Tau-targeted treatment strategies in Alzheimer's disease.

With populations ageing worldwide, the need for treating and preventing diseases associated with high age is pertinent. Alzheimer's disease (AD) is reaching epidemic proportions, yet the currently available therapies are limited to a symptomatic relief, without halting the degenerative process that characterizes the AD brain. As in AD cholinergic neurons are lost at high numbers, the initial strategies were limited to the development of acetylcholinesterase inhibitors, and more recently the NMDA receptor antagonist memantine, in counteracting excitotoxicity. With the identification of the prot…

Effect of amyloid peptides on the increase in TrkA receptor expression induced by nicotine in vitro and in vivo.

The ability of nicotine to induce a cytoprotective or neuroprotective action occurs through several downstream mechanisms. One possibility i … ions of Abeta1-42 can prevent nicotine-induced TrkA receptor expression in brain regions containing cholinergic neurons susceptible to the neurotoxicity associated with Alzheimer's disease.

Behavioral effects of anti-muscarinic, anti-serotonergic, and anti-NMDA treatments: hippocampal and neocortical slow wave electrophysiology predict the effects on grooming in the rat.

Previous research has shown that hippocampal and neocortical activation accompanies the postural changes occurring during self-grooming in rats but is absent or reduced during the stereotyped components of grooming, including head-washing and licking or biting. Since electrocortical activation is dependent on ascending cholinergic and serotonergic projections, we hypothesized that central muscarinic and serotonergic blockade would disrupt grooming by degrading cerebral control of changes in posture. Consistent with this, we find that systemic injections of scopolamine: (a) markedly reduce the

Evaluation of human astrocytoma and glioblastoma cell lines for nerve growth factor release.

Nerve growth factor (NGF) prevents degeneration of cholinergic neurons in the central nervous system (CNS), and has potential as a therapeutic treatment for Alzheimer's disease. The inability of NGF to cross the blood-brain barrier has prompted pharmacological approaches investigating peripherally administered compounds that stimulate release of endogenous NGF. This study describes the NGF-releasing properties of six human astrocytoma and glioblastoma cell lines (SW 1088, SW 1783 and CRL 1718 astrocytomas, and U-138, U-373, and T98G glioblastomas). Using a highly specific two-site ELISA for hu…

Involvement of brain catecholamines and acetylcholine in growth hormone hypersecretory states. Pathophysiological, diagnostic and therapeutic implications.

Secretion of growth hormone (GH) is excessive in acromegaly, but also in a number of other pathological states such as anorexia nervosa, insulin-dependent diabetes mellitus (IDDM), liver cirrhosis, depression, renal failure and GH-insensitivity syndrome. Abnormalities in the neuroendocrine control of GH secretion and/or a state of insensitivity to GH contribute to hypersecretion of GH in these states, with the possible exception of acromegaly, which appears to be a primary pituitary disease. GH hypersecretion may also occur in neonates or adolescents with tall stature, thus reflecting particul

Contralateral sound suppresses distortion product otoacoustic emissions through cholinergic mechanisms.

Presentation of an acoustic signal to one ear can suppress sound-evoked activity recorded at the opposite ear. The suppression appears to be mediated by medial olivocochlear (MOC) efferent neurons synapsing with outer hair cells (OHCs) and acting through the MOC neurotransmitter, acetylcholine (ACh). The purpose of the present investigation was to study the suppression of distortion product otoacoustic emissions (DPOAEs) by contralateral sound and to examine whether the suppression could be blocked by known antagonists of olivocochlear (OC) efferent activity. Urethane-anesthetized guinea pigs

Pharmacological treatment strategies in Alzheimer type dementia.

Substitution of cholinergic drugs in the treatment of Alzheimer type dementia (AD/SDAT) has hitherto not been very successful. Accumulating data indicate that monoamines are disturbed in AD/SDAT patients. The selective 5-hydroxytryptamine re-uptake blocker, Citalopram, has proven effective in reducing overactivity in the hypothalamic-pituitary-adrenal axis and in improving emotional disturbances in patients with dementia. Efforts to substitute the dopaminergic system have hitherto been negative. Neither has treatment with neuropeptides made any breakthrough in the treatment of demented patient

Another convulsant barbiturate, S(+)-1-methyl-5-phenyl-5-propyl barbituric acid [S(+)-MPP …

[Role of leptin receptor in the dorsal vagal complex in the inhibition of food intake in obese mice by electroacupuncture at "Zusanli" (ST36)].

OBJECTIVES: To observe the role of leptin receptor (LepR) in the dorsal vagal complex (DVC) in electroacupuncture(EA) -mediated regulation of food intake, so as to explore its central mechanism underlying improvement of metabolic disorders. METHODS: Ten C57BLKS/JGpt mice were randomly selected as the normal group, and the remaining mice were fed with high-fat diet (HFD) to establish a obesity model. Successfully modeled mice were randomly divided into the HFD group and EA group, with 10 mice in each group. The EA group received EA stimulation (2 mA, 2 Hz /15 Hz) at bilateral "Zusanli" (ST36)

Impaired dynein function preserves spinal interneuron survival and positioning in an ALS-like mouse model.

Impaired cytoplasmic dynein function has been implicated in amyotrophic lateral sclerosis (ALS) pathogenesis, yet the contributions of spinal interneurons to disease phenotypes remain unclear. We tested the hypothesis that hypomorphic dynein function in cholinergic neurons disrupts the development, survival, or positioning of inhibitory interneuron populations in the lumbar spinal cord. Using ChAT-Cre recombination, we generated four mouse genotypes with graded reductions in dynein activity in ChAT+ cells: Dync1h1+/+ (wildtype), Dync1h1-/+ (hemizygous wildtype), Dync1h1+/Loa (heterozygous Loa …

The first 25 years of the Northwestern University SuperAging Program.

During late life, "average" does not mean "intact." For example, cross-sectional data from a common word list learning test show that average delayed word recall raw score at age 80 (5/15) is approximately half that at age 56 to 66 (9/15). Cognitive and neurobiological dissolution is therefore implicitly incorporated into concepts of the aging brain. This position is being challenged through investigations on "superaging," a term that was coined at the Northwestern Alzheimer's Disease Research Center (ADRC) to define persons ≥ 80 years with delayed word recall raw scores at least equal to thos

M1 cholinergic signaling in the brain modulates cytokine levels and splenic cell sub-phenotypes following cecal ligation and puncture.

BACKGROUND: The contribution of the central nervous system to sepsis pathobiology is incompletely understood. In previous studies, administration of endotoxin to mice decreased activity of the vagus anti-inflammatory reflex. Treatment with the centrally-acting M1 muscarinic acetylcholine (ACh) receptor (M1AChR) attenuated this endotoxin-mediated change. We hypothesize that decreased M1AChR-mediated activity contributes to inflammation following cecal ligation and puncture (CLP), a mouse model of sepsis. METHODS: In male C57Bl/6 mice, we quantified basal forebrain cholinergic activity (immunos

Self-organizing human heart assembloids with autologous and developmentally relevant cardiac neural crest-derived tissues.

UNLABELLED: Neural crest cells (NCCs) are a multipotent embryonic cell population of ectodermal origin that extensively migrate during early … ories known to occur in the human heart. They demonstrated extensive migration, differentiated into cholinergic neurons capable of generating nerve impulses, and formed mature glial cells. Additionally, they contributed to the mesenchymal populations of the developing outflow tract. Through transcriptomic analysis, we revealed that NCCs acquire molecular features of …

Targeting oxidative stress and neurodegeneration: the role of Putranjiva roxburghii in Alzheimer's.

AD is a complex neurodegenerative disease that leads to progressive memory loss, worsening cognitive abilities, and synaptic dysfunction. The pathophysiology of the disease is driven by oxidative stress and neuroinflammation, which cause neuronal damage and may facilitate amyloid-beta aggregation and tau hyperphosphorylation. Current treatment strategies provide symptomatic improvement but do not address the multifactorial causes of the disease; therefore, multi-targeted approaches are critical. Putranjiva roxburghii is a medicinal plant with a wealth of ethnomedicinal literature from India. I
