Diversity of Calliphoridae (Diptera) in Brazil's Tinguá Biological Reserve.

The Calliphoridae are flies of great ecological, medical and sanitary importance because they are decomposers of organic matter, mechanical vectors of pathogenic agents, and causers of myiasis. This paper attempts to ascertain the diversity of Calliphoridae in the Tinguá Biological Reserve and correlate meteorological data (temperature, relative humidity of the air, and precipitation) with the occurrence of these flies. The study was conducted at a site in the Tinguá Biological Reserve, located in the municipality of Nova Iguaçu, state of Rio de Janeiro, Brazil. Four traps were set up using ch

[Definition of appropriate temperature and storage conditions in the detection of Leishmania DNA with PCR in phlebotomine flies].

For epidemiological studies and control programs of leishmaniasis, taxonomic identification of the etiologic agent of the disease in the insect vector is of critical importance. The implementation of molecular techniques such as the polymerase chain reaction (PCR) has permitted great advances in the efficacy and sensitivity of parasite identification. Previously, these investigations involved labor-intensive dissections and required expert personnel. The present work evaluates the effects of storage methods of phlebotomine samples in the optimization of PCR identification of Leishmania. Female

Experimentally induced infection with bluetongue virus serotype 11 in cows.

The consequences of inoculation of bluetongue virus (BTV) serotype 11 into 16 susceptible cows either at the time of breeding or at specified stages of pregnancy were studied. The cows were free of BTV or epizootic hemorrhagic disease virus, and none had antibodies to BTV before virus inoculation. A group of 4 cows was mated naturally to a bull reported to shed BTV-11 (CO75B300 strain) in the semen. The bull was suspected of infecting cows at mating with BTV-11, which subsequently transplacentally infected the developing fetuses and induced persistently infected and congenitally malformed prog

Tomato secondary metabolites as natural regulators of Bemisia tabaci behavior and performance: current applicability and prospects.

The sweetpotato whitefly (Bemisia tabaci) is one of the most damaging insect pests of tomato (Solanum lycopersicum), often causing severe yield losses and transmitting viral pathogens such as Tomato Yellow Leaf Curl Virus. Although chemical insecticides are commonly used for whitefly management, the rapid development of insecticide resistance in B. tabaci poses a major challenge to sustainable control. In contrast, wild tomato genotypes exhibit superior resistance, primarily attributed to the production of secondary metabolites (SMs), such as terpenoids, phenolic compounds, and acylsugars. The

Bulk segregant mapping and transcriptome analyses reveal the molecular mechanisms of spinetoram resistance in Spodoptera frugiperda.

The evolution of resistance to insecticides poses a significant threat to pest management programs. Understanding the molecular mechanisms underlying insecticide resistance is essential to design sustainable pest control and resistance management programs. The fall armyworm, Spodoptera frugiperda, is an important insect pest of many crops and has a remarkable ability to evolve resistance to insecticides. In this study, we employed bulk segregant analysis (BSA) combined with DNA and RNA sequencing to characterize the molecular basis of spinetoram resistance in S. frugiperda. Analysis of genomic

A Novel Cuticular Protein-like Cpr21L Is Essential for Nymph Survival and Male Fecundity in the Brown Planthopper.

Cuticular proteins (CPs) are a large family and perform a variety of functions. However, the physiological roles of cuticle protein 21-like (Cpr21L) in the brown planthopper (Nilaparvata lugens, BPH), one of the most destructive insect pests of rice, are largely unclear. In this study, Cpr21L was revealed to be expressed in both BPH nymphs and adults, and the mRNA expression level was much higher in male adults than female adults. Spatially, the expression of Cpr21L in the testis was higher than in the ovary. The RNA interference (RNAi) of Cpr21L seriously decreased nymph survival, and no indi

Function analysis of CYP321A9 from Spodoptera frugiperda (Lepidoptera: Noctuidae) associated with emamectin benzoate, and a novel insecticide, cyproflanilide detoxification.

The fall armyworm, Spodoptera frugiperda, is an invasive agricultural pest that is a serious threat to agricultural production and global food security. Chemical control is the most effective method for preventing outbreaks of S. frugiperda. However, insecticide resistance often develops as a result of prolonged pesticide use, and the molecular mechanisms involved in insecticide resistance remain unclear. Insect cytochrome P450 monooxygenases play an important role in the detoxification of insecticides and insecticide resistance in Lepidoptera. In our study, the LC50 of a novel insecticide (cy

Rapid adaptation of Chironomus yoshimatsui to acetylcholinesterase inhibitors (pyraclofos and pirimicarb) in a multi-generation study.

We evaluated the real effects of pollutants through a multi-generation study. We tested whether short-term exposure (48 h) of successive (first and second) generations of Chironomus yoshimatsui neonates (<24-h-old) to two acetylcholinesterase inhibitor insecticides, pyraclofos, and pirimicarb, would change insecticide sensitivity and life-cycle parameters over four generations. Additionally, we tested whether acetylcholinesterase (AChE) activity levels would be associated with this sensitivity change. Sensitivities (48 h EC50 value, using immobility as the endpoint) in chironomids (<24-h-old)

Protein electrophoresis in cranes with presumed insect bite.

BACKGROUND: Serum protein electrophoresis (SPE) has emerged as a potentially valuable diagnostic tool in avian medicine; yet, there is limited information regarding SPE in cranes. Since 2000, 20 cases of unilateral periocular or facial soft tissue swelling, blepharitis, feather loss, and ocular or nasal discharge attributed to insect bite hypersensitivity were observed in cranes from a captive breeding center. SPE may be useful for evaluating these lesions. OBJECTIVE: The aim of this study was to characterize the inflammatory response in cranes with hypersensitivity reactions using SPE. METH

Wolbachia as a potential tool for suppressing filarial transmission.

There is currently a great deal of interest in Wolbachia because of their wide distribution in arthropods and filarial nematodes and their striking effects on the biology of their hosts, including a possible role in speciation. They manipulate the reproduction of arthropod hosts through various effects on their hosts' biology, particularly cytoplasmic incompatibility (CI), to increase the proportion of infected individuals in the population, often to the point of fixation. This ability of Wolbachia to sweep through host populations indicates several potential applications of Wolbachia in the c

Wireworms (Coleoptera: Elateridae) Use Root Volatiles and CO2 to Discriminate Among Host Plants.

In the absence of visual signals, subterranean arthropods rely on olfactory and tactile cues to navigate toward resources. Here, in a series of pairwise dual-choice bioassays, we investigated the Limonius californicus (Col., Elateridae) larva response to wheat, pea, and bean seedlings in sand-filled olfactometers. We then quantified volatile organic compounds (VOC) emitted from roots. Wireworm preference for beans compared to wheat was attributed to the higher CO2 emission. Wireworm preference for peas compared to wheat was attributed to the higher amounts (µg/hr) of hexanal emitted from pea r

RNAi-Mediated FoxO Silencing Inhibits Reproduction in Locusta migratoria.

FoxO is a downstream target gene of cellular nutrient and growth factors, oxidative stress responses, and insulin signaling pathways. It play a crucial role in insect growth, development, and reproduction. Locusta migratoria is a significant agricultural pest; therefore, the identification of novel control targets for its management is of significant importance. After injecting dsRNA to interfere with FoxO expression, we observed changes in the reproduction-related gene expression and ovary development through RT-qPCR and morphological observation. Simultaneously, the trehalose and glycogen co

Altered gene expression profile of Wolbachia pipientis wAlbB strain following transinfection from its native host Aedes albopictus to Aedes aegypti cells.

Wolbachia is an obligate intracellular bacterial symbiont prevalent among arthropods and nematodes. To survive and reproduce, Wolbachia interacts with and modifies host subcellular structures, while sensing and responding to changes within the cellular environment. In mutualistic associations, Wolbachia may provision the host with metabolites, or help to maintain the chemical homeostasis of the host cell. Some strains can rapidly invade insect populations by manipulating host reproductive biology, while also preventing viral replication, allowing their use in vector control of arthropod-borne

Integrated transcriptomic and metabolomic analysis reveals differential molecular mechanisms underlying resistance of native and invasive Phytolacca americana against Spodoptera litura herbivory.

BACKGROUND: In response to herbivory from insects, plants have evolved complex defense strategies. Alien plants often escape control by specialist insects in their introduced range. This release from herbivory pressure may lead to a reallocation of the defensive strategies of alien plants, thereby promoting their invasiveness. However, the molecular mechanisms underlying this resource reallocation process remain unclear. In the present study, we used Phytolacca americana as a model species, and combined transcriptomic and metabolomic analyses to investigate the molecular basis of its invasion

Lethal and Sublethal Effects of Three Novel Insecticides on Biological Parameters of Pink Bollworm, Pectinophora gossypiella (Lepidoptera: Gelechiidae).

Pink bollworm (Pectinophora gossypiella) is a serious pest of cotton that causes huge economic losses in major cotton-growing regions. The development of resistance to transgenic Cry1Ac and Cry2Ab in India has led to long-term reliance on insecticides, thus enables the larvae to experience sublethal effects due to its concealed feeding behavior. This study investigated the impact of sublethal concentrations (LC10 and LC30) of emamectin benzoate, spinetoram, and spinosad on P. gossypiella. The results showed that emamectin benzoate was more toxic to P. gossypiella (LC50-2.12 mgL-1) than spineto

Brain and antennal transcriptome changes are associated with a colonization-mediated behavioural switch in an environmentally transmitted symbiosis.

Many organisms engage in symbiosis with microbial partners that are not passed on directly to offspring but are acquired from the environment each generation. Hosts can minimize the risk of relying on such horizontal transmission of beneficial symbionts by engaging in specialized acquisition behaviours. Squash bugs, Anasa tristis, locate their Caballeronia symbiont through olfactory cues released from adult frass. After ingesting the symbiont, the nymph's behaviour shifts from symbiont searching to host-plant feeding. To begin elucidating the underlying changes behind this behavioural switch,
