TMEM16A Maintains Acrosomal Integrity Through ERK1/2, RhoA, and Actin Cytoskeleton During Capacitation.

Mammalian spermatozoa undergo a series of physiological and biochemical changes in the oviduct that lead them to acquire the ability to fertilize eggs. During their transit in the oviduct, spermatozoa face a series of environmental changes that can affect sperm viability. A series of ion channels and transporters, as well as the sperm cytoskeleton, allow spermatozoa to remain viable and functional. Cl- channels such as TMEM16A (calcium-activated chloride channel), CFTR (cystic fibrosis transmembrane conductance regulator), and ClC3 (chloride voltage-gated channel 3) are some of the ion transpo

Exosc10 deficiency in the initial segment is dispensable for sperm maturation and male fertility in mice.

EXOSC10 is an exosome-associated ribonuclease that degrades and processes a wide range of transcripts in the nucleus. The initial segment (IS) of the epididymis is crucial for sperm transport and maturation in mice by affecting the absorption and secretion that is required for male fertility. However, the role of EXOSC10 ribonuclease-mediated RNA metabolism within the IS in the regulation of gene expression and sperm maturation remains unknown. Herein, we established an Exosc10 conditional knockout (Exosc10 cKO) mouse model by crossing Exosc10 F/F mice with Lcn9-Cre mice which expressed recomb…

Industrialization possibilities of purified pig sperm hyaluronidase.

The goals of the present study were to develop a simple method for obtain highly purified pig sperm hyaluronidase (pHyase) and to assess its activity, function, and safety. In mammals, sperm-specific glycophosphatidylinositol (GPI)-anchored Hyase assists sperm penetration through the cumulus mass surrounding the egg and aids in the dispersal of the cumulus-oocyte complex. Recently, Purified bovine sperm hyaluronidase (bHyase) has been shown to enhance therapeutic drug transport by breaking down the hyaluronan barrier to the lymphatic and capillary vessels, thereby facilitating tissue absorptio

Long-term segmentation-free assessment of head-flagellum movement and intracellular calcium in swimming human sperm.

Human spermatozoa are the archetype of long-term self-organizing transport in nature and are critical for reproductive success. They utilize coordinated head and flagellar movements to swim long distances within the female reproductive tract in order to find and fertilize the egg. However, to date, long-term analysis of the sperm head-flagellar movements, or indeed those of other flagellated microorganisms, remains elusive due to limitations in microscopy and flagellar-tracking techniques. Here, we present a novel methodology based on local orientation and isotropy of bio-images to obtain long

Effect of zinc on boar sperm liquid storage.

Storage and transport of liquid boar sperm for artificial insemination (AI) requires the addition of solutions called extenders, which increase the volume of the ejaculate and help preserve its functional characteristics. Yet, the quality of sperm decreases over time primarily due to the increased production of reactive oxygen species (ROS) that damage the plasma membrane. Many commercial extenders are supplemented with additives that mitigate this effect. In semen, zinc is supplied at high concentration on the seminal plasma and helps protect the plasma membrane of sperm. However, zinc in the

Treatment with cholesterol just after thawing maintains the fertility of bull sperm.

Freezing and thawing diminish sperm motility and fertility by disrupting the cholesterol balance in sperm plasma and organelle membranes. The aim of this study was to elucidate the mechanisms through which exogeneous cholesterol treatment enhances the quality of frozen-thawed bull sperm. The incorporation of cholesterol was investigated using boron-dipyrromethene (BODIPY)-cholesterol, and BODIPY signals were detected not only in the plasma membrane but also in the midpiece region immediately after thawing. The positive signal of cholesterol in the midpiece region was inhibited by a scavenger r

Complementary role of CNNM2 in sperm motility and Ca(2+) influx during capacitation.

Ca(2+) plays a central role in the regulation of sperm motility. We recently reported an unexpected role of CNNM4, a Mg(2+) transporter, in this process by demonstrating perturbed Ca(2+) influx and gradual loss of motility of Cnnm4-deficient sperm. However, Cnnm4-deficient male mice were not entirely infertile, and a significant Ca(2+) response was still observed in their sperm. In the present study, we generated Cnnm4-deficient mice harboring a non-functional Cnnm2 allele (Cnnm2(Δ)), to examine whether CNNM2 compensates for the lost function of CNNM4 in sperm. Cnnm2(+/Δ); Cnnm4(Δ/Δ) mice were

Penis size and sperm quality, are all bats grey in the dark?

Penises play a key role in sperm transport and in stimulating female genitals. This should impact post-copulatory competition, and expose penis characteristics to sexual selective pressures. Studies of male genitalia have repeatedly reported negative static allometries, which mean that, within species, large males have disproportionally small genitals when compared with smaller individuals. Males of some sperm-storing bat species may stand as an exception to such a pattern by arousing from hibernation to copulate with torpid females. The selection for large penises might take place, if a long …

Chloride Intracellular Channel 4 (CLIC4) controls volume regulation in sperm development via Protein Kinase C (PKC).

BACKGROUND: Chloride ions (Cl) regulate sperm physiology, influencing spermatogenesis, volume regulation, capacitation, and fertilization processes. They contribute to the maintenance of membrane potential and intracellular pH, both of which are critical for sperm motility and capacitation. Any deviation in Cl homeostasis causes impaired sperm function and male infertility. Although several Cl channels and transporters have been implicated in the Cl homeostasis and osmoregulation of sperm cells, the precise mechanisms and molecular components governing volume regulation during sperm developmen

Circadian clock and output genes are rhythmically expressed in extratesticular ducts and accessory organs of mice.

Circadian clocks regulate multiple rhythms in mammalian tissues. In most organs core clock gene expression is oscillatory, with negative components Per and Cry peaking in antiphase to Bmal1. A notable exception is the testis, where clock genes seem nonrhythmic. Earlier mammalian studies, however, did not examine clock expression patterns in accessory ductal tissue required for sperm maturation and transport. Previous studies in insects demonstrated control of sperm maturation in vas deferens by a local circadian system. Sperm ducts express clock genes and display circadian pH changes controlle

Oocyte-sperm interactions.

The penetration of the zona pellucida is a crucial step during fertilization. Spermatozoa that are unable to recognize and bind to the zona pellucida glycoproteins or respond to the zona pellucida by undergoing the acrosome reaction fail to fertilize the egg. In most mammalian species, after entering the fallopian tube sperm are stored in the isthmic part of the oviduct under conditions that maintain sperm viability and synchronize both sperm transport and the process of acquisition of fertilizing ability, called capacitation. Only capacitated sperm are enabled to recognize the oocyte and resp…

Epididymal protein markers and fertility.

The last stages of male gamete differentiation occur outside the gonad in a specific environment controlled by the epididymal epithelium. All the fundamental characteristics of a fertile spermatozoon are acquired sequentially during transit through the epididymal tubule. Full understanding of the mechanisms involved in these gamete modifications is a key to understanding and controlling such important stages in male fertility. With the development of new large scale technologies, large amounts of information give hope of identifying the fundamental elements involved in such cellular events and

Effect of seminal plasma fractions on stallion sperm survival after cooled storage.

This study aimed to evaluate stallion sperm survival after 24 h of cooled storage in the presence of seminal plasma (SP) derived from the sperm-rich fractions (SRF) or sperm-poor fractions(SPF) of the ejaculate, without SP, or in the presence of SP from other stallions. Ejaculates were collected from four stallions using an automated phantom, which separated the semen into five cups. Centrifuged and washed spermatozoa from cup 2 (SRF) were mixed with skim milk extender to a concentration of 100 x 10(6) sperm/ml and then 1:1 (v/v) with SP from the stallion's own or another stallions' second (SP

A novel homozygous mutation in WDR19 induces disorganization of microtubules in sperm flagella and nonsyndromic asthenoteratospermia.

BACKGROUND: Asthenoteratospermia with multiple morphological abnormalities in the sperm flagella (MMAF) is a significant cause of male infertility. WDR19 is a core component in the IFT-A complex and has a critical role in intraflagellar transport. However, the role of WDR19 mutations in male infertility has yet to be examined. METHODS AND RESULTS: We performed whole exome sequencing (WES) for 65 asthenoteratospermia individuals and identified a proband who carried a homozygous WDR19 (c.A3811G, p.K1271E) mutation from a consanguineous family. Systematic examinations, including CT scanning and

Delayed translation and posttranslational processing of cyritestin, an integral transmembrane protein of the mouse acrosome.

This paper presents data on the cellular localization of the testis-expressed mouse Cyrn gene product, cyritestin. This cysteine-rich protei … has an apparent molecular weight of 110,000 Daltons, but is subject to processing during epididymal sperm transport, resulting in a shorter molecule lacking approximately 55 kDa from the N-terminal half. We conclude that cyritestin becomes exposed on the sperm surface after successful acrosome reaction and thus may play a role in sperm function rather than in tes …

Diallyl Trisulfide, an Active Substance from Garlic, Inhibits Female Oviposition by Decreasing the Expression of the OCT Gene, which is Highly Expressed in the Spermathecal Gland of Sitotroga cerealella (Oliver).

Diallyl trisulfide (DAT) effectively inhibits the fecundity of Sitotroga cerealella. Organic cation transporter (OCT) was most highly expressed in the spermathecal gland of female moths and was significantly decreased after DAT fumigation. However, the function of OCT in insect reproduction has been rarely reported. In this study, after silencing OCT in female adults, the mating rate, oviposition, and number of sperm transferred to females were decreased significantly, and transfer time of sperm in the female was delayed. Meanwhile, the long and short waves formed during sperm movement became

Effects of different extenders, storage temperatures, and antioxidant supplementation on chilled semen quality: a review.

The successful preservation of ram semen is essential to promote genetic variability, ensure semen transportation, and inseminate multiple ewes. Currently, either animal or plant-based lipoprotein-based extenders are used for semen preservation. Animal product-based extenders include milk and egg yolk, while soybean lecithin is a plant-based extender. Although extenders containing products of animal origin better preserve the quality of chilled semen, the in vivo efficacy after 24 h of storage is still of great concern. Storage temperature is another important and effective factor in preservin

Perspectives in porcine semen conservation (liquid and frozen).

In pigs, breeding primarily relies on Artificial Insemination (AI) with preserved semen. Maintaining sperm under proper conditions is thus vital for optimal reproductive performance. Currently, pig sperm can be preserved either in a liquid state at 15-20°C or 4-5°C, or cryopreserved, with most AI being conducted with liquid-stored semen at 15-20°C. This article reviews the state of the Assisted Reproductive Technology (ART) in preserving pig semen, focusing on the most recent advances. It describes the differences between conventional (15-20°C) and hypothermic (4-5°C) storage, and discusses th

Disruption of testis-enriched cytochrome c oxidase subunit COX6B2 but not COX8C leads to subfertility.

Mammalian sperm flagellum contains the midpiece characterized by a mitochondrial sheath that packs tightly around the axoneme and outer dense fibers. Mitochondria are known as the "powerhouse" of the cell, and produce ATP through the tricarboxylic acid (TCA) cycle and oxidative phosphorylation (OXPHOS). However, the contribution of the TCA cycle and OXPHOS to sperm motility and male fertility is less clear. Cytochrome c oxidase (COX) is an oligomeric complex localized within the mitochondrial inner membrane, and the terminal enzyme of the mitochondrial electron transport chain in eukaryotes. B

Introduction to the pathways involved in the activation and regulation of sperm motility: A review of the relevance of ion channels.

To participate in sperm-oocyte fusion, spermatozoa need to be motile. In the testes, spermatozoa are immotile, although these gametes acquire the capacity for motility during the transit through the epididymis. During the period of epididymal transport from the male genital tract to the female genital tract, spermatozoa exhibit various types of motility that are regulated by complex signalling and communication mechanisms. Because motility is very dynamic, it can be affected by small changes in the external or internal environment of spermatozoa within a very short time. This indicates that re
