N6-methyladenosine (m6A) RNA modification plays an important role in plant development and environmental stress responses. However, whether m6A demethylation modulates flowering under low-temperature (LT) stress in tomatoes remains unclear. Here, we investigated whether ectopic expression of FTO, a well-characterized human m6A demethylase, influences flowering and post-transcriptional behaviour in tomato (Solanum lycopersicum) under LT conditions. Flowering of transgenic tomato plants expressing FTO was analyzed under LT and normal conditions (NC), and the impacts of FTO on transcripts-specifi

BACKGROUND: Plant viruses have evolved adaptations that enable them to alter host cues, thereby facilitating their replication and efficient transmission by insect vectors. Satellite RNAs (satRNAs), which accompany certain plant RNA viruses and are dependent on them for replication and transmission, can change the progression of pathogenesis and the expression of disease symptoms. This study aimed to analyse how the change in the course of infection by satRNA (exacerbation or mitigation of pathogenesis) impacts the subsequent stages of virus transmission. We hypothesised that satRNAs influence

Post-transcriptional gene silencing (PTGS) is an evolutionarily conserved plant defense mechanism against viruses. This paper aimed to evaluate a dsDNA construct (77 bp) as a template for in vitro production of virus-derived artificial small hairpin RNAs (shRNAs) and test for their potential to trigger the RNAi mechanism in Nicotiana benthamiana plants against CMV after their foliar infiltration. This approach allowed for the production of significant amounts of shRNAs (60-mers) quickly and easily. The gene silencing was confirmed using polymerase chain reaction (PCR), immunological-based assa

As sessile organisms, plants rely on environmental cues to make key developmental decisions. These decisions are rarely based on a single time point but instead emerge through the integration of past and present experiences, a process broadly referred to as molecular memory. Canonical epigenetic mechanisms operating at the chromatin and RNA levels are often viewed as major drivers of plant molecular memory, yet whether plants also exploit changes in protein state remains an open and compelling question. In this opinion piece, we speculate that prion-like proteins (PrLPs) may function as memory

RNAs that are able to prevent degradation by the 5'-3' exoribonuclease Xrn1 have emerged as crucial structures during infection by an increasing number of RNA viruses. Several plant viruses employ the so-called coremin motif, an Xrn1-resistant RNA that is usually located in 3' untranslated regions. Investigation of its structural and sequence requirements has led to its identification in plant virus families beyond those in which the coremin motif was initially discovered. In this study, we identified coremin-like motifs that deviate from the original in the number of nucleotides present in th

Arbuscular mycorrhizal (AM) fungi are obligate biotrophs whose molecular biology has long been difficult to investigate due to limited genomic resources and the occurrence, in experimental samples, of a high quantity of host plant material. High-throughput short-read RNA sequencing has become a key approach for characterizing AM fungal transcriptomes, but requires tailored bioinformatic strategies to overcome challenges such as mixed-species datasets, low fungal RNA abundance, and the absence of high-quality reference AM fungal genomes. This chapter presents a robust pipeline for AM fungal tra

The separate, advanced treatment of hospital wastewater might be a promising approach to prevent the dissemination of residual compounds of high environmental concern, like pharmaceuticals, viruses and pathogenic microorganisms. This study investigates the performance of a full-scale, on-site treatment plant, consisting of a membrane bioreactor and a subsequent ozonation, at a German hospital. We analysed the elimination of pharmaceutical residues, microbiological parameters and SARS-CoV-2 RNA fragments. Additionally, we conducted an orienting study on the practicability of implementing target

During infection, Citrus tristeza virus (CTV) produces a non-coding subgenomic RNA referred to as low-molecular-weight tristeza 1 (LMT1), which for a long time has been considered as a by-product of the complex CTV replication machinery. In this study, we investigated the role of LMT1 in the virus infection cycle using a CTV variant that does not produce LMT1 (CTV-LMT1d). We showed that lack of LMT1 did not halt virus ability to replicate or form proper virions. However, the mutant virus demonstrated significantly reduced invasiveness and systemic spread in Nicotiana benthamiana as well as an

Benzothiadiazole (BTH) is a novel chemical activator of disease resistance in tobacco, wheat and other important agricultural plants. In this report, it is shown that BTH works by activating SAR in Arabidopsis thaliana. BTH-treated plants were resistant to infection by turnip crinkle virus, Pseudomonas syringae pv 'tomato' DC3000 and Peronospora parasitica. Chemical treatment induced accumulation of mRNAs from the SAR-associated genes, PR-1, PR-2 and PR-5. BTH treatment induced both PR-1 mRNA accumulation and resistance against P. parasitica in the ethylene response mutants, etr1 and ein2, and

Nowadays, the focus of mycovirology research has expanded from plant pathogenic fungi and mycovirus mediated hypovirulence to include insect and human pathogenic fungi together with a range of mycovirus mediated phenotypes, such as hypervirulence, control of endophytic traits, regulation of metabolite production and drug resistance. In fungus-mycovirus-environmental interactions, the environment and both abiotic and biotic factors play crucial roles in whether and how mycovirus mediated phenotypes are manifest. Mycovirus infections result in alterations in the host transcriptome profile, via p

Plant mitochondria share many features with animal mitochondria but also possess unique traits, including presence of both circular and linear mitochondrial DNA, RNA editing, a branched electron transport chain, and metabolic pathways supporting photosynthesis. An accurate mitochondrial proteome is crucial for studying these conserved and lineage-specific functions. Here, we introduce a novel approach to defining plant mitoproteomes that combines serial mitochondrial enrichment, mass spectrometry, and protein correlation profiling. This approach allows us to experimentally define a mitochondri

RNA covalent modifications (RCMs) influence RNA stability and translation efficiency, and they thus play critical roles in eukaryotic growth and development. However, their role in regulating plant performance under abiotic stress remains largely unexplored. Here, we integrated multi-omics data in 6 Sorghum bicolor accessions under water-limiting conditions in the field to explore the relationship between RCMs and drought response. Within a stress- and photosynthesis-associated gene co-expression module, we identified SbDUS2, a member of a family of enzymes conserved across eukaryotes, that ca

Soft rot is a major disease affecting the production of colored calla lily, significantly compromising its ornamental, commercial, and market value. In this study, we identified potential long non-coding RNAs (lncRNAs) in colored calla lily (Zantedeschia elliottiana) leaves infected with soft rot via transcriptome sequencing. Among them, long non-coding RNA 88490 (LNC88940) was identified as a differentially expressed lncRNA that was significantly upregulated following soft rot infection. The results revealed that LNC88940 functions as an endogenous target mimic (eTM) for miR528, reducing the

Calla lily (Zantedeschia spp.), native to southern Africa, is a very attractive plant and is used as cut flowers, potted flowering plants, and bedding plants. It can be distinguished into two groups depending on the color of the flower. The chloroplast genome of the hybrid variety 'Silky White' (Z. aethiopica × Z. aethiopica), a white-flowered calla lily, was sequenced using Illumina sequencing technology. The total length of the chloroplast genome was 168,880 bp with a GC content of 35.59%. It comprised a large single copy (LSC) region of 89,391 bp, a small single copy (SSC) region of 31,849

Witches' broom disease of blue palo verde (Parkinsonia florida) was reported more than sixty years ago. Characteristic symptoms consist of dense clusters of shortened, brittle branches and stunted leaves. The suspect causal agent has been identified as palo verde broom virus (PVBV), genus, Emaravirus, family, Fimoviridae. Here, the first complete PVBV genome sequence was determined, and virus small interfering RNAs (vsiRNAs), primary metabolites, and phytohormone profiles were characterized from infected palo verde leaves, adventitious shoots, flowers, and seeds. Based on pairwise distances, P

Plant mitochondrial genomes (mitogenomes) vary markedly in size and architecture despite generally slow rates of sequence evolution. Phoebe is an ecologically and economically valuable genus of Lauraceae, yet its mitogenome diversity remains poorly characterized. In this study, we newly sequenced, assembled, and annotated the mitogenomes of three nationally protected Class II wild plants (P. bournei, P. chekiangensis, P. zhennan) from China and compared their mitogenomic characteristics. The three assemblies were resolved into representative circular configurations ranging from 808 to 864 kb,

Advances in synthetic biology now offer spatiotemporal, conditional, and precise control of plant metabolism, enabling engineering of pathways that have resisted constitutive approaches. Plant cell wall biosynthesis and metabolite glycosylation are industrially valuable pathways that are also acutely sensitive to constitutive perturbation. We use these pathways as a lens to examine the potential impact of emerging programmable tools, including synthetic, tissue-specific, and inducible promoter design; RNA-, protein-, and pathway-level regulatory switches; and CRISPR-based systems. We then exam

In animals and fungi, small cysteine-rich proteins called metallothioneins (MTs) play a role in heavy metal tolerance. MT genes have been isolated in plants, but their function remains to be elucidated. We have isolated two distinct Vicia faba MT genes that belong to the type 1 group of plant MT genes in contrast to a MT gene we previously isolated that belongs to type 2. We found similarities and differences between the V. faba MT genes. The RNA expression patterns differed and this was most pronounced in roots, which contained high MT1 but very low MT2 RNA levels. Like MT2, MT1 transcript le

Medicinal plants, as a core constituent of traditional medicine, provide an essential foundation for preventing and treating human diseases. Research on their pharmacologically active constituents has long focused on secondary metabolites, including alkaloids, terpenoids, flavonoids, and other compounds. However, with the rapid advancement of molecular biology techniques, nucleic acids have gradually become a research focus as a novel class of active components in medicinal plants that possess both nutritional and regulatory functions. This paper systematically reviews the types, characteristi

Nucleic acid aptamers are short single-stranded DNA/RNA (ssDNA/RNA) oligonucleotides that can selectively bind to the targets. They are widely used in medicine, biosensing, and diagnostic assay. They have also been identified and extensively used for various targets in agriculture. In this review we summarize the progress of nucleic acid aptamers on pesticides (herbicides, insecticides, and fungicides), toxins, specific biomarkers of crops, and plant growth regulators in agricultural field in recent years. The basic process of aptamer selection, the already identified DNA/RNA aptamers and the
