Developing Methods for Assessing Mental Activity Using Human-Smartphone Interactions: Comparative Analysis of Activity Levels and Phase Patterns in General Mental Activities, Working Mental Activities, and Physical Activities.

BACKGROUND: Human biological rhythms are commonly assessed through physical activity (PA) measurement, but mental activity may offer a more substantial reflection of human biological rhythms. OBJECTIVE: This study proposes a novel approach based on human-smartphone interaction to compute mental activity, encompassing general mental activity (GMA) and working mental activity (WMA). METHODS: A total of 24 health care professionals participated, wearing wrist actigraphy devices and using the "Staff Hours" app for more than 457 person-days, including 332 workdays and 125 nonworkdays. PA was meas

Machine learning analyses reveal circadian clock features predictive of anxiety among UK biobank participants.

Mood disorders, including depression and anxiety, affect almost one-fifth of the world's adult population and are becoming increasingly prevalent. Mutations in circadian clock genes have previously been associated with mood disorders both directly and indirectly through alterations in circadian phase, suggesting that the circadian clock influences multiple molecular pathways involved in mood. By targeting previously identified single nucleotide polymorphisms (SNPs) that have been implicated in anxiety and depressive disorders, we use a combination of statistical and machine learning techniques

The Evolution and Biology of SARS-CoV-2 Variants.

Our understanding of the still unfolding severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) pandemic would have been extremely limited without the study of the genetics and evolution of this new human coronavirus. Large-scale genome-sequencing efforts have provided close to real-time tracking of the global spread and diversification of SARS-CoV-2 since its entry into the human population in late 2019. These data have underpinned analysis of its origins, epidemiology, and adaptations to the human population: principally immune evasion and increasing transmissibility. SARS-CoV-2, despi

Comparative outcomes and prognosis in patients with ureteral upper tract urothelial carcinoma undergoing segmental ureterectomy versus radical nephroureterectomy: a multicenter cohort study.

BACKGROUND: Although radical nephroureterectomy (RNU) with bladder cuff excision remains the gold standard treatment for upper tract urothelial carcinoma (UTUC), segmental ureterectomy (SU) may offer a nephron-sparing alternative. Studies comparing oncologic outcomes and renal functional outcomes between SU and RNU have yielded controversial, often conflicting results. Furthermore, investigations specifically involving ureteral UTUC patients in northern China are scarce. METHODS: We retrospectively enrolled 546 ureteral urothelial carcinoma patients (282 SU and 264 RNU cases) from three hospi

Are Current Survival Prediction Tools Useful When Treating Subsequent Skeletal-related Events From Bone Metastases?

BACKGROUND: Bone metastasis in advanced cancer is challenging because of pain, functional issues, and reduced life expectancy. Treatment planning is complex, with consideration of factors such as location, symptoms, and prognosis. Prognostic models help guide treatment choices, with Skeletal Oncology Research Group machine-learning algorithms (SORG-MLAs) showing promise in predicting survival for initial spinal metastases and extremity metastases treated with surgery or radiotherapy. Improved therapies extend patient lifespans, increasing the risk of subsequent skeletal-related events (SREs).

[Development of a prediction model for levetiracetam plasma concentration in children with epilepsy based on pharmacokinetic characteristics: A comparison of multiple linear regression and complex algorithms].

OBJECTIVES: Levetiracetam (LEV) is mainly eliminated unchanged through the kidneys, and its steady-state serum concentration is associated with dose, body weight, renal function, and concomitant medications. However, marked concentration variability remains in children with epilepsy due to growth and developmental changes and interindividual pharmacokinetic differences. This study aims to compare the performance of multiple linear regression and complex machine learning algorithms in predicting steady-state trough concentrations of LEV in children with epilepsy, and evaluate the influence of i

A novel three-CpG signature for lung adenocarcinoma diagnosis: integrating multi-omics, machine learning, and clinical validation.

BACKGROUND: Lung adenocarcinoma (LUAD) is a leading cause of cancer-related mortality worldwide. The identification of non-invasive biomarkers for early detection is critical to improving patient outcomes. Integrative analysis of DNA methylation and gene expression represents a promising approach for identifying cancer-specific diagnostic markers. This study aimed to identify and validate a LUAD-specific DNA methylation signature for the early detection of LUAD. METHODS: An integrated analysis of DNA methylation data from the Illumina HumanMethylation450K array, including 455 tumor samples, 3

Optimising Pharmacovigilance Efficiency with MLIT (Machine Learning for Intelligent Triage): A Tool for Statistical Safety Alerts.

BACKGROUND AND AIM: Pharmacovigilance is essential to ensuring patient safety by enabling timely identification of adverse reactions in increasingly complex and voluminous data. Routine quantitative signal detection methods generate statistical alerts for product-event pairs based on predefined criteria; however, most alerts do not warrant further investigation, creating inefficiencies and significant time demands for pharmacovigilance teams. Manual triage of these alerts is often resource-intensive, prone to variability, and challenging to audit, highlighting the need for more reliable, trans

TyG-ABSI and TyG-WWI as insulin-resistance body shape markers of mortality in middle-aged and older adults within the cardiovascular-kidney-metabolic framework: A multi-cohort observational study with external survival-model validation.

Insulin resistance, adiposity, kidney dysfunction, and cardiovascular disease are closely interconnected in middle-aged and older adults and are central to the cardiovascular-kidney-metabolic (CKM) framework. Triglyceride-glucose (TyG)-related indices combine routine lipid, glucose, and anthropometric information, but their cross-population consistency and incremental prognostic value for mortality-risk assessment remain uncertain. We analyzed harmonized participant-level data from National Health and Nutrition Examination Survey (NHANES), English Longitudinal Study of Ageing (ELSA), China Hea

A Tangible Multimodal Sensing Interface for Collaborative Task Engagement in Children With Autism Spectrum Disorder: Cross-Sectional Mixed Methods Feasibility Study.

BACKGROUND: Social engagement is critical for the developmental progress of children with autism spectrum disorder (ASD). While tangible technologies and serious games (SGs) show promise, a dual-purpose platform that both facilitates collaborative interaction and enables sensing of collaborative task engagement is required. Existing tangible interfaces often lack integrated sensing capabilities, while devices designed for complex input are typically not optimized for the sensory and interaction needs of this population. OBJECTIVE: This study aims to present InteractiSense, a tangible prototyp

Quantifying the Predictive Power of Social Determinants of Health in Cardiovascular Disease and Type 2 Diabetes Progression Using XGBoost: Retrospective Cohort Study.

BACKGROUND: Cardiovascular diseases (CVDs) and type 2 diabetes (DM2) are influenced not only by biomedical risk factors but also by social determinants of health (SDOH). While the inclusion of SDOH in predictive models is increasingly advocated, few studies have quantified their specific contribution in a high-risk clinical cohort using robust statistical and machine-learning approaches. OBJECTIVE: This study aims to quantify the added predictive value of SDOH in predicting CVD or DM2 disease onset within 5 years, within 10 years, and at any time during follow-up among individuals already at

From a Multi-Omics Signature to a Therapeutic Candidate: Computational Prediction and Experimental Validation in Liver Fibrosis.

Background: Advanced liver fibrosis (LF) is a major determinant of prognosis across chronic liver diseases. Current biomarkers are often etiology-specific and lack cross-cohort robustness. Shared molecular drivers across etiologies remain incompletely defined, and effective anti-fibrotic therapies are limited. Methods: We developed a multi-algorithm consensus machine-learning framework to derive a robust LF progression signature. In the training non-alcoholic fatty liver disease (NAFLD) cohort GSE213621 (n = 368), samples were formulated as a binary classification task (mild fibrosis, F0-F2; a
