Effects of low-dose γ-radiation on skeletal muscle stem cells.

Introduction: Exposure to low dose ionizing radiation (LDIR) has been associated with aging related health effects. The aging related decline in the regenerative capacity of muscle stem cells in elderly individuals, as well as other myopathy conditions, represents a major health issue. Effects of LDIR exposures, such as those from routine medical CT scans on functional status of muscle stem cells, are not known. Methods: We investigated how a single acute 60Co γ-irradiation (10 and 100 mGy) affected myogenesis into mature muscle fibers in cultures of mouse C2C12 myoblasts and biopsy-derived hum

Radioprotection or radiosensitization? Unraveling caffeine's bifurcated effects on radiation-induced damage.

Caffeine, a widely consumed phytochemical, exhibits a dual and paradoxical role in modulating cellular responses to radiation, acting as both a radioprotector and radiosensitizer depending on context. This comprehensive review synthesizes evidence from in vitro, in vivo, and clinical studies to elucidate caffeine's multifaceted interactions with ionizing (gamma rays, X-rays) and non-ionizing (UV) radiation. Mechanistically, caffeine influences DNA repair pathways-notably inhibiting ATM/ATR checkpoint kinases-overrides G2/M cell cycle arrest, and modulates apoptosis through p53-dependent and in

Inverse dose protraction effects of low-LET radiation: Evidence and significance.

Biological effects of ionizing radiation vary not merely with total dose but also with temporal dose distribution. Sparing dose protraction effects, in which dose protraction reduces effects of radiation have widely been accepted and generally assumed in radiation protection, particularly for stochastic effects (e.g., solid cancer). In contrast, inverse dose protraction effects (IDPEs) in which dose protraction enhances radiation effects have not been well recognized, nor comprehensively reviewed. Here, we review the current knowledge on IDPEs of low linear energy transfer (LET) radiation. To …

New perforated radiation shield for anesthesiologists: Monte Carlo simulation of effects.

Catheterization for structural heart disease (SHD) requires fluoroscopic guidance, which exposes health care professionals to radiation exposure risk. Nevertheless, existing freestanding radiation shields for anesthesiologists are typically simple, uncomfortable rectangles. Therefore, we devised a new perforated radiation shield that allows anesthesiologists and echocardiographers to access a patient through its apertures during SHD catheterization. No report of the relevant literature has described the degree to which the anesthesiologist's radiation dose can be reduced by installing radiatio

Short-term and bystander effects of radiation on murine submandibular glands.

Many patients treated for head and neck cancers experience salivary gland hypofunction due to radiation damage. Understanding the mechanisms of cellular damage induced by radiation treatment is important in order to design methods of radioprotection. In addition, it is crucial to recognize the indirect effects of irradiation and the systemic responses that may alter saliva secretion. In this study, radiation was delivered to murine submandibular glands (SMGs) bilaterally, using a 137Cs gamma ray irradiator, or unilaterally, using a small-animal radiation research platform (SARRP). Analysis at

Age-dependent effects on radiation-induced carcinogenesis in the rat thyroid.

Childhood radiation exposure is a known thyroid cancer risk factor. This study evaluated the effects of age on radiation-induced thyroid carcinogenesis in rats irradiated with 8 Gy X-rays. We analyzed cell proliferation, cell death, DNA damage response, and autophagy-related markers in 4-week-old (4W) and 7-month-old (7M) rats and the incidence of thyroid tumors in 4W, 4-month-old (4M), and 7M rats 18 months after irradiation. Cell death and DNA damage response were increased in 4W rats compared to those in controls at 1 month post-irradiation. More Ki-67-positive cells were observed in 4W rat

Early side effects of radiation treatment for head and neck cancer.

Purpose of this review of medical literature is to present the immediate side effects of radiation therapy for head and neck cancer and their treatment. The likelihood and severity of these immediate side effects depends on a number of factors, including the total dose of radiation delivered, over what time it was delivered and what parts of the head and neck received radiation. Early side effects include: inflammation of the oropharyngeal mucosa (mucositis), painful swallowing (odynophagia), difficulty swallowing (dysphagia), hoarseness, lack of saliva (xerostomia), orofacial pain, laryngeal

Radiation-induced DNA damage and repair effects on 3D genome organization.

The three-dimensional structure of chromosomes plays an important role in gene expression regulation and also influences the repair of radiation-induced DNA damage. Genomic aberrations that disrupt chromosome spatial domains can lead to diseases including cancer, but how the 3D genome structure responds to DNA damage is poorly understood. Here, we investigate the impact of DNA damage response and repair on 3D genome folding using Hi-C experiments on wild type cells and ataxia telangiectasia mutated (ATM) patient cells. We irradiate fibroblasts, lymphoblasts, and ATM-deficient fibroblasts with

Early Tissue Effects of Stereotactic Body Radiation Therapy for Spinal Metastases.

PURPOSE: Stereotactic body radiation therapy (SBRT) is a highly effective and potentially ablative treatment for complex spinal metastases. Recent data have suggested radiobiologic effects of SBRT that expand beyond the traditional concept of DNA damage. Antitumor immunity, vascular damage leading to tumor necrosis, and increased rates of tumor apoptosis have been implied; however, in-human evidence remains scarce. The present study reports unique pathologic confirmation of SBRT-induced biological effects within spinal metastases treated with preoperative SBRT. METHODS AND MATERIALS: Ten pati

Preventive and therapeutic effects of Smad7 on radiation-induced oral mucositis.

We report that K5.Smad7 mice, which express a Smad7 transgene under the control of a keratin 5 promoter, were resistant to radiation-induced oral mucositis, a painful oral ulceration. In addition to nuclear factor κB (NF-κB) activation, which is known to contribute to oral mucositis, we found activated transforming growth factor β (TGF-β) signaling in cells from this condition. Smad7 dampened both pathways to attenuate inflammation, growth inhibition and apoptosis. Additionally, Smad7 promoted oral epithelial migration to close the wound. Further analyses revealed that TGF-β signaling Smads an

Beneficial effects of UV radiation other than via vitamin D production.

Most of the positive effects of solar radiation are mediated via ultraviolet-B (UVB) induced production of vitamin D in skin. However, several other pathways may exist for the action of ultraviolet (UV) radiation on humans as focused on in this review. One is induction of cosmetic tanning (immediate pigment darkening, persistent pigment darkening and delayed tanning). UVB-induced, delayed tanning (increases melanin in skin after several days), acts as a sunscreen. Several human skin diseases, like psoriasis, vitiligo, atopic dermatitis and localized scleroderma, can be treated with solar radia

Protective effects of hominis placenta hydrolysates on radiation enteropathy in mice.

The aim of this study was to investigate whether the hominis placenta hydrolysates (HPhs) have a protective effect against radiation-induced enteropathies. HPh (1-10 mg kg⁻¹, i.p.) was treated to C57BL/6 mice, once daily for 5 days. Mice were irradiated (10 Gy) 1 h after the last injection. Cell damage was investigated at 24 and 72 h by haematoxylin-eosin staining, and the apoptotic index was determined at 24 h by deoxynucleotidyl transferase-mediated dUTP nick end labelling staining. The results showed that the HPh alleviated radiation-induced damage of crypts and suppressed apoptosis dose de
