Ewing sarcoma is the second most common pediatric bone tumor, with three cases per million worldwide. In clinical terms, Ewing sarcoma is an aggressive, rapidly fatal malignancy that mainly develops not only in osseous sites (85%) but also in extra-skeletal soft tissue. It spreads naturally to the lungs, bones, and bone marrow with poor prognosis in the two latter cases. Bone lesions from primary or secondary (metastases) tumors are characterized by extensive bone remodeling, more often due to osteolysis. Osteoclast activation and subsequent bone resorption are responsible for the clinical fea

Williams syndrome (WS) is a relatively rare multisystem neurodevelopmental disorder caused by a hemizygous deletion of contiguous genes on chromosome 7q11.23. Although WS does not predispose carriers to cancers, alterations of chromosome 7 are common in several human neoplasms. We report here a patient with WS and two different cancers, Burkitt lymphoma and Ewing sarcoma. Array-CGH analysis of the patient blood revealed a constitutive 1.4 million base pair deletion at 7q11.23, compatible with WS diagnosis.

Extra skeletal Ewing Sarcoma (EES) is a rare entity which predominantly occurs in adolescents and young adults. It usually arises from the soft tissues of the trunk or the extremities. We present a case of EES arising from the left visceral pleura in a 21 year old female patient who presented to the emergency room of our institution with fever, productive cough and sternal pain radiating to the back for the last 3 days. Chest radiograph was firstly performed, followed by chest CT examination. Finally open lung biopsy revealed a small round cell malignancy. The mass was resected and the histolo

The Ewing sarcoma (ES) family of tumors is characterized by nonrandom chromosomal translocations involving the EWSR1 gene on chromosome 22 with one of the members of the ETS family of transcription factors. The majority of ES tumors are characterized by a balanced translocation t(11;22)(q24;q12), which results in the fusion of the 5' portion of EWSR1 gene with the 3'end of the FLI1 gene. Fusions with ERG, another member of the ETS family, occur in less than 10% of ES tumors, and can arise through complex chromosomal rearrangements. Here, we report a case of a 5-year-old female with an ES tumor

Second malignant neoplasms (SMNs) are being increasingly recognized. This report describes a case of a 7-year-old girl with a history of acute lymphoblastic leukemia (ALL) who presented with a mass in her humerus that was diagnosed as Ewing sarcoma. Second malignant neoplasms are relatively rare in survivors of ALL treated without radiation. Even more unusual is the development of Ewing sarcoma as the SMN.

PURPOSE: To compare MR imaging features of stress fractures, simulating malignancies, and Ewing sarcomas.
MATERIALS AND METHODS: MR imaging studies of 4 patients with longitudinal stress fractures of the tibia (n = 2) and the femur (n = 2) simulating malignancy were retrospectively compared with the MRI scans of 10 patients with histologically proven Ewing sarcoma (femur n = 5; tibia n = 3, fibula n = 1, humerus n = 1). The diagnosis of stress fractures was confirmed by follow-up examinations. An additional biopsy was performed in two patients.
RESULTS: Despite negative x-ray examinations, M

Roentgenographic review of the first 210 cases of Ewing sarcoma (ES) in the Intergroup Ewing Sarcoma Study revealed that 37.6% of cases had evidence of diffusely increased intraosseous density or diffuse sclerosis (DS). In these cases the sclerosis was usually mixed with various patterns of lysis and/or combined with a periosteal reaction. A radiograph blinded histologic review of selected biopsies showed an 83% incidence of dead bone compared to 23% in those without DS. Ten percent of the cases with DS had appositional new bone formation on dead bone whereas none of the cases without DS showe

We report on a twenty-four-year-old woman with extraosseous Ewing sarcoma in the right retroperitoneum. The tumor blocked the common iliac vein, and because of an intervening pregnancy, the blood flow was shunted through the uterus and left ovarian vein back into the vena cava. She died within nine months despite radical surgery, radiotherapy, and chemotherapy.

High-quality single-cell analyses critically depend on standardized tissue processing workflows that preserve cellular integrity and minimize dissociation-induced artifacts. Here, we describe a robust and reproducible protocol for the isolation, cryopreservation, and downstream processing of single-cell suspensions from Ewing sarcoma (EwS) tumors, including the bioinformatic analyses and tools to interrogate the resulting data. The workflow is optimized for strict cold-chain handling and employs a cold-active Bacillus licheniformis protease-based digestion strategy coupled with gentle mechanic

Renal Ewing sarcoma (EWS) is an exceptionally rare pediatric malignancy that may mimic neuroblastoma on conventional imaging. We report a 6-year-old boy presenting with a left suprarenal mass suspicious for neuroblastoma. The initial biopsy was inconclusive. I-131 metaiodobenzylguanidine (mIBG) scintigraphy demonstrated no tracer uptake within the mass. Subsequent Ga-68 DOTANOC PET/CT revealed only mild somatostatin receptor expression, whereas F-18 FDG PET/CT showed intense hypermetabolism. Given the discordant molecular imaging profile, repeat biopsy with fluorescence in situ hybridization c

PURPOSE: PARP inhibitors (PARPi) proved effective in Ewing sarcoma cells in preclinical studies. In clinical evaluation, however, the PARPi olaparib failed to elicit substantial responses, suggesting an unknown mechanism of resistance to PARPi in Ewing sarcoma. Since autophagy has been identified as a PARPi resistance mechanism in other tumours, this study aimed at exploring the impact of autophagy on PARPi effectiveness in Ewing sarcoma cells.
METHODS: Effects of the PARPi olaparib and veliparib were assessed by flow cytometry in the Ewing sarcoma cell lines WE-68 and SK-ES-1. Autophagy leve

Subpopulations of cancer-associated fibroblast (CAF)-like tumor cells deposit extracellular matrix (ECM) proteins that support Ewing sarcoma (EwS) progression and metastasis. We previously showed a hallmark of CAF-like EwS cells is their hybrid transcriptional state wherein the driver fusion oncogene, EWS::FLI1, maintains activation of proliferative programs but loses capacity to repress mesenchymal genes. Here, we studied primary patient tumors and cell line models to identify molecular drivers of this hybrid state. Our data reveal that hybrid EwS cells are induced and maintained by a transfo

BACKGROUND Paravertebral extraskeletal Ewing sarcoma located in the extradural region is uncommon. Muscle denervation can be caused by nerve injury, destruction of the neuromuscular junctions, and aging-related loss of motor neurons. Psoas denervation caused by spinal extraskeletal Ewing sarcoma is extremely rare. CASE REPORT A 38-year-old Chinese man presented to our hospital with back pain lasting for 1 month. He presented thoracolumbar mobility limitations. Magnetic resonance imaging revealed a well-defined margin extradural soft tissue mass involving the intervertebral foramen and paravert

Extraskeletal Ewing sarcoma (EES) is a rare subtype within the Ewing sarcoma family of tumors (ESFT), typically presenting as a large soft tissue mass. Imaging features of EES are generally non-specific and vary depending on the tumor's location and the tissues involved. Magnetic resonance imaging (MRI) plays a crucial role in the initial evaluation, local staging, and ongoing management. A definitive diagnosis requires core needle biopsy, immunochemistry, and histopathological confirmation. Treatment typically involves systemic chemotherapy combined with surgical excision when feasible. Due t

Adamantinoma-like Ewing sarcoma (ALES) of the parotid gland is an exceptionally rare neoplasm. We report the case of a 57-year-old male with a history of p16-positive tonsillar squamous cell carcinoma who presented 2 years after remission with excessive mucus production and neck stiffness. Imaging identified a left parotid mass, and diagnosis was confirmed through surgical pathology, with molecular testing revealing a Fused in Sarcoma-Fifth Ewing Variant (FUS-FEV) fusion gene. Treatment included radical parotidectomy, adjuvant external beam radiation, and systemic chemotherapy following the Ew

Primary extraosseous intracranial Ewing sarcoma (ES) is an extremely rare disease, limited to the pediatric population, that primarily originates in the skull. Here, we present an unusual case of adult Ewing's sarcoma originating from the brain parenchyma. The 50-year-old male patient visited our hospital with severe headache lasting 3 weeks. MRI presented 6.1×6.2×5.2 cm sized heterogeneously enhanced mass containing peritumoral edema in the right frontal lobe. The patient underwent right frontal craniotomy, at which time the gray and red masses adhered to the surrounding brain parenchyma. The
