Hipk2 cooperates with p53 to suppress γ-ray radiation-induced mouse thymic lymphoma.

A genome-wide screen for genetic alterations in radiation-induced thymic lymphomas generated from p53+/- and p53-/- mice showed frequent loss of heterozygosity (LOH) on chromosome 6. Fine mapping of these LOH regions revealed three non-overlapping regions, one of which was refined to a 0.2 Mb interval that contained only the gene encoding homeobox-interacting protein kinase 2 (Hipk2). More than 30% of radiation-induced tumors from both p53+/- and p53-/- mice showed heterozygous loss of one Hipk2 allele. Mice carrying a single inactive allele of Hipk2 in the germline were susceptible to inducti

Unravelling UVA-induced mutagenesis.

Ultraviolet A (UVA) radiation represents more than 90% of the solar UV radiation reaching Earth's surface. Exposure to solar UV radiation is a major risk in the occurrence of non-melanoma skin cancer. Whole genome sequencing data of melanoma tumors recently obtained makes it possible also to definitively associate malignant melanoma with sunlight exposure. Even though UVB has long been established as the major cause of skin cancer, the relative contribution of UVA is still unclear. In this review, we first report on the formation of DNA damage induced by UVA radiation, and on recent advances o

Cross-species identification of genomic drivers of squamous cell carcinoma development across preneoplastic intermediates.

Cutaneous squamous cell carcinoma (cuSCC) comprises 15-20% of all skin cancers, accounting for over 700,000 cases in USA annually. Most cuSCC arise in association with a distinct precancerous lesion, the actinic keratosis (AK). To identify potential targets for molecularly targeted chemoprevention, here we perform integrated cross-species genomic analysis of cuSCC development through the preneoplastic AK stage using matched human samples and a solar ultraviolet radiation-driven Hairless mouse model. We identify the major transcriptional drivers of this progression sequence, showing that the ke

[The prospects of using transgenic insects in biocontrol programs].

Methods of biocontrol are widely used for suppression of pests and human disease vectors. One of the key methods is insects sterilization (sterile insect technique--SIT), which currently is accomplished by irradiation. Radiation-exposed insects have reduced fitness so theis competitive abilities are diminished as compared to insects from wild populations. Modern bioengineering is capable of producing transgenic insects with predetermined traits, and by now the schemes for getting sterile insects without exposure to radiation are developed. Another area of modern studies is producing insects th

Hesperidin reduces genomic instability in glioblastoma cells and is associated with reduced HNRNPA2B1/m6A levels.

Genomic instability contributes to tumor progression and limits the effectiveness of anticancer therapy. In this study, we investigated whether the citrus flavonoid hesperidin influences cytogenetic damage and epitranscriptomic markers in tumor cells exposed to ionizing radiation or maintained under long-term culture conditions. In human glioblastoma cell lines U87MG and LN229, hesperidin reduced radiation-induced micronucleus formation and decreased the spontaneous micronucleus frequency during prolonged passaging with repeated supplementation. Immunocytochemical analysis demonstrated that lo

Radiation-induced late effects in two affected individuals of the Lilo radiation accident.

Radiation exposure leads to a risk for long-term deterministic and stochastic late effects. Two individuals exposed to protracted photon radiation in the radiological accident at the Lilo Military site in Georgia in 1997 received follow-up treatment and resection of several chronic radiation ulcers in the Bundeswehr Hospital Ulm, Germany, in 2003. Multi-parameter analysis revealed that spermatogenetic arrest and serum hormone levels in both patients had recovered compared to the status in 1997. However, we observed a persistence of altered T-cell ratios, increased ICAM1 and beta1-integrin expr

Frequent T:A-->G:C transversions in X-irradiated mouse cells.

Ionizing radiation is a known carcinogen and teratogen. However, the point mutations produced by ionizing radiation in mammalian cells have not been fully characterized. Determination of a characteristic spectrum of X-ray-induced mutations in mammalian cells could provide clues to cellular repair processes and could serve as a marker of individual exposure to radiation. Mouse fibroblasts containing in their genome multiple copies of a recoverable lambda phage shuttle vector were used to detect and analyze radiation-induced point mutations in the supF mutation reporter gene. Following fractiona

The essential role of the Deinococcus radiodurans ssb gene in cell survival and radiation tolerance.

Recent evidence has implicated single-stranded DNA-binding protein (SSB) expression level as an important factor in microbial radiation resistance. The genome of the extremely radiation resistant bacterium Deinococcus radiodurans contains genes for two SSB homologs: the homodimeric, canonical Ssb, encoded by the gene ssb, and a novel pentameric protein encoded by the gene ddrB. ddrB is highly induced upon exposure to radiation, and deletions result in decreased radiation-resistance, suggesting an integral role of the protein in the extreme resistance exhibited by this organism. Although expres

Oxidative genomic damage in humans exposed to high indoor radon levels in Northeast Brazil.

Radon gas inhalation is the main source of exposure to ionizing radiation by humans. There is still lack in knowledge concerning the chronic and indirect effects of exposure to this carcinogenic factor. Therefore, the aim of this work is to analyze the levels of oxidative genomic damage in inhabitants of a medium-high background radiation area (HBRA) (N = 82) in Northeastern Brazil and compare them with people living in a low background radiation area (LBRA) (N = 46). 8-hydroxy-2-deoxyguanosine (8-OHdG) was quantified in urine, Ser326Cys polymorphism was determined in the hOGG1 gene and indoor

Gentiana lutea Root Extract Attenuates Radiation-Induced Damage in Human PBMCs in vitro.

BACKGROUND: Ionizing radiation (IR) has the potential to harm cellular macromolecules, raising concerns across a range of exposure contexts. This study investigates the radioprotective potential of Gentiana lutea root extract (GRE), a natural product known for its antioxidant properties. RESEARCH DESIGN: The GRE composition was analysed using ultra-performance liquid chromatography (UPLC). Peripheral blood mononuclear cells were treated with 0.25-2 mg/mL GRE 24 hours prior to 0.5 and 2 Gy gamma radiation exposure (Co-60 source). Oxidative stress and antioxidant responses were evaluated via li

The Role of STAG2, CDKN2A, and TP53 Genes in Ewing Sarcoma: A Report of a Rare Case of Primary Ewing Sarcoma of the Kidney With Favorable Outcome.

BACKGROUND: Primary renal Ewing sarcoma (EWS) is a rare but aggressive pediatric malignancy with poor outcomes and limited knowledge of its behavior and biology. The use of molecular markers as prognostic factors and local control strategies with surgical resection and radiation is not well-described for renal EWS. OBSERVATION: We document a case of stage IV renal EWS with 4.5 years of durable remission. CONCLUSION: We hypothesize the favorable outcome may be associated with the lack of specific genomic signatures (STAG2, CDKN2A, and TP53) and the use of upfront surgical resection followed b

Role of DNA damage in cardiovascular disease.

Patients with some progeroid syndromes, such as Werner syndrome, exhibit atherosclerotic cardiovascular disease (CVD) at a young age as a manifestation of premature aging. Recent studies have revealed that most progeroid syndromes are caused by genetic defects in specific molecules involved in the DNA damage response, a cornerstone of genome stability. Ionizing radiation is one of the most potent genotoxic stimuli and causes various kinds of DNA damage. Further, there is increasing evidence that therapeutic radiation treatments can cause cardiovascular complications. Here, we describe the DNA

The FOXE1 locus is a major genetic determinant for radiation-related thyroid carcinoma in Chernobyl.

Papillary thyroid cancer (PTC) among individuals exposed to radioactive iodine in their childhood or adolescence is a major internationally recognized health consequence of the Chernobyl accident. To identify genetic determinants affecting individual susceptibility to radiation-related PTC, we conducted a genome-wide association study employing Belarusian patients with PTC aged 0-18 years at the time of accident and age-matched Belarusian control subjects. Two series of genome scans were performed using independent sample sets, and association with radiation-related PTC was evaluated. Meta-ana

The characterization and transmissibility of chromosome aberrations induced in peripheral blood lymphocytes by in vitro alpha-particle radiation.

Peripheral blood lymphocytes were irradiated in vitro with (213)Bi alpha particles at doses of 0, 10, 20, 50, 100, 200 and 500 mGy. Chromosome analysis was performed on 47-h cultures using single-color fluorescence in situ hybridization (FISH) to paint chromosomes 1, 3 and 5. The whole genome was analyzed for unstable aberrations to derive aberration frequencies and determine cell stability. The dose response for dicentrics was 33.60 +/- 0.47 x 10(-2) per Gy. A more detailed analysis revealed that the majority of aberrations scored as dicentrics were part of complex/multiple aberrations, with

Localized movement and morphology of UBF1-positive nucleolar regions are changed by γ-irradiation in G2 phase of the cell cycle.

The nucleolus is a well-organized site of ribosomal gene transcription. Moreover, many DNA repair pathway proteins, including ATM, ATR kinases, MRE11, PARP1 and Ku70/80, localize to the nucleolus (Moore et al., 2011 ). We analyzed the consequences of DNA damage in nucleoli following ultraviolet A (UVA), C (UVC), or γ-irradiation in order to test whether and how radiation-mediated genome injury affects local motion and morphology of nucleoli. Because exposure to radiation sources can induce changes in the pattern of UBF1-positive nucleolar regions, we visualized nucleoli in living cells by GFP-

Pan-Cancer Gene Expression Predicts Per-Cancer Radiation Therapy Response.

PURPOSE: Although radiation therapy response involves biological processes shared across cancers, most biomarker studies rely on small, disease-specific sample sizes that limit the reliability of prognostic models. This study evaluated whether pan-cancer data can increase the information available for estimating genomic associations with progression after radiation therapy, based on Molecular Signatures Database hallmark gene sets applied to The Cancer Genome Atlas patients. METHODS AND MATERIALS: Cross-cancer predictive potential was evaluated in cancer types with at least 100 patients who r

NADPH Oxidase as a Target for Modulation of Radiation Response; Implications to Carcinogenesis and Radiotherapy.

BACKGROUND: Radiotherapy is a treatment modality for cancer. For better therapeutic efficiency, it could be used in combination with surgery, chemotherapy or immunotherapy. In addition to its beneficial therapeutic effects, exposure to radiation leads to several toxic effects on normal tissues. Also, it may induce some changes in genomic expression of tumor cells, thereby increasing the resistance of tumor cells. These changes lead to the appearance of some acute reactions in irradiated organs, increased risk of carcinogenesis, and reduction in the therapeutic effect of radiotherapy. DISCUSSI

The Influence of Micronutrients and Environmental Factors on Thyroid DNA Integrity.

Micronutrients and environmental factors are key exogenous agents influencing thyroid DNA integrity. Micronutrients act as cofactors in DNA replication, repair, and antioxidant defence, while environmental exposure, such as radiation, heavy metals, and endocrine-disrupting chemicals, can directly damage DNA, leading to genomic instability. Although many studies have confirmed the link between micronutrient status and thyroid health, the effects of nutrient imbalances and environmental stressors on thyroid DNA remain underexplored. This narrative review examines how these factors may compromise
