This report describes the first isolation of Sporothrix globosa from a Brazilian patient. A 77-year-old woman was examined for sporotrichosis infection. Histopathological examination of skin biopsy revealed chronic granulomatous infiltrate with microabcess. Furthermore, S. schenckii-like yeasts were evident as demonstrated by PAS and Grocott stains. A case of cryptococcosis due to capsule-deficient Cryptococcus is presented, in which culturing of the clinical specimen and tests for capsular antigen in cerebrospinal fluid and serum were negative. The histopathological techniques evaluated were hematoxylin-eosin, Grocott methenamine silver, Mayers mucicarmine and Fontana-Masson. The diagnosis of cryptococcosis due to capsule-deficient Cryptococcus was confirmed by means of the Fontana-Masson technique and by direct immunofluorescence. The potential of the histochemical techniques is discussed.

Microbiological research of 59 patients with extrapulmonary localization tuberculosis was conducted for exposition of tuberculosis mycobacterium. It was determined that the method of microscopy of smear painted by Ziel-Nilsen with preliminary treatment of pathological material in the way of "enrichment" using sodium bicarbonate gives more effective result, than the method of direct microscopy of smear painted according to Ziel-Nilsen. It indicates the possibility to use this method for early diagnostics of extrapulmonary tuberculosis, for exposing mycobacteria at its urinary-sexual location.

Fine-needle biopsy is used in the thyroid pathology especially for the differential diagnosis concerning malignant lesions. From a total of 59 cases of thyroiditis, in 12 was possible to proceed to fine-needle biopsy, in large goiters and nodules larger than 1 cm in diameter; in these cases were obtained smears, stained in May-Grünwald-Giemsa technique. We report here results of modern staining techniques including anti-prion protein (PrP) immunocytochemistry to a set of archival brain specimens of a 16 year-old male who died from kuru in 1967. The multi-sections, which were stained by Fite method, of skin biopsies taken from twelve active BT cases were examined under the guidance of special stains for demonstration of nerve components. The method of differential staining of muscles and connective tissue in the genitourinary organs (urinary bladder, ureters, renal pelvis) and gastrointestinal tract is suggested. Biopsy material from 42 cases of florid hyperplastic cystic disease of the breast were reviewed and were found to have foci of apocrine metaplasia. This appearance was limited to the ductular epithelium of hyperplastic lobules. It is believed that this scattered change is an initial feature in relation to the genesis of apocrine-lined and fully developed cysts.
