The reactions of monoclonal antibodies with structural proteins of mumps virus.

Mouse hybridomas producing antibodies against structural proteins of mumps virus were established by fusion of FO or SP 2/0 myeloma cells with spleen cells from BALB/c mice immunized with purified preparations of egg-grown mumps virus. Ascites fluids collected after i.p. inoculation of mice were characterized by different serologic tests. By immune precipitation tests with [35S]methionine-labeled mumps virus polypeptides, 17 clones were found to produce antibodies against the nucleocapsid protein (NP), 11 against the polymerase (P) protein, 10 against the membrane (M) protein, 12 against the f

Broad-spectrum inhibition of influenza A virus replication by blocking the nuclear export of viral ribonucleoprotein complexes.

UNLABELLED: Influenza A virus (IAV) remains a major threat to global health. The emergence of IAV strains resistant to commonly used anti-influenza medicines has made the need for novel broad-spectrum antiviral agents more urgent. The IAV nucleoprotein (NP) is one of the most conserved viral proteins encoded by IAV and is essential for multiple processes in the lifecycle of IAVs. In this study, we screened a high-affinity nanobody, Nb7, which specifically recognizes NP from different subtypes of IAV and broadly inhibits the replication of various IAVs. Mechanistically, Nb7 blocked the nuclear

Cross-Reactive Antibodies to SARS-CoV-2 and MERS-CoV in Pre-COVID-19 Blood Samples from Sierra Leoneans.

Many countries in sub-Saharan Africa have experienced lower COVID-19 caseloads and fewer deaths than countries in other regions worldwide. Under-reporting of cases and a younger population could partly account for these differences, but pre-existing immunity to coronaviruses is another potential factor. Blood samples from Sierra Leonean Lassa fever and Ebola survivors and their contacts collected before the first reported COVID-19 cases were assessed using enzyme-linked immunosorbent assays for the presence of antibodies binding to proteins of coronaviruses that infect humans. Results were com

Insights on 3D Structures of Potential Drug-targeting Proteins of SARS-CoV-2: Application of Cavity Search and Molecular Docking.

The emergence of the COVID-19 has caused public health problems worldwide and there is no effective pharmacological treatment for this disease. Research on 3D models of proteins and the search for active molecular sites are important tools to assist in the discovery of effective antiviral drugs to combat COVID-19. To address this problem, the 3D protein structures of SARS-CoV 2 were analyzed and submitted to cavities research, evaluation of their druggabillity and liganbility, and applied to molecular docking studies with potential ligand candidates actually assayed against COVID-19. Eight dru

Protein and modified vaccinia virus Ankara-based influenza virus nucleoprotein vaccines are differentially immunogenic in BALB/c mice.

Because of the high variability of seasonal influenza viruses and the eminent threat of influenza viruses with pandemic potential, there is great interest in the development of vaccines that induce broadly protective immunity. Most probably, broadly protective influenza vaccines are based on conserved proteins, such as nucleoprotein (NP). NP is a vaccine target of interest as it has been shown to induce cross-reactive antibody and T cell responses. Here we tested and compared various NP-based vaccine preparations for their capacity to induce humoral and cellular immune responses to influenza v

Identification of a novel compound targeting the nuclear export of influenza A virus nucleoprotein.

Although antiviral drugs are available for the treatment of influenza infection, it is an urgent requirement to develop new antiviral drugs regarding the emergence of drug-resistant viruses. The nucleoprotein (NP) is conserved among all influenza A viruses (IAVs) and has no cellular equivalent. Therefore, NP is an ideal target for the development of new IAV inhibitors. In this study, we identified a novel anti-influenza compound, ZBMD-1, from a library of 20,000 compounds using cell-based influenza A infection assays. We found that ZBMD-1 inhibited the replication of H1N1 and H3N2 influenza A

Adaptive Mutations That Occurred during Circulation in Humans of H1N1 Influenza Virus in the 2009 Pandemic Enhance Virulence in Mice.

UNLABELLED: During the 2009 H1N1 influenza pandemic, infection attack rates were particularly high among young individuals who suffered from pneumonia with occasional death. Moreover, previously reported determinants of mammalian adaptation and pathogenicity were not present in 2009 pandemic H1N1 influenza A viruses. Thus, it was proposed that unknown viral factors might have contributed to disease severity in humans. In this study, we performed a comparative analysis of two clinical 2009 pandemic H1N1 strains that belong to the very early and later phases of the pandemic. We identified mutati

Antibodies to influenza nucleoprotein cross-react with human hypocretin receptor 2.

The sleep disorder narcolepsy is linked to the HLA-DQB1*0602 haplotype and dysregulation of the hypocretin ligand-hypocretin receptor pathway. Narcolepsy was associated with Pandemrix vaccination (an adjuvanted, influenza pandemic vaccine) and also with infection by influenza virus during the 2009 A(H1N1) influenza pandemic. In contrast, very few cases were reported after Focetria vaccination (a differently manufactured adjuvanted influenza pandemic vaccine). We hypothesized that differences between these vaccines (which are derived from inactivated influenza viral proteins) explain the associ

Genetic diversity of perch rhabdoviruses isolates based on the nucleoprotein and glycoprotein genes.

Despite the increasing impact of rhabdoviruses in European percid farming, the diversity of the viral populations is still poorly investigated. To address this issue, we sequenced the partial nucleoprotein (N) and complete glycoprotein (G) genes of nine rhabdoviruses isolated from perch (Perca fluviatilis) between 1999 and 2010, mostly from France, and analyzed six of them by immunofluorescence antibody test (IFAT). Using two rabbit antisera raised against either the reference perch rhabdovirus (PRhV) isolated in 1980 or the perch isolate R6146, two serogroups were distinguished. Meanwhile, ba

Apoptotic cells are generated at every division of in vitro cultured T cell lines.

Long- and short-term T cell lines form the backbone of many assays for T cell function and also represent important tools for use in human immunotherapy. Despite much study concerning the requirements for T cell activation and growth in culture there is relatively little information about the kinetics of proliferation and cell death in such cultures. Here we studied these parameters in a long-term CD8(+) T cell line using a tetrameric MHC reagent and the fluorescent dye CFSE. We observed proliferation of the T cells within 24 h of restimulation with antigen and IL-2 and the cells continued to

A soluble inhibitor of T lymphocyte function induced by HIV-1 infection of CD4+ T cells: characterization of a cellular protein and its relationship to p15E.

Soluble suppressor factor (SSF), first described in association with HIV-1 infection in vivo, is a molecule(s) capable of inhibiting T cell-dependent immune reactivity. Its relationship to human immunodeficiency virus (HIV) was further defined as supernatants of mononuclear cell cultures from HIV-1-seropositive carriers, CD4+ T lymphocytes infected with HIV-1 in vitro, and a T cell hybridoma incorporating CD4+ lymphocytes from an HIV-1-seropositive individual were shown to elaborate factors with similar activity profiles. These factors were recognized antigenically by certain antibodies direct

Cohort profile: the SEROCoV-KIDS population-based cohort study and biobank on the impact of the COVID-19 pandemic in children and adolescents in Geneva, Switzerland.

PURPOSE: The COVID-19 pandemic raised major concerns about its potential effects on children and adolescents, particularly through disruptions to their health, social and educational environments and the exacerbation of existing inequalities. In response to the limited evidence-based knowledge available at the time, the SEROCoV-KIDS study was launched in 2021 as a prospective cohort and biobank to assess the pandemic's effects on youth health and well-being. It focuses on the general population of Geneva, Switzerland, as well as subgroups with vulnerabilities, including prior SARS-CoV-2 infect

Discontinuous template switching generates coronavirus subgenomic RNAs from the 3' viral genome end by 5' to 3' transcription.

In coronavirus (CoV)-infected cells, several structural and accessory proteins are synthesized from subgenome RNAs (sgRNA) containing a common genomic 5'-leader followed by a given open reading frame (ORF). We report that the abundance of these sgRNAs varies with distance from the 3'-end of the genome. Thus, there are more sgRNAs encoding nucleocapsid (N) than spike (S), presumably the results from discontinuous 5'-3' transcription template switch mediated by the viral replication and transcription complex (RTC). We optimized the circular polymerase extension reaction (CPER) methodology to gen

Arsenic trioxide impacts hepatitis B virus core nuclear localization and efficiently interferes with HBV infection.

UNLABELLED: The key to a curative treatment of hepatitis B virus (HBV) infection is the eradication of the intranuclear episomal covalently closed circular DNA (cccDNA), the stable persistence reservoir of HBV. Currently, established therapies can only limit HBV replication but fail to tackle the cccDNA. Thus, novel therapeutic approaches toward curative treatment are urgently needed. Recent publications indicated a strong association between the HBV core protein SUMOylation and the association with promyelocytic leukemia nuclear bodies (PML-NBs) on relaxed circular DNA to cccDNA conversion. Accuracy of Real-Time Polymerase Chain Reaction in COVID-19 Patients.

Coronavirus disease 2019 (COVID-19) is a mild to severe respiratory illness caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). The diagnostic accuracy of the Centers for Disease Control and Prevention (CDC)- or World Health Organization (WHO)-recommended real-time PCR (RT-qPCR) primers in clinical practice remains unproven. We conducted a prospective study on the accuracy of RT-qPCR using an in-house-designed primer set (iNP) targeting the nucleocapsid protein as well as various recommended and commercial primers. The accuracy was assessed by culturing or seroconversion. W

The Long-Term Characteristics of Immunity Conferred by COVID-19 Using Antibody Tests.

Introduction Since December 2019, more than 184 Million cases and 3.97 Million COVID-19 related deaths have been reported around the world. Since these statistics are laboratory-based confirmed cases, the true burden of disease may be underestimated. Many populations like those who are regularly visiting health care facilities and those with end-stage renal disease (ESRD) for visiting dialysis units, patients with malignancies, on regular chemo and radiotherapy, and healthcare workers (HCW) are considered high risk for nosocomial COVID-19 re-infections. Objective To understand the long-term be

Pharmacoinformatics-based identification of potential bioactive compounds against Ebola virus protein VP24.

BACKGROUND: The impact of Ebola virus disease (EVD) is devastating with concomitant high fatalities. Currently, various drugs and vaccines are at different stages of development, corroborating the need to identify new therapeutic molecules. The VP24 protein of the Ebola virus (EBOV) plays a key role in the pathology and replication of the EVD. The VP24 protein interferes with the host immune response to viral infections and promotes nucleocapsid formation, thus making it a viable drug target. This study sought to identify putative lead compounds from the African flora with potential to inhibit

Dimerization of the SP1 Region of HIV-1 Gag Induces a Helical Conformation and Association into Helical Bundles: Implications for Particle Assembly.

UNLABELLED: HIV-1 immature particle (virus-like particle [VLP]) assembly is mediated largely by interactions between the capsid (CA) domains of Gag molecules but is facilitated by binding of the nucleocapsid (NC) domain to nucleic acid. We previously investigated the role of SP1, a "spacer" between CA and NC, in VLP assembly. We found that small changes in SP1 drastically disrupt assembly and that a peptide representing the sequence around the CA-SP1 junction is helical at high but not low concentrations. We suggested that by virtue of such a concentration-dependent change, this region could a

[Investigation of West Nile virus seroprevalence in Hacettepe University Hospital blood donors and confirmation of the positive results by plaque reduction neutralization test].

West Nile virus (WNV), a member of Flaviviridae family, is an enveloped, icosahedral, single-stranded positive-sense RNA virus. WNV is transmitted to humans by infected mosquitoe (especially Culex spp.) bites and cause a variety of clinical outcomes, ranging from asymptomatic infection to severe meningoencephalitis. The aims of this study were to determine and confirm the WNV seroprevalence in a chosen healthy population and to provide epidemiological data for Turkey about the recent status of the infection at our region. A total of 1200 serum samples collected from blood donors (400 were fema

Binding of rabies virus polymerase cofactor to recombinant circular nucleoprotein-RNA complexes.

In rabies virus, the attachment of the L polymerase (L) to the viral nucleocapsids (NCs)-a nucleoprotein (N)-RNA complex that serves as template for RNA transcription and replication-is mediated by the polymerase cofactor, the phosphoprotein (P). P forms dimers (P(2)) that bind through their C-terminal domains (P(CTD)) to the C-terminal region of the N. Recombinant circular N(m)-RNA complexes containing 9 to 12 protomers of N (hereafter, the subscript m denotes the number of N protomers) served here as model systems for studying the binding of P to NC-like N(m)-RNA complexes.
