Argonaute proteins regulate the timing of the spermatogenic transcriptional program.

Argonaute proteins are best known for their role in microRNA-mediated post-transcriptional gene silencing. Here, we show that AGO3 and AGO4, but not AGO2, localize to the sex chromatin of pachytene spermatocytes where they are required for transcriptional silencing of XY-linked genes, known as Meiotic Sex Chromosome Inactivation (MSCI). Using an Ago413 -/- mouse, we show that AGO3 and AGO4 are key regulators of spermatogenesis, orchestrating expression of meiosis-related genes during prophase I while maintaining silencing of spermiogenesis genes. Premature overexpression of spermiogenesis gene

Therapeutic Repair of Sperm Quality Decline Caused by Polytetrafluoroethylene.

The alarming prevalence of environmental microplastics has raised global concerns about fertility. However, the detriment of polytetrafluoroethylene (PTFE, Teflon), a widely used microplastic in non-stick cookware, to sperm quality remains unclear. Here, a high detection rate (46.62%) and bioaccumulation of PTFE in the male urogenital system are reported and the mechanisms of PTFE exposure on male fertility are investigated in both humans and mice and potential therapeutic strategies are explored. These findings reveal that PTFE exposure delays the development of spermatogonia and spermatocyte

Meiotic transcriptional reprogramming mediated by cell-cell communications in humans and mice revealed by scATAC-seq and scRNA-seq.

Meiosis is a highly complex process significantly influenced by transcriptional regulation. However, studies on the mechanisms that govern transcriptomic changes during meiosis, especially in prophase I, are limited. Here, we performed single-cell ATAC-seq of human testis tissues and observed reprogramming during the transition from zygotene to pachytene spermatocytes. This event, conserved in mice, involved the deactivation of genes associated with meiosis after reprogramming and the activation of those related to spermatogenesis before their functional onset. Furthermore, we identified 282 t

PRSS50-mediated inhibition of MKP3/ERK signaling is crucial for meiotic progression and sperm quality.

Serine protease 50 (PRSS50/TSP50) is highly expressed in spermatocytes. Our study investigated its role in testicular development and spermatogenesis. Initially, PRSS50 knockdown was observed to impair DNA synthesis in spermatocytes. To further explore this, we generated PRSS50 knockout ( Prss50 -/- ) mice ( Mus musculus), which exhibited abnormal spermatid nuclear compression and reduced male fertility. Furthermore, dysplastic seminiferous tubules and decreased sex hormones were observed in 4-week-old Prss50 -/- mice, accompanied by meiotic progression defects and increased apoptosis of sperm

Abnormal spermatogenesis and male infertility in testicular zinc finger protein Zfp318-knockout mice.

Zfp318, a mouse gene with a Cys2/His2 zinc finger motif, is mainly expressed in germ cells in the testis. It encodes two alternative transcripts, which regulate androgen receptor-mediated transcriptional activation or repression by overexpression of them. However, the role of Zfp318 is still obscure in vivo, especially in spermatogenesis. To elucidate the role of Zfp318 during gamete production, we established a knockout mouse line. Zfp318-null male mice exhibited infertility, whereas Zfp318-null female mice displayed normal fertility. ZFP318 was expressed during multiple stages of spermatogen

Germ cell removal after induction of cryptorchidism in adult rats.

Cryptorchidism is associated with male infertility due to germ cell loss in response to elevated temperature. However, there is a great deal of contradictory information prevalent on the status of germ cells and their process of removal in the cryptorchid testis. In the present study, we investigate the cell removal from cryptorchid rat testis by the methods of morphology and stereology. The testis weight is reduced according to previous reports after surgical induction of cryptorchidism. Interestingly, the epididymal weight is significantly increased in 7 days after surgery, and the caput epi

Inducible 70 kDa heat shock protein does not protect spermatogenic cells from damage induced by cryptorchidism.

Accumulation of inducible heat shock proteins (e.g. Hsp70i) during cellular stress confers thermotolerance, reduces the consequences of damage and facilitates cellular recovery, while abrogation of Hsp70i expression renders sensitivity to apoptosis. Testis translocation into abdominal cavity, which results in temperature elevation, does not induce expression of the Hsp70i proteins. Despite constitutive expression of testis-specific Hsp70 proteins, spermatocytes are very sensitive to damage at elevated temperatures. To test whether Hsp70i protein could protect testes from heat-induced damage, w

Heat shock transcription factor 1 is involved in quality-control mechanisms in male germ cells.

Quality-control mechanisms in spermatogenesis are important to eliminate injured or abnormal cells, thereby protecting the organism from abnormal development in the next generation. The processes of spermatogenesis are highly sensitive to high temperatures; however, the mechanisms by which injured germ cells are eliminated remain unclear. Here, we found that heat shock proteins are not induced in male germ cells in response to thermal stress, although heat shock transcription factor 1 (HSF1) is activated. Using HSF1-null mice, we showed that apoptosis of pachytene spermatocytes was markedly in

Different nuclear/cytoplasmic distributions of RET finger protein in different cell types.

The RET finger protein (RFP), which belongs to the B box zinc finger protein family, has a tripartite motif consisting of a Ring finger, a B box finger and a coiled-coil domain. The RET finger protein becomes oncogenic when its tripartite motif is fused with the tyrosine kinase domain of the RET protein. This study examined the RFP expression in normal and tumor tissues by immunohistochemistry. RFP was detected in the nuclei of various cells, including peripheral and central neurones, hepatocytes, adrenal chromaffin cells and male germ cells. Among them, RFP was expressed at high levels in mal

Expression of heat shock proteins in developing and degenerating rat testes.

In the testis, several types of heat shock proteins (HSPs) have been identified and characterized, although the cellular basis of the HSPs remains elusive. In the present study, alterations in the cellular localization of HSPs, including HSP 25, 60, 70, and 90, were studied during the developing and degenerating periods in the rat testis using immunohistochemistry and Western blotting. HSP25 was expressed in neither germ cells nor somatic cells on all days examined. In contrast, HSP 60 was expressed in Leydig cells during neonatal and prepuberty periods, and only in spermatogonia and primary s

Relationship between germinal cells function and dynamics of FSH secretion.

Tubular histology was correlated to FSH levels assayed under basal conditions (8 cases) or in response to Gn-RH (24 cases) in 32 infertile males, in whom a testicular biopsy was indicated as part of the work-up. A good correlation was found between number of spermatogonia and primary spermatocytes and levels of FSH, under basal conditions or, even more, in response to Gn-RH; correlation was less close with respect to spermatids 1 and 2 and nonexistent with respect to Serotoli cells. In spite of those findings, when taking into account only mean counts of spermatogonia and primary spermatocytes

Increased Total mtDNA Copy Number Cures Male Infertility Despite Unaltered mtDNA Mutation Load.

Mutations of mtDNA cause mitochondrial diseases and are implicated in age-associated diseases and aging. Pathogenic mtDNA mutations are often present in a fraction of all mtDNA copies, and it has been widely debated whether the proportion of mutant genomes or the absolute number of wild-type molecules determines if oxidative phosphorylation (OXPHOS) will be impaired. Here, we have studied the male infertility phenotype of mtDNA mutator mice and demonstrate that decreasing mtDNA copy number worsens mitochondrial aberrations of spermatocytes and spermatids in testes, whereas an increase in mtDNA

Ectopic expression of Sonic Hedgehog in a cryptorchid man with azoospermia: a case report.

A 30-year-old man presented with a left undescended testis, right testicular deficiency and azoospermia. Testicular biopsy revealed an absence of spermatocytes and increased numbers of Leydig cells in the undescended testis. Additional comparative analyses were undertaken to explore Sonic Hedgehog (Shh) immunostaining in the testis of juvenile and adult mice, in the testis of the patient with cryptorchidism, and in archival testicular tissue from a patient with obstructive azoospermia and a patient with prostate cancer. Shh immunostaining was demonstrated in spermatocytes in juvenile and adult

Metabolic adaptation mechanisms of glycogen reduction and lipid accumulation in testicular protection in Daurian ground squirrels during hibernation.

The role of glycogen and lipid metabolism in the testes of Daurian ground squirrels (Spermophilus dauricus) during different stages of the hibernation cycle and their influence on reproductive function remain poorly understood. This study examined testicular morphology across hibernation stages and investigated potential molecular mechanisms. Results showed that: (1) Spermatocyte density was reduced in the torpor group compared to the pre-hibernation (PRE) group, suggesting a suppression of spermatogenesis during torpor. In the post-hibernation (POST) group, reduced spermatocyte density was sp

The complex role of transcription factor GAGA in germline death during Drosophila spermatogenesis: transcriptomic and bioinformatic analyses.

The GAGA protein (also known as GAF) is a transcription factor encoded by the Trl gene in D. melanogaster. GAGA is involved in the regulation of transcription of many genes at all stages of fly development and life. Recently, we investigated the participation of GAGA in spermatogenesis and discovered that Trl mutants experience massive degradation of germline cells in the testes. Trl underexpression induces autophagic death of spermatocytes, thereby leading to reduced testis size. Here, we aimed to determine the role of the transcription factor GAGA in the regulation of ectopic germline cell d

Primary malignant melanoma of the bladder.

AIM: The aim of the present study was to elucidate the role of high temperature requirement A2 (HtrA2) in germ cell loss in the heat-stressed testis. METHODS: We examined the expression of HtrA2, caspase-9 activity and proteolytic activity of HtrA2 in the rat testis, and their in vivo responses to experimental cryptorchid treatment. RESULTS: Northern analysis revealed the expression of HtrA2 mRNA peaked at days 1 and 7 after cryptorchid treatment. While expression of HtrA2 mRNA was seen in the spermatogonium, spermatocytes and some spermatids in normal adult rat testis, experimental cryptorc

O-GlcNAc transferase governs spermatogenic mitotic-to-meiotic transition and progression by coordinating transcription and alternative splicing programs.

BACKGROUND: O-GlcNAcylation is a post-translational modification (PTM) uniquely catalyzed by O-GlcNAc transferase (OGT), which has been linked to tumorigenesis and neurodegeneration. However, its roles in mammalian spermatogenesis remain unexplored. This study aims to elucidate the functional mechanisms of OGT in spermatogenesis and male fertility. METHODS: We employed immunoprecipitation-mass spectrometry (IP-MS) to identify candidate O-GlcNAcylated substrates of OGT in juvenile mouse testes. To explore the physiological roles of OGT and O-GlcNAcylation, we constructed a mouse model with pos

Dual modes of contractility tailor cytokinesis to the requirements of diverse tissues.

The actomyosin network generates contractile forces underlying cell shape changes. While actomyosin equatorial contractility is essential for cytokinesis, less is known about the importance of contractility at the poles. This question is pertinent in multicellular organisms where cytokinesis must adapt to the constraints imposed by diverse cell types. Here, we manipulated contractility at the equator and poles during cytokinesis genetically in Drosophila. We examined the consequences of these alterations on cytokinesis in two different cell types: non-polarized and non-adherent large spermatoc

Gametogenesis and fertilization in Rhabdias ranae Walton 1929: I. The parasitic hermaphrodite.

The somatic, diploid, and haploid chromosome numbers were determined in the hermaphrodite (24, 12, eggs 6, spermatozoa 5 or 6). Oocytes in the ovary synapsis zone contain 6 tetrads. The tetrads seem to disappear in the growth zone and reappear in the oocyte just prior to fertilization. After sperm penetration, the tetrads move to the periphery and form 2 polocytes by rapid meiotic divisions. Spermatogenesis occurs in the testis zone. Primary spermatocytes show 5 large tetrads and 2 small heterochromosomal dyads which lag on the spindle during division. Each resultant secondary spermatocyte has
