Utilization of endogenous urea secreted into the gastrointestinal tract of the sheep.

The possibility of the utilization of endogenous urea secreted into the gastrointestinal tract was studied in sheep to which a single dose of 3 g 15N urea (50.4% enrichment) was administered by slow infusion via a permanent surgical cannula into the caecum. During a 7 days' experiment, 31.15 +/- 1.08% of the dose was excreted in the urine and 19.65 +/- 1.08% in the faeces, while 49.20 +/- 1.49% was retained in the organism. The maximum 15N values in blood from the jugular vein were found 10 h after starting the infusion and a high concentration was maintained for a further 24 h; sixty hours af…

Treatment of the syndrome of inappropriate secretion of antidiuretic hormone by urea.

Recent data have shown the role of urea in the urinary concentrating mechanism. We studied the effects of exogenous urea administration in hyponatremia associated with the syndrome of inappropriate secretion of antidiuretic hormone (SIADH). In 20 patients with SIADH, we observed a positive correlation between serum sodium and blood urea levels (r = 0.65; p less than 0.01). In one patient with an oat cell carcinoma and SIADH-induced hyponatremia, we observed the same positive correlation (r = 0.80; p less than 0.01) but also a negative one between the excreted fraction of filtered sodium and ur…

Brain imaging as a prognostic biomarker in urea cycle disorders.

The Urea cycle disorders (UCDs) represent a group of rare inborn errors of metabolism that have impaired nitrogen handling from the breakdown of protein, with ensuing acute episodic or chronic hyperammonemia and neurotoxicity. Although advances in newborn screening, molecular diagnosis, and therapeutic interventions have improved survival, neurocognitive, behavioral, and psychiatric sequelae remain prevalent in those with both neonatal onset as well as later onset partial deficiencies. The mechanisms of brain injury in UCDs are complex and multifactorial, and in some cases are not fully known.…

The Role of Urea Cycle Functional Studies in Preclinical Research.

Inborn errors of metabolism affecting the urea cycle are rare severe conditions caused by impaired nitrogen detoxification, leading to hyperammonemia and neurological morbidity across a broad clinical spectrum. Current biochemical diagnostics largely rely on quantification of metabolites. While indispensable for diagnosis and acute management, these snapshot readouts are highly dependent on external factors and often fail to reflect urea cycle function in metabolically compensated states, limiting their usefulness for disease stratification, therapeutic monitoring, and preclinical evaluation o…

Urea Transporters in Cancer: Emerging Roles and Their Clinical Implications.

Background/Objective: The SLC14A gene family in mammals encodes the urea transporters UT-A and UT-B (UTs), whose primary function is urea transport. In recent years, increasing research has shown that UTs are involved in tumor formation and progression. The purpose of this article is to review the current knowledge in the potential of urea transporters as tumor biomarkers and therapeutic targets. Methods: An extensive review of the literature was performed utilizing both PubMed and Web of Science databases focusing on articles published within the last ten years. Results: UTs are significantly…

Energy-Dependent Urea Transports in Mammals and their Functional Consequences.

In lower organisms (bacteria, fungi, yeast), some species that express the enzyme urease take up urea from the surrounding medium as a source of nitrogen, by energy-dependent urea transporters. In contrast, in mammals, urea is an endproduct of nitrogen metabolism, and the energy-dependent urea transports are associated with either the need to excrete nitrogen efficiently, in the case of excess nitrogen intake, or the need to conserve nitrogen and re-use it, in the case of low nitrogen supply.Three different energy-dependent urea transports have been characterized functionally in the mammalian …

Hyperammonemia in urea cycle disorders: A toxic metabolite for the brain.

Ammonia can easily cross the blood-brain barrier in its non-ionized form, leading to significant accumulation in the brain during severe hyperammonemia. Excess ammonia is highly toxic to the brain and can cause neurological dysfunctions, such as tremors, ataxia, seizures, coma, and even death. Hyperammonemia is often the primary manifestation of urea cycle disorders (UCDs), a group of inherited metabolic disorders characterized by impaired nitrogen detoxification due to dysfunctions in urea cycle-related enzymes and transporters. Patients with complete deficiencies of urea cycle enzymes typica…

Effect of oral urea supplementation on the endometrial transcriptome of mares.

An intravenous large dose of protein led to an increased blood urea nitrogen (BUN), resulting in a lesser uterine pH and altered uterine gene expression in mares. The objective of the present study was to evaluate effects of a more physiological methodology to increase BUN on the endometrium of mares. Mares were fed hay and a treatment or control diet (n = 11 mares/treatment) in a crossover design starting at time of ovulation detection (D0) and continuing until D7. Mares of the treated group were fed urea (0.4 g/kg BW) with sweet feed and molasses, and those of the control group were fed swee…

Design, Synthesis, and Evaluation of Isoquinoline Ureas as TRPV1 Antagonists.

BACKGROUND: The inhibition of transient receptor potential vanilloid receptor 1 (TRPV1) has emerged as a novel approach for the treatment of various pain states. Pyrrolidinyl urea, SB 705498 with pKb = 7.3 in guinea pig TRPV1 receptor has been investigated in Phase II clinical trials for pain and chronic cough. Another heteroaryl urea derivative, A-425619 1, has been reported to be a potent and selective TRPV1 antagonist of capsaicin-evoked receptor activation with an IC50 value of 4 nM in hTRPV1. OBJECTIVE: A series of thirteen A-425619 1 analogues with modifications centered around the Creg…

Hydrolysis of particleboard bonded with urea-formaldehyde resin for recycling.

In this study, the removal of urea-formaldehyde (UF) resin adhesives from waste wood particleboards (PBs) via hydrolysis was discussed, particularly the use of this application to combat environmental issues often encountered in recycling projects. Herein, the conditions required for producing PBs with poor binding properties were examined. Additionally, we determined the appropriate formaldehyde: urea (F/U) mole ratios, namely, 0.95, 1.05, and 1.15, required for generating UF resins that can be characterized and used as PB binders. The resulting values were compared with those obtained for a …

Determination of salivary urea and uric acid of patients with halitosis.

BACKGROUND: Halitosis is the presence of unpleasant or foul smelling breath. The origin of halitosis may be related to both systemic and oral conditions, but a large percentage of cases, about 90%, is generally related to an oral cause. The aim of this study was to compare the concentration of urea and uric acid in patients with halitosis and people without halitosis. MATERIALS AND METHODS: In this case-control study, concentration of urea and uric acid was compared between two groups: (1) persons suffering halitosis (2) control group without halitosis. Each group includes fifty patients. Uns…

The emerging physiological roles of the SLC14A family of urea transporters.

In mammals, urea is the main nitrogenous breakdown product of protein catabolism and is produced in the liver. In certain tissues, the movement of urea across cell membranes is specifically mediated by a group of proteins known as the SLC14A family of facilitative urea transporters. These proteins are derived from two distinct genes, UT-A (SLC14A2) and UT-B (SLC14A1). Facilitative urea transporters play an important role in two major physiological processes - urinary concentration and urea nitrogen salvaging. Although UT-A and UT-B transporters both have a similar basic structure and mediate t…
