A selected review of current retinal research and study.

The present paper reviews recent studies of retinal pathology from the American and British Journals of Ophthalmology during a one-year period (1973-1974). Selection is on the basis of potential value to clinical optometrists. Certain underlying factors in several types of retinal degeneration are first discussed, followed by characteristics of diabetic maculopathy and of other types of macular degeneration including that due to aging. Some material is presented on retinopathies related to sickle cell traits, to viral and worm infections, and to injury. Following a review of certain recent stu…

Evaluation of nucleated red blood cell count by Sysmex XE-2100 in patients with thalassaemia or sickle cell anaemia and in neonates.

BACKGROUND: Current haematology analysers have variable sensitivity and accuracy for counting nucleated red blood cells in samples with low values and in all those conditions characterised by altered sensitivity of red blood cells to the lysing process, such as in beta-thalassaemia or sickle-cell diseases and in neonates. The aim of our study was to evaluate the performance of the automated analyser XE-2100 at counting nucleated red blood cells in the above-mentioned three categories of subjects with potentially altered red blood cell lysis sensitivity and yet a need for accurate nucleated red

[Hemoglobin beta S haplotype in the Kebili region (southern Tunisia)].

Sickle cell anemia is a monogenic hereditary disease characterized by a mutation in the beta globin gene. Five major haplotypes associated with the beta S mutation have been defined: Benin, Bantu, Senegalian, Camerounian, and Arabo-Indian. Previous studies in northern Tunisia showed that sickle cell anemia was of Benin origin in this region. Patients from the south of Tunisia, mainly from the Kebili region, were not previously concerned. In this study, we have determined the beta S haplotype and evaluated phenotypical expression of the disease in 14 patients from this latter region. The use of

The triumph of good over evil: protection by the sickle gene against malaria.

The mechanisms underlying Plasmodium falciparum resistance in persons with sickle trait have been under active investigation for more than a half century. This Perspective reviews progress in solving this challenging problem, including recent studies that have exploited the genomics and proteomics of the parasite. The formation of Hb S polymer in the parasitized AS RBC leads to impaired parasite growth and development along with enhanced clearance from the circulation and reduced deposition in deep postcapillary vascular beds. Enhanced generation of reactive oxygen species in sickled AS RBCs i

Resignifying the sickle cell gene: Narratives of genetic risk, impairment and repair.

Connecting theoretical discussion with empirical qualitative work, this article examines how sickle cell became a site of public health intervention in terms of 'racialised' risks. Historically, sickle cell became socio-politically allied to ideas of repair, in terms of the state improving the health of a neglected ethnic minority population. Yet, we elucidate how partial improvements in care and education arose alongside preventative public health screening efforts. Using qualitative research based in the United Kingdom, we show how a focus on collective efforts of repair can lie in tension w

[One year of neonatal screening for sickle-cell disease in Emma Children's Hospital/Academic Medical Center in Amsterdam].

OBJECTIVE: Early detection of children with sickle cell disease, determination of carriership frequency as well as evaluation of the knowledge regarding this haemoglobinopathy in various ethnic risk groups. DESIGN: Prospective. METHODS: From 1 November 1998 through to 31 October 1999, the ethnic background was recorded for consecutive pregnant women under care of the Academic Medical Centre, Amsterdam, the Netherlands, and the presence of carriership for sickle cell disease was evaluated. Carriers were asked about their knowledge of sickle cell disease. A diagnostic blood test of cord blood

Amplification of intermediate-size DNA sequences from formalin and B-5 fixed tissue by polymerase chain reaction.

Retrospective analysis of DNA from paraffin-embedded fixed bone marrow biopsy specimens is possible if preceded by amplification of the DNA sequences of interest by the polymerase chain reaction (PCR). These fixed specimens yield degraded DNA that may not be suitable for direct analysis by conventional digestion and hybridization methods. This limitation is circumvented by PCR amplification and subsequent analysis of the amplified products. The model used in this study is the amplification of a 725 base-pair (bp) beta-globin gene sequence encompassing the sickle-cell anemia point mutation, fol

Bone infarction in children with sickle cell disease: early diagnosis and differentiation from osteomyelitis.

An early differential diagnosis between bone infarction and osteomyelitis in sickle cell patients is practically impossible using routine laboratory methods. Twenty radioisotope studies in sickle cell patients during vaso-occlusive crises, were analyzed. A three stage process can be described. In the first stage a decreased uptake can be demonstrated by Tc 99m methylene diphosphonate (MDP) bone scanning. In osteomyelitis, an increased uptake area is usually seen at this early stage, corresponding to increased uptake in Ga-67 citrate scanning. At the second stage, approximately a week later, no

Haematological factors associated with proliferative retinopathy in sickle cell-haemoglobin C disease.

In a selected sample of patients with sickle cell-haemoglobin C (SC) disease, proliferative retinopathy (PSR) occurred in 90/243 (37%) patients, developed most frequently between the ages of 20 and 30 years, and affected 68% patients aged 45 years or over. Comparison of haematological indices in patients with and without PSR, after age-related effects were allowed for, indicated significant relationships with high mean cell volume in males and with low fetal haemoglobin in both sexes. A highly significant relationship with total haemoglobin level in males (as previously reported in SS disease)

[Prevalence of abnormal hemoglobins in newborns in Natal, Rio Grande do Norte, Brazil].

Hemoglobinopathies are among the most prevalent hereditary diseases in humans. Studies in different areas of Brazil have identified the prev … al hemoglobin. Some 37 (1.91%) of the newborns presented hemoglobinopathies, as follows: 29 (1.50%) sickle cell trait (Hb FAS), 6 (0.31%) heterozygous Hb C (Hb FAC), one (0.05%) homozygous Hb S (Hb FS), and one (0.05%) Hb Barts suggestive of alpha thalassemia. The results show the need to implement screening for hemoglobinopathies in the neonatal population.

[Acute silicosis caused by intentional inhalation of scouring powder].

A case of acute silicosis due to intentional inhalation of the scouring powder Ajax in a female patient suffering a sickle-cell trait (30% of S haemoglobin) is reported. It consisted of an acute immuno-allergic pneumonitis with fever and a respiratory distress syndrome. It was associated with a severe biological inflammatory syndrome and immunological disturbances with circulating immune complexes and antinuclear antibodies. The assessment of respiratory function demonstrated the presence of a restrictive syndrome with hypoxaemia and decreased CO transfer. The anatomopathological and mineralog

Improved method for automated red cell exchange in sickle cell disease.

An improved method for intermittent-flow erythrocytapheresis in patients with sickle cell disease is reported. The method, a modification of the standard red cell exchange procedure for the Haemonetics 30S unit, dilutes with physiologic saline the patient's blood as it flows from the draw line and before it reaches the centrifugation bowl. The blood dilution (approximately 1.6 parts saline to 1 part blood) is used only during the first two passes, when the proportion of sickle erythrocytes in the patient's blood is still high. Only that amount of bowl supernatant (saline-diluted plasma) necess
