Serum sialic acid levels in Vibrio cholera serotype Hikojima infected Nigerian patients.

BACKGROUND AND AIMS: Vibro cholera neuraminidase is known to cleave sialic acid in the gut to expose receptors for cholera enterotoxin. This study determined if cholera neuraminidase crosses significantly into the circulation of patients with cholera to cause cleavage of sialic acid on circulating blood cells. METHODS: Systemic cholera neuraminidase activity was determined by measuring serum sialic acid levels (thiobarbituric method) in 20 consecutive adult cholera patients (12 females, 8 males) with severe diarrhea before rehydration was commenced at Ahmadu Bello University Teaching Hospital

Sialic acid is a cellular receptor for coxsackievirus A24 variant, an emerging virus with pandemic potential.

Binding to target cell receptors is a critical step in the virus life cycle. Coxsackievirus A24 variant (CVA24v) has pandemic potential and is a major cause of acute hemorrhagic conjunctivitis, but its cellular receptor has hitherto been unknown. Here we show that CVA24v fails to bind to and infect CHO cells defective in sialic acid expression. Binding of CVA24v to and infection of corneal epithelial cells are efficiently inhibited by treating cells with a sialic acid-cleaving enzyme or sialic acid-binding lectins and by treatment of the virus with soluble, multivalent sialic acid. Crystal structure of species D adenovirus fiber knobs and their sialic acid binding sites.

Adenovirus serotype 37 (Ad37) belongs to species D and can cause epidemic keratoconjunctivitis, whereas the closely related Ad19p does not. Primary cell attachment by adenoviruses is mediated through receptor binding of the knob domain of the fiber protein. The knobs of Ad37 and Ad19p differ at only two positions, Lys240Glu and Asn340Asp. We report the high-resolution crystal structures of the Ad37 and Ad19p knobs, both native and in complex with sialic acid, which has been proposed as a receptor for Ad37. Overall, the Ad37 and Ad19p knobs are very similar to previously reported knob structure

Brucella sp. bind to sialic acid residues on human and animal red blood cells.

We report that Brucella abortus and Brucella melitensis agglutinate human (A+ and B+), hamster and rabbit erythrocytes, a heretofore undescribed feature in this genus. This activity was associated with a 29-kDa surface protein (SP29) that bound selectively to these erythrocytes and this binding was inhibited by rabbit anti-SP29 antibodies. Hemagglutination was inhibited by pretreatment of erythrocytes with neuraminidase and by preincubation of B. abortus with chondroitin sulfate, N-acetylneuraminic acid and N-acetylneuramin-lactose.

Infantile sialic acid storage disease: a rare cause of cytoplasmic vacuolation in pediatric patients.

We report a case of infantile sialic acid storage disease (ISSD) in a black infant presenting in utero with nonimmune hydrops, ascites, and anemia requiring intrauterine transfusion. Upon birth, the patient had prominent edema, large anterior fontanelle, partial absence of the rectus abdominis, clubbing of the left foot, gingival hypertrophy, short first metatarsals, prominent scrotal raphe, right heart dilatation, and left ventricular dysfunction. Radiographs showed pulmonary hypoplasia and epiphyseal stippling. He died of respiratory failure at day 2. Autopsy demonstrated capillary hemangiom

Fetal ascites and oligohydramnios: prenatal diagnosis of a sialic acid storage disease (index case).

In a 20-year-old primiparous patient, a routine ultrasound scan performed at 28 weeks revealed fetal ascites, bilateral talipes, and oligohydramnios. This woman, married to possibly her first cousin, was at risk for an autosomal recessive disease, a metabolic disorder. At 29 weeks, an amniotic fluid biochemical study revealed the presence of an abnormal band of free sialic acid, leading to a diagnosis of a congenital form of sialic acid storage disease. Termination of pregnancy was performed at 30 weeks. Measurement of free sialic acid in cultured fetal skin fibroblasts confirmed the diagnosis

Electrokinetic behaviour and surface sialic acid status of blood platelets in essential thrombocythaemia (ET).

We have previously reported, for platelets from patients with idiopathic thrombocytopenic purpura, that there was in increased electrophoretic mobility that was related to their increased sialic acid content. Heterogeneity in the circulating platelet pool of patients with essential thrombocythaemia (platelet counts 600-1000 x 10(9)/l) has also been investigated by preparative continuous flow electrophoresis (CFE). The surface charge-dependent separation profiles of all the patients' platelets showed marked anodal shifts compared with the profiles of platelets from age- and sex-matched control

Moving boundary electrophoresis and sialic acid content of normal and polycythaemic red blood cells.

Moving boundary electrophoresis has been used for the assessment of the negative charge on erythrocyte membranes. The equipment is easy to construct and simpler to use than the microelectrophoresis technique. There is a direct relationship between the electrophoretic velocity measured by the moving boundary method and the sialic acid content of the red cell membrane. Polycythaemic erythrocytes have a higher electrophoretic mobility and higher sialic acid content than normals. Separation of the erythrocytes according to their densities shows that old polycythaemic erythrocytes have a higher ele

Sialic acids cleavage induced by elastin-derived peptides impairs the interaction between insulin and its receptor in adipocytes 3T3-L1.

The insulin receptor (IR) plays an important role in insulin signal transduction, the defect of which is believed to be the root cause of ty … tivity of the insulin receptor. We demonstrate by molecular dynamics approaches that the absence of sialic acids on one of these two sites is sufficient to generate local and general modifications of the structure of the IR. Biochemical approaches highlight a decrease in the interaction between insulin and its receptor when ERC sialidase activity is induced by …

The specificity of viral and bacterial sialidases for alpha(2-3)- and alpha(2-6)-linked sialic acids in glycoproteins.

The anomeric specificity of six sialidases (Vibrio cholerae, Arthrobacter ureafaciens, Clostridium perfringens, Newcastle disease virus, fowl plague virus and influenza A2 virus sialidases) was assessed with sialylated antifreeze glycoprotein, ovine submandibular gland glycoprotein and alpha 1-acid glycoprotein, resialylated specifically in alpha(2-3) or alpha(2-6) linkage with N-acetylneuraminic acid or N-glycolylneuraminic acid using highly purified sialyltransferases. The rate of release of sialic acid from these substrates was found to correlate well with the specificity observed earlier w

The infectious salmon anemia virus esterase prunes erythrocyte surfaces in infected Atlantic salmon and exposes terminal sialic acids to lectin recognition.

Many sialic acid-binding viruses express a receptor-destroying enzyme (RDE) that removes the virus-targeted receptor and limits viral interactions with the host cell surface. Despite a growing appreciation of how the viral RDE promotes viral fitness, little is known about its direct effects on the host. Infectious salmon anemia virus (ISAV) attaches to 4-O-acetylated sialic acids on Atlantic salmon epithelial, endothelial, and red blood cell surfaces. ISAV receptor binding and destruction are effectuated by the same molecule, the haemagglutinin esterase (HE). We recently discovered a global lo…

Studies of naturally occurring modifications of sialic acids by fast-atom bombardment-mass spectrometry. Analysis of positional isomers by periodate cleavage.

A variety of modifications of sialic acids have been described in nature. There are currently many difficulties in the detection and quantitation of these modified sialic acids from biological sources. We report here that fast-atom bombardment-mass-spectrometry (FAB-MS) of native sialic acids provides specific detection and quantitation of many previously known compounds. Derivatization of the sialic acids by reduction and peracylation under acidic conditions prior to FAB-MS provides further confirmation of their identity and improves the sensitivity of detection. Samples containing as little …

Purification and properties of a Ca2+-independent sialic acid-binding lectin from human placenta with preferential affinity to O-acetylsialic acids.

A Ca2+-independent sialic acid-specific lectin from two developmental stages of human placenta was similarly purified to apparent homogeneity by DEAE-cellulose chromatography, affinity chromatography on bovine submaxillary mucin, and gel filtration. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis and gel filtration disclosed a molecular mass of 53 kDa. The specificity of the lectin for O-acetylsialic acids was substantiated by the dependence of hemagglutination on the presence of acetylated sialic acids on the surface of mammalian erythrocytes of various sources, by hapten inhibition…

Perspectives of Natural Products and Sialic Acid Mimetics as Modulators of Human Sialidases NEU1-3 in Atherosclerosis.

This narrative review summarizes current data on the role of desialylation of low-density lipoproteins (LDL) as a key initiating mechanism of atherogenesis that precedes oxidative modification. The structural and functional significance of sialic acid in apolipoprotein B-100 is considered, and the enzymes of the sialidase (neuraminidase) family responsible for its cleavage are characterized. The pathogenic role of increased sialidase activity in atherosclerosis, type 2 diabetes mellitus, and other diseases is discussed. The advantages of using dietary supplements based on natural sialidase inh

Sialic acid-binding immunoglobulin-like lectin-6 (SIGLEC6) is increased by hypoxia and inflammation in trophoblasts.

OBJECTIVES: SIGLEC6, a human-specific transmembrane receptor, is highly expressed in placenta. SIGLEC6 is elevated in maternal circulation preceding preeclampsia and in women with preeclampsia, correlating with disease severity. This study aimed to investigate the regulatory mechanisms of placental SIGLEC6 and its involvement in processes associated with preeclampsia pathogenesis. STUDY DESIGN: To determine cell source, SIGLEC6 was measured in (cyto)trophoblasts, syncytiotrophoblasts, and extravillous trophoblasts. We then assessed the impact of hypoxia (1% vs 8% Oxygen), inflammatory cytokin

Sialic Acid in Neurodegenerative and Psychiatric Disorders: From Molecular Regulation to Targeted Nanocarrier-Based Therapy.

In recent years, the exploration of molecular and cellular mechanisms underlying central nervous system (CNS) disorders has expanded beyond classical neurotransmitter- and receptor-based approaches toward a more integrated view including immune, metabolic, and glycosylation processes. Among these, sialic acid and its derivatives have emerged as critical regulators of neuronal communication, immune modulation, and synaptic plasticity. Their involvement ranges from maintaining neurochemical homeostasis under physiological conditions to contributing to the onset and progression of neurodegenerati

A sialic acid-binding protein in Toxoplasma gondii contains a conserved globular domain in apicomplexan parasites.

BACKGROUND: Apicomplexan protozoans employ an intricate invasion mechanism involving dynamic interactions with host cells, characterized by sequential secretion of adhesins and lectins. Our laboratory previously identified TgSABP1, a novel Toxoplasma gondii adhesin, demonstrating specific binding affinity for sialic acid (SA) receptors on host cell surfaces. However, the structural determinants governing SA recognition by this adhesin remain undefined. METHODS: Three-dimensional structural predictions of TgSABP1 and homologous proteins were generated using AlphaFold2. Bio-layer interferometry

Targeted Delivery of Arctigenin Using Sialic Acid Conjugate-Modified Liposomes for the Treatment of Breast Cancer.

Arctigenin (ATG) is a broad-spectrum antitumor drug with an excellent inhibitory effect on malignant tumors such as breast cancer, glioblastoma, liver cancer, and colon cancer. However, the clinical application of ATG is limited by its poor water solubility and quick hydrolysis in the liver, intestine, and plasma, which might hinder its application. Sialic acid (SA) recognizes selectin receptors overexpressed on the surface of tumor-associated macrophages. In this study, SA was conjugated with octadecylamine (ODA) to prepare SA-ODA, which was employed to prepare SA functionalized nanoliposomes
