Trimethoprim-sulfamethoxazole (TMP-SMX) prophylaxis in HIV-uninfected, exposed (HUE) children variably reduces clinical malaria burden despite antifolate resistance, but data regarding achieved serum levels and adherence are lacking. Serum samples from 70 HUE children aged 3-12 months from Rakai, Uganda, enrolled in an observational study were assayed for random SMX levels using a colorimetric assay. Adherence with TMP-SMX prophylaxis data (yes/no) was also collected. Of 148 visits with concurrent SMX levels available, 56% had self-reported adherence with TMP-SMX therapy. Among these 82 visits

Juvenile hormone (JH) controls many biological activities in insects, including development, metamorphosis, and reproduction. In the Aedes aegypti mosquito, a vector of dengue, yellow fever, chikungunya, and zika viruses, the metabolic tissue (the fat body, which is an analogue of the vertebrate liver) produces yolk proteins for developing oocytes. JH is important for the fat body to acquire competence for yolk protein production. However, the molecular mechanisms of how JH promotes mosquito reproduction are not completely understood. In this study we show that stimulation of the JH receptor m

We investigated pyrethroid resistance mechanisms in Tetranychus urticae strains from Greece. Combined bioassay, biochemical and synergistic data indicated that although P450 mono-oxygenase activities were associated with the trait, target site insensitivity was the major resistance component. A 3.3 kb cDNA fragment of the T. urticae para sodium channel gene encompassing segment 4 of domain II to segment 6 of domain IV was obtained by a degenerate PCR strategy. The T. urticae sequence showed highest identity (56%) to the scabies mite, Sarcoptes scabiei, and was phylogenetically classified withi

BACKGROUND: Malaria vector control with synthetic insecticides can cause resistance, environmental toxicity, and harm to non-target animal species. To address these issues, it is critical to investigate safe and environmentally friendly botanical extraction methods for mosquito control. This study investigates the effects of crude root solvent extracts from Impatiens rothii and Salvia officinalis on Anopheles stephensi eggs and larvae under controlled environments.
METHODS: Fresh roots of S. officinalis and I. rothii were collected separately, allowed to air dry, ground into a powder, and the

Dengue outbreaks have occurred in Burkina Faso since 2013, but the country experienced its first chikungunya outbreak in Pouytenga in 2023. Thus, a field investigation was undertaken to determine entomological risk factors associated with the chikungunya outbreak to inform decision-making and respond to the outbreak. Field entomological investigations were performed during the chikungunya outbreak in Pouytenga in September-October 2023. Adult mosquitoes were collected indoors and outdoors using Prokopack aspirators in selected households, while BG-sentinel traps were deployed in public places

Aedes aegypti, the main urban vector of dengue fever, represents a growing public health problem in Nouakchott, the capital of Mauritania. Identifying the factors influencing the distribution and productivity of its breeding sites is essential for the development of effective control strategies. From May 2023 to April 2024, physico-chemical characteristics were recorded and mosquito larvae were collected, using a standard dipping method, from 60 water collections each month during the dry season and twice a month during the rainy season, totaling 294 observations. The larval positivity of wate

Bed bugs have become resistant to traditional insecticides, and the demand for effective drugs to eradicate bed bugs on poultry farms (especially table-egg layer) and breeder farms (especially broiler breeders) is growing. Fluralaner has been shown to be effective in treating bed bug infestation in poultry. However, there is no product containing fluralaner labeled for use in chickens in the United States. In the present study, we prepared medicated drinking water using fluralaner dog chews (Bravecto®) and diethylene glycol monoethyl ether (Transcutol V). The present study aimed to determine t

The Aedes aegypti mosquito is generally associated with arboviruses that cause yellow fever, dengue, zika and chikungunya. The most efficient way to control their populations is through application in breeding sites of highly toxic insecticides that can also impact non-target organisms and generate resistant populations. Therefore, the use of compounds is desirable. Morin hydrate has broad pharmacological applications based on its antioxidant potential, in addition to not having negative effects on mammals. Therefore, the objective of the present study was to investigate the effects of morin h

BACKGROUND: Malaria transmission in Tanzania is driven by mosquitoes of the Anopheles gambiae complex and Anopheles funestus group. The latter includes An. funestus s.s., an anthropophilic vector, which is now strongly resistant to public health insecticides, and several sibling species, which remain largely understudied despite their potential as secondary vectors. This paper provides the initial results of a cross-country study of the species composition, distribution and malaria transmission potential of members of the Anopheles funestus group in Tanzania.
METHODS: Mosquitoes were collecte

Type 2 diabetes mellitus (T2DM) is usually depicted by relative insulin deficiency, raised blood glucose levels, and the predominant risk factor, insulin resistance. Hence, the development of insulin sensitizer drugs targeting PPAR-γ receptors has expanded enormous interest as an attractive choice for T2DM treatment. Thiazolidinediones (TZD) enhance insulin sensitivity either by directly functioning on gene transcription of the PPARγ receptor related to glucose homeostasis or by systemic sensitization of insulin and, therefore, improved hyperglycemia in a wide range of patients. However, sever

BACKGROUND: Aedes aegypti, the primary vector of dengue, chikungunya, and Zika viruses, poses a significant public health threat worldwide. Traditional control methods using insecticides are increasingly challenged by resistance and environmental concerns. The sterile insect technique (SIT) offers an eco-friendly alternative that has been successfully applied to other insect pests. This article aims to briefly review Ae. aegypti management in Cuba, highlighting the accomplishments, challenges, and future directions of the SIT.

East Coast fever (ECF) in cattle is caused by the protozoan parasite Theileria parva, transmitted by Rhipicephalus appendiculatus ticks. In cattle ECF is often fatal, causing annual losses >$500 million across its range. The African buffalo (Syncerus caffer) is the natural host for T. parva but the transmission dynamics between wild hosts and livestock are poorly understood. This study aimed to determine the prevalence of T. parva in cattle, in a 30 km zone adjacent to the Serengeti National Park, Tanzania where livestock and buffalo co-exist, and to ascertain how livestock keepers controlled

Light is known to excite photosensitizers (PS) to produce cytotoxic reactive oxygen species (ROS) in the presence of oxygen. This modality is attractive for designing control measures against animal diseases and pests. Many PS have a proven safety record. Also, the ROS cytotoxicity selects no resistant mutants, unlike other drugs and pesticides. Photodynamic therapy (PDT) refers to the use of PS as light activable tumoricides, microbicides and pesticides in medicine and agriculture. Here we describe "photodynamic vaccination" (PDV) that uses PDT-inactivation of parasites, i.e. Leishmania as who

Head louse infestations (pediculosis) are prevalent worldwide. In developed countries, the infestation rate of 4- to 13-year-old children remains high despite preventive efforts. This is due to the existence of numerous ineffective pediculicides, the incorrect use of the effective agents, toxicological concerns and the development of louse strains resistant to insecticides. One of the most effective tools for the prevention and control of lice is the louse comb, which should be used regularly for the detection of living lice at an early stage of infestation, and as an accessory to any treatmen

In Germany, the last period of the Second World War and the following years were characterized by deficiencies of hygiene which had not occurred previously in Middle Europe during the 20th century. There were focuses of typhus, typhoid fever, tuberculosis, diphtheria, scarlet fever, and meningitis. Insufficiencies in the removal of faeces caused high incidences of shigellosis, hepatitis A, and ascariasis. As a result of insufficient body care, many people were infested with fleas, lice and scabies. The migration of large proportions of the population resulted in an increasing prevalence of syp

BACKGROUND: Chlorfenapyr-based indoor residual spraying (IRS) has been prequalified for malaria control. Residual efficacy (mosquito mortality) of IRS applied to houses is typically assessed using forced-contact cone bioassays. However, chlorfenapyr is a pro-insecticide that requires mosquito metabolic activity for conversion to its toxic metabolite, tralopyril. Cone bioassays consistently underestimate mortality compared to free-flying bioassays. This study evaluated whether increasing mosquito activity during cone bioassays could better capture chlorfenapyr-induced mortality on mud surface u

The plant parasitic nematode, Aphelenchoides besseyi, is a serious pest causing severe damage to various crop plants and vegetables. The Bacillus thuringiensis (Bt) strains, GBAC46 and NMTD81, and the biological strain, FZB42, showed higher nematicidal activity against A. besseyi, by up to 88.80, 82.65, and 75.87%, respectively, in a 96-well plate experiment. We screened the whole genomes of the selected strains by protein-nucleic acid alignment. It was found that the Bt strain GBAC46 showed three novel crystal proteins, namely, Cry31Aa, Cry73Aa, and Cry40ORF, which likely provide for the safe
