac

Cucurbitacin B from Cucurbitaceae Plants: Treating Pancreatic Cancer via Inducing Mitophagy, Inhibiting Glycolysis, and Enhancing Immune Function.

BACKGROUND: Cucurbitacin B (CuB) is a relatively unique and valuable component in plants of the Cucurbitaceae family due to its diverse and remarkable physiological activities, but its specific mechanisms in regulating tumor metabolism and immune response remain unclear. The hypoxic tumor microenvironment (TME) of pancreatic cancer induces metabolic reprogramming in cancer cells, causing them to rely on glycolysis for energy. LDHA, a key enzyme in glycolysis, can suppress glycolysis and tumor growth when inhibited. OBJECTIVE: The objective of this study was to investigate the mechanism of CuB

A novel plant-derived recombinant COBRA infectious bronchitis virus spike protein can elicit a strong immune response in chickens.

Infectious bronchitis virus (IBV) causes an acute respiratory disease in chickens of all ages, and is an economic burden on the global poultry industry. In severe cases, this virus can spread from the respiratory tract to urinary and reproductive organs, leading to kidney damage, poor egg quality, and high mortality rate of chickens. Among IBV glycoproteins, spike (S) is the major determinant of viral attachment to host receptors and induction of neutralizing antibodies. Rapid mutations were found within the S gene of numerous IBV strains presenting in multiple geographical locations. Since th

Plant-derived recombinant macromolecular PAP-IgG Fc as a novel prostate cancer vaccine candidate eliciting robust immune responses.

Prostatic acid phosphatase (PAP) is a specific protein that is highly expressed in prostate cancer. In this study, we constructed two recombinant PAP fusion genes: PAP fused to the immunoglobulin G (IgG) Fc fragment (designated PAP-Fc) and PAP-Fc fused to the endoplasmic reticulum retention sequence KDEL (designated PAP-FcK). Transgenic Nicotiana tabacum plants expressing these recombinant macromolecular proteins (MPs) were generated using Agrobacterium-mediated transformation, and the presence of both genes was confirmed through genomic PCR. Western blot analysis validated the expression of P

Influence of Temperature on Salicylate-Induced Components in Tomato Plants Immune System under Invasion with Root-Knot Nematode Meloidogyne incognita.

The tomato gene Mi-1,2 is currently the only commercially available source of resistance to the root-knot nematode Meloidogyne incognita, which loses activity when soil temperatures exceed 28°C. The study aimed to investigate the components of the tomato immune system to root-knot nematode associated with salicylic acid (SA) at elevated (34°C) and normal (25°C) temperatures. The obtained results showed that the Mi-1,2-mediated immune response is disrupted at 34°C. At elevated temperatures in plants infested with nematodes, the synthesis of salicylic acid and the activity of phenylalanine ammon

DEAD-box RNA helicase RH20 positively regulates RNAi-based antiviral immunity in plants by associating with SGS3/RDR6 bodies.

RNA silencing plays a crucial role in defending against viral infections in diverse eukaryotic hosts. Despite extensive studies on core components of the antiviral RNAi pathway such as DCLs, AGOs and RDRs proteins, host factors involved in antiviral RNAi remain incompletely understood. In this study, we employed the proximity labelling approach to identify the host factors required for antiviral RNAi in Nicotiana benthamiana. Using the barley stripe mosaic virus (BSMV)-encoded γb, a viral suppressor of RNA silencing (VSR), as the bait protein, we identified the DEAD-box RNA helicase RH20, a br

Distinct profiles of plant immune resilience revealed by natural variation in warm temperature-modulated disease resistance among Arabidopsis accessions.

Elevated temperature suppresses the plant defence hormone salicylic acid (SA) by downregulating the expression of master immune regulatory genes CALMODULIN BINDING PROTEIN 60-LIKE G (CBP60g) and SYSTEMIC ACQUIRED RESISTANCE DEFICIENT1 (SARD1). However, previous studies in Arabidopsis thaliana plants have primarily focused on the accession Columbia-0 (Col-0), while the genetic determinants of intraspecific variation in Arabidopsis immunity under elevated temperature remain unknown. Here we show that BASIC HELIX LOOP HELIX 059 (bHLH059), a thermosensitive SA regulator at nonstress temperatures,
