BK Polyomavirus Genotypes in Two Patients after Hematopoietic Cell Transplant.

BK polyomavirus (BKPyV) infection can lead to nephropathy and hemorrhagic cystitis (HC). We evaluated BKPyV genotypes in two individuals after hematopoietic cell transplant (HCT). The first case developed HC and was infected with genotype Ib-2, while the second did not develop HC and was infected with genotype Ia.

Caveolin- and clathrin-independent entry of BKPyV into primary human proximal tubule epithelial cells.

BK polyomavirus (BKPyV) is a human pathogen that causes polyomavirus-associated nephropathy and hemorrhagic cystitis in transplant patients. Gangliosides and caveolin proteins have previously been reported to be required for BKPyV infection in animal cell models. Recent studies from our lab and others, however, have indicated that the identity of the cells used for infection studies can greatly influence the behavior of the virus. We therefore wished to re-examine BKPyV entry in a physiologically relevant primary cell culture model, human renal proximal tubule epithelial cells. Using siRNA kno

PD-1 regulates CD4+ T cell-mediated CD8+ T cell responses in the brain to balance viral control and neuroinflammation.

Programmed cell death protein 1 (PD-1) is expressed by T cells during progressive multifocal leukoencephalopathy (PML), a life-threatening brain disease caused by the human-only JC polyomavirus (JCPyV). PD-1 checkpoint immunotherapy has benefited some PML patients, but reasons for its variable outcomes are unclear. Using mouse polyomavirus (MuPyV), we show that PD-1 loss acts in a brain-autonomous manner to increase the magnitude of brain-infiltrating CD4+ and CD8+ T cells and the function of virus-specific CD8+ T cells; in concert, brain virus levels decline and neuroinflammation increases. High expression of JC polyomavirus-encoded microRNAs in progressive multifocal leukoencephalopathy tissues and its repressive role in virus replication.

JC polyomavirus (JCPyV, JCV) causes progressive multifocal leukoencephalopathy (PML) in immunocompromised hosts. JCPyV replicates in oligodendrocytes within the brain tissue of patients with PML. The JCPyV genome encodes a microRNA (miRNA) in the region encoding the large T antigen. JCPyV-encoded miRNA (miR-J1) has been detected in the tissue and cerebrospinal fluid samples of patients with PML; however, there are no reports describing the localization of polyomavirus-encoded miRNA in histological samples of patients with virus-associated diseases. In the present study, we detected high miR-J1

Incidence and diversity of circoviruses, polyomaviruses, and adenoviruses in exotic free-roaming rosy-faced lovebirds from Phoenix, Arizona, USA.

Free-roaming rosy-faced lovebirds (Agapornis roseicollis) were first documented in the greater Phoenix area (Arizona, USA) in the mid-1980s and have since established small populations. In blood and cloacal swab samples from 69 lovebirds at four locations, we identified the presence of beak and feather disease virus (BFDV), aves polyomavirus (APyV), and psittacine adenovirus 5 (PsAdV-5). Additionally, we identified a novel aviadenovirus, hereby referred to as psittacine adenovirus 12 (PsAdV-12). PsAdV-12 is most closely related to members of the species Aviadenovirus senegalense and Aviadenovi

Transplant centers' prophylaxis and monitoring strategies: a key determinant of current herpes and polyomavirus incidences - results from the DZIF kidney transplant cohort.

BACKGROUND: Herpes- and polyomaviruses are major opportunistic pathogens after renal transplantation. Despite established guidelines, there is limited data on transplant centers' prophylaxis and monitoring strategies and centers' adherence to these guidelines and their impact on infection rates and patient outcomes. METHODS: This multicenter cohort study, conducted by the German Center for Infection Research, included 1035 kidney transplant recipients from five centers (01/2014-02/2021), focusing on herpes- and polyomavirus viremia within the first year and adherence to prophylaxis strategies

Derivation of a JC virus-resistant human glial cell line: implications for the identification of host cell factors that determine viral tropism.

JC virus (JCV) is a common human polyomavirus that infects 70-80% of the population worldwide. In immunosuppressed individuals, JCV infects oligodendrocytes and causes a fatal demyelinating disease known as progressive multifocal leukoencephalopathy (PML). The tropism of JCV is restricted to oligodendrocytes, astrocytes, and B lymphocytes. Several mechanisms may contribute to the restricted tropism of JCV, including the presence or absence of cell-type-specific transcription and replication factors and the presence or absence of cell-type-specific receptors. We have established a system to inv

Persistent JC virus (JCV) infection is demonstrated by continuous shedding of the same JCV strains.

The polyomavirus JC virus (JCV), the causative agent of progressive multifocal leukoencephalopathy, is ubiquitous in the human population, infecting children asymptomatically. JCV is often detected in normal renal tissue and in the urine of healthy individuals. We demonstrate that renal JCV represents that which persists after primary infection.

Herpesviruses and polyomaviruses in renal transplantation.

Viral infections remain a significant cause of morbidity and mortality among transplant patients despite recent advances in early detection and treatment. Herpesviruses and polyomaviruses are the most relevant viruses post-transplant as they establish latency in immunocompetent individuals and frequently reactivate in the immunosuppressed transplant recipient. Although we have made significant strides in the early diagnosis and treatment of viral infections in renal transplant recipients over the past five years, many questions remain. Optimization of screening and prophylactic/preemptive prot

[BK virus nephropathy after kidney transplantation].

JC and BK viruses are strains of the polyomavirus group with pathogenic potential in humans. BK is the most frequent pathogenic agent of polyomavirus nephropathy (BKVN) in kidney transplant patients, which is only exceptionally caused by JC virus. Asymptomatic BK virus infection is often acquired in childhood and the virus persists in urothelium and kidneys of healthy individuals, where it can be reactivated under immunosuppression. Up to 10% of transplanted kidneys are affected by BKVN, while the risk of transplant failure due to BKVN exceeds 50% in some publications. In kidney biopsies BKVN

Seroprevalence and stability of IgG antibodies to WU polyomavirus and human polyomavirus 6 among Finnish couples during a prospective follow-up.

Although WU polyomavirus (WUPyV) and human polyomavirus 6 (HPyV6) are widespread in adults, the long-term serological responses to these infections have not been systematically characterized. To address this gap, we assessed seroprevalence and IgG antibody stability of WUPyV and HPyV6 using data from the Finnish Family HPV (FFHPV) study, initiated in 1998. The cohort included 318 women in their third trimester of pregnancy and 133 male spouses, with follow-up samples available from 281 women and 119 men at 12, 24 and 36 months. IgG antibodies against the VP1 capsid proteins of both viruses wer

Merkel cell carcinoma: an update.

Merkel cell carcinoma (MCC) is an uncommon primary cutaneous neuroendocrine carcinoma associated with an adverse prognosis. In recent years, our understanding of MCC biology has markedly progressed. Since the discovery of the Merkel cell polyomavirus, it has become clear that MCC represents an ontogenetically dichotomous group of neoplasms with overlapping histopathology. Specifically, most MCCs arise secondary to viral oncogenesis, while a smaller subset is the direct result of UV-associated mutations. The distinction of these groups bears relevance in their immunohistochemical and molecular

Acute Kidney Injury and BK Polyomavirus in Urine Sediment Cells.

Polyomaviruses are widespread, with BK viruses being most common in humans who require immunosuppression due to allotransplantation. Infection with BK polyomavirus (BKV) may manifest as BK virus-associated nephropathy and hemorrhagic cystitis. Established diagnostic methods include the detection of polyomavirus in urine and blood by PCR and in tissue biopsies via immunohistochemistry. In this study, 79 patients with pathological renal retention parameters and acute kidney injury (AKI) were screened for BK polyomavirus replication by RNA extraction, reverse transcription, and virus-specific qPC

JC virus enters human glial cells by clathrin-dependent receptor-mediated endocytosis.

The human polyomavirus JC virus (JCV) is the etiologic agent of a fatal central nervous system (CNS) demyelinating disease known as progressive multifocal leukoencephalopathy (PML). PML occurs predominantly in immunosuppressed patients and has increased dramatically as a result of the AIDS pandemic. The major target cell of JCV infection and lytic replication in the CNS is the oligodendrocyte. The mechanisms by which JCV initiates and establishes infection of these glial cells are not understood. The initial interaction between JCV and glial cells involves virus binding to N-linked glycoprotei

Evaluation of Trichodysplasia Spinulosa Polyomavirus in Skin Biopsies, United States.

Trichodysplasia spinulosa polyomavirus (TSPyV) has been identified in human endothelial cell tumors. We evaluated 25 clinical cases of various inflammatory dermatoses in California, USA, for TSPyV by using RNAscope in situ hybridization. We did not detect TSPyV in any of the biopsy samples, suggesting that endothelial TSPyV is uncommon in inflammatory dermatoses.

MCPyV-Negative Merkel Cell Carcinoma In Situ Associated With Immunosuppression.

Merkel cell carcinoma (MCC) is a rare and aggressive cutaneous neoplasm that is almost always intradermal. Immunosuppression increases the risk of MCC, which is believed to be due to increased susceptibility to Merkel cell polyomavirus (MCPyV). Intraepidermal MCC, or MCC in situ (MCCis), is extremely rare and usually associated with other cutaneous lesions. Here, we describe a case of MCPyV-negative MCCis arising in an immunocompromised patient. This case adds to only 9 previously reported cases of MCCis without a coexisting neoplasm and suggests that immunosuppression can lead to MCCis by mec
