The emerging role of Deubiquitinases (DUBs) in parasites: A foresight review.

Before the discovery of the proteasome complex, the lysosomes with acidic proteases and caspases in apoptotic pathways were thought to be the only pathways for the degradation of damaged, unfolded, and aged proteins. However, the discovery of 26S and 20S proteasome complexes in eukaryotes and microbes, respectively, established that the degradation of most proteins is a highly regulated ATP-dependent pathway that is significantly conserved across each domain of life. The proteasome is part of the ubiquitin-proteasome system (UPS), where the covalent tagging of a small molecule called ubiquitin

Targeting SLC1A5-mediated glutamine metabolism overcomes PARP inhibitor resistance in ovarian cancer.

Poly(ADP-ribose) polymerase inhibitors (PARPi), such as olaparib, have significantly improved outcomes in ovarian cancer (OC); however, therapy resistance remains a major clinical challenge. Here, we showed that glutamine metabolism was upregulated in PARPi-resistant OC cells and identified the glutamine transporter SLC1A5 as a key mediator of this metabolic adaptation. Inhibition of SLC1A5 increased the sensitivity of OC cells to olaparib both in vitro and in vivo. Mechanistically, we demonstrated that the deubiquitinating enzyme USP14 directly interacted with and stabilized SLC1A5 by removin

A wheat-specific proline-rich protein regulates grain weight through a ubiquitin-mediated delay of endosperm cellularization.

Grain weight is a key determinant of wheat yield, yet the developmental mechanisms that define this trait remain poorly understood. Here, we identify TaPGW1-A1, a wheat-specific proline-rich gene that regulates grain weight. TaPGW1-A1 is a recently evolved gene found only in wheat. It is highly expressed in early developing grains, and its overexpression delays endosperm cellularization and prolongs the syncytial phase, resulting in an 8% increase in field-based grain weight. TaPGW1-A1 protein directly interacts with the ubiquitin receptor TaDSK2a (Dominant Suppressor of KAR2 homolog) via its

Host OTUB2 and viral PLpro stabilize NSP8 to promote SARS-CoV-2 replication.

The ubiquitin-proteasome system (UPS) plays a central role in antiviral defense but is also frequently hijacked by viruses to facilitate their replication. Here, we demonstrate that the host deubiquitinase OTUB2 stabilizes the viral replication factor NSP8 of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) through a dual-track mechanism. OTUB2 directly removes polyubiquitin chains from NSP8 to prevent its degradation. In parallel, OTUB2 stabilizes the viral papain-like protease (PLpro), which further promotes NSP8 stability through deubiquitination. Together, these effects preserv

Role of ubiquitin-specific protease 5 in the inflammatory response of chronic periodontitis.

BACKGROUND: The systemic inflammatory response caused by chronic periodontitis is a risk factor for multiple diseases. Ubiquitin-specific protease 5 (USP5) is a kind of deubiquitinase which mainly responsible for dissociating unanchored polyubiquitin. However, the functions of USP5 in chronic periodontitis have not been reported. METHODS: Chronic periodontitis patients were recruited, and their periodontal samples were collected. The levels of USP5, tumor necrosis factor-alpha (TNF-α), interleukin-6 (IL-6), and interleukin-1β (IL-1β) in gingival crevicular fluid were evaluated by ELISA. The e

A TRIM32-AMBRA1-ULK1 complex initiates the autophagy response in atrophic muscle cells.

The Ser/Thr protein kinase ULK1 is an upstream macroautophagy/autophagy regulator that is rapidly activated to ensure a proper adaptive response to stress conditions. Signaling pathways modulating ULK1 activity have been extensively characterized in response to nutrient/energy shortage, which mainly act by mediating ULK1 post-translational modifications, such as phosphorylation, acetylation and ubiquitination. Less characterized is how tissue-specific stress signals are able to activate ULK1 to induce autophagy. Our recent study has uncovered the E3 ubiquitin ligase TRIM32 as a novel ULK1 acti

lncRNA HULC promotes the growth of hepatocellular carcinoma cells via stabilizing COX-2 protein.

Highly upregulated in liver cancer (HULC), a lncRNA overexpressed in hepatocellular carcinoma (HCC), has been demonstrated to be involved in the carcinogenesis and progression of HCC. However, the mechanisms of HULC promoting the abnormal growth of HCC cells are still not well elucidated. In the present study, we for the first time demonstrated that HULC promoted the growth of HCC cells through elevating COX-2 protein. Moreover, the study of the corresponding mechanism by which HULC upregulated COX-2 showed that HULC enhanced the level of ubiquitin-specific peptidase 22 (USP22), which decrease

Assessing ubiquitination of viral proteins: Lessons from flavivirus NS5.

Ubiquitin (Ub) conjugation to a substrate protein is a widely used cellular mechanism for control of protein stability and function, modulation of signal transduction pathways and antiviral responses. Identification and characterization of ubiquitinated viral proteins is an important step in understanding novel mechanisms of viral protein regulation as well as elucidating cellular antiviral strategies. Here we describe a protocol to easily detect and characterize the ubiquitination status of a viral substrate protein expressed either during infection or ectopically expressed as a fusion with a

Accumulation of polyubiquitylated proteins in response to Ala-Ala-Phe-chloromethylketone is independent of the inhibition of Tripeptidyl peptidase II.

In the present study we have addressed the issue of proteasome independent cytosolic protein degradation. Tripeptidyl peptidase II (TPPII) has been suggested to compensate for a reduced proteasome activity, partly based on evidence using the inhibitor Ala-Ala-Phe-chloromethylketone (AAF-cmk). Here we show that AAF-cmk induces the formation of polyubiquitin-containing accumulations in osteosarcoma and Burkitt's lymphoma cell lines. These accumulations meet many of the landmarks of the aggresomes that form after proteasome inhibition. Using a combination of experiments with chemical inhibitors a

Growth hormone decreases muscle glutamine production and stimulates protein synthesis in hypercatabolic patients.

We determined the effects of 24-h recombinant human growth hormone (rhGH) infusion into a femoral artery on leg muscle protein kinetics, amino acid transport, and glutamine metabolism in eight adult hypercatabolic trauma patients. Metabolic pathways were assessed by leg arteriovenous catheterization and muscle biopsies with the use of stable amino acid isotopes. Muscle mRNA levels of selected enzymes were determined by competitive PCR. rhGH infusion significantly accelerated the inward transport rates of phenylalanine and leucine and protein synthesis, whereas the muscle protein degradation ra

Vepdegestrant: First Approval.

Vepdegestrant (VEPPANU; formerly ARV 471) is a novel PROteolysis TArgeting Chimera (PROTAC) that is able to simultaneously bind the estrogen receptor (ER) and an E3 ubiquitin ligase complex, resulting in polyubiquitination of the ER and its subsequent degradation via a proteasome. It is being developed by Arvinas and Pfizer primarily as a treatment for ER+, HER2- breast cancer. In May 2026, vepdegestrant received its first global approval in the USA for adults with ER+, HER2-, ESR1-mutated advanced or metastatic breast cancer, as detected by an FDA-authorized test, with disease progression fol

Serine 403-phosphorylated p62/SQSTM1 immunoreactivity in inclusions of neurodegenerative diseases.

Protein inclusions in neurodegenerative diseases are associated with p62, which has an important role in autophagic clearance of polyubiquitinated proteins. Selective autophagy is regulated by S403-phosphorylation of p62, and S403-phosphorylated p62 (S403-phos-p62) accumulates in Atg5 conditional knockout (Atg5CKO) mice in which autophagosome formation is impaired. We performed immunohistochemical tests for the presence of S403-phos-p62 in postmortem brain of neurodegenerative disease cases, and found accumulations in amyotrophic lateral sclerosis and Alzheimer's disease tissues. In Atg5CKO an

The Edwardsiella T3SS effector EseJ enhances NF-κB signaling by stabilizing p65 and promoting TAK1 phosphorylation.

Edwardsiella piscicida PPD130/91 is an important pathogen that infects both fish and humans. Upon infection, E. piscicida induces strong inflammatory responses through its type III secretion system (T3SS). The underlying mechanisms remain to be defined. Here, we demonstrate that E. piscicida T3SS effector EseJ activates the NF-κB pathway in macrophages, leading to increased transcription and secretion of multiple pro-inflammatory cytokines (e.g., IL-1β and IL-6). EseJ orchestrates NF-κB signaling by interfering with two ubiquitin-dependent mechanisms that are mediated by separate functional do

A toxic mutant huntingtin species is resistant to selective autophagy.

Protein misfolding is a common theme in neurodegenerative disorders including Huntington's disease (HD). The HD-causing mutant huntingtin protein (mHTT) has an expanded polyglutamine (polyQ) stretch that may adopt multiple conformations, and the most toxic of these is the one recognized by antibody 3B5H10. Here we show that the 3B5H10-recognized mHTT species has a slower degradation rate due to its resistance to selective autophagy in human cells and brains, revealing mechanisms of its higher toxicity.

SH2B3 Promotes PD-L1-Mediated Immune Evasion in Glioblastoma via BRCC3-Dependent IGF2BP2 Stabilization.

Glioblastoma (GBM) is characterized by aggressive progression and profound immune evasion, yet the underlying mechanisms remain incompletely understood. Here, we identify the adaptor protein SH2B3 as a critical driver of immune evasion in glioblastoma and show that elevated SH2B3 expression is associated with higher glioma grade and poorer survival in public datasets and clinical data. Although SH2B3 modulation did not significantly affect GBM cell proliferation, migration, or invasion in vitro assays, it significantly altered tumorigenesis in vivo, with overexpression enhancing and knockdown

Hepatokine fibrinogen-like protein 1 drives liver-kidney crosstalk to promote renal fibrosis.

The concept of organ-organ communication in systemic diseases has gained significant traction. Despite accumulating evidence of liver-kidney crosstalk, the underlying regulatory mechanisms remain poorly understood. Here, we identify hepatokine fibrinogen-like protein 1 (FGL1) as a pivotal mediator driving liver-kidney communication in renal fibrosis. Integrative RNA sequencing and clinical analyses revealed FGL1 upregulation in the liver post-renal injury, with subsequent selective accumulation in fibrotic kidneys. Hepatocyte-specific knockout of FGL1 improved renal morphology, attenuated coll

Reciprocal regulation between JOSD2 and CREB1 drives aerobic glycolysis and progression of renal cell carcinoma.

Dysregulated aerobic glycolysis represents a defining metabolic feature of renal cell carcinoma (RCC), yet the regulatory networks driving this metabolic shift remain incompletely elucidated. In the present study, we found that the deubiquitinase Josephin domain-containing protein 2 (JOSD2) is necessary for both glycolytic and proliferative activities in RCC. Furthermore, JOSD2 is significantly upregulated in RCC and its high expression correlates with poor clinical outcomes. Functional studies demonstrated that JOSD2 enhances RCC cell growth, metastatic capacity, and glycolytic activity in vi

OTUD6A-Mediated Deubiquitination of PRDX1 Protects Against Oral Ulcer by Restoring Mitochondrial Function.

Oral ulcers (OU), as the most highly prevalent and recurrent oral mucosal lesion, have an unclear pathogenesis that hampers the development of effective treatments. While emerging evidence suggests that deubiquitinating enzymes (DUBs) may play a role in oral diseases, their functions in OU remain unclear. In this study, we screened the expression of DUBs in tongue tissues from mice with OU and identified OTU domain-containing protein 6A (OTUD6A) as the most substantially downregulated DUB during OU progression. The functional studies demonstrated that OTUD6A deficiency exacerbated OU pathology

Adaptor AP4M1 promotes hepatocellular carcinoma progression by inhibiting ferroptosis through USP15-mediated deubiquitination and MINK1-dependent phosphorylation.

Hepatocellular carcinoma (HCC) is a highly aggressive malignancy with a poor prognosis, highlighting the urgent need for novel diagnostic and therapeutic targets. In this study, through integrated proteomic profiling, we identify AP4M1 as a potential therapeutic vulnerability for HCC and characterize its upstream regulatory and downstream effector mechanisms. We show that AP4M1 is markedly upregulated in HCC, correlates with unfavorable prognosis, and is functionally involved in driving HCC progression. Mechanistically, the deubiquitinase USP15 stabilizes AP4M1 by removing K11- and K48-linked
