Exclusion of bioactive contaminations in Streptococcus pyogenes erythrogenic toxin A preparations by recombinant expression in Escherichia coli.

The streptococcal erythrogenic exotoxin A (SPEA) belongs to the family of bacterial superantigens and has been implicated in the pathogenesis of a toxic shock-like syndrome and scarlet fever. Concerning its biological activity, mainly T-cell-stimulatory properties, conflicting data exist. In this study, we show that most of the SPEA preparations used so far contain biologically active contaminations. Natural SPEA from the culture supernatant of Streptococcus pyogenes NY-5 and recombinant SPEA purified from the culture filtrate of S. sanguis are strongly contaminated with DNases. We show that n

[Value of MRI with injection of gadolinium in the diagnosis of mitral ring abscess. Apropos of a case].

Transoesophageal echocardiography is the investigation of choice for the diagnosis of valvular vegetations in infectious endocarditis but the diagnosis of an associated ring abscess is sometimes difficult especially in patients with valvular prostheses or calcifications which are in important source of artefacts. The authors report a case of mitral annular abscess confirmed at surgery and comment on the diagnostic and prognostic value of cardiac MRI. MRI showed an abnormal cavity directly related to the mitral annulus and separated from the left ventricle by a low intensity signal which increa

Microbial pathogenicity in oral soft tissue diseases.

Recent work implicating microbial factors in the pathogenesis of several oral soft tissue diseases is discussed. These conditions include recurrent aphthous ulceration, oral cancer, Sjogren's syndrome, and the oral lesions of AIDS (the acquired immune deficiency syndrome). This paper reviews some recent work on these topics, with emphasis on investigations in our laboratory.

Effect of bacterial products on inflammatory reactions in the dental pulp.

Culture filtrates (extracellular components) and material obtained from disintegrated cells (intracellular components) of cultured plaque bacteria were studied for their capacity to induce inflammatory reactions in the dental pulp. Class V cavities were prepared on the buccal surface of 94 teeth: 42 test and 52 control teeth in six adult monkeys. lyophilized bacterial components were sealed into the test cavities either alone or following an 8-h topical application of a solution of the same components in phosphate-buffered saline (PBS). Culture medium and PBS were applied in two sets of contro

Brain abscess in a patient with psoriatic arthritis treated with adalimumab: A case report.

RATIONALE: In patients receiving biological therapies, serious infections are a major concern. Infections associated with anti-tumor necrosis factor antibody therapy include tuberculosis, viral, fungal, and bacterial infections. Likewise, severe infections of the upper and lower respiratory tract, lung, skin and soft tissue, urinary tract, gastrointestinal tract, joint, and bone have also been reported previously. However, infections involving the central nervous system are rare, especially an intracranial infection caused by odontogenic infection. To date, only few cases have been reported of

A tissue-dependent hypothesis of dental caries.

Current understanding of dental caries considers this disease a demineralization of the tooth tissues due to the acid produced by sugar-fermenting microorganisms. Thus, caries is considered a diet- and pH-dependent process. We present here the first metagenomic analysis of the bacterial communities present at different stages of caries development, with the aim of determining whether the bacterial composition and biochemical profile are specific to the tissue affected. The data show that microbial composition at the initial, enamel-affecting stage of caries is significantly different from that

Increased interleukin-6, interferon-gamma and tumour necrosis factor-alpha production by tonsillar mononuclear cells stimulated with alpha-streptococci in patients with pustulosis palmaris et plantaris.

In vitro cytokine production by tonsillar mononuclear cells under culture conditions with or without lyophilized streptococcal antigens was measured by enzyme-linked immunosorbent assay (ELISA). Under culture conditions without any stimulus, a certain amount of interferon (IFN)-gamma, tumour necrosis factor (TNF)-alpha, and interleukin (IL)-6, was detected in the supernatant culture fluids from all 29 donors tested and just detectable levels of IL-1alpha and IL-2 were identified in 5 of 12 donors tested; but IL-4 and IL-5 were not detected in any supernatants from 21 donors tested. In 16 patie

Anterior chest wall abscess--an unusual complication of an odontogenic infection.

INTRODUCTION: A majority of odontogenic infections can be successfully managed with routine incision and drainage and empirical antibiotics. Sometimes an odontogenic infection may spread to contiguous spaces and descend along the neck causing significant morbidity and rarely even mortality. CASE REPORT: We report a case of an odontogenic infection which had an unusual spread to the anterior chest wall.

The use of DNA probes to examine the distribution of subgingival species in subjects with different levels of periodontal destruction.

The present investigation examined the distribution of 14 subgingival species at a total of 2299 sites in 90 subjects with different levels of periodontal destruction. Subgingival plaque samples taken from the mesial aspect of each tooth were anaerobically dispersed, diluted and plated on non selective media. After anaerobic incubation, colonies were lifted to nylon filters and specific species detected using digoxygenin-labeled whole chromosomal DNA probes. The mean total viable count for all sites in all subjects was 8.3 x 10(6). The probes accounted for an average of 27.8% of the total viab

Effect of antimicrobial agents on root surface caries, alveolar bone loss, and microflora in rice rats.

Two antimicrobial agents, 9-aminoacridine (0.2%) and minocycline (0.2%), were evaluated for their efficacy in inhibiting root surface caries, bone loss, and microflora in rice rats. A solution of 5000 ppm fluoride was used as a positive control for the inhibition of root surface caries, and double-distilled water was used as a negative control group. Each rat was treated by having its molar teeth swabbed 2 X per day with the prescribed agent in its group for nine weeks. Root caries reduction in the minocycline and fluoride groups was not significantly different, but the reduction was significa

Haemophilus segnis polymicrobial and monomicrobial bacteraemia identified by 16S ribosomal RNA gene sequencing.

This paper reports a case of Haemophilus segnis polymicrobial bacteraemia and a case of H. segnis monomicrobial bacteraemia identified by 16S ribosomal RNA gene sequencing. In the first case, a gram-negative aerobic coccobacillus was isolated with Streptococcus intermedius and S. sanguis from the blood culture of a 32-year-old intravenous drug addict with left thoracic empyema. In the second case, a gram-negative aerobic coccobacillus was isolated from the blood culture of an 82-year-old woman with Clostridium difficile colitis and septicaemic shock. Both gram-negative coccobacilli grew on cho

Coaggregation of human oral Cytophaga species and Actinomyces israelii.

A total of 19 strains of oral Cytophaga sp. obtained from subgingival plaque deposits were tested for their ability to coaggregate with strains of Actinomyces israelii, A. viscosus, A. naeslundii, Streptococcus sanguis, S. mutans, S. salivarius, and S. mitis. Coaggregation was observed only with A. israelii. Based on their coaggregation patterns with eight A. israelii strains, the Cytophaga strains were distributed among three distinct groups: those that coaggregated with A. israelii PK16 but not with A. israelii W1011 (ATCC 29322), those that coaggregated with A. israelii ATCC 29322 but not w…

Streptococcus sanguinis meningitis following endoscopic ligation for oesophageal variceal haemorrhage.

We report a case of acute purulent meningitis caused by Streptococcus sanguinis after endoscopic ligation for oesophageal variceal haemorrhage in a cirrhotic patient without preceding symptoms of meningitis. Initial treatment with flomoxef failed. The patient was cured after 20 days of intravenous penicillin G. This uncommon infection due to S. sanguinis adds to the long list of infectious complications among patients with oesophageal variceal haemorrhage.
