Gardenia jasminoides Ellis polysaccharides improve gut microbiota disorder and glucose homeostasis in type 2 diabetic mice.

Type 2 diabetes mellitus (T2DM) is the most prevalent metabolic disorder. Gardenia jasminoides Ellis is a traditional herbal medicine used in metabolic disease treatment; however, the effects of Gardenia jasminoides Ellis polysaccharides (GPS) on T2DM remain unknown. This study aimed to investigate the antidiabetic effect of GPS in a T2DM mouse model induced by high-fat diet (HFD) and streptozotocin administration, and to elucidate the potential molecular mechanism. Treatment with low and high doses (200 and 400 mg/kg) of GPS significantly reduced the fasting blood glucose level of T2DM mice b

Linking the relationship between drug-induced osteoporosis and the gut microbiota.

Drug-induced osteoporosis is considered secondary osteoporosis caused by pharmacological drugs that can alter the diversity and function of the gut microbiota (GM), which in turn may be associated with the development or exacerbation of osteoporosis. This review uncovers the relationship between drug-induced osteoporosis and GM based on preclinical and clinical studies. In this context, we focused on secondary osteoporosis induced by glucocorticoids, aromatase inhibitors, proton pump inhibitors, antiretroviral drugs, antiepileptic drugs, antipsychotics, antidepressants, and subsequent alterati

Structure and protection of Cichorium glandulosum polysaccharides against sarcopenic obesity through activating mitophagy via butyrate-GPR43-AMPK pathway.

Sarcopenic obesity (SO) is a major complication of type 2 diabetes with limited therapeutic options. This study characterized CGP-A, a novel branched fructan (6.722 kDa) from Cichorium glandulosum. Its backbone consists of →1)-β-D-Fruf-(2→ and →6)-α-D-Glcp-(1→ residues, interspersed with →1,6)-β-D-Fruf-(2→ branching points. The side chains consist of terminal β-D-Fruf-(2→ units attached to the C-6 position of the fructofuranosyl residues in the backbone. In db/db mice, CGP-A dose-dependently ameliorated insulin resistance, hepatic steatosis, muscle loss and intestinal barrier dysfunction. Impo

Structural design and delivery performance of resistant starch-containing delivery systems: A comprehensive review from material fundamentals to colon-targeted applications.

Resistant starch (RS) is a dietary fiber that escapes small-intestinal digestion and reaches the colon, offering inherent colon-targeting potential, bioactive protection, and prebiotic benefits. However, native RS often shows limited mechanical strength and high hydrophilicity, driving the development of modified and composite delivery systems. This review summarizes the functional advantages of RS and evaluates representative RS-containing delivery systems according to carrier architecture and the functional role of RS, including particulate carriers, Pickering emulsions, hydrogels, nanoparti

Dates, Polyunsaturated Fatty Acids, Dietary Fiber, and Probiotics as Potential Complementary Modulators of Gut Microbiota and Oxidative Stress: An Integrative Review.

This integrative review examines the potential of dates, omega-3 (ω-3) and omega-6 (ω-6) polyunsaturated fatty acids (PUFAs), dietary fibers (DFs), and probiotic bacteria to support gut health through complementary nutritional pathways. The literature was searched in PubMed, Web of Science, Scopus, and Google Scholar for English-language studies published through June 2026. Dates contain fermentable substrates, including DFs and non-digestible oligosaccharides, that can serve as growth substrates for beneficial gut microbes. However, the whole fruit has not been formally established as a prebi

Novel oat β-glucan nanoparticles for diabetes-associated colitis therapy.

The comorbidity of Inflammatory Bowel Disease and Diabetes Mellitus presents a formidable clinical challenge, as these conditions engage in vicious cycles mediated by intestinal barrier dysfunction, microbiota dysbiosis, and chronic inflammatory stress. This complex pathophysiological interplay substantially limits the efficacy of conventional monotherapeutic approaches. Therefore, we developed an innovative nanotherapeutic platform (BG@EGCG-Mn) that harnesses the synergistic potential of natural bioactive compounds. This platform combines the complementary biological activities of oat β-gluca

Arabinoxylan ameliorates memory deficits and amyloid pathology in male 5 × FAD mice via modulation of gut microbiota structure.

Alzheimer's disease (AD), a prevalent neurodegenerative disorder, is primarily characterized by β-amyloid (Aβ) deposition. Current therapies alleviate symptoms but lack agents capable of modifying disease progression. Meanwhile, cross-regional studies indicate that AD patients exhibit disrupted gut microbiota composition, which is closely associated with cerebral molecular dysregulation. Building on this gut-brain connection, this study aimed to attenuate AD progression by targeting gut microbiota through microbially metabolized carbohydrates. Specifically, using 5 × FAD mice modeling AD patho

The impact of a superfood chocolate beverage on indices of peripheral vascular function.

Elevated blood lipids and reduced vascular health contribute to cardiovascular disease. Diets and/or interventions high in antioxidants, polyphenols, and anti-inflammatory properties mitigate this risk. This study hypothesized that daily consumption of a beverage high in antioxidants, polyphenols, and anti-inflammatory properties would improve vascular function, blood lipids, and arterial blood pressure. Vascular function was assessed as brachial artery flow mediated vasodilation (%FMD) and following 5 min of ischemia. Brachial artery blood pressure was assessed, and blood samples were collect

A Novel Phosphatase Reverses the Leloir Pathway to Promote Tagatose Synthesis from Glucose.

d-Tagatose is a natural, low-calorie, rare monosaccharide that has gained substantial interest as an alternative sweetener. It offers numerous benefits, including a low glycemic index, prebiotic properties, and the ability to lower blood sugar levels. Currently, tagatose is primarily produced industrially through the isomerization of galactose, utilizing both chemical and enzymatic catalysis. While these established processes can produce tagatose, they are inefficient and expensive. Recent works have demonstrated alternative biosynthetic routes to produce d-tagatose but suffer from low theoret

The Akkermansia muciniphila-tryptophan metabolism-aromatic hydrocarbon receptor axis mediates the protective effect of Schisandra chinensis pectin polysaccharide against colitis.

Current therapies for ulcerative colitis (UC) remain inadequate, necessitating novel treatment strategies. This study isolated SCPII-1, a homogeneous pectin polysaccharide (Mw: 278.153 kDa, DM: 57.21 %) from Schisandra chinensis, primarily composed of →4)-α-D-GalpA-(1 → units. In dextran sulfate sodium (DSS)-induced colitis mice, SCPII-1 alleviated symptoms, repaired mucosal ultrastructure (microvilli, mitochondria), and enhanced barrier integrity. Mechanistically, SCPII-1 ameliorated gut dysbiosis in UC mice, notably through the selective enrichment of the beneficial bacterium Akkermansia muc

Konjac mannan oligosaccharides as a sustainer of fasting-associated gut microbiome signature after discontinuation of intermittent fasting in overweight individuals: A protocol for an open-label randomized clinical trial.

BACKGROUND: Our previous studies show that the improvement in cardiometabolic traits by intermittent fasting is associated with remodeling the gut microbiome, with short-chain fatty acids (SCFA) producing bacteria being evident. These effects, however, are largely lost when intermittent fasting is discontinued. Konjac mannan oligosaccharides (KMOS) are a commercial nature-deprived prebiotic, which has been reported to increase the levels of intestinal SCFA-producing bacteria. However, the capacity of continued KMOS consumption to maintain intermittent fasting-provoked levels of SCFA-producing

The gut-bone axis in osteoporosis: a multifaceted interaction with implications for bone health.

With the accelerated aging of the population, degenerative orthopedic diseases, particularly osteoporosis, have become a major public health challenge, threatening bone health and affecting the quality of life. Existing anti-osteoporosis regimens remain rather unitary or poorly adhered, which also limits the maintenance of bone health to some extent. Given the increasingly elucidated prominence of gut-related factors in osteoblasts/osteoclasts and bone formation/metabolism/maintenance, focusing on intestinal microecology and then targeting the distal bone tissue via the gut-bone axis have been

NF-κB pathway activation by Octopus peptide hydrolysate ameliorates gut dysbiosis and enhances immune response in cyclophosphamide-induced mice.

Cyclophosphamide (CTX) is an anticancer medication that suppresses host immunity as well as adversely affects mucosal inflammation and gut microflora dysbiosis. The gut microflora is recognized as a substantial factor in host metabolism and immunological homeostasis. To improve immunity and inhibit cytotoxic and homeostatic imbalances triggered by CTX, it is essential to monitor immunoregulators. In this research, we assessed the impact of Octopus peptide hydrolysate (OPH) on immune modulation, intestinal integrity, and gut microbial composition in CTX-induced immune-deficient mice. The result

Treating chronic stress and chronic pain by manipulating gut microbiota with diet: can we kill two birds with one stone?

Background: Chronic stress and chronic pain are closely linked by the capacity to exacerbate each other, sharing common roots in the brain and in the gut. The strict intersection between these two neurological diseases makes important to have a therapeutic strategy aimed at preventing both to maintain mental health in patients. Diet is an modifiable lifestyle factor associated with gut-brain axis diseases and there is growing interest in its use as adjuvant to main therapies. Several evidence attest the impact of specific diets or nutrients on chronic stress-related disorders and pain with a g

Effects of a laminarin-rich algal extract on caecal microbiota composition, leukocyte counts, parasite specific immune responses and growth rate during Eimeria tenella infection of broiler chickens.

Coccidiosis, infection with protozoan parasites of genus Eimeria, is a major problem in poultry husbandry world-wide. The disease is currently managed by coccidiostats and live vaccines, but these approaches are not sustainable. Hence, it is important to identify new means to control the infection and/or ameliorate its detrimental effects on gut health. Laminarin, a β-glucan found in marine brown algae, has prebiotic and bioactive properties that could be beneficial in coccidiosis control. The present study aimed to examine the potential of laminarin as an immunostimulatory and microbiota-regu

Extraction and characterization of hepatoprotective polysaccharides from Anoectochilus roxburghii against CCl4-induced liver injury via regulating lipid metabolism and the gut microbiota.

Anoectochilus roxburghii polysaccharides exhibit notable hepatoprotective effects, but the underlying substance basis and mechanisms remain unknown. In this study, four new polysaccharides named ARP-1a, ARP-1b, ARP-2a and ARP-2b, were isolated from A. roxburghii. Their structural characteristics were systematically analyzed using HPGPC, HPLC, GC-MS, IR and NMR analysis. ARP-1a, the leading polysaccharide isolated from A. roxburghii, was further evaluated for its hepatoprotective effects on acute liver injury mice induced by CCl4. ARP-1a significantly reduced the serum ALT, AST, TNF-α, IL-1β an
