[Science and law in assisted reproductive medicine -- two sides of the same token].

Polycystic kidney diseases (autosomal dominant and autosomal recessive) are progressive renal tubular cystic diseases, which are characterised by cyst expansion and loss of normal kidney structure and function. Autosomal dominant polycystic kidney disease (ADPKD) is the most common life- threatening, hereditary disease. ADPKD is more prevalent than Huntington's disease, haemophilia, sickle cell disease, cystic fibrosis, myotonic dystrophy and Down's syndrome combined. Early diagnosis and treatment of hypertension with inhibitors of the renin-angiotensin-aldosterone system (RAAS) and its potent

Promoting effect of beta-mercaptoethanol on in vitro development under oxidative stress and cystine uptake of bovine embryos.

The effects of beta-mercaptoethanol (beta-ME) on in vitro development under oxidative stress and cystine uptake of bovine embryos were investigated. Bovine 1-cell embryos obtained by in vitro fertilization were cultured in TCM-199 or synthetic oviductal fluid (SOF) in 20% O(2) supplemented with beta-ME. Addition of beta-ME significantly (P < 0.01) promoted embryo development when cultured in both TCM-199 and SOF under high levels of O(2), to almost the same rates when they were cultured in 5% O(2). To investigate whether the growth-promoting effect of beta-ME was related to cystine uptake, whi

An infectious basis for multiple sclerosis: perspectives on the role of Chlamydia pneumoniae and other agents.

The aetiology of multiple sclerosis (MS) remains unknown. Epidemiological, clinical and pathological data support the theory that MS is a complex disease/syndrome with many factors affecting its development and progression. It may be appropriate to regard MS as a syndrome with differing clinical and pathological features occurring along a spectrum. Patients with MS are more likely to have an affected relative than are individuals without MS, which suggests that there is a genetic component to this illness. Despite this genetic susceptibility, 85% of MS patients do not have an affected relative

Heat-induced release of CGRP from isolated rat skin and effects of bradykinin and the protein kinase C activator PMA.

In the skin, noxious heating induces an axon reflex response which is commonly accepted to be due to the release of vasodilatory neuropeptides from polymodal nociceptors. In the present study, the quantitative assessment of calcitonin gene-related peptide (CGRP) release from rat skin serves as an integrative measure of primary afferent activation by noxious heat and the presumed sensitising action of bradykinin and an activator of protein kinase C (PKC). The isolated rat hairy skin of either hind paw was mounted on acrylic rods and exposed for 5 min periods to synthetic interstitial fluid of e

The role of newer macrolides in the treatment of community-acquired respiratory tract infection. A review of experimental and clinical data.

The macrolide class of antibiotics is well established and often recommended for use in the treatment of community-acquired respiratory tract infection (RTI). The newer agents clarithromycin and azithromycin are frequently prescribed as first- or second-line therapy, and have been considered as superior to erythromycin in microbiological activity and clinical efficacy. In-vitro data show that clarithromycin and azithromycin have good activity (MIC < or = 0.5 microg/ml) against certain RTI pathogens. However the activity of both compounds is intrinsically low against Haemophilus influenzae whil

A homologue of the axonally secreted protein axonin-1 is an integral membrane protein of nerve fiber tracts involved in neurite fasciculation.

Axonin-1 is a glycoprotein that is released from axons of cultured neurons (Stoeckli, E. T., P. F. Lemkin, T. B. Kuhn, M. A. Ruegg, M. Heller, and P. Sonderegger. 1989. Eur. J. Biochem. 180:249-258). It has recently been purified from the ocular vitreous fluid of the chicken embryo (Ruegg, M. A., E. T. Stoeckli, T. B. Kuhn, M. Heller, R. Zuellig, and P. Sonderegger. 1989. EMBO (Eur. Mol. Biol. Organ.) J. 8:55-63). Immunohistochemistry localized axonin-1 prevalently in developing nerve fiber tracts. The presence of anti-axonin-1 Fab fragments during axon growth in vitro resulted in antibody bin

Chemoreceptors and their reflexes with special reference to the fetus and newborn.

Such evidence as there is indicates that hypoxia excites peripheral chemoreceptors in the adult by causing a fall in the rate of oxidative phosphorylation and that this process is potentiated by a rise in PCO2 which most probably acts by altering intracellular pH. What is unclear is whether this occurs in Type I cells or in sensory fibres and whether either should be regarded as the primary receptor. There is also uncertainty about the role of noradrenaline and dopamine, which are stored and presumably secreted in the chemoreceptor complex: are they, respectively, excitatory and inhibitory tra

Pancreatic acinar cells: ionic dependence of the membrane potential and acetycholine-induced depolarization.

1. Intracellular recordings of membrane potentials have been made in vitro from the exocrine acinar cells of the mouse pancreas using glass micro-electrodes.2. The mean membrane potential of the acinar cells during superfusion with Krebs-Henseleit solution was -39.2 mV. Increasing [K](o) tenfold decreased the membrane potential by 28 mV when [K](o) was above 10 mM. This depolarization was not affected by atropine (1.4 x 10(-6)M). Strophanthin-G (10(-3)M) slowly depolarized the cells at about 10 mV hr(-1).3. Brief exposure to acetylcholine (ACh), 5.5 x 10(-5)M, or pancreozymin resulted in a sho

Ionomics powered single-cell mapping reveals targetable iron dysregulation in endometriosis associated macrophages.

BACKGROUND: Endometriosis affects approximately 10% of women of reproductive age worldwide. Although elemental components of refluxed menstrual blood have been implicated in disease progression, comprehensive ionomic profiling of the endometriotic microenvironment remains limited. Characterizing the localized elemental landscape may generate new hypotheses regarding disease mechanisms. METHODS: We employed inductively coupled plasma mass spectrometry (ICP-MS) to profile 63 elements in endometriotic cyst fluid. Single-cell RNA sequencing (scRNA-seq) was performed to explore cellular subpopulat

Ferroptotic memory of the peritoneal niche: Mechanisms, cross-disease and cardiovascular-reproductive parallels, and targetable vulnerabilities.

Menstrual retrograde hemorrhage deposits heme-rich, erythrocyte-laden fluid into the peritoneal cavity, creating a spatially concentrated and recurring source of labile Fe(II) that is chemically poised to catalyze Fenton chemistry and seed polyunsaturated-lipid peroxidation. Current human and experimental data document co-occurrence of peritoneal iron accumulation, oxidized phospholipids, platelet activation and neutrophil extracellular traps, but the causal chain that converts cyclical iron exposure into durable tissue reprogramming and lesion persistence remains unresolved. Integrative bioinformatics analysis identifies APOE as a candidate link between lipid dysregulation and macrophage activation in inborn errors of metabolism.

BACKGROUND: Inborn errors of metabolism (IEM) are a heterogeneous group of genetic disorders characterized by metabolic dysregulation and high mortality. Despite extensive genetic discoveries, the molecular mechanisms underlying severe disease progression remain incompletely understood. Increasing evidence suggests that host metabolic states, particularly lipid dysregulation, can profoundly influence immune cell function and may contribute to impaired responses against intracellular pathogens. METHODS: We integrated genomic data from four high-confidence databases (GWAS Catalog, CTD, Open Tar

Desflurane post-conditioning inhibits HMGB1 nucleocytoplasmic translocation via the SIRT1/HMGB1 axis to attenuate hepatic ischemia-reperfusion-induced acute lung injury in young rats.

Acute lung injury (ALI) is a common clinical complication in pediatric patients undergoing perioperative liver transplantation, and therapeutic approaches are limited. While desflurane exhibits organ-protective potential, its specific efficacy against hepatic ischemia-reperfusion (HIR)-induced ALI in the developing lung, as well as the underlying molecular mechanisms, remain poorly understood. This study aimed to investigate the protective effects of desflurane and delineate the regulatory pathways involved. 2 week-old Sprague-Dawley rats were subjected to 70% warm HIR to simulate pediatric li

HLA class I-naturally presented synovial tissue peptides are recognized by CD8+ T lymphocytes from rheumatoid arthritis patients.

INTRODUCTION: Rheumatoid arthritis (RA) is an autoimmune disease resulting from a response driven by self-reactive CD4+ T cells that recognize autoantigenic peptides presented by antigen-presenting cells (APCs). Recent evidence suggests that CD8+ T cells are also important players in this process. This study aims to define the immunopeptidome of HLA class I molecules from RA synovial tissue (ST)- APCs and synovial fluid (SF)-pulsed monocyte-derived dendritic cells (DCs), and to prove the suitability of this approach to identify peptides recognized by CD8+ T cells from RA patients. A previously uncharacterized R881S variant of transporter NBCe1 exhibits intracellular retention and virtually no plasma membrane expression.

Mutations in the Na+/[Formula: see text] cotransporter NBCe1 (SLC4A4) cause proximal renal tubular acidosis (pRTA) and extrarenal symptoms including glaucoma, band keratopathy, migraine, growth disorder, and abnormal tooth enamel. From the NCBI dbSNP database, we recently identified a previously uncharacterized single-nucleotide variant (SNV), R881S, in NBCe1, located in hydrophilic helix 4. R881S NBCe1-A showed intracellular retention in human embryonic kidney 293 (HEK293) cells, and its plasma membrane expression was profoundly reduced in polarized Madin-Darby carine kidney (MDCK) cells. Muscle Strength and Functional Performance as Predictors of Metabolic and Body Composition Improvement in Anorexia Nervosa.

Background/Objectives: Anorexia nervosa (AN) is a severe psychiatric disorder characterized by energy restriction and associated with profound metabolic and body composition alterations. The loss of Body Cell Mass (BCM) leads to impaired muscle strength and functional capacity. Traditional monitoring based on body mass index (BMI, kg/m2) and weight primarily captures quantitative recovery, failing to reflect early qualitative metabolic and functional restoration. This study evaluated the longitudinal associations between improvements in physical performance and metabolic and structural recover

Pedunculoside alleviates lipopolysaccharide-induced acute lung injury/acute respiratory distress syndrome by inhibiting NF-κB pathway.

Acute lung injury (ALI) is a severe respiratory disease characterized by various clinical manifestations, including intractable hypoxemia, alveolar hypertension, parenchymal edema, and progressive respiratory distress. If left untreated, ALI can progress into acute respiratory distress syndrome (ARDS). A promising therapeutic option for ALI is Pedunculoside (PE), a major bioactive flavonoid found in Butcher's broom (Ruscus aculeatus). PE has been shown to possess significant anti-inflammatory and antioxidant effects. This study aimed to investigate the therapeutic effects of PE on ALI in mice

Hypocretin/orexin peptide release occurs mostly extrasynaptically, is self-controlled, and specifically enhanced in female rats.

The hypothalamic hypocretinergic/orexinergic (Hcrt/Ox) system comprises two neuropeptides -Hcrt1/OxA and Hcrt2/OxB- and two receptors -Hcrt/OxR1 and Hcrt/OxR2- which perform multiple modulatory actions. Its neurotransmission mechanism remains poorly understood despite its malfunction entails narcolepsy and low cerebrospinal fluid (CSF)-Hcrt1/OxA levels is the most specific biomarker of the disease. This work examines: (1) synaptic and volume Hcrt/Ox transmission types; (2) Hcrt/Ox receptors involvement in Hcrt/Ox-peptide release/synthesis; and (3) Hcrt/Ox system sexual dimorphism. Axonal centr

Passive water exchange between multiple sites can explain why apparent exchange rate constants depend on ionic and osmotic conditions in gray matter.

Porous materials, such as biological tissue, often have heterogeneous microstructures where imbibed fluid experiences distinct environments on short timescales, but can exchange among different environments over long timescales. Nuclear magnetic resonance (NMR) methods such as diffusion exchange spectroscopy (DEXSY) can measure this exchange in water under steady-state and equilibrium conditions; however, modeling becomes more complex when more than two exchanging environments are involved. This complexity is particularly relevant in the central nervous system (CNS), where water diffusion and
