ATP sulfurylase atypical leucine zipper interacts with Cys3 and calcineurin A in the regulation of sulfur amino acid biosynthesis in Cryptococcus neoformans.

Fungal pathogens are a major cause of death, especially among immunocompromised patients. Therapies against invasive fungal infections are restricted to a few antifungals; therefore, novel therapies are necessary. Nutritional signaling and regulation are important for pathogen establishment in the host. In Cryptococcus neoformans, the causal agent of fungal meningitis, amino acid uptake and biosynthesis are major aspects of nutritional adaptation. Disruptions in these pathways lead to virulence attenuation in an animal model of infection, especially for sulfur uptake and sulfur amino acid bios

Sequential detection of hypochlorous acid and sulfur dioxide derivatives by a red-emitting fluorescent probe and bioimaging applications in vitro and in vivo.

Hypochlorous acid (HOCl) and sulfur dioxide derivatives (SO3 2-/HSO3 -) play critical roles in complex signal transduction and oxidation pathways. Therefore, it is meaningful and valuable to detect both HOCl and SO2 derivatives in biosystems by a fluorescence imaging assay. In this work, we developed a red-emitting fluorescent probe (DP) by the condensation of malononitrile and phenothiazine derivatives through a C[double bond, length as m-dash]C double bond. DP was designed with a donor-π-acceptor (D-π-A) structure, which enables absorption and emission in the long wavelength region. In the p

Direct NMR evidence for the dissociation of sulfur-dioxide-bound acetaldehyde under acidic conditions: Impact on wines oxidative stability.

SO2 reaction with electrophilic species present in wine, including in particular carbonyl compounds, is responsible for the reduction of its protective effect during wine aging. In the present study, direct 1H NMR profiling was used to monitor the reactivity of SO2 with acetaldehyde under wine-like oxidation conditions. The dissociation of acetaldehyde bound SO2 was evidenced suggesting that released free SO2 can further act as an antioxidant. EPR and DPPH assays showed an increasing antioxidant capacity of wine with the increase in the concentration of acetaldehyde sulfonate. The presence of

Alleviation of alcoholic liver injury by betaine involves an enhancement of antioxidant defense via regulation of sulfur amino acid metabolism.

Previous studies suggested that the hepatoprotective activity of betaine is associated with its effects on sulfur amino acid metabolism. We examined the mechanism by which betaine prevents the progression of alcoholic liver injury and its therapeutic potential. Rats received a liquid ethanol diet for 6 wk. Ethanol consumption elevated serum triglyceride and TNFα levels, alanine aminotransferase and aspartate aminotransferase activities, and lipid accumulation in liver. The oxyradical scavenging capacity of liver was reduced, and expression of CD14, TNFα, COX-2, and iNOS mRNAs was induced marke

Ferredoxins from the photosynthetic purple non-sulfur bacterium Rhodopseudomonas palustris. Isolation and amino acid sequence of ferredoxin I.

Two ferredoxins, ferredoxins I and II, were prepared from Rhodopseudomonas palustris. They were separated on a Sephadex column after carboxymethylation and ferredoxin I, the major component, was subjected to an amino acid sequence study. The protein was composed of 63 amino acid residues and the sequence was as follows: (sequence; see text). The molecular weight was calculated to be 6,718, excluding iron and sulfur atoms. The distribution of the nine cysteine residues was similar to but clearly distinct from those of ferredoxins of other photosynthetic bacteria. Comparison of this ferredoxin w

Positively charged sulfur quantum dots: an outstanding fluorescent nanoprobe for sensitive and selective detection of 2,4,6-trinitrophenol (picric acid).

2,4,6-Trinitrophenol (TNP), a toxic and explosive nitroaromatic industrial pollutant, necessitates low-cost, heavy-metal-free fluorescent probes with superior water dispersibility for sensitive aqueous monitoring. Herein, positively charged amine-functionalized sulfur quantum dots (A-SQDs) are fabricated via a one-pot solvothermal strategy, serving as a novel fluorescent nanoprobe for sensitive and selective TNP detection. The resultant A-SQDs exhibit a uniform particle size of about 2.47 nm, persistent positive surface charge over a wide pH window of 3-10, and a high photoluminescence quantum

Iron-sulfur cofactors in nucleic acid metabolism and protein synthesis: Assembly, delivery, and putative roles in cellular and viral systems.

Composed of iron (Fe) and inorganic sulfur (S), iron-sulfur clusters (ISCs) are ancient cofactors present across all domains of life and in some viruses. Over the past 3 decades, cytosolic and nuclear Fe-S proteins have emerged as integral components of DNA replication and repair machineries, telomere maintenance pathways, transcriptional processes, cell cycle regulation, and protein synthesis. More recently, ISCs were identified in viral proteins, including multiple components of the SARS-CoV-2 replication and transcription complex (RTC), which collectively host seven experimentally verified

Cysteine, methionine, and pantothenic acid remodel the Saccharomyces cerevisiae transcriptome and volatile sulfur compound metabolome during alcoholic fermentation.

Yeast nitrogen and vitamin nutrition are fundamental levers for managing wine quality, yet the specific mechanisms linking nutrient availability to sulfur aroma formation remain poorly understood. This study explored the impact of methionine, cysteine, and pantothenic acid (vitamin B5)-nutrients with direct, pivotal roles in sulfur metabolic pathways-on the Saccharomyces cerevisiae transcriptome and VSC metabolome during fermentation. Our findings reveal that methionine and cysteine catabolic routes act as isolated compartments; the genes responsible for bridging these two pathways remain tran

α-Lipoic acid increases the viability of nephrocytes and elevates sulfane sulfur level in plasma of patients with chronic kidney disease.

BACKGROUND: Kidney diseases are a major global health problem affecting millions of people. Despite this, there is as yet no effective drug therapy improving outcome in patients with renal disease. The aim of this study was to examine the nephroprotective effect of α-lipoic acid (ALA) in vitro and to examine the effect of ALA administered in vivo on the production of reactive sulfur species (RSS), including hydrogen sulfide (H2S) and compounds containing sulfane sulfur. METHODS: The effect of ALA was studied in vitro by determining the viability of human embryonic kidney cells (HEK293) in nor

Boosting Catalytic and Anti-fluorination Performance of the Ru/Vanadia-Titania Catalyst for the Oxidative Destruction of Freon by Sulfuric Acid Modification.

Chlorofluorocarbons (CFCs) exert a strong greenhouse effect and constitute the largest contributor to ozone depletion. Catalytic removal is considered an effective pathway for eliminating low-concentration CFCs under mild conditions. The key issue is the easy deactivation of the catalysts due to their surface fluorination. We herein report a comparative investigation on catalytic dichlorodifluoromethane (CFC-12) removal in the absence or presence of water over the sulfuric-acid-modified three-dimensionally ordered macroporous vanadia-titania-supported Ru (S-Ru/3DOM VTO) catalysts. The S-Ru/3DO

Effects of dietary restriction on hepatic sulfur-containing amino acid metabolism and its significance in acetaminophen-induced liver injury.

Dietary restriction (DR) has been revealed to have health benefits as it induces reduction in oxidative stress. Glutathione (GSH), an important cellular antioxidant, is increased in rodent livers owing to DR; however, the exact mechanism and clinical relevance of DR are yet to be fully understood. In this study, male C57BL/6 mice were administered a 50% restricted diet for 7 d, and the hepatic sulfur-containing amino acid (SAA) metabolism was determined to assess the biosynthesis of GSH. The hepatic methionine level was found to decrease, while the homocysteine, cysteine, and GSH levels were i

Use of sulfuric acid-carbonization materials from grape pulp for the removal of hexavalent chromium (Cr(VI)): mechanism and characterization.

This study investigated the reduction of hexavalent chromium (Cr(VI)) with sulfur dioxide (SO2) and adsorption of Cr(VI) onto dried grape pulp carbonized with sulfuric acid. Cr(VI) reduction capacities of SO2 were determined. The filtrate was titrated with NaOH solution after shaking and filtering the carbonized material to retain unreacted sulfuric acid (H2SO4). Simple washing recovered 25-38% of the experimental acid at low concentrations. The carbonized material was washed twice with distilled water and then dried at 105 °C and weighed. The carbonized material had a yield of 56.6% (grape pu

Sulfur-Induced Resistance against Pseudomonas syringae pv. actinidiae via Triggering Salicylic Acid Signaling Pathway in Kiwifruit.

Sulfur has been previously reported to modulate plant growth and exhibit significant anti-microbial activities. However, the mechanism underlying its diverse effects on plant pathogens has not been elucidated completely. The present study conducted the two-year field experiment of sulfur application to control kiwifruit canker from 2017 to 2018. For the first time, our study uncovered activation of plant disease resistance by salicylic acid after sulfur application in kiwifruit. The results indicated that when the sulfur concentration was 1.5-2.0 kg m-3, the induced effect of kiwifruit canker

Responses of gaseous sulfuric acid and particulate sulfate to reduced SO2 concentration: A perspective from long-term measurements in Beijing.

SO2 concentration decreased rapidly in recent years in China due to the implementation of strict control policies by the government. Particulate sulfate (pSO42-) and gaseous H2SO4 (SA) are two major products of SO2 and they play important roles in the haze formation and new particle formation (NPF), respectively. We examined the change in pSO42- and SA concentrations in response to reduced SO2 concentration using long-term measurement data in Beijing. Simulations from the Community Multiscale Air Quality model with a 2-D Volatility Basis Set (CMAQ/2D-VBS) were used for comparison. From 2013 to

Catalytic conversions of atmospheric sulfur dioxide and formation of acid rain over mineral dusts: Molecular oxygen as the oxygen source.

Sulfur dioxide (SO2) ranks as a major air pollutant and is likely to generate acid rain. When molecular oxygen is the oxygen source, the regular surfaces of gibbsite (one of the most abundant mineral dusts) show no reactivity for SO2 conversions to H2SO4, while the partially dehydrated (100) surface with coordination-unsaturated Al sites becomes catalytically effective. Because of the easy availability of molecular oxygen, results manifest that acid rain can form under all atmospheric conditions and may account for the high conversion ratio of atmospheric SO2. The (100) and (001) surfaces show

Intrauterine growth restriction leads to changes in sulfur amino acid metabolism, but not global DNA methylation, in Yucatan miniature piglets.

Intrauterine growth restriction (IUGR), in both animals and humans, has been linked to metabolic syndrome later in life. There has been recent evidence that perturbations in sulfur amino acid metabolism may be involved in this early programming phenomenon. Methionine is the precursor for cellular methylation reactions and for the synthesis of cysteine. It has been suggested that the mechanism behind the "fetal origins" of adult diseases may be epigenetic, involving DNA methylation. Because we have recently demonstrated the fetal origins phenomenon in Yucatan miniature swine, we hypothesized th

Current therapeutics, their problems, and sulfur-containing-amino-acid metabolism as a novel target against infections by "amitochondriate" protozoan parasites.

The "amitochondriate" protozoan parasites of humans Entamoeba histolytica, Giardia intestinalis, and Trichomonas vaginalis share many biochemical features, e.g., energy and amino acid metabolism, a spectrum of drugs for their treatment, and the occurrence of drug resistance. These parasites possess metabolic pathways that are divergent from those of their mammalian hosts and are often considered to be good targets for drug development. Sulfur-containing-amino-acid metabolism represents one such divergent metabolic pathway, namely, the cysteine biosynthetic pathway and methionine gamma-lyase-me

Simultaneous measurement of volatile sulfur compounds using ascorbic acid for oxidant removal and gas chromatography-flame photometric detection.

A method for the simultaneous measurement of volatile sulfur compounds (COS, H2S, CS2, CH3SH, DMS) is established with preconcentration and GC-flame photometric detection (FPD). Prior to preconcentration of ambient air, it was necessary to remove SO2, water vapor and atmospheric oxidant. SO2 and water vapor were removed using a glass fiber filter and a cooled PTFE water trap loop, respectively. In order to remove atmospheric oxidant, the efficiency of an ascorbic acid scrubber was examined. It was found that an ascorbic acid scrubber enabled measurement of volatile sulfur compounds without ads
