Contextual gating of whisker-evoked responses by frontal cortex supports flexible decision making.

Context-dependent sensory processing underlies important aspects of flexible behavior. Here, we investigate how mice can use a briefly-presented auditory contextual Go or Nogo cue after a delay period to gate the transformation of a whisker deflection into licking for reward. Spatiotemporally-specific optogenetic inactivation demonstrated an important contribution of various cortical regions during distinct trial epochs, with only whisker secondary motor cortex (wM2) contributing strongly to all task epochs including auditory cue, delay and whisker stimulus. Electrophysiological recordings rev

Pragmatic representations of self- and others' action in the monkey putamen.

Social coordination in primates relies on parieto-frontal networks encoding self- and others' actions. These areas send convergent projections to the putamen, but its role in representing self- and others' actions remains unknown. We recorded neuronal activity from anatomically characterized putamen regions of two male macaques during a Mutual Action Task (MAT), where a monkey and a human took turns grasping a multi-affordance object based on sensory cues. Cortico-striatal synaptic input, indexed by local field potentials, mirrored known cortical dynamics during sensory instructions and moveme

Linking sensory experience to bodily states in the insular cortex.

The brain must learn to associate sensory experiences with delayed physiological consequences in order to generate adaptive behavior. The insular cortex is uniquely positioned to support this function, integrating gustatory, visceral, immune, pain, affective, and metabolic information into representations of bodily states. In this review, we examine evidence, primarily from mouse and rat studies and with reference to human work, that insular circuits couple external cues to predicted bodily states. We focus on conditioned taste aversion and conditioned immune responses as complementary models

A review of the neural basis underlying the acoustic startle response with a focus on recent developments in mammals.

The startle response consists of whole-body muscle contractions, eye-blink, accelerated heart rate, and freezing in response to a strong, sudden stimulus. It is evolutionarily preserved and can be observed in any animal that can perceive sensory signals, indicating the important protective function of startle. Startle response measurements and its alterations have become a valuable tool for exploring sensorimotor processes and sensory gating, especially in the context of pathologies of psychiatric disorders. The last reviews on the neural substrates underlying acoustic startle were published a…

Interaction between Spatial and Feature Attention in Posterior Parietal Cortex.

Lateral intraparietal (LIP) neurons encode a vast array of sensory and cognitive variables. Recently, we proposed that the flexibility of feature representations in LIP reflect the bottom-up integration of sensory signals, modulated by feature-based attention (FBA), from upstream feature-selective cortical neurons. Moreover, LIP activity is also strongly modulated by the position of space-based attention (SBA). However, the mechanisms by which SBA and FBA interact to facilitate the representation of task-relevant spatial and non-spatial features in LIP remain unclear. We recorded from LIP neur

Abnormal tactile discrimination and somatosensory plasticity in familial primary hyperhidrosis.

Abnormal sensory processing seems to be involved in hyperhidrosis. To test this hypothesis, we investigated tactile acuity and cortical plastic changes in patients with primary hyperhidrosis (PH) and their asymptomatic relatives. We studied thirteen subjects belonging to two families with PH and thirteen age-matched healthy controls using Johnson-Van Boven-Phillips domes before and after 45min of transient visual deafferentation. Spatial discrimination thresholds (SDTs) were lower in controls than in the familial group (1.08+/-0.25 vs 1.59+/-0.71; p=0.0032). After 45min of light deprivation an

The Rise and Fall of TRP-N, an Ancient Family of Mechanogated Ion Channels, in Metazoa.

Mechanoreception, the sensing of mechanical forces, is an ancient means of orientation and communication and tightly linked to the evolution of motile animals. In flies, the transient-receptor-potential N protein (TRP-N) was found to be a cilia-associated mechanoreceptor. TRP-N belongs to a large and diverse family of ion channels. Its unusually long N-terminal repeat of 28 ankyrin domains presumably acts as the gating spring by which mechanical energy induces channel gating. We analyzed the evolutionary origins and possible diversification of TRP-N. Using a custom-made set of highly discrimin

Stem cell-derived sensory neurons modelling inherited erythromelalgia: normalization of excitability.

Effective treatment of pain remains an unmet healthcare need that requires new and effective therapeutic approaches. NaV1.7 has been genetically and functionally validated as a mediator of pain. Preclinical studies of NaV1.7-selective blockers have shown limited success and translation to clinical studies has been limited. The degree of NaV1.7 channel blockade necessary to attenuate neuronal excitability and ameliorate pain is an unanswered question important for drug discovery. Here, we utilize dynamic clamp electrophysiology and induced pluripotent stem cell-derived sensory neurons (iPSC-SNs

Combined prior chronic methamphetamine treatment and gp120 expression reduce PPI in aged male but not female mice.

Methamphetamine (METH) use disorder is highly prevalent among people with HIV and is a significant public health problem. Furthermore, people with HIV are living longer and using drugs such as METH even into old age, making it important to understand the effects of METH use and aging in this population. HIV, METH, and aging negatively impact a variety of brain functions, including sensorimotor gating (i.e. - automatic, pre-conscious information processing). Sensorimotor gating is often measured using prepulse inhibition (PPI), a paradigm that can be conducted in animals, thereby allowing for p

The Role of Voltage-Gated Sodium Channels in Pain Signaling.

Acute pain signaling has a key protective role and is highly evolutionarily conserved. Chronic pain, however, is maladaptive, occurring as a consequence of injury and disease, and is associated with sensitization of the somatosensory nervous system. Primary sensory neurons are involved in both of these processes, and the recent advances in understanding sensory transduction and human genetics are the focus of this review. Voltage-gated sodium channels (VGSCs) are important determinants of sensory neuron excitability: they are essential for the initial transduction of sensory stimuli, the elect

A vibrissa pathway that activates the limbic system.

Vibrissa sensory inputs play a central role in driving rodent behavior. These inputs transit through the sensory trigeminal nuclei, which give rise to the ascending lemniscal and paralemniscal pathways. While lemniscal projections are somatotopically mapped from brainstem to cortex, those of the paralemniscal pathway are more widely distributed. Yet the extent and topography of paralemniscal projections are unknown, along with the potential role of these projections in controlling behavior. Here, we used viral tracers to map paralemniscal projections. We find that this pathway broadcasts vibri

Acoustic trauma increases inhibitory effects of amygdala electrical stimulation on thalamic neurons in a rat model.

Acoustic trauma (AT) induced hearing loss elicits plasticity throughout the central auditory pathway, including at the level of the medial geniculate nucleus (MGN). Hearing loss also results in altered neuronal responses in the amygdala, which is involved in sensory gating at the level of the MGN. However, whether these altered responses in the amygdala affect sensory gating at the level of the MGN requires further evaluation. The current study aimed to investigate the effects of AT-induced hearing loss on the functional connectivity between the amygdala and the MGN. Male Sprague-Dawley rats w…

Multisensory dysfunction across sleep disorders: A systematic review of 31 human modalities.

Sleep disorders are increasingly understood as systems-level conditions in which the brain and body reweight sensory evidence from the external world and internal milieu. We systematically reviewed human evidence for sensory and multisensory dysfunction across sleep disorders and experimental sleep loss. Searches identified 25,777 records; after removal of 7235 duplicates, 18,542 records were screened, 4452 full records were assessed for eligibility, and 377 studies met inclusion criteria. Evidence was organised across 31 sensory modalities in seven domains: olfaction, gustation, vision, audit

Multimodal Integration Across Spatiotemporal Scales to Guide Invertebrate Locomotion.

Locomotion is a hallmark of organisms which has enabled adaptive radiation to an extraordinarily diverse class of ecological niches, and allows animals to move across vast distances. Sampling from multiple sensory modalities enables animals to acquire rich information to guide locomotion. Locomotion without sensory feedback is haphazard; therefore, sensory and motor systems have evolved complex interactions to generate adaptive behavior. Notably, sensory-guided locomotion acts over broad spatial and temporal scales to permit goal-seeking behavior, whether to localize food by tracking an attrac

Are voltage-gated sodium channels on the dorsal root ganglion involved in the development of neuropathic pain?

Neuropathic pain is a common clinical condition. Current treatments are often inadequate, ineffective, or produce potentially severe adverse effects. Understanding the mechanisms that underlie the development and maintenance of neuropathic pain will be helpful in identifying new therapeutic targets and developing effective strategies for the prevention and/or treatment of this disorder. The genesis of neuropathic pain is reliant, at least in part, on abnormal spontaneous activity within sensory neurons. Therefore, voltage-gated sodium channels, which are essential for the generation and conduc

What do we really know and what do we need to know: some controversies, perspectives, and surprises.

TRP channels comprise one of the most rapid growing research topics in ion channel research, in fields related to ion channels including channelopathies and translational medicine. We provide here a critical survey on our current knowledge of TRP channels and highlight some of the still open or controversial questions. This comprises questions related to evolution of TRP channels; biophysics, i.e., permeation; pore properties and gating; modulation; the still-elusive 3D structure; and channel subunits but also their role as general sensory channels and in human diseases. We will conclude that

Sensorimotor gating in boys with Tourette's syndrome and ADHD: preliminary results.

Deficits in sensorimotor gating, defined by prepulse inhibition (PPI), have been associated with subcortical dopaminergic overactivity in animal and clinical studies. Utilizing supraorbital nerve electrical stimulation, we produced adequate blink responses and measured decreases in amplitude resulting from electric prestimuli just above sensory threshold. Seven boys comorbid for attention-deficit hyperactivity disorder (ADHD) and a tic disorder had significantly reduced PPI, compared to 14 screened controls and seven boys with ADHD alone. If independently replicated, these results may reflect
