Angiotensin II-induced natriuresis is attenuated in knockout mice lacking the receptors for tumor necrosis factor-α.

Intravenous infusion of relatively higher doses of angiotensin II (AngII) elicits natriuresis as opposed to its usual anti-natruretic response. As AngII can induce tumor necrosis factor-α (TNFα) production which elicits natriuresis via its action on TNFα receptor type 1 (TNFR1), we hypothesize that the concomitant release of TNFα contributes to the natriuretic response to AngII. Responses to AngII infusion (1 ng min-1  g-1 for 75 min, iv) were evaluated in anesthetized knockout (KO) mice lacking TNFR1 (n = 6) and TNFR2 (TNFα receptor type 2; n = 6) and compared these responses with those in wi

P2X7 receptor blockade during Cryptococcus neoformans infection promotes a Th2-associated immune profile and enhances fungal dissemination to the brain.

Cryptococcosis is a systemic fungal infection caused by Cryptococcus species, predominantly Cryptococcus neoformans. The disease affects the lungs and can disseminate to the Central Nervous System (CNS), leading to high-mortality meningoencephalitis. Purinergic signaling, driven by extracellular nucleotides and nucleosides, plays a key role in immune regulation. Among purinergic receptors, P2X7 receptors modulate immune cell activity and cytokine release. However, its role in cryptococcosis remains unclear. Here, we evaluated the effects of Brilliant Blue G (BBG, 50 mg/kg), a widely used pharm

Contributory Role of BLT2 in the Production of Proinflammatory Cytokines in Cecal Ligation and Puncture-Induced Sepsis.

BLT2 is a low-affinity receptor for leukotriene B4, a potent lipid mediator of inflammation generated from arachidonic acid via the 5-lipoxygenase pathway. The aim of this study was to investigate whether BLT2 plays any role in sepsis, a systemic inflammatory response syndrome caused by infection. A murine model of cecal ligation and puncture (CLP)-induced sepsis was used to evaluate the role of BLT2 in septic inflammation. In the present study, we observed that the levels of ligands for BLT2 (LTB4 [leukotriene B4] and 12(S)-HETE [12(S)-hydroxyeicosatetraenoic acid]) were significantly increas

A 5-Lipoxygenase Inhibitor, Zileuton, Modulates Host Immune Responses and Improves Lung Function in a Model of Severe Acute Respiratory Syndrome (SARS) Induced by Betacoronavirus.

Exacerbated inflammatory responses are a hallmark of severe coronavirus disease 2019 (COVID-19). Zileuton (Zi) is a selective inhibitor of 5-lipoxygenase, an enzyme involved in the production of several inflammatory/pro-resolving lipid mediators. Herein, we investigated the effect of Zi treatment in a severe acute respiratory syndrome (SARS) model. Mouse hepatitis virus (MHV)3-infected mice treated with Zi significantly improved the clinical score, weight loss, cardiopulmonary function, and survival rates compared with infected untreated animals. The protection observed in Zi-treated mice was

Luteolin ameliorates rat model of metabolic syndrome-induced cardiac injury by apoptosis suppression and autophagy promotion via NR4A2/p53 regulation.

BACKGROUND: Reduced cardiac autophagy, inflammation, and apoptosis contribute to cardiovascular complications caused by metabolic syndrome (MetS). It is documented that the nuclear receptor 4A2 (NR4A2) could modulate autophagy and apoptosis in cardiac complications. The aim of this investigation was to assess the therapeutic potential of luteolin, with documented beneficial properties, against MetS-associated cardiac injury. METHODS: Forty male albino Wistar rats were divided into 5 groups randomly as controls, MetS, and MetS animals treated with luteolin (25, 50, 100 mg/kg ip). The animal's

Inflammation, genetic polymorphisms in proinflammatory genes TNF-A, RANTES, and CCR5, and risk of pancreatic adenocarcinoma.

Adenocarcinoma of the exocrine pancreas is the fourth leading cause of cancer-related death in men and women in the U.S. Cytokines and other proinflammatory mediators have been implicated in inflammatory pancreatic diseases including pancreatitis and cancer. We analyzed cytokine gene polymorphisms as risk factors for pancreatic cancer using questionnaire data obtained by in-person interviews and germ line DNA collected in a population-based case-control study of pancreatic cancer (532 cases and 1,701 controls) conducted in the San Francisco Bay Area. We used mass spectrometry and gel-based met

Modulation of the HMGB1/TLR4 pathway by montelukast in PTZ-induced epilepsy: alleviating neuroinflammatory oxidative stress and seizure severity.

Background: Epilepsy is a multifactorial neurological disorder often associated with neuroinflammatory processes that contribute to the persistence of recurrent seizures. Conventional antiepileptic drugs (AEDs) frequently fail to achieve complete seizure control, highlighting the urgent need for alternative therapeutic strategies. Recent studies have explored the potential of montelukast, a leukotriene receptor antagonist, in the management of neurological disorders, including epilepsy. Methods: In the present study, the neuroprotective and antiepileptic effects of montelukast were evaluated u

Study of the effector mechanism involved in the production of haemorrhagic necrosis of the small intestine in rat passive anaphylaxis.

1. The effector mechanism of intestinal necrosis in rat anaphylaxis was studied following several complementary approaches: (i) the use of monoclonal antibodies (mAb) belonging to different classes (IgG1, IgG2b and IgE anti-DNP), (ii) the assay of mediators, and (iii) the use of pharmacological tools. 2. Lethality and haemorrhagic necrosis of the small intestine were observed in IgE-sensitized rats, whereas IgG mAb produced milder physiological disturbances. 3. Inhibition of leukotriene biosynthesis reduced the drop of systemic blood pressure (BP) and the extent of protein-rich plasma exudatio

SARS-CoV-2 Delta, not Omicron, drives Transferrin receptor-1 upregulation, inflammatory activation, and viral persistence in macrophages.

SARS-CoV-2 variants imprint distinct immune and metabolic signatures in host cells. Here, we show that the Delta and Omicron BA.4/BA.5 variants differentially regulate proinflammatory responses, oxidative stress, and iron metabolism in human primary M1 macrophages. Exposure to Delta - but not Omicron - strongly upregulated interleukin (IL)-6, IL-1β, and tumor necrosis factor alpha (TNF-α), increased reactive oxygen species (ROS) production, suppressed IL-10, and impaired viral clearance. Exposure of macrophages with Delta was further associated with disrupted iron homeostasis, marked by concur

Epstein-Barr virus LMP1 inhibits the expression of SAP gene and upregulates Th1 cytokines in the pathogenesis of hemophagocytic syndrome.

The primary infection of Epstein-Barr virus (EBV) may result in fatal infectious mononucleosis or hemophagocytic syndrome (HPS) in 2 diseases; that is, X-linked lymphoproliferative disorder (XLP) and hemophagocytic lymphohistiocytosis (HLH). XLP is linked to mutations of the SAP/SH2D1A gene with dysregulated T-cell activation in response to EBV infection. Patients with sporadic HLH, however, usually have no mutation of the SAP/SH2D1A gene, and EBV latent membrane protein-1 (LMP1) can up-regulate Th1 cytokines in EBV-infected T cells. Since both diseases share common manifestations of HPS, it i

Montelukast alleviates thioacetamide-induced hepatic encephalopathy in rats.

Hepatic encephalopathy (HE) is a serious neuropsychiatric dysfunction associated with acute and chronic liver disease. Montelukast (Mon) is a cysteinyl leukotriene receptor 1 (CysLT1R) antagonist approved as adjuvant therapy for asthma. The antioxidant and anti-inflammatory effects of montelukast have been reported in previous studies. To the best of our knowledge, Mon therapeutics' efficacy against thioacetamide-induced HE has not been investigated. This study aims to detect the protective effects of Mon (5 and 10 mg/kg, orally for seven consecutive days) on TAA (200 mg/kg, i.p., at three alt

Lack of soluble tumor necrosis factor alpha receptor 1 and 2 and interleukin-1beta compartmentalization in lungs of mice after a single intratracheal inoculation with live Porphyromonas gingivalis.

Porphyromonas gingivalis aspiration pneumonia induces local and systemic cytokine responses, but the dynamic of the immune response following lung exposure to live P. gingivalis is poorly understood. Groups of 50 12-week-old male BALB/c mice were inoculated intratracheally with live P. gingivalis ATCC 33277 using low dose (2 x 10(5) colony-forming units [CFU]), high dose (2.9 x 10(9) CFU), or phosphate-buffered saline (PBS; sham-inoculated), and the 3 groups were sacrificed at 2, 6, 24, 72, 168 hours. Lung and serum samples were collected for tumor necrosis factor alpha (TNF-alpha), soluble TN

Mitigation and Management of Common Toxicities Associated with the Administration of CAR-T Therapies in Oncology Patients.

Chimeric antigen receptor T-cell (CAR-T) therapies are one of the main approaches among targeted cellular therapies. Despite the potential benefit and durable responses observed in some patients receiving CAR-T therapies, serious and potentially fatal toxicities remain a major challenge. The most common CAR-T-associated toxicities include cytokine release syndrome (CRS), neurotoxicity, cytopenias, and infections. While CRS and neurotoxicity are generally managed with tocilizumab and corticosteroids, respectively, high-grade toxicities can be life-threatening. Close postinfusion monitoring and
