Effect of CB-154 (2 Br-a-ergocryptine) on serum follicle stimulating hormone, luteinzing hormone and prolactin in women with the amenorrhoea-galactorrhoea syndrome.

Four women with the amenorrhoea-galactorrhoea syndrome were treated with 2-Br-a-ergocryptine (CB-154). In 3 patients, lactation was inhibited and ovulation was induced. Furthermore, pregnancy was achieved in one of them. In the remaining case, neither was lactation inhibited nor ovulation induced. Serum follicle stimulating hormone (FSH), luteinizing hormone (LH) and prolactin (PRL) were measured sequentially in the 4 CB- 154 treated women. They showed high serum PRL levels and acyclic patterns of serum FSH and LH. CB-154 suppressed PRL secretion and stimulated FSH and LH secretion in 3 cases

Analysis of changes in sodium and chloride ion transport in the skin.

The measurement of electric potential and resistance reflect the transport of sodium and chloride ions which take place in keratinocytes and is associated with skin response to stimuli arising from external and internal environment. The aim of the study was to assess changes in electrical resistance and the transport of chloride and sodium ions, under iso-osmotic conditions and following the use of inhibitors affecting these ions' transport, namely amiloride (A) and bumetanide (B). The experiment was performed on 104 fragments of rabbit skin, divided into three groups: control (n = 35), A-inhi

Influence of dynorphin on estradiol- and cervical stimulation-induced prolactin surges in ovariectomized rats.

Prolactin is an anterior pituitary hormone necessary for fertility, pregnancy maintenance, lactation, and aspects of maternal behavior. In rodents, there is a surge of prolactin on the afternoon of proestrus, and a semi-circadian pattern of prolactin surges during early pregnancy, with a diurnal and nocturnal surge every day. Both of these patterns can be replicated in ovariectomized rats. A prior study demonstrated that central antagonism of κ-opioid receptors, the target of dynorphin, largely abolished the nocturnal prolactin surge in pregnant rats. We build on this to determine whether dyno

The amygdala central nucleus is required for acute stress-induced bladder hyperalgesia in a rat visceral pain model.

Chronic stress has been implicated in the pathogenesis of chronic visceral pain conditions, such as interstitial cystitis (IC), and bouts of acute stress exacerbate clinical urological pain. Studies using animal models have shown that exposure to chronic footshock stress augments reflex responses to urinary bladder distension (UBD) in animal models, however acute effects in animal models are largely unknown, as are the central nervous system mechanisms of stress-related increases in nociception. The amygdala is a salient structure for integration of sensory and cognitive/emotional factors. The

The influence of the ventrolateral medulla on thermoregulatory circulations in the rat.

The rostral ventrolateral medulla (RVLM) was stimulated electrically and chemically (0.1-0.2 microliter, 0.4 M DL-homocysteic acid) in urethane-anaesthetised rats. Changes in the vasomotor tone of the cutaneous circulation of the tail, front and hind feet, the nose and the fur covered areas of the back, proximal hind leg and neck were assessed indirectly by measurement of skin temperatures. Electrical stimulation of the RVLM at 25 Hz (2-5 min) produced sustained decreases in skin temperatures and differed from the effect induced by stimulation at 100 Hz or chemical stimulation, in that the con

Spino-vagal nonadrenergic inhibition of gastric motility elicited by abdominal nociceptive stimulation in the cat.

Reflex inhibition of gastric motility in response to intestinal nociception and afferent nerve stimulation was studied in anesthetized cats. Mechanical stimulation of the small and large intestines elicited marked gastric inhibition, which was imitated by direct electrical stimulation of mesenteric or splanchnic afferents. The reflex response was resistant to atropine, guanethidine, and adrenalectomy. Spinal cord transection at the cervical level or spinal anesthesia completely blocked the reflex, as did vagotomy or vagal cold blockade. Chemical peritoneal stimulation by hydrochloric acid indu

A new concept for risk assessment of the hazards of non-genotoxic chemicals--electronmicroscopic studies of the cell surface. Evidence for the action of lipophilic chemicals on the Ca2+ signaling system.

Recently, we presented evidence for the localization of components of the cellular Ca2+ signaling pathway in microvilli. On stimulation of this pathway, microvilli undergo characteristic morphological changes which can be detected by scanning electron microscopy (SEM) of the cell surface. Here we show that both receptor-mediated (vasopressin) and unspecific stimulation of the Ca2+ signaling system by the lipophilic tumor promoters thapsigargin (TG) and phorbolmyristateacetate (PMA) are accompanied by the same type of morphological changes of the cell surface. Since stimulated cell proliferatio

Antigastritis effects of Armillariella tabescens (Scop.) Sing. and the identification of its anti-inflammatory metabolites.

OBJECTIVES: This study demonstrates the biological and chemical analysis of the mushroom Armillariella tabescens (Scop.) Sing. (Tricholomataceae). METHODS: Chemical structures of the isolates were determined by 1D and 2D NMR, and ESI-MS, as well as comparison with previously reported data. All isolates were tested for anti-inflammatory effects based on their ability to inhibit LPS-stimulated nitric oxide (NO) production in RAW264.7 cells. KEY FINDINGS: We found that the MeOH extract of the fruiting bodies of A. tabescens showed antigastritis activity against ethanol-induced gastric damage in

On participation of the autonomic nervous system in the mechanisms of chemical carcinogenesis.

With the view of studying the role of autonomic nervous system in chemical carcinogenesis mechanism in outbred white male rats, chronically treated with NDEA, there has been investigated the modifying effect of some neurotropic pharmacological drugs which cause either stimulation or inhibition of: 1. adrenergic processes (noradrenalin, isoproterenol, clonidine, pyrroxane and propranolol); 2. cholinergic processes (proserine and atropine); 3. the function of the central nervous system (CNS)--caffeine and ethanol. Pharmacological activation of alpha-adrenoreceptors or blocking of both beta-adren

Interindividual differences in chemosensory perception: Toward a better understanding of perceptual ratings during chemical exposures.

Perceptions that arise from stimulation of olfactory and trigeminal receptors in the nasal cavity guide the evaluation of chemical environment in humans. Strong interindividual differences in these assessments may be attributed to nonsensory factors such as gender, anxiety, and chemical sensitivity. Knowledge regarding the influence of these factors originates mainly from basic odor research using short-term exposure scenarios. In situations with continuous chemical exposures-common in the working environment-their impact is less clear. To investigate their role during the exposure to workplac

Central pain mechanisms following combined acid and capsaicin perfusion of the human oesophagus.

Visceral afferents originating from different gut-segments converge at the spinal level. We hypothesized that chemically-induced hyperalgesia in the oesophagus could provoke widespread visceral hypersensitivity and also influence descending modulatory pain pathways. Fifteen healthy volunteers were studied at baseline, 30, 60 and 90 min after randomized perfusion of the distal oesophagus with either saline or 180 ml 0.1M HCl+2mg capsaicin. Electro-stimulation of the oesophagus, 8 cm proximal to the perfusion site, rectosigmoid electrical stimulation and rectal mechanical and heat stimulations w

Receptive properties of afferent nerve fibres associated with the rat saphenous vein.

We tested afferent fibres isolated from the saphenous nerve for their responses to mechanical, osmotic and chemical stimuli applied to a vascularly isolated segment of the saphenous vein in the rat. Fifteen units were excited by local mechanical stimulation and many of them were also activated by distension of the venous segment, however exclusively by presumably noxious pressures. Most mechanosensitive fibres were excited by one of the three pain-producing substances, hyperosmotic saline (HS), potassium chloride (KCl) and bradykinin. Three fibres which were insensitive to mechanical stimulati

Innervation territories of mechano-insensitive C nociceptors in human skin.

Microneurographic recordings were obtained in the peroneal nerve from 20 mechano-insensitive units (CMi) and six mechano-heat responsive C units (CMH) in healthy human subjects. Their innervation territories in the skin of the leg or foot were assessed by transcutaneous electrical stimulation with a pointed probe at intensities of 10 to 100 mA (0.2 ms) and, when applicable, by mechanical von Frey hair stimulation. Electro-receptive fields (eRFs) of CMH units had a median area of 1.95 cm(2) when mapped with 10 mA that coincided approximately with mechano-receptive fields (mRFs) as mapped with a

Simultaneous voltammetric and chemical monitoring of dopamine release in situ.

The validity of voltammetric monitoring of stimulated dopamine (DA) release was tested by simultaneous chemical assay of the extracellular fluid region next to the tip of a selective Nafion-coated graphite voltammetric electrode. Micro alumina probes were positioned next to the recording electrode tip to adsorb released catecholamines. The catecholamines were desorbed into microvolumes of acid and analyzed by very sensitive HPLC assays to provide unequivocal chemical verification of the electrochemical signals. The results fully confirm that selective voltammetric electrodes can detect release
