Cytomegalovirus shedding in seropositive healthy women of reproductive age in Tianjin, China.

Cytomegalovirus (CMV) enters latency after primary infection and can reactivate periodically with virus excreted in body fluids which can be called shedding. CMV shedding during the early stage of pregnancy is associated with adverse pregnancy outcome. The shedding pattern in healthy seropositive women who plan to have babies has not been well characterised. Vaginal swabs, urine and blood were collected from 1262 CMV IgG-positive women who intended to have babies and tested for CMV DNA by fluorogenic quantitative PCR method. Serum IgM was also detected. The association between sociodemographic

Genetic characterization of the HIV-1 reservoir after Vacc-4x and romidepsin therapy in HIV-1-infected individuals.

OBJECTIVE: Therapeutic HIV-1 immunization followed by latency reversal has been suggested as a strategy to eradicate HIV-1. Here we investigate the phylogenetic composition of the HIV-1 regions targeted by the therapeutic HIV-1 peptide vaccine Vacc-4x in participants in a clinical trial. DESIGN: Seventeen participants on suppressive antiretroviral therapy were vaccinated with six doses of Vacc-4x followed by three doses of romidepsin. Seven study participants were selected for sequencing analysis. All participants underwent an analytical treatment interruption. METHODS: Single-genome/provira

Current and future effects of varicella and herpes zoster vaccination in Germany - Insights from a mathematical model in a country with universal varicella vaccination.

Varicella zoster virus (VZV) is primarily known for causing varicella in childhood, but can reactivate again as herpes zoster (HZ) after a period of latency, mainly in persons older than 50 years. Universal varicella vaccination was introduced in Germany in 2004, while HZ vaccination has not been recommended yet. We aimed to quantify the potential long-term effects of universal childhood varicella vaccination and HZ vaccination of the elderly on varicella and HZ incidence in Germany over a time horizon of 100 years, using a transmission model calibrated to pre-vaccination data and validated ag

High-risk human papillomavirus reactivation in human immunodeficiency virus-infected women: risk factors for cervical viral shedding.

OBJECTIVE: To evaluate the presence of and estimate risk factors for reactivation of latent high-risk human papillomavirus (HPV) cervical infection in human immunodeficiency virus (HIV)-infected and HIV-uninfected women. METHODS: Data from 898 women in the HIV Epidemiology Research Study (HERS) were used to evaluate cervical HPV latency and reactivation. Prior exposure to HPV types (16, 18, 31, 35, and 45) was determined by serologic testing at enrollment, and cervical shedding of HPV was detected by polymerase chain reaction at 6-month intervals. Human papillomavirus cervical shedding and se

DNA damage in brain cells and behavioral deficits in mice after treatment with high doses of amantadine.

Amantadine (AMA) is an uncompetitive antagonist of the N-methyl-d-aspartate receptor, with clinical application, acting on treatment of influenza A virus and Parkinson's disease. It has been proposed that AMA can indirectly modulate dopaminergic transmission. In high doses, the central nervous system is its primary site of toxicity. To examine deleterious effects on CNS induced by AMA, this study evaluated possible neurobehavioral alterations induced by AMA such as stereotyped behavior, the effects on locomotion and memory and its possible genotoxic/mutagenic activities. Adult male CF-1 mice w

Post-infectious acute cerebellar ataxia in children.

Acute cerebellar ataxia is a relatively common neurologic disorder among children. Our aim was to characterize the clinical picture, etiology, and prognosis of acute cerebellar ataxia. The medical records of all children with a diagnosis of acute cerebellar ataxia hospitalized in our center and Hasharon Medical Center from 1990 to 2001 were reviewed. The diagnosis of acute cerebellar ataxia was based on the following criteria: acute onset of ataxia with or without nystagmus; absence of known genetic predisposing factors, such as familial degenerative disorders; and absence of drug intoxication

Latent reservoirs in memory T cell subsets are linked to poor immune recovery in people living with HIV.

Highly active antiretroviral therapy (HAART) suppresses viral loads in 71% of people living with HIV globally, while failing to bring adequate immune reconstitution in nearly one-third of them. We hypothesize that the persistence of latent HIV reservoirs in specific memory T cell subsets contributes to impaired immune recovery. We conducted a case-control study to estimate differences in the HIV-1 proviral DNA across memory CD4-positive T cell subsets between participants with CD4 counts of over 500 cells/µL (immune responders or IRs) and those with counts of less than 350 cells/µL (immune non

A Single Protein S-acyl Transferase Acts through Diverse Substrates to Determine Cryptococcal Morphology, Stress Tolerance, and Pathogenic Outcome.

Cryptococcus neoformans is an opportunistic yeast that kills over 625,000 people yearly through lethal meningitis. Host phagocytes serve as the first line of defense against this pathogen, but fungal engulfment and subsequent intracellular proliferation also correlate with poor patient outcome. Defining the interactions of this facultative intracellular pathogen with host phagocytes is key to understanding the latter's opposing roles in infection and how they contribute to fungal latency, dissemination, and virulence. We used high-content imaging and a human monocytic cell line to screen 1,201

[Molecular mechanisms of Epstein-Barr virus-mediated carcinogeneis].

Epstein-Barr virus (EBV), a ubiquitous human double stranded DNA virus, is associated with a variety of malignancies including Burkitt's lymphoma, Hodgkin's lymphoma, nasopharyngeal carcinoma (NPC) and gastric carcinoma (GC). These EBV-associated cancers are characterized by the proliferation of monoclonal EBV-infected cells, and viral gene expression in these cells is limited to a subset of latent genes, indicating that EBV latent genes contribute to carcinogenesis. Here I describe the mechanisms of carcinogenesis by EBV, focusing on the function of two EBV latent gens, latent membrane protei

IFI16 Mediates Deacetylation of KSHV Chromatin via Interaction with NuRD and Sin3A Co-Repressor Complexes.

IFI16 is a well-characterized nuclear innate immune DNA sensor that detects foreign dsDNA, including herpesviral genomes, to activate the inflammasome and interferon pathways. Beyond immune signaling, IFI16 also functions as an antiviral restriction factor, promoting the silencing of invading viral genes through transcriptional and epigenetic mechanisms. We recently demonstrated another role of IFI16, in which it interacts with and recruits the class I histone deacetylases, HDAC1 and 2, to the KSHV latency protein LANA, modulating its acetylation and function. In this study, we asked whether t

Human herpesvirus types 6 and 7 infection in pediatric hematopoietic stem cell transplant recipients.

BACKGROUND: The immunosuppressed state in patients undergoing hematopoietic stem cell transplantation (HSCT) predisposes them to activation of latent viral infections such as herpesvirus types 6 and 7 (HHV6 and HHV7). We aimed to evaluate the incidence, manifestations, outcomes, and risk factors of HHV 6 and HHV 7 infections after pediatric hematopoietic stem cell transplantation. MATERIAL AND METHODS: We performed 106 HSCTs (86 allogeneic and 20 autologous) on 94 children (56 boys, median age 7.1 years) during 2006-2012. Patients suspected to have HHV6 or HHV7 infections were tested using po

Systems Pharmacology of Human Oncoviruses: Network-Based Drug Discovery for Virus-Driven Cancers.

Human oncoviruses, including high-risk HPV, EBV, HBV, HCV, MCPyV, KSHV, and HTLV-1, account for approximately 15%-20% of global human malignancies. Rather than acting through isolated pathways, viral oncoproteins systematically rewire host regulatory networks, rendering conventional single-target oncology paradigms vulnerable to rapid resistance and clinical failure. Systems pharmacology offers a transformative, network-centric framework that views virus-driven cancers as systemic perturbations of host protein-protein interaction networks. This review synthesises computational methodology, net

SIRT2 deacylase modulators control B cell metabolic reprogramming in EBV infection and mitogenic activation.

Sirtuin-2 (SIRT2) is a cellular deacylase, regulating cell cycle progression and metabolic homeostasis. Recently, SIRT2 has emerged as a target with both anticancer and antiviral potential. However, the role and targetability of SIRT2 in viral-driven cancers remains unexplored. Epstein-Barr virus (EBV) is a ubiquitous herpesvirus with oncogenic potential that establishes latency in B lymphocytes and is typically controlled by a robust T cell immune response. In settings that compromise this response, such as immune suppression following transplant, EBV can cause B cell lymphomas. With broad im

The Complex Interplay of Malaria and EBV in Burkitt Lymphoma.

Burkitt lymphoma (BL) is an aggressive B-cell lymphoma endemic in children in regions of sub-Saharan Africa, where its incidence geographically overlaps holoendemic Plasmodium falciparum malaria and poorly controlled childhood Epstein-Barr virus (EBV) infection. Despite decades of research, the precise mechanistic synergy between these two pathogens remains incompletely defined. This review synthesizes current epidemiological, immunological, and molecular evidence to propose an integrated model for the etiology of endemic BL. We outline a paradoxical, dual-edged relationship wherein EBV infect

Multiancestry Genome-Wide Association Study Identifies Variants in a Zinc Finger Gene Cluster as Novel Regulators of HIV Control.

BACKGROUND: Human immunodeficiency virus 1 (HIV-1) remains a global health challenge, emphasizing the need to identify new host factors that influence pathogenesis to serve as drug targets. Identifying genetic determinants of HIV-1 control across diverse populations remains a top priority. METHODS: We conducted a multiancestry genome-wide association study (GWAS) of HIV-1 set-point viral load (spVL) leveraging genotype and viral load data from 10 723 individuals with HIV-1. To prioritize candidate genes and mechanisms, we integrated GWAS findings with transcriptome-wide association study (TWA

In vivo effect of anti-PD-1 on HIV-specific CD8+T-cells in men with HIV.

One approach to achieve a cure for HIV infection or allow for people with HIV to safely stop antiretroviral therapy is to enhance HIV-specific immune function by reversing immune exhaustion. Immune checkpoint blockade with anti-PD1 can enhance virus-specific T-cell function ex vivo and in animal models but the effects of anti-PD1 on HIV-specific T-cells in vivo has not been explored. We examine samples from men with HIV and cancer on antiretroviral therapy who receive nivolumab (anti-PD1) every two weeks as part of a prospective clinical trial. We show that a single dose of anti-PD-1 increases

Avian HEV triggered egg decline: Viral-host, immune-neuroendocrine mechanisms in layers.

Avian hepatitis E virus (aHEV), a member of the Hepeviridae family, poses substantial threats to poultry health and productivity, particularly through its detrimental impact on egg production in laying hens. This review elucidates the multifaceted mechanisms underlying aHEV-induced egg production decline in laying hens, focusing on direct viral-host interactions, immune dysregulation, and neuroendocrine disruption. aHEV exhibits strong tropism for reproductive organs, with active replication in ovarian tissues causing follicular apoptosis, structural damage, and hormonal imbalance. Curcumin derivative C210 induces Epstein-Barr virus lytic cycle and inhibits virion production by disrupting Hsp90 function.

Lytic induction therapy was devised to selectively combat malignancies associated with Epstein-Barr virus (EBV) by triggering viral reactivation from latency. At present, the major challenges of lytic induction therapy are to maximize reactivating efficiencies and meanwhile minimize infectious virion production. C210, a novel curcumin derivative with potent Hsp90 inhibitory activity, was explored for EBV-reactivating and virion-producing effects in EBV-positive nasopharyngeal carcinoma (NPC) and gastric carcinoma (GC) cell lines. And the molecular mechanisms underlying these effects were deter
