Prevalence of Porphyromonas gingivalis and Bacteroides forsythus in chronic periodontitis by multiplex PCR.

The present research decided to study prevalence of Porphyromonas gingivalis and Bacteroides forsythus in chronic periodontitis patient by use of Multiplex PCR. The subgingival plaque samples from 61 patients suffering from chronic periodontitis with probing depth PD > or = 6 and 40 healthy controls were collected by sterile curette. In this study we used two species-specific Forward primers in combination with a single Reverse primer. These primers target variable and conserved region of 16S rRNA gene, respectively. The study included 61 patients (34 women, 27 men; 24-69 years of age; mean 43…

Proteomics-based analysis of a counter-oxidative stress system in Porphyromonas gingivalis.

Porphyromonas gingivalis is a Gram-negative anaerobic pathogen associated with chronic periodontitis. Although anaerobic, P. gingivalis exhibits a high degree of aerotolerance, which enables it to survive within periodontal pockets. The aim of the present study was to examine the effect of oxidative stress on protein expression in P. gingivalis to obtain a better understanding of the mechanism underlying its aerotolerance. To accomplish this, P. gingivalis cells were grown under conditions of hemin limitation (0.01 microg/mL) to avoid the oxygen protective effect of hemin on oxidative stress. …

Fibrinogen-neutrophil interactions in response to fMLP and Porphyromonas gingivalis fimbrial peptides.

Porphyromonas gingivalis (P.g) is the primary bacterial agent in many forms of chronic periodontitis. Since polymorphonuclear leukocytes (PMNs) are first-line responders to P.g.- induced inflammation, and fibrinogen is important for in vivo PMN in this disease, we have studied the effect of N-formyl-methionyl-leucyl-phenylalanine (fMLP) (an inflammatory stimulus), P.g. fimbriae and fimbrial peptides (based on FimA, the main structural protein of P.g. fimbriae) on PMN-fibrinogen interactions. Freshly isolated human PMNs were allowed to react with FITC-Fibrinogen and various fimbrial peptides (d…

The relationship between Porphyromonas gingivalis infection and local and systemic factors in children.

BACKGROUND: Porphyromonas gingivalis (P. gingivalis) is a major pathogen related to periodontitis. There are few reports on the prevalence and effect of P. gingivalis in children. The purpose of the present study was to describe in children the level of DNA-P gingivalis in subgingival plaque and the IgG serum reactivity to P. gingivalis (IgG-Pg), and examine their relationship to demographic, systemic, and oral variables. METHODS: Thirty-four children, aged 1.83 to 13.42 years, were included in the study; 16 with no systemic disease and 18 with systemic conditions. The demographic data for ca…

Localization of Porphyromonas gingivalis-carrying fimbriae in situ in human periodontal pockets.

Fimbriae, which are involved in adherence, constitute an important pathogenic factor of Porphyromonas gingivalis. In vivo, however, the distribution of P. gingivalis-carrying fimbriae is unknown. The localization of P. gingivalis-carrying fimbriae was examined in situ. From 19 patients with severe periodontitis and P. gingivalis, we obtained 20 teeth with periodontal tissue attached, with and without immunolocalized fimbriae. Eleven teeth were subjected to light microscopy, 9 to electron microscopy. In 6 of the 11 samples examined, we detected positive reactions with an anti-P. gingivalis-fimb…

Characterization and expression of a novel Porphyromonas gingivalis outer membrane protein, Omp28.

We report the characterization of a Porphyromonas gingivalis gene, designated omp28, encoding a protein that we have previously purified and characterized as a 28-kDa outer membrane protein. The deduced amino acid sequence of the omp28 open reading frame displayed an outer membrane leader sequence and lipoprotein attachment site but did not exhibit any significant overall sequence identity with protein sequences in the databases. A small stretch of amino acids (19 residues) exhibits 50% sequence identity with a segment of a fimbrial protein from Dichelobacter nodosus involved in adhesion, sugg…

Role of gingipains in growth of Porphyromonas gingivalis in the presence of human serum albumin.

Porphyromonas gingivalis, a bacterium associated with active chronic periodontitis lesions, produces several proteolytic enzymes that are thought to be involved in host colonization, perturbation of the immune system, and tissue destruction. The aim of the present study was to investigate the contribution of Arg- and Lys-gingipains produced by P. gingivalis to its growth. Although all of the proteins studied were degraded by P. gingivalis, only human serum albumin and transferrin supported growth during serial transfers in a chemically defined medium (CDM). Growth studies with site-directed gi…

Oral ecology and person-to-person transmission of Actinobacillus actinomycetemcomitans and Porphyromonas gingivalis.

The ecological characteristics of the oral cavity are dissimilar for A. actinomycetemcomitans and for P. gingivalis, as judged by differences in their colonization preferences and patterns, associations with periodontal disease parameters, relationships with the subgingival microbiota and the type of periodontitis and their clonal persistence in the oral cavity. These features also suggest that as a periodontal pathogen, A. actinomycetemcomitans is different from P. gingivalis. Probably in most infected individuals, low levels of A. actinomycetemcomitans can persist for years in equilibrium wi

Demonstration of a bimodal coaggregation reaction between Porphyromonas gingivalis and Treponema denticola.

This study demonstrates a strong coaggregation reaction between 2 suspected periodontopathogens: Porphyromonas gingivalis and Treponema denticola. Other black-pigmented oral bacterial species tested did not coaggregate with T. denticola. This specific interbacterial aggregation was bimodal, since heating of both cell types was required to completely eliminate the reaction. The coaggregation reaction occurred between pH 4 and 9. Under some conditions, arginine and D-galactosamine were effective in preventing the coaggregation. The heat-sensitive receptor on P. gingivalis was found to be loosely…

Pathogenesis of Porphyromonas gingivalis peptidylarginine deiminase: from periodontal microenvironment to systemic diseases.

Periodontitis is a globally prevalent chronic inflammatory disease that exerts profound effects on both oral and systemic health. Porphyromonas gingivalis is a key pathogenic bacterium in periodontitis. Its secreted peptidylarginine deiminase (PPAD) is a unique protein-citrullinating enzyme. It catalyzes the citrullination of arginine residues in proteins, thereby altering protein structure and function. This review systematically integrates current evidence to elucidate the enzymatic properties, secretion regulatory mechanisms, and pathogenic roles of PPAD in the periodontal microenvironment …

Ozone ultrafine bubble water sterilizes Porphyromonas gingivalis and neutralizes its virulence factors.

Periodontitis is caused by Gram-negative bacteria. Porphyromonas gingivalis, a major Gram-negative periodontal pathogen, produces virulence factors such as gingipains, lipopolysaccharide (LPS), and lipoproteins, which contribute to tissue destruction. Ozone ultrafine bubble water (OUFBW) has been studied for its antimicrobial effects against various bacteria and toxin protein inactivation. This present study aimed to explore the effects of OUFBW on P. gingivalis and its virulence factors. OUFBW was generated and applied to P. gingivalis to assess bactericidal activity. OUFBW effects on morphol…

Effects and mechanisms of Porphyromonas gingivalis outer membrane vesicles induced cardiovascular injury.

BACKGROUND: The outer membrane vesicles (OMVs) derived from Porphyromonas gingivalis (P. gingivalis) have long been acknowledged for their crucial role in the initiation of periodontitis. However, the implications of P. gingivalis OMVs in the context of cardiovascular disease (CVD) remain incompletely understood. This study aimed to clarify both the impact and the underlying mechanisms through which P. gingivalis OMVs contribute to the propagation of distal cardiovascular inflammation and trauma. METHODS: In this study, various concentrations (0, 1.25, 2.5, and 4.5 µg/µL) of P. gingivalis OMV…

Porphyromonas gingivalis outer membrane vesicles in cerebral ventricles activate microglia in mice.

OBJECTIVE: Porphyromonas gingivalis (Pg) is thought to be involved in the progression of Alzheimer's disease (AD). Whether Pg or its contents can reach the brain and directly affect neuropathology is, however, unknown. Here, we investigated whether outer membrane vesicles (OMVs) of Pg translocate to the brain and induce the pathogenic features of AD. MATERIAL AND METHODS: Pg OMVs were injected into the abdominal cavity of mice for 12 weeks. Pg OMV translocation to the brain was detected by immunohistochemistry using an anti-gingipain antibody. Tau protein and microglial activation in the mous…

Porphyromonas gingivalis Promotes Oral Squamous Cell Carcinoma Progression in an Immune Microenvironment.

Increasing evidence has revealed a significant association between microorganisms and oral squamous cell carcinoma (OSCC). Porphyromonas gingivalis, the keystone pathogen in chronic periodontitis, is considered an important potential etiologic agent of OSCC, but the underlying immune mechanisms through which P. gingivalis mediates tumor progression of the oral cancer remain poorly understood. Our cohort study showed that the localization of P. gingivalis in tumor tissues was related to poor survival of patients with OSCC. Moreover, P. gingivalis infection increased oral lesion multiplicity and…

Antibacterial activity of plant species used for oral health against Porphyromonas gingivalis.

Porphyromonas gingivalis is the keystone pathogen of periodontitis, a chronic inflammatory disease which causes tooth loss and deterioration of gingiva. Medicinal plants have been traditionally used for oral hygiene and health and might play a role as antibacterial agents against oral pathogens. In this work, we aimed to evaluate the antibacterial activity of plants used for oral hygiene or symptoms of periodontitis against P. gingivalis. We first reviewed the literature to identify plant species used for oral hygiene or symptoms of periodontitis. Then, we cross-checked this species list with …
