Molecular cloning and expression of a cDNA encoding the rabbit ileal villus cell basolateral membrane Na+/H+ exchanger.

A cDNA clone encoding a rabbit ileal villus cell Na+/H+ exchanger was isolated and its complete nucleotide sequence was determined. The cDNA is 4 kb long and contains 322 bp of 5'-untranslated region, 2451 bp of open reading frame and 1163 bp of 3'-untranslated area, with 70%, 91% and 40% identity to the human sequence, respectively. Amino acid sequence deduced from the longest open reading frame indicated a protein of 816 residues (predicted Mr 90,716) which exhibits 95% amino acid identity to the human Na+/H+ exchanger. The two putative glycosylation sites in the human Na+/H+ exchanger are c

The MHC class I associated beta 2-microglobulin (beta 2m) light chain is expressed in a molar excess over HLA-ABC and CD1 on the membrane of leukaemic B cells but not leukaemic T cells: evidence for further beta 2m-associated molecules.

Beta 2-microglobulin (beta 2m) constitutes the common light chain of both the MHC-encoded HLA-ABC molecules and a group of structurally related glycoproteins recognized by antibodies of the first cluster of differentiation (CD1a, CD1b and CD1c). These CD1 antigens appear similar to murine T1 and Qa molecules in terms of structure and tissue distribution, although the question of inter-species homology is controversial. A further group of alloantigens expressed predominantly on T cells has been reported however, with immunogenetic characteristics more closely analogous to the murine T1/Qa syste

Tissue-specific expression of murine keratin K13 in internal stratified squamous epithelia and its aberrant expression during two-stage mouse skin carcinogenesis is associated with the methylation state of a distinct CpG site in the remote 5'-flanking region of the gene.

Under normal conditions, the expression of the murine type-I keratin K13 is restricted to the suprabasal, differentiating cell layers of internal stratified squamous epithelia that line the oral cavity and the upper digestive tract. It is, however, also expressed aberrantly but constitutively in only the differentiating parts of 7,12-dimethylbenz[alpha]anthracene/12.0-tetradecanoyl-phorbol-13-acetate (DMBA/TPA) induced malignant epidermal tumors of the back skin of mice, whereas its likewise suprabasal expression in papillomas is highly variable [27]. In an approach to unravel regulatory DNA s

Lipid dependence of surface conformations of protein kinase C.

The change of conformation of protein kinase C interacting with the surface of a mercury electrode directly from a solution or through a lipid monolayer was inferred from the number of cystine residues exposed and reduced on the electrode and from their reduction potentials. Soluble protein kinase C was estimated to have 5-6 disulfide bonds which could potentially react with the mercury electrode. Two major reduction peaks of cystine at different microenvironments within the protein molecule adsorbed to a mercury surface. They were observed in a.c. polarograms and cyclic voltamograms at two di

Biosynthesis of paf-acether. Activators of protein kinase C stimulate cultured mast cell acetyltransferase without stimulating paf-acether synthesis.

Antigen stimulation of cultured bone-marrow-derived mast cells sensitized with specific monoclonal IgE induced cell degranulation and paf-acether (paf; 1-O-alkyl-2-acetyl-sn-glycero-3-phosphocholine) biosynthesis via the deacylation/acetylation (remodelling) pathway. Phorbol myristate acetate (PMA; 20-100 ng/ml) triggered only acetyltransferase activation, without concomitant lyso-paf (1-O-alkyl-sn-glycero-3-phosphocholine) and paf formation. A low concentration of PMA (5 ng/ml) potentiated antigen-induced degranulation, acetyltransferase activation and paf formation by about 30% but did not c

Serotonin-induced acute desensitization of serotonin2 receptors in human platelets via a mechanism involving protein kinase C.

Serotonin (5-HT)-induced changes in the levels of intracellular Ca++ were analyzed in human platelets, using the Ca+(+)-sensitive dye 1-(2-(5'-carboxyoxazol-2'-yl)-6-aminobenzofuran-5-oxy)-2-(2' -amino-5'- methylphenox)-ethane-N,N,N',N'-tetraacetic acid, pentaacetoxymethyl ester, to investigate the regulation of 5-HT2 receptor function. Serotonin mobilized intracellular Ca++ in a dose-dependent fashion from basal level of 98 +/- 2.7 and up to 211 +/- 5.8 nM with an EC50 value for 5-HT of 0.2 microM. Ketanserin, a 5-HT2 antagonist, reversed the 5-HT (10 microM)-induced Ca++ increase in a dose-d

Negative feedback mechanisms: evidence that desensitization of pineal alpha 1-adrenergic responses involves protein kinase-C.

alpha 1-Adrenergic stimulation of the pinealocyte translocates protein kinase-C, which, in turn, has an important positive effect on pineal cell function; translocation amplifies beta-adrenergic stimulation of both cAMP and cGMP. In the present report negative feedback effects of protein kinase-C are described, including inhibition of alpha 1-adrenergically induced increases in cytosolic Ca2+ ([Ca2+]i), phosphatidylinositol hydrolysis, and cGMP in beta-adrenergically stimulated cells. Time-course studies of cGMP and [Ca2+]i responses indicated that the onset of inhibition by the protein kinase

Triggering of neoplastic B cells via surface IgM and the cell surface antigens CD20 and CDw40. Responses differ from normal blood B cells and are restricted to certain morphologic subsets.

By raising monoclonal antibodies (MAbs) against B cells, a number of cell surface molecules have recently been identified which after binding by their specific antibody can trigger B cells, either alone or in co-operation with antibodies to surface immunoglobulin (sIg). The anti-CD20 (Bp35) MAb IF5 can deliver a strong activation signal to resting normal B cells, and the anti-CDw40 (Bp50) MAb G28-5 can promote activated G1 B cells to enter S phase. These antibodies were tested for their functional effects in vitro on suspended cells from 17 follicle-center-cell (FCC) lymphomas, 5 cases of chro

Sex-specific effects of 17-β-estradiol and bisphenol A on neutrophil function and phenotype - does centrifugation matter?

BACKGROUND: Neutrophils (polymorphonuclear leukocytes, PMNs) have long been underestimated in the context of autoimmunity. To elucidate their role from a sex-specific perspective, an immunoendocrinological framework offers a compelling investigative approach. We hypothesize that the sex hormone 17-β-estradiol (E2) and the endocrine-disrupting chemical bisphenol A (BPA) modulate PMN functions in a sex- and concentration-dependent manner. In addition, we consider the potential confounding effects of centrifugation protocols employed during PMN isolation. METHODS: PMNs from healthy women and men

Major Traumatic and Severe Thermal Injuries Lead to Immediate and Persistent Elevations in Circulating Concentrations of Resistin That Are Associated with Poor Clinical Outcomes and Impaired Innate Immune Responses.

Major trauma induces innate immune suppression, yet the underlying mechanisms are poorly understood. Resistin is an immunosuppressive molecule that is systemically elevated post-injury. However, its role in trauma-induced immune dysfunction and clinical outcomes is poorly defined. Here, we acquired blood samples from 147 adult trauma patients (≤1, 4-12, 48-72 h post-injury) and 95 burns patients (days 1, 3, 7, 14, 28 post-burn). We measured plasma resistin concentrations, studied resistin gene expression in peripheral blood mononuclear cells (PBMCs) and neutrophils, and measured resistin produ

METTL3 promotes neutrophil extracellular trap formation via SYK/ERK/MEK signaling during acute lung injury.

INTRODUCTION: Acute lung injury (ALI) is a life-threatening condition characterized by overwhelming pulmonary inflammation and neutrophil infiltration. Neutrophils play a central role in ALI pathogenesis by releasing proteolytic enzymes, reactive oxygen species, and neutrophil extracellular traps (NETs). Although NETs contribute to pathogen clearance, excessive NET formation exacerbates tissue injury and amplifies inflammation. N6-methyladenosine (m6A) is the most abundant internal modification of eukaryotic mRNA and plays a pivotal role in immune cell regulation by modulating RNA stability an

Impact of rifampicin on P-glycoprotein (ABCB1) expression in M1 and M2 macrophages derived from the THP-1 monocytic cell line or peripheral blood mononuclear cells.

Rifampicin induces ABCB1 (coding for P-glycoprotein; P-gp) in enterocytes or hepatocytes, lowering drug levels and efficacy. However, there is no induction data for macrophages, the primary site of rifampicin's anti-tuberculosis action. THP-1 cells were differentiated (200 nM phorbol 12-myristate-13-acetate; 72 h) and polarized to M1 macrophages (50 ng/mL lipopolysaccharide (LPS), 20 ng/mL interferon-gamma (IFNγ); 48 h) or M2 macrophages (20 ng/mL interleukin 4 (IL-4) and interleukin 13 (IL-13) each; 48 h). Peripheral blood mononuclear cells (PBMC) from a healthy volunteer were differentiated

PC3/Tis21/BTG2 and BTG1 genes: regulators of the cell cycle and neurogenesis, as well as tumor suppressors in malignant brain tumors.

PC3/Tis21/BTG2 and BTG1, prototype members of the BTG/Tob family, are antiproliferative transcriptional cofactors discovered 35 years ago as genes induced by nerve growth factor and phorbol 12-myristate 13-acetate or associated with lymphocytic leukemia. They are today known to serve as developmental regulators in several tissues, including neural cells. Biological functions such as cell division, transcriptional control, DNA repair, and mRNA stability, have been linked to their protein products. We will focus in this review on the effects of PC3/Tis21/BTG2 and BTG1 on brain tumorigenesis and

Targeting iNOS and associated proteins in neutrophils: An in-silico and ex-vivo approach for anti-sepsis therapy.

BACKGROUND: Previous reports from the lab have demonstrated the role of inducible nitric oxide synthase (iNOS) and associated proteins in sepsis-induced neutrophilic hyperinflammation. This study aimed to identify FDA-approved drugs that inhibit iNOS and associated proteins to attenuate neutrophil-induced excessive inflammation. METHODS: An in-silico approach involving STRING and Cytoscape was used to map protein-protein interactions (PPI). The FDA-approved drugs that target these proteins were retrieved from DrugBank. Candidate drugs were identified from the DrugBank database using a target

Er Miao San and its main components phellodendrine and atractylenolide-I exert anti-rheumatoid arthritis effects by inhibiting PAD4 and thereby reducing the formation of NETs.

BACKGROUND: Er Miao San (EMS) is an important Chinese herbal formula for the treatment of damp-heat underflow syndrome, and has been clinically proven to be effective in the treatment of rheumatoid arthritis (RA). Previous studies have shown that EMS can regulate the function of T cells and dendritic cells, affect the polarisation of macrophages, and inhibit the abnormal activation and angiogenesis of fibroblast-like synoviocytes (FLSs). However, it is unclear whether the inhibitory effect of EMS on RA is related to neutrophil dysfunction. PURPOSE: In this study, we investigated whether EMS a

Bioinformatics and system biology approach to discover the common pathogenetic processes between COVID-19 and chronic hepatitis B.

INTRODUCTION: The ongoing coronavirus disease 2019 (COVID-19) pandemic, caused by the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), presents a significant global public health threat. Concurrently, hepatitis B virus (HBV) remains a significant public health challenge. While previous studies have indicated an association between COVID-19 and chronic hepatitis B, the common underlying pathogenesis of these diseases remains incompletely understood. METHODS: To investigate the shared molecular mechanisms between chronic HBV infection and COVID-19, a comprehensive investigation was

Anti-inflammatory control of human skin keratinocytes by targeting nuclear transport checkpoint.

BACKGROUND: In the two common inflammatory skin diseases, Atopic Dermatitis (AD) and Psoriasis (Ps), keratinocytes (KCs) respond to immune insults through activation of proinflammatory transcription factors (TFs) and their translocation to the cell's nucleus. Therein, the TFs induce expression of genes encoding mediators of skin inflammation. The Nuclear Transport Checkpoint Inhibitors (NTCIs) were developed to regulate nuclear translocation of activated TFs, the essential step of inflammatory response. This new class of cell-penetrating peptide therapeutics controls inflammation caused by all

Nicotine regulates abnormal macrophage polarization and trophoblast invasion associated with preterm labor via the α7nAChR/SIRT1 axis.

INTRODUCTION: Preterm birth (PTB) frequently results from the syndrome of preterm labor (PTL). PTL is linked to an atypical maternal inflammatory response, as well as intrauterine inflammation and/or infection. In this study, we explored the mechanisms involved in nicotine-mediated abnormal macrophage polarization and trophoblast invasion associated with PTL. METHODS: First, THP-1-M0 macrophages were generated by treating the human monocytic leukemia cell line (THP-1) with phorbol 12-myristate 13-acetate for a duration of 24 h. Afterward, nicotine treatment was administered, followed by cocul

Photobiomodulation reduces neuropathic pain after spinal cord injury by downregulating CXCL10 expression.

BACKGROUND: Many studies have recently highlighted the role of photobiomodulation (PBM) in neuropathic pain (NP) relief after spinal cord injury (SCI), suggesting that it may be an effective way to relieve NP after SCI. However, the underlying mechanisms remain unclear. This study aimed to determine the potential mechanisms of PBM in NP relief after SCI. METHODS: We performed systematic observations and investigated the mechanism of PBM intervention in NP in rats after SCI. Using transcriptome sequencing, we screened CXCL10 as a possible target molecule for PBM intervention and validated the

Ca2+-regulated expression of high affinity methylaminoisobutryic acid transport in hippocampal neurons inhibited by riluzole and novel neuroprotective aminothiazoles.

High affinity methylaminoisobutyric acid(MeAIB)/glutamine(Gln) transport activity regulated by neuronal firing occurs at the plasma membrane in mature rat hippocampal neuron-enriched cultures. Spontaneous Ca2+-regulated transport activity was similarly inhibited by riluzole, a benzothiazole anticonvulsant agent, and by novel naphthalenyl substituted aminothiazole derivatives such as SKA-378. Here, we report that spontaneous transport activity is stimulated by 4-aminopyridine (4-AP) and that phorbol-myristate acetate (PMA) increases high K+ stimulated transport activity that is inhibited by sta
