CD95 (Fas) may control the expansion of activated T cells after elimination of bacteria in murine listeriosis.

CD95 (Fas) is known to mediate activation-induced T-cell death by apoptosis. To understand the role of CD95 during the course of bacterial infection, we examined the kinetics of alphabeta and gammadelta T cells in the peritoneal cavities and livers of 5-week-old CD95-defective MRL/lpr mice after an intraperitoneal infection with Listeria monocytogenes. The number of bacteria in the spleen decreased to an undetectable level by day 10 after infection with 7 x 10(3) Listeria cells similar to the number in MRL/+/+ mice. The number of alphabeta T cells expressing CD44 and CD95 reached a maximum in

Evidence for antigenic selection of large granular lymphocytes in a patient with Wiskott-Aldrich syndrome.

It is now recognized that CD3+ large granular lymphocyte (LGL) proliferations may be clonally derived from their normal CD3+LGL+ counterpart, but the nature of the pressure responsible for the proliferation of these cells remains unclear. We approached this problem by analyzing the diversity of the T-cell receptor repertoire of LGL developed in different clinical settings. Two of our patients had typical lymphoproliferative disorders. The third case was much more unusual, as the LGL proliferation was associated with a Wiskott-Aldrich syndrome. Our data relative to the patients with the lymphop

Circulating levels of cytokines and soluble cytokine receptors in various T-cell malignancies.

BACKGROUND: Cytokines, interleukin (IL)-4, IL-6, interferon-gamma (IFN-gamma), tumor necrosis factor-alpha (TNF-alpha), soluble CD23 (sCD23), and soluble IL-2 receptors (sIL-2R) are mediators of inflammation and immune response. Alterations in immune status of patients with various cancers may result in release of cytokines in circulation. The authors measured the circulating levels of IL-4, IL-6, IFN-gamma, TNF-alpha, sCD23, and sIL-2R from patients with T-cell chronic lymphocytic leukemia (T-CLL), T-cell acute lymphoblastic leukemia (T-ALL) and peripheral T-cell lymphoma (PTCL) to determine

Immune defects in families and patients with xeroderma pigmentosum and trichothiodystrophy.

Xeroderma pigmentosum (XP) is a rare autosomal recessive disease characterized by photosensitivity, a high incidence of cancer in sun-exposed portions of the skin and a reduced capacity to repair the u.v.-induced DNA damage. One of the XP mutations (XP-D) has also been identified in patients affected by trichothiodystrophy (TTD), a rare autosomal recessive disease characterized by brittle hair, mental and physical retardation, peculiar face and ichthyosis. However, in these patients there is no evidence of increased skin tumour incidence. Since an impairment of cell-mediated immunity has been

Cytotoxic T lymphocytes specific for self tumor immunoglobulin express T cell receptor delta chain.

CTL are thought to play a role in the elimination of transformed cells in vivo. The effectiveness of such CTL is in part dependent on recognition of tumor specific antigens. Among the best characterized tumor-specific antigens are the unique or idiotypic determinants on the Ig of B cell lymphomas. Here we describe the generation and properties of human CTL specific for the idiotype on autologous B cell tumors. These cells are CD3+,CD4-,CD8- and express the delta chain of the TCR. Such cells may prove useful in tumor-specific adoptive therapy.

Clonal expansion of CD8+ effector T cells in patients with calcific aortic valve disease.

AIMS: Calcific aortic valve disease (CAVD) is a complex cardiovascular pathology, culminating in aortic stenosis and heart failure with no effective treatment strategy, except invasive surgery. While inflammation may contribute to valvular fibro-calcific pathology and T cells have been identified within the leaflets, their contribution to CAVD pathogenesis remains unclear. METHODS & RESULTS: Disease-staged whole-tissue proteomics (n=16) confirmed a pro-inflammatory T cell driven signature, with immunohistochemistry (n=10) and immunofluorescence (n=8) conducted on control and CAVD valves to co

Genetic Evidence for Immune Cell-Lipid Mediation Pathways Linked to Esophageal Cancer and Associated Throat/Chest Pain Phenotypes.

PURPOSE: Previous studies showed complex associations between immune cells and esophageal cancer (EC). EC-related pain is closely linked to immune dysregulation and abnormal lipid metabolism, but their synergistic regulatory mechanisms remain unclear. This study aimed to investigate causal relationships between immune cell traits and EC, explore plasma lipids' mediating role, supplement genetic evidence for core immune factors-lipids-pain associations, and provide targets for EC's combined " Antineoplastic-analgesic" intervention. METHODS: Genome-wide association study summary data of 731 imm

Effect of compound kushen injection on immune function in patients with primary liver cancer: a systematic review and meta-analysis.

BACKGROUND: Primary liver cancer (PLC) is the third leading cause of cancer mortality worldwide. The heterogeneity of PLC and the complex immunosuppressive microenvironment make it difficult for single treatment regimens to meet patient needs. Therefore, it is crucial to find effective combination treatment strategies that enhance immune function in PLC therapy. OBJECTIVE: To systematically evaluate the effect of compound kushen injection (CKI) on enhancing immune function in PLC patients and its role in treatment efficacy and related adverse reactions. METHODS: Relevant Chinese and English

Atomistic TCR-ligand interactions instruct memory T-cell differentiation.

Memory T cells endow the mammalian host with an essential form of adaptive immunity that generates durable and rapid protection following re-exposure to an infectious pathogen or a cancerous transformation1-4. How naive CD8+ T cells develop into memory T cells that bifurcate into long-lived central (TCM) versus effector (TEM) subsets after such an antigen encounter remains ill-defined at the molecular level. To address this gap, here we dissect the functional T-cell receptor (TCR) repertoire of 242 murine CD8+ TCRαβ clonotypes specific for the peptide-major histocompatibility complex molecule

HIV-negative primary effusion lymphoma: a series of seven cases.

AIMS: Primary effusion lymphoma (PEL) is a rare aggressive, human herpesvirus-8 (HHV8)-associated neoplasm of post-germinal centre B cell origin. It usually presents as a serous effusion in human immunodeficiency virus (HIV)-positive patients. PEL is rarely reported in HIV-negative patients. METHODS AND RESULTS: wWe report seven cases of HIV-negative elderly men diagnosed with PEL in a single institution. Clinical information and laboratory characteristics were collected. All patients were men, with a mean age of 76 years (range = 60-93) and presented with pleural effusions (n = 6), pericardi

Human iPSC-Derived Endothelial Cells Exhibit Reduced Immunogenicity in Comparison With Human Primary Endothelial Cells.

Human induced pluripotent stem cell (iPSC)-derived endothelial cells (ECs) have emerged as a promising source of autologous cells with great potential to produce novel cell therapy for ischemic vascular diseases. However, their clinical application still faces numerous challenges including safety concerns such as the potential aberrant immunogenicity derived from the reprogramming process. This study investigated immunological phenotypes of iPSC-ECs by a side-by-side comparison with primary human umbilical vein ECs (HUVECs). Three types of human iPSC-ECs, NIBSC8-EC generated in house and two c

Becoming aware of γδ T cells.

The discovery that B cells and αβ T cells exist was predictable: These cells gave themselves away through their products and biological effects. In contrast, there was no reason to anticipate the existence of γδ T cells. Even the accidental discovery of a novel TCR-like gene (later named γ) that did not encode TCR α or β proteins did not immediately change this. TCR-like γ had no obvious function, and its early expression in the thymus encouraged speculation about a possible role in αβ T cell development. However, the identification of human PBL-derived cell-lines which expressed CD3 in comple

T lymphocytes in skin lesions of psoriasis and mycosis fungoides express B7-1: a ligand for CD28.

The activation of T cells requires two distinct signals. One signal involves interaction of the antigen-specific T-cell receptor with major histocompatibility complex molecules plus antigenic peptide; a second signal, which is antigen nonspecific, is the interaction of CD28 with its natural ligands B7-1 and B7-2/B70. CD28 is expressed on 80% of T cells, is upregulated after activation, and binds to B7 gene-family members, found on antigen-presenting cells. Because of our interest in the immunologic basis of benign and malignant T-cell-mediated disorders of the skin, we investigated the cellula

Mucosal cytokine response after short-term gluten challenge in celiac disease and non-celiac gluten sensitivity.

OBJECTIVES: In celiac disease (CD), gluten induces both adaptive and innate immune responses. Non-celiac gluten sensitivity (NCGS) is another form of gluten intolerance where the immune response is less characterized. The aim of our study was to explore and compare the early mucosal immunological events in CD and NCGS. METHODS: We challenged 30 HLA-DQ2(+) NCGS and 15 CD patients, all on a gluten-free diet, with four slices of gluten-containing bread daily for 3 days. Duodenal biopsy specimens were collected before and after challenge. The specimens were examined for cytokine mRNA by quantitat

The effect of the peritoneal tumor microenvironment on invasion of peritoneal metastases of high-grade serous ovarian cancer and the impact of NEOADJUVANT chemotherapy.

Peritoneal metastases of high-grade serous ovarian cancer (HGSOC) are small-sized deposits with superficial growth toward the peritoneal cavity. It is unknown whether integrity of the peritoneal elastic lamina (PEL) correlates with the peritoneal tumor microenvironment (pTME) and whether neoadjuvant chemotherapy (NACT) affects the pTME. We explored integrity of PEL, composition of pTME, effects of NACT, and the prognostic implications in patients with extensive peritoneal metastases of HGSOC. Peritoneal samples (n = 69) were collected during cytoreductive surgery between 2003 and 2016. Clinica

Combination exposure of melamine and cyanuric acid is associated with polyuria and activation of NLRP3 inflammasome in rats.

The molecular mechanisms of melamine-induced renal toxicity have not been fully understood. The purpose of the study aimed to investigate whether melamine and cyanuric acid induced NOD-like receptor family pyrin domain-containing 3 (NLRP3) inflammasome activation in the kidney, which may contribute to abnormal water and sodium handling in a rat model. Wistar rats received melamine (Mel; 200 mg·kg body wt-1·day-1), cyanuric acid (CA; 200 mg·kg body wt-1·day-1), or Mel plus CA (Mel + CA; 100 mg·kg body wt-1·day-1, each) for 2 wk. Mel + CA caused damaged tubular epithelial structure and organelle

Increased Expression of Cytotoxic T-Lymphocyte-Associated Protein 4 by T Cells, Induced by B7 in Sera, Reduces Adaptive Immunity in Patients With Acute Liver Failure.

BACKGROUND & AIMS: Patients with acute liver failure (ALF) have defects in innate immune responses to microbes (immune paresis) and are susceptible to sepsis. Cytotoxic T-lymphocyte-associated protein 4 (CTLA4), which interacts with the membrane receptor B7 (also called CD80 and CD86), is a negative regulator of T-cell activation. We collected T cells from patients with ALF and investigated whether inhibitory signals down-regulate adaptive immune responses in patients with ALF. METHODS: We collected peripheral blood mononuclear cells from patients with ALF and controls from September 2013 thr

Impact of the Oral Commensal Flora on Alveolar Bone Homeostasis.

Homeostasis of healthy periodontal tissues is affected by innate and adaptive immunosurveillance mechanisms in response to the normal oral flora. Recent comparisons of germ-free (GF) and normal specific-pathogen-free (SPF) mice have revealed the impact of host immunosurveillance mechanisms in response to the normal oral flora on alveolar bone height. Prior reports that alveolar bone height is significantly less in normal SPF mice compared with their age- and strain-matched GF counterparts suggest that naturally occurring alveolar bone loss is a normal component of healthy periodontal tissue ho
