Effects of dietary protein restriction and oil type on the early progression of murine polycystic kidney disease.

A paucity of research data exists on the potential for early dietary modification to directly retard cystic growth and proliferation in polycystic kidney disease (PKD). We have therefore examined the relative effects of dietary protein levels and oil type on the progression of disease in a murine model of PKD. In the first study, weanling DBA/2FG-pcy (pcy) mice were fed either a normal (NP), 25%, or low (LP), 6%, casein diet with 10% of either sunflower seed oil (SO) (containing n-6 fatty acids), or fish oil (FO) (containing n-3 fatty acids), in a 2 x 2 design. At the end of the dietary treatm

Characterization of human neutrophil ecto-protein kinase activity released by kinase substrates.

Although most studies of protein phosphorylation have focused on intracellular protein kinases, evidence for protein kinase activity on the surface of several types of cells has been described. Evidence was recently provided for the existence of ecto-protein kinase activity on the surface of human neutrophils. Evidence for three distinct ecto-protein kinase activities was detected, one that phosphorylates endogenous surface proteins, one that phosphorylates exogenous substrates in a cAMP-independent manner and is released in the presence of substrate, and a low level of activity of one that ph

Diet calcium carbonate, phosphorus and acidifying and alkalizing salts as factors influencing silica urolithiasis in rats fed tetraethylorthosilicate.

Three experiments were conducted to determine the effects of excess dietary calcium carbonate, phosphorus and urine acidifying and alkalizing salts on silica urolith formation in a model using rats fed dextrose-based diets containing 2% tetraethylorthosilicate (TES). Diets containing 2% TES lowered weight gains to 91-95% of gains made by rats fed non-TES diets. Urine silica concentrations of rats fed TES were generally in the range of 50-60 mg/dl. In experiment 1, rats fed TES with no additional dietary calcium carbonate had a silica urolith incidence of 35%. With additions of 1 and 2% calcium

The effects of nutrition on unconjugated plasma bilirubin concentrations in squirrel monkeys.

Bolivian squirrel monkeys (Saimiri sciureus) have fasting unconjugated hyperbilirubinemia (males: 2.0 +/- 0.14; females: 3.0 +/- 0.26 mg per dl) which resembles that of humans with Gilbert's syndrome. Closely related Brazilian squirrel monkeys have fasting levels (males: 0.29 +/- 0.045; females: 0.36 +/- 0.073 mg per dl) similar to normal people. The purpose of this study was to identify the underlying mechanisms and the nutritional factors involved. Both Bolivian and Brazilian squirrel monkeys had higher plasma bilirubin concentrations after an 18 hr fast than 4 hr after feeding. The developm

Rapid development of oedema and defective brain histidine metabolism in young guinea pigs fed protein-energy deficient diet.

Male weanling guinea pigs were fed ad libitum a 3% pelleted casein diet supplemented with ascorbic acid while control animals were pair-fed a standard guinea-pig chow. Oedema was observed in some of the former group as early as the 2nd week and became very prominent by the end of the 3rd week. The malnourished group was characterized by reduced plasma albumin level and some disaggregation of hepatic polyribosomes but these showed no correlation in time with development of oedema. The pool of plasma essential amino acids were markedly reduced as early as Day 7, and most prominently affected wer

The effects of excess amino acids on maintenance of pregnancy and fetal growth in rats.

The effects of diets, each with an excess of one essential amino acid, on the maintenance of pregnancy and fetal growth were investigated in rats. Rats were fed on 6% casein diet containing 5% threonine, methionine, valine, isoleucine, leucine, tyrosine, phenylalanine, tryptophan, or lysine from day 1 to day 14 or 21 of pregnancy. Excess methionine and leucine diets resulted in complete and 80% loss of fetuses, respectively. This fetal wastage was prevented by daily injection of 0.5 microgram of estrone and 4 mg of progesterone. Judging from the total food consumptions and body weight gains du

Protein Substitutes in PKU; Their Historical Evolution.

Protein substitutes developed for phenylketonuria (PKU) are a synthetic source of protein commonly based on L-amino acids. They are essential in the treatment of phenylketonuria (PKU) and other amino acid disorders, allowing the antagonistic amino acid to be removed but with the safe provision of all other amino acids necessary for maintaining normal physiological function. They were first formulated by a chemist and used experimentally on a 2-year-old girl with PKU and their nutritional formulations and design have improved over time. Since 2008, a bioactive macropeptide has been used as a ba

Isolation of a proteinase with plasminogen-activating activity from Lachesis muta muta (bushmaster) snake venom.

A plasminogen activator enzyme (LV-PA) from Lachesis muta muta venom was purified to homogeneity using gel filtration and anion exchange chromatography. SDS-PAGE under reducing conditions showed a single protein band with an Mr of 33,000 Da. It is an acidic glycoprotein which activates plasminogen to plasmin indirectly, functioning via prior formation of a molecular complex, known as plasminogen activator. The purified preparation catalyzes the hydrolysis of several p-nitroanilide peptide substrates containing Lys at the scissile bond. In contrast, no hydrolysis was detected on the synthetic s

Serum IgG subclass antibodies to a variety of food antigens in patients with coeliac disease.

Levels of serum IgA, IgG, and IgG subclass antibodies to a variety of dietary antigens were determined by enzyme linked immunosorbent assays in 14 adults with untreated coeliac disease and in 10 disease controls selected because of raised total IgG activities. The untreated coeliacs showed somewhat higher total IgG activity (p approximately 0.05) and significantly raised IgA and IgG1 + IgG3 activities to gliadin but reduced IgG4 activity (p less than 0.02) compared with the controls. High IgA and IgG1 + IgG3 activities were positively correlated (r = 0.67, p less than 0.01), and so were IgG an

Metabolic complexity and nutritional reductionism. Deconstructing the single active ingredient paradigm in protein and amino acid supplements.

Metabolism is an adaptive and highly interconnected system in which biological outcomes emerge from dynamic interactions among genes, proteins, metabolites, signaling pathways, microbiota, and environmental factors. Nevertheless, nutritional research has traditionally relied on a reductionist paradigm, attributing biological effects to isolated compounds acting on specific molecular targets through linear cause-effect relationships. While this approach has been instrumental in elucidating biochemical mechanisms and addressing nutrient deficiencies, its explanatory and predictive power is limit

Dietary Protein Source Mediates Colitis Pathogenesis Through Bacterial Modulation of Bile Acids.

BACKGROUND & AIMS: Evidence-based dietary recommendations for individuals with inflammatory bowel diseases are limited. Red meat consumption is associated with increased inflammatory bowel disease incidence and relapse in patients, suggesting that switching to a plant-based diet may limit gut inflammation. However, the components and mechanisms underlying the differential effects of these diets remain poorly understood. This study investigated the differential impact of dietary protein source on experimental colitis and related mechanisms. METHODS: Isocaloric synthetic diets containing protei

Immunorestorative effects of Limosilactobacillus reuteri LR6 in protein-energy malnourished mice under pathogen and allergen challenge.

Protein-energy malnutrition (PEM) is the most severe form of malnutrition, often linked with impaired immunity and increased susceptibility to infections. In this study, mice were initially fed a normal semi-synthetic well-nourished diet for one week and then divided into five groups. The animals in Group C (control) continued on the normal well-nourished diet, while Groups M (malnourished), SM (skim milk), PFM (probiotic fermented milk), and BS (probiotic suspension) received a protein- and energy-restricted diet (2% casein, 315.2 kcal) until PEM was established. Later, the groups SM, PFM and

Tea, coffee, and cocoa delay milk gastric coagulation in a biomimetic in vitro system (NERDT™) by hindering both acid- and pepsin-induced milk gelation.

This study investigated the effect of mixing whole milk with hot water extract of cocoa, coffee, or tea on milk intragastric coagulation during dynamic in vitro digestion. Milk diluted with water in the same proportion (70:30 v/v) was used as the control. The effect of adding cocoa, coffee, or tea extracts on acid- and pepsin-induced milk coagulation was first studied using a rheometer. The control exhibited a significantly shorter (P < 0.0001) gelation time in acid-induced coagulation than the milk mixtures containing extracts. The delaying effect of extracts on pepsin-induced coagulation was
