Stem cells for urinary tract regeneration.

Regeneration of the urinary bladder is a complicated task, due to organ dimensions and diseases (cancer, interstitial cystitis) when autologous bladder cells cannot be used. Cancer is the most frequent indication for bladder removal (cystectomy). Stem cells can be used with the guarantee of the sufficient cell number for the in vitro construction of the urinary bladder wall. Tissue engineering techniques hold great promise for regeneration of dysfunctional urinary sphincter. Denervation following surgical procedures or injuries results in weakness of the urethral sphincter and stress urinary i…

Septal perforation repair: mucosal regeneration technique.

A novel method for repair of septal perforations. Fifteen volunteers with symptomatic septal perforations were recruited. Open technique rhinoplasty approach was preferred: auricular conchal cartilage graft with intact perichondrium on both sides was harvested and shaped to fit the perforated site and attached to the septum with absorbable sutures. All margins of the graft were covered with nasal mucosa. The severity of patient symptoms was assessed at preoperation, 3 and 6 months postoperatively via visual analogue scale (VAS). Crust formation, whistling, nasal blockage, epistaxis and overall

Endoderm specification, liver development, and regeneration.

The endoderm is the innermost germ layer that gives rise to the lining of the gut, the gills, liver, pancreas, gallbladder, and derivatives of the pharyngeal pouch. These organs form the gastrointestinal tract and are involved with the absorption, delivery, and metabolism of nutrients. The liver has a central role in regulating these processes because it controls lipid metabolism, protein synthesis, and breakdown of endogenous and xenobiotic products. Liver dysfunction frequently leads to significant morbidity and mortality; however, in most settings of organ injury, the liver exhibits remarka

Augmenting nerve regeneration with electrical stimulation.

OBJECTIVE: Poor functional recovery after peripheral nerve injury is generally attributed to irreversible target atrophy. In rats, we addressed the functional outcomes of prolonged neuronal separation from targets (chronic axotomy for up to 1 year) and atrophy of Schwann cells (SCs) in distal nerve stumps, and whether electrical stimulation (ES) accelerates axon regeneration. In carpal tunnel syndrome (CTS) patients with severe axon degeneration and release surgery, we asked whether ES accelerates muscle reinnervation. METHODS: Reinnervated motor unit (MUs) and regenerating neuron numbers wer…

Bioengineered nerve regeneration and muscle reinnervation.

The peripheral nervous system has the intrinsic capacity to regenerate but the reinnervation of muscles is often suboptimal and results in limited recovery of function. Injuries to nerves that innervate complex organs such as the larynx are particularly difficult to treat. The many functions of the larynx have evolved through the intricate neural regulation of highly specialized laryngeal muscles. In this review, we examine the responses of nerves and muscles to injury, focusing on changes in the expression of neurotrophic factors, and highlight differences between the skeletal limb and laryng

Expression of survivin during liver regeneration.

Survivin functions to suppress cell death and regulate cell division, and is observed uniquely in tumor cells and developmental cells. However, the expression and regulation of survivin in non-transformed cells are not well elucidated. Therefore, we investigated the expression of survivin in a murine liver regeneration model after partial hepatectomy and intraperitoneal carbon tetrachloride (CCl(4)) injection. We found that the expression of survivin transcript and protein were markedly elevated with the onset of DNA synthesis and remained elevated during G2 and M phases during liver regenerat…

Periodontal regeneration with polypeptide growth factors.

Attempts at periodontal regeneration have been plagued by inconsistent results. The failures associated with these procedures are likely due to an insufficient number of cells themselves lacking sufficient activity to produce the supporting structures of the periodontium. Molecular studies have provided insight into the regenerative process by identifying locally acting mediators expressed during healing. These mediators, termed growth factors, may stimulate a wide variety of cellular events, including chemotaxis, proliferation, differentiation, and production of extracellular matrix proteins.…

Guided tissue regeneration in mucogingival surgery.

The use of expanded polytetrafluoroethylene (ePTFE) membranes in the treatment of mucogingival problems such as gingival recession, has been recently described. The major issues raised dealt with creating and maintaining space under the membrane and designing a flap that could be coronally positioned and at the same time capable of maintaining a good blood supply. The authors proposed the use of a trapezoidal flap and some technical space-making solutions, such as the use of a suturing technique and different metal structures to support the membranes. J Periodontol 1993; 64:1184-1191.

Periodontal regeneration via selective cell repopulation.

The purpose of this investigation was to determine the histogenesis of periodontal regeneration using the principle of selective and guided cell repopulation of the root surface. A fenestration model, developed in the squirrel monkey, made it possible to exclude gingival epithelium and connective tissue and promote cell repopulation of the denuded root surface from the periodontal ligament and alveolar bone. Experimental and sham-operated control sites were compared histometrically after 3, 7, 14 and 35 days of healing. The results indicated that new cementum, bone and periodontal ligament for…

Mechanical analysis of tracheal anastomosis reinforcement through cartilage regeneration: a finite element study on regeneration site configuration.

Postoperative bronchial fistula and anastomotic stenosis remain significant concerns in lung cancer surgery requiring bronchial reconstruction. Previous studies have shown that basic fibroblast growth factor (bFGF)-induced cartilage regeneration could stabilize anastomosis sites and replace conventional tissue coverage. To inform the optimal configuration of bFGF-based protocols, this study evaluated the relative mechanical effects of varying the number and arrangement of regeneration sites. We developed a mechanically equivalent two-dimensional finite element model based on rabbit tracheal an…

Atf3 mutant mice show reduced axon regeneration and impaired regeneration-associated gene induction after peripheral nerve injury.

Axon injury in the peripheral nervous system (PNS) induces a regeneration-associated gene (RAG) response. Atf3 (activating transcription factor 3) is such a RAG and ATF3's transcriptional activity might induce 'effector' RAGs (e.g. small proline rich protein 1a (Sprr1a), Galanin (Gal), growth-associated protein 43 (Gap43)) facilitating peripheral axon regeneration. We provide a first analysis of Atf3 mouse mutants in peripheral nerve regeneration. In Atf3 mutant mice, facial nerve regeneration and neurite outgrowth of adult ATF3-deficient primary dorsal root ganglia neurons was decreased. Usin…

The Role of Guided Bone Regeneration in Enhancing Dental Implant Success : Guided Bone Regeneration in Dental Implant.

Guided Bone Regeneration (GBR) has become an essential technique in dental implantology, particularly for cases with compromised bone volume that impact implant success. This narrative review examines the role of GBR in enhancing dental implant outcomes, focusing on its applications, biological principles, materials, and clinical protocols. GBR utilizes barrier membranes and bone graft materials to foster bone regeneration in deficient areas, creating a stable foundation for implant placement by preventing soft tissue invasion and promoting osteogenic activity. Through a literature review of r…

Theoretical study on the catalytic mechanism of oxyluciferin hydrolysis by luciferin-regenerating enzyme in firefly bioluminescence substrate regeneration.

Firefly bioluminescence (BL) is widely utilized in bioimaging due to its high quantum yield; however, its performance in long-term monitoring is limited by rapid signal decay resulting from product inhibition by oxyluciferin (oLu). In nature, fireflies employ the luciferin-regenerating enzyme (LRE) to catalyze oLu hydrolysis, thereby removing inhibition and initiating substrate regeneration for subsequent emission. While previous studies have established the three-step regeneration pathway-LRE-catalyzed oLu hydrolysis, CHBT condensation with L-Cys, and L/D-luciferin isomerization-the initiatin…

Differential expression of unconventional myosins in apoptotic and regenerating chick hair cells confirms two regeneration mechanisms.

Hair cells of the inner ear are damaged by intense noise, aging, and aminoglycoside antibiotics. Gentamicin causes oxidative damage to hair cells, inducing apoptosis. In mammals, hair cell loss results in a permanent deficit in hearing and balance. In contrast, avians can regenerate lost hair cells to restore auditory and vestibular function. This study examined the changes of myosin VI and myosin VIIa, two unconventional myosins that are critical for normal hair cell formation and function, during hair cell death and regeneration. During the late stages of apoptosis, damaged hair cells are ej…

Deferoxamine Promotes Peripheral Nerve Regeneration by Enhancing Schwann Cell Function and Promoting Axon Regeneration of Dorsal Root Ganglion.

Deferoxamine (DFO) is a potent iron chelator for clinical treatment of various diseases. Recent studies have also shown its potential to promote vascular regeneration during peripheral nerve regeneration. However, the effect of DFO on the Schwann cell function and axon regeneration remains unclear. In this study, we investigated the effects of different concentrations of DFO on Schwann cell viability, proliferation, migration, expression of key functional genes, and axon regeneration of dorsal root ganglia (DRG) through a series of in vitro experiments. We found that DFO improves Schwann cell …

Pancreatic duct ligation after almost complete β-cell loss: exocrine regeneration but no evidence of β-cell regeneration.

There has been great interest in the extent of β-cell regeneration after pancreatic duct ligation (PDL) and whether α- to β-cell conversion might account for β-cell regeneration after near-complete β-cell loss. To assess these questions, we established a PDL-model in adult male rats after almost complete beta-cell depletion achieved by giving a single high dose of streptozocin (STZ) in the fasted state. Because of the resultant severe diabetes, rats were given islet cell transplants to allow long-term follow-up. Although animals were followed up to 10 months, there was no meaningful β-cell reg…
