This study evaluated the effect of dietary calcium (Ca) levels and deoxynivalenol (DON) contamination on Ca and phosphorus (P) utilization and bone mineralization in piglets. During an initial 13-d depletion phase, 64 piglets (15.7 ± 0.7 kg) received a control (DON-) or DON-contaminated treatment (DON+, 2.7 mg DON/kg) with either a low Ca (Ca-, 0.39%) or normal Ca level (Ca+, 0.65%) with a constant digestible P level (0.40%). A second group of 16 piglets received DON- or DON+ treatments for 9 d for gene expression analysis. During the subsequent 14-d repletion phase, all piglets were fed a Ca+

T-2 toxin and deoxynivalenol (DON) are two prevalent mycotoxins that cause cartilage damage in Kashin-Beck disease (KBD). Cartilage extracellular matrix (ECM) degradation in chondrocytes is a significant pathological feature of KBD. It has been shown that the Hippo pathway is involved in cartilage ECM degradation. This study aimed to examine the effect of YAP, a major regulator of the Hippo pathway, on the ECM degradation in the hiPS-derived chondrocytes (hiPS-Ch) model of KBD. The hiPS-Ch injury models were established via treatment with T-2 toxin/DON alone or in combination. We found that T-

Fungal contamination is a concern for the food industry. Fungal spores resist food sterilization treatments and produce mycotoxins that are toxic for animals and humans. Technologies that deactivate spores and toxins without impacting food quality are desirable. This study demonstrates the efficiency of a high voltage atmospheric cold plasma (HVACP) technology using air to generate reactive oxygen (ROS) and nitrogen (RNS) species for the degradation of Aspergillus flavus cultures and the deoxynivalenol (DON) mycotoxin. Optical emission and absorption spectroscopy demonstrate ionization of hydr

Contamination of food/feedstuffs by mycotoxins is a serious problem worldwide, causing severe economic losses and serious health problems in animals/humans. Deoxynivalenol (DON) is a major mycotoxin contaminant and is known to impair intestinal barrier function. Grapes and red wine are rich in polyphenols, such as resveratrol (RES), which has striking antioxidant and anti-inflammatory activities. RES is a food-derived component; therefore, it may be simultaneously present with DON in the gastrointestinal tract. The aim of this study was to explore in vitro protective effects of RES against DON

Deoxynivalenol (DON) is the most commonly detected mycotoxin contaminant of cereal crops and cereal based food products in temperate regions of the world. DON causes adverse health effects in animals, passes through to the foetus and causes foetal abnormalities in animals. Biomonitoring for DON has revealed frequent human exposure. This study reports on DON transfer across the human placenta. Firstly, in vitro studies with the BeWo b30 clone were used as a rapid screening model showing transfer of DON through a stable confluent cell monolayer. Five term placentas were then used to study DON tr

Histological observation combined with determination of the serum glucose level and histochemical detection of liver glycogen was undertaken to examine the acute toxicity of fusarenon-X (FX) in mice. Mice intraperitoneally injected with a sublethal dose of the toxin showed rapidly developed hypoglycemia followed by depletion of liver glycogen. Mitotic inhibition was observed in many visceral organs and most markedly in the intestinal crypt cells, where the mitotic figures completely disappeared prior to the increase in number of the degenerated and nucrotic cells. No glycosuria was found. The

T-2 toxin is the most toxic as a type A trichothecenes, which could contaminate grains, especially in wheat and corn. It can cause immune suppression, neurotoxicity, the apoptosis of cells and even induce tumorigenesis. Recent studies have indicated that selenium (Se) have protective effect against mycotoxins-induced toxicity. The present studies was designed to investigate the protective role of Selenomethionine (SeMet) on T-2 toxin-induced toxicity in rabbit's jejunum. 50 New Zealand rabbits were divided into five group (Control group, T-2 group, low-dose Se + T-2 group, medium-dose + T-2 gr…

Chronic exposure to deoxynivalenol (DON)-contaminated food and feed poses significant hepatotoxic, enterotoxic, and immunotoxic risks to humans and animals. Nuclear factor erythroid 2-related factor 2 (Nrf2), a crucial cellular protective factor, plays a pivotal role in oxidative stress, inflammatory responses, and apoptosis. This review demonstrates that modulation of the Nrf2 signaling pathway can mitigate oxidative damage, suppress inflammatory responses and ferroptosis, reduce apoptosis, and alleviate endoplasmic reticulum stress and DON-induced injury, thereby protecting organisms from DO

Deoxynivalenol (DON), a prevalent Fusarium mycotoxin, poses a significant threat to intestinal health in piglets. This study established a deoxynivalenol (DON) intoxication model in piglets, revealing compromised intestinal barrier function accompanied by inflammatory responses and oxidative stress. Using liquid chromatography-tandem mass spectrometry (LC-MS/MS) on intestinal samples from piglets (n = 6 per group), we identified 8,084 proteins, of which 124 showed significantly altered expression following DON exposure. Bioinformatic analysis indicated that the differentially expressed protein

The mycotoxin deoxynivalenol (DON) is of high importance among feed contaminants because of its frequent occurrence in toxicologically relevant concentrations worldwide. Cereal crops, the main component of chicken diet, are commonly contaminated with DON, resulting in frequent exposure of chickens to DON. Likewise, Campylobacter (C.), a pathogen of major public and animal health concern, is frequently found in chicken flocks and poses a threat to the One Health approach. Campylobacter colonizes the gastrointestinal (GI) tract of poultry with a high bacterial load in the caeca. However, the mec

Mycotoxins, antibiotic-resistant bacteria (ARB), and antibiotic resistance genes (ARGs) are significant environmental pollutants that pose considerable threats to environmental health and human safety through the food chain. This study is the first to investigate the impact of deoxynivalenol (DON), the most common mycotoxin, on antibiotic resistance dynamics in gut microbiota, demonstrating that DON exposure significantly selects for ARB and ARGs. Results indicated that 80.69 % of the ARGs with the highest increase in the DON group were exclusive to gram-positive bacteria, particularly those r

Fusarium graminearum colonizes the maize ear, causing Gibberella ear rot (GER) and producing harmful mycotoxins, including deoxynivalenol (DON) and zearalenone (ZEA). The disease can be managed in part by breeding and planting resistant maize cultivars. Resistance to GER is a quantitative and complex trait. Evaluation of diverse germplasm to identify regions and candidate genes associated with resistance may be useful for crop improvement efforts. Screening for GER is time-consuming and costly. Thus, identifying other traits that may serve as a proxy for GER resistance may accelerate resistanc

Deoxynivalenol and Alternaria toxins are common food contaminants posing significant threats to human and animal health. Although their individual toxicities have been extensively studied, the combined effects of co-exposure remain largely unexplored. Our findings revealed combined toxins displayed concentration-dependent synergistic and antagonistic interactions on human hepatocellular carcinoma cells. Co-exposure significantly reduced mitochondrial activity, increased intracellular ROS levels, and activated the mitochondrial-dependent caspase signaling pathway, ultimately leading to enhanced

Mycotoxin occurrence in food worldwide is estimated to increase due to climate change. Moreover, studies on how these food contaminants interfere with medications and especially anticancer therapies are rare. With the rise of anticancer immunotherapies, particularly mycotoxins with immunomodulatory activity, such as alternariol (AOH) or deoxynivalenol (DON), are of great concern. Both mycotoxins interfere with the pro-inflammatory nuclear factor kappa B (NF-κB) pathway in myeloid cells. This pathway not only plays an important role in the anticancer immune response but also inflammatory side e

As a common mycotoxin, deoxynivalenol (DON) contaminates cereal grains and feed in field or during processing and storage. DON elicits a spectrum of adverse effects in animals including anorexia and growth retardation. Especially, the presence of DON has also been detected in muscle, suggesting that DON may has the potential to affect the development of muscle. However, the relevant research is very rare and the molecular mechanism remains unclear. Myoblasts differentiation into multinucleated myotubes is one of the crucial steps of skeletal muscle development. In the present study, we investi

Based on the fact that mycotoxins and the food-borne bacteria coexist in the natural environment and pose a significant health hazard to humans and animals, it is important to investigate the immunosuppressive mechanism of ZEA (zearalenone), DON (deoxynivalenol), and their combination in bacterial infections. In this study, we established a mouse model of mycotoxin low-dose exposure combined with Listeria monocytogenes infection and investigated the effects of ZEA, DON and their combination on Th1-mediated anti-intracellular bacterial infection based on CD4+ T cell activation and differentiati

There is a lack of information available on the anorexic action of fusarenon-x (FX), which is a sesquiterpenoid mycotoxin. In this study, we investigated the changes in the hypothalamus and small intestine related to appetite after oral FX exposure. The time-course change of food intake after oral FX exposure (0.5, 1.0, and 2.5 mg/kg bw) in B6C3F1 mice showed that 2.5 mg/kg bw of FX significantly suppressed food intake during 3-6 h compared to the control. Furthermore, the total food intake for 24 h was lower in the group exposed to FX than in the control. The FX exposure (2.5 mg/kg bw for 3 h
