Exercise-induced bronchoconstriction.

While mainstream therapies for androgenetic alopecia (AGA), including pharmacological treatments, mesotherapy, low-level light therapy, and hair transplantation, often fall short of clinical needs due to limited efficacy, side effects, or invasiveness, regenerative medicine based on stem cells shows promise yet faces challenges related to immunogenicity, standardization, and safety. Against this backdrop, extracellular vesicles (EVs)-nanoscale signaling vesicles enclosed by a lipid bilayer-have rapidly emerged as an innovative "cell-free" therapeutic paradigm. By delivering bioactive "cargo" s

[Preparation of dual-functional composite magnetic nanomaterials modified with different metals/aptamers and their performance in exosome enrichment].

Exosomes, which are extracellular vesicles with sizes of 30-150 nm, contain proteins, lipids, RNA, etc., which can reflect important information about parental cells. They also have unique structures and can perform characteristic biological functions. Although the release of exosomes is a normal process, tumor cells release more exosomes, and the contents can induce cancer progression. Exosomes are widely distributed in body fluids at high concentrations and are easy to obtain; hence, the collection of exosomes released by tumor cells has become one of the main directions in tumor liquid biop

Potentiation of intraocular absorption and drug metabolism of N-acetylcarnosine lubricant eye drops: drug interaction with sight threatening lipid peroxides in the treatment for age-related eye diseases.

Cataract is the dominant cause of blindness worldwide. Studies of the morphological structure and biophysical changes of the lens in human senile cataracts have demonstrated the disappearance of normal fiber structure in the opaque region of the lens and the disintegration of the lens fiber plasma membrane in the lens tissue. Morphological and biochemical techniques have revealed the regions in human cataractous lenses in which the plasma membrane derangement occurs as the primary light scattering centers which cause the observed lens opacity. Human cataract formation is mostly considered to b

Biological interactions of graphene-family nanomaterials: an interdisciplinary review.

Graphene is a single-atom thick, two-dimensional sheet of hexagonally arranged carbon atoms isolated from its three-dimensional parent material, graphite. Related materials include few-layer-graphene (FLG), ultrathin graphite, graphene oxide (GO), reduced graphene oxide (rGO), and graphene nanosheets (GNS). This review proposes a systematic nomenclature for this set of Graphene-Family Nanomaterials (GFNs) and discusses specific materials properties relevant for biomolecular and cellular interactions. We discuss several unique modes of interaction between GFNs and nucleic acids, lipid bilayers,…

Serum and uterine fluid extracellular vesicles and prostaglandin E metabolite concentrations during diestrus and early pregnancy in mares.

Prostaglandins produced by endometrial and conceptus tissues have an established role in maintaining luteal function in early pregnancy and initiating luteolysis during diestrus in mares. The cellular mechanisms controlling the secretion and delivery of prostaglandins have not been fully elucidated. We hypothesized that nanosized lipid bilayer extracellular vesicles (EV) that carry a variety of biological molecules would be involved in these key regulatory processes. Serum and uterine fluid samples were collected from diestrus and pregnant mares on day (D) 10 and D14 post-ovulation, and concep

Cascade-responsive HA-decorated liposome/mesoporous silica nanoplatform co-delivering BMF-219 and 5-FU enhances gastric cancer chemotherapy through Menin- and p53-associated apoptotic signaling.

BACKGROUND: Our preliminary study found that Men1 is an oncogene and a potential therapeutic target in gastric cancer (GC). Chemotherapy remains a common therapeutic regimen for GC, but is frequently limited by multidrug resistance and poor intratumoral coordination of combination drugs. Targeting Menin may enhance chemotherapy sensitivity, but efficient co-delivery of Menin inhibitors and chemotherapeutics is hindered by the profound spatiotemporal pharmacokinetic decoupling of the drugs in vivo. METHODS: We first evaluated the synergistic antitumor effect and Menin/p53-associated mechanism

Bacterial Membrane Vesicles as Versatile Platforms for Systemic and Mucosal Vaccines.

Bacterial membrane vesicles (BMVs), encompassing outer membrane vesicles (OMVs) released from Gram-negative bacteria and extracellular vesicles (EVs) released from Gram-positive bacteria, have emerged as promising vaccine platforms owing to their intrinsic immunostimulatory properties and capacity to deliver a wide range of antigens. Although conventional vaccines effectively prevent infectious diseases, their long-term efficacy is often limited by antigenic variation and reliance on a restricted number of licensed adjuvants. BMVs, as self-adjuvanting systems, enable both antigen delivery and

Vesicle Nucleation Peptide Fusion Induced Extracellular Vesicles Are Distinct From Escherichia coli Outer Membrane Vesicles, and Provide an Enhanced Platform for Protein Production and Purification.

Bacterial outer membrane vesicles (OMVs) are nano-sized, spherical structures released by Gram-negative bacteria that play diverse roles in bacterial physiology, including communication, nutrient acquisition and host interactions. These vesicles bud from the bacterial outer membrane and contain lipopolysaccharides, periplasmic proteins, nucleotides and other biomolecules. The vesicle nucleating peptide (VNp) is a short peptide tag that, when fused to the amino terminus of a protein of interest, promotes the formation of bespoke recombinant extracellular vesicles (EVs) in Escherichia coli, enab

Discovery of Novel SARS-CoV-2 Fusion Inhibitors-Posaconazole-Polyarginine Conjugates.

Objectives: The ongoing evolution of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) and the current treatment limitations-particularly the emergence of drug resistance and the reduced efficacy of some existing drugs against new variants-highlight the need for novel antiviral strategies with novel action mechanisms. Fusion inhibitors that disrupt six-helix bundle (6-HB) formation during viral entry represent a promising approach. Posaconazole, an antifungal agent, has been identified as a weak fusion inhibitor, but suffers from poor membrane permeability and modest activity. This

Clinical variability of ATP11C-related hemolytic anemia: expanding the phenotypic and diagnostic spectrum.

The ATP11C gene, localized on the X chromosome, encodes the major phosphatidylserine flippase in human red blood cells (RBCs). Flippases actively transport phospholipids from the outer to the inner leaflet of the lipid bilayer, establishing and maintaining phospholipid asymmetry crucial for cell survival. Variants in ATP11C have been recently associated with a novel form of X-linked congenital hemolytic anemia. In this study, we identified 10 individuals from 7 unrelated families harboring 6 rare, novel variants in the ATP11C gene. Three of the variants were further characterized and functiona

Calcium Nanoliposome Improves Glycemic Control in a Mouse Diabetes Mellitus Model.

INTRODUCTION: Intracellular calcium in pancreatic beta cells plays a crucial role in insulin synthesis and secretion. Diabetes impairs this calcium-mediated action, necessitating an effective delivery system such as liposomes to facilitate calcium uptake. METHODS: Calcium lactate nanoliposomes (6.25 mg/mL) were prepared via the thin-film hydration method using lecithin and cholesterol as bilayer lipids. Their glucose-lowering efficacy was tested in hyperglycemic mice induced by oral glucose (1 g/kg) and intraperitoneal streptozotocin (45 mg/kg). Pancreatic calcium levels were measured using X

Successful Control of Refractory Coccidioides Meningitis With MAT2203.

BACKGROUND: Coccidioides meningitis is a life-threatening complication with Coccidioides spp that requires lifelong antifungal therapy. While some cases respond to azoles, others are refractory and require periodic treatment with intravenous and/or intrathecal (IT) amphotericin B. MAT2203 is a novel formulation of amphotericin B that uses a rolled phosphatidylserine lipid nanocrystal bilayer that is orally absorbed. MAT2203 has been successfully used to treat cryptococcal meningitis and Histoplasma meningitis. CASE PRESENTATION: In 2017 a previously healthy construction worker developed cocci

Molecular Mechanisms of Basil (Ocimum basilicum L.) Polyphenol Extracts as Bio-Based Cryoprotectants for Streptococcus thermophilus: Chemical Profiling, DFT, Molecular Dynamics and Cell Viability.

Natural plant extracts rich in polyphenols are increasingly being studied as multifunctional food ingredients with antioxidant and stabilizing properties. In this study, Ocimum basilicum L. extracts were evaluated as biological cryoprotective agents for Streptococcus thermophilus. The extracts contained high levels of phenolic compounds (~1350-2200 mg GAE equivalents/L) and exhibited strong antioxidant activity (up to 5.6 mM Trolox equivalents). Density functional theory calculations showed low O-H bond dissociation energies (~72-74 kcal/mol in ethanol) for key components, including luteolin a

Functional properties of Candida albicans extracellular vesicles released in the presence of the antifungal drugs amphotericin B, fluconazole and caspofungin.

The prevalence of diseases caused by pathogenic fungi of the Candida genus is currently a significant problem, particularly due to the emerging antifungal drug resistance and increasing number of immunocompromised individuals susceptible to opportunistic infections. Recently, it has been shown that fungal extracellular vesicles (EVs) - nanometre-sized structures of cellular origin, equipped with varied cargo enclosed by lipid bilayer - may play a vital role in the response of pathogen cells to antifungal treatment. In this work, we demonstrated that Candida albicans yeast cells grown at the su

NIR-II fluorescence membrane probes for rapidly labelling hybrid of probiotic outer membrane vesicles and anti-PD-L1 scFv over-expressing cellular vesicles with targeted photothermal-immunotherapy of colon cancer.

Lipid-bilayer structured nanovectors (LBSNs) (e.g., liposomes and biomimetic vesicles) are burgeoning for diagnosis and therapy, and fluorescence labelling is often conducted for biodistribution tracing with visible/NIR-I fluorescence membrane probes (MPs). As NIR-II fluorescence imaging exhibits significantly improved fidelity, we synthesize NIR-II emissive donor-acceptor structured BODIPY-based MPs (BMP1-2) to label LBSNs composed of immunogenic probiotic outer membrane vesicles and cellular nanovesicles over-expressing anti-PD-L1 scFv, yielding BMP@Hybrid with tandem-amplified immunotherapy

ABCB4 disease-causing variants S242R, S346I, T437I and T1077M significantly impair its function and display differential sensitivity to potentiators.

The hepato-canalicular adenosine triphosphate (ATP)-binding cassette transporter ABCB4/MDR3 is responsible of the secretion of phosphatidylcholine (PC) into bile. Variations in ABCB4 gene induce a spectrum of cholestatic liver diseases, the most severe form is progressive familial intrahepatic cholestasis type 3 (PFIC3). The purpose of our study was to investigate the impact and potential rescue of four ABCB4 missense variants identified in patients, two of which (T437I and T1077M) affect homologous amino acids in the Walker A motifs in the two nucleotide-binding sites NBS2 and NBS1, respectiv

Keratinocyte-derived extracellular vesicles in painful diabetic neuropathy.

Painful diabetic neuropathy (PDN) is a challenging complication of diabetes with patients experiencing a painful and burning sensation in their extremities. Existing treatments provide limited relief without addressing the underlying mechanisms of the disease. PDN involves the gradual degeneration of nerve fibers in the skin. Keratinocytes, the most abundant epidermal cell type, are closely positioned to cutaneous nerve terminals, suggesting the possibility of bi-directional communication. Extracellular vesicles are lipid-bilayer encapsulated nanovesicles released from many cell types that med
