Does KarXT (xanomeline-trospium) represent a novel approach to schizophrenia management? A GRADE-assessed systematic review and meta-analysis of randomized controlled clinical trials.

BACKGROUND: Schizophrenia is a complex psychiatric disorder characterized by positive, negative, and cognitive symptoms. KarXT, a novel combination of xanomeline and trospium, offers potential therapeutic benefits for schizophrenia treatment by targeting muscarinic receptors and avoiding dopamine receptor blockade. We conducted a systematic review and meta-analysis to evaluate the efficacy and safety of KarXT. METHODS: PubMed, Scopus, Web of Science, and Cochrane databases were systematically searched for relevant randomized controlled trials (RCTs) up to October 2024. Studies involving adult

Digital Interventions and Mental Health Outcomes in Patients With Cancer: Systematic Review and Meta-Analysis.

BACKGROUND: Rising cancer rates have amplified psychiatric and psychosocial burdens, with 35%-40% of patients exhibiting diagnosable psychiatric disorders. While digital mental health interventions (DMHIs) present potential solutions for improving emotional well-being in this population, evidence remains fragmented and lacks clarity regarding optimal implementation strategies. This study evaluates the efficacy of digital interventions on mental health outcomes in patients with cancer, with particular focus on intervention duration and stakeholder involvement as moderating factors. OBJECTIVE:

Social-Media-Based Mental Health Interventions: Meta-Analysis of Randomized Controlled Trials.

BACKGROUND: Compared with other forms of online mental health interventions, programs delivered through social media apps may require less training and be more acceptable and accessible to various populations. During and after the pandemic, both the number of social media users and the prevalence of social-media-based mental health interventions increased significantly. However, to the best of the authors' knowledge, no meta-analysis so far has focused on rigorous social-media-based mental health interventions for general populations. OBJECTIVE: This preregistered meta-analysis synthesized fi

Impact of Interventions on Peri-Intubation Hypoxemia and Hypotension in Critically Ill Patients: Systematic Review and Meta-Analysis.

INTRODUCTION: Emergent endotracheal intubation is common in critically ill patients. Underlying pathophysiologic derangements puts these patients at increased risk of peri-intubation major adverse events (MAE) and have been associated with higher morbidity and mortality. Investigating the impact of interventions in the peri-intubation period on the rate of peri-intubation hypoxemia and hypotension can help improve management of emergent airways. METHODS: We searched PubMed, Embase, and Scopus databases from their beginning through April 2024 to identify randomized controlled trials (RCT) eval

Evaluating Minimal Residual Disease Negativity as a Surrogate Endpoint for Treatment Efficacy in Multiple Myeloma: A Meta-Analysis of Randomized Controlled Trials.

This meta-analysis examined the association between minimal residual disease (MRD) negativity and survival outcomes in 15 304 patients with multiple myeloma (MM) enrolled in randomized controlled trials published until June 2, 2024. Overall, there was a significant, negative and strong association between MRD negativity odds ratios and survival hazard ratios (β_PFS = -0.20, p < 0.001, β_OS = -0.12, p = 0.023). These associations remained significant for newly diagnosed patients (β_PFS = -0.35, p < 0.001), and they were consistent but not significant for relapsed/refractory patients (β_PFS = -0

Weighting methods for truncation by death in cluster-randomized trials.

Patient-centered outcomes, such as quality of life and length of hospital stay, are the focus in a wide array of clinical studies. However, participants in randomized trials for elderly or critically and severely ill patient populations may have truncated or undefined non-mortality outcomes if they do not survive through the measurement time point. To address truncation by death, the survivor average causal effect has been proposed as a causally interpretable subgroup treatment effect defined under the principal stratification framework. However, the majority of methods for estimating the surv
