Clinical and Biological Features of Cutibacterium (Formerly Propionibacterium) avidum, an Underrecognized Microorganism.

The recent description of the genus Cutibacterium has altered the taxonomy of Propionibacterium species. These organisms still belong to the genera of the skin coryneform group, and the most-studied species remains Cutibacterium acnes. Cutibacterium avidum is also a known skin commensal. This underrecognized microorganism can, however, act as a pathogen after bacterial seeding and can be considered opportunistic, causing either superficial or deep/invasive infections. It can cause numerous infections, including but not limited to breast infections, skin abscesses, infective endocarditis, and d

Vibrio cholerae O1 epidemic variants in Angola: a retrospective study between 1992 and 2006.

Cholera is still a major public health concern in many African countries. In Angola, after a decade of absence, cholera reemerged in 1987, spreading throughout the country until 1996, with outbreaks recurring in a seasonal pattern. In 2006 Angola was hit by one of the most severe outbreaks of the last decade, with ca. 240,000 cases reported. We analyzed 21 clinical strains isolated between 1992 and 2006 from several provinces throughout the country: Benguela, Bengo, Luanda, Cuando Cubango, and Cabinda. We used two multiplex PCR assays to investigate discriminatory mobile genetic elements (MGE)

Microbial Characteristics and Genomic Analysis of an ST11 Carbapenem-Resistant Klebsiella pneumoniae Strain Carrying bla KPC-2 Conjugative Drug-Resistant Plasmid.

BACKGROUND: The sequence type 11 (ST11) carbapenem-resistant Klebsiella pneumoniae (CRKP) carrying bla KPC-2 has been widespread all over the world, and it has been reported frequently in China. The bla KPC-2 located on the mobile genetic element brings tremendous pressure to control the spread and outbreak of resistant bacteria. Whole-genome sequencing (WGS) technology can comprehensively and in-depth display the molecular characteristics of drug-resistant bacteria, providing a basis for evaluating the genetic diversity within the CRKP genome. METHODS: The ST11 CRKP in this study was collect

New perspectives on bacterial chlorine resistance: Phages encoding chlorine resistance genes improve bacterial adaptation.

Bacterial resistance to chlorine disinfectant reduces its effectiveness in killing pathogenic bacteria and poses a severe threat to environmental and health safety. The interaction between bacteria and phages is the most frequent biological activity in Earth's biosphere, but little is known about what role and mechanism phages play in the resistance of bacterial communities to chlorine disinfectants. Here, we investigated the changes in the abundance, activity and function of the bacterial-phage community under the effect of chlorine disinfectants in a 92-day running anaerobic-anoxic-oxic syst

Molecular Characterization and Epidemiologic Study of NDM-1-Producing Extensively Drug-Resistant Escherichia coli.

The emergence and dissemination of NDM-1 (New Delhi metallo-β-lactamase-1)-producing Enterobacteriaceae have resulted in a worldwide public health risk. This study described a high incidence and endemic spread of NDM-1-producing extensively drug-resistant Escherichia coli in a teaching hospital in Zhejiang province, China. We recovered six nonduplicated NDM-1-producing E. coli isolates from May 2014 to August 2014 with positive modified Hodge test and EDTA synergistic test. These isolates were highly resistant to β-lactam antimicrobials, aminoglycosides, and quinolones. PCR and DNA sequences a

Members of the Enterobacteriaceae family produce a diffusible signal factor that modulates enteric pathogen virulence.

UNLABELLED: Diffusible signal factors (DSFs) are a gammaproteobacterial family of quorum-sensing signals that mediate the transition between sessile and planktonic lifestyles. In addition to coordinating behavior within producing species, DSFs can be sensed by surrounding organisms, enabling interspecies "eavesdropping." Several enteric pathogens that do not produce DSFs use these environmental signals to modulate their virulence, allowing them to conserve energy when expression of pathogenesis genes is not advantageous. Although DSFs have been detected in the gut, the microbial origins of the

Antibiotic factories on the move: Synthetic bacteria survive drinking water treatment and continuously produce antibiotics.

Synthetic biology enables the creation of high-yield synthetic antibiotic-producing bacteria (HYSAPB) for industrial biomanufacturing. However, HYSAPB may be inadvertently released into the environment. The fate of HYSAPB in natural water and subsequent the whole drinking water supply systems remains unexplored. This study found that a HYSAPB, S. lividans LJ1018, when released into source water, not only survived but retained its engineered advances, including multidrug resistance, enhanced environmental adaptability, and persistent actinomycin D (Act-D) production. Unlike wild-type strains, S

Iron speciation and microbial functional responses shape sludge dewatering and phase-specific biological risks during Fe2+/ peracetic acid treatment.

The disposal of waste activated sludge (WAS) is challenged by poor dewaterability and environmental risks associated with antibiotic resistance genes (ARGs). Although advanced oxidation is promising for sludge conditioning, whether prolonged oxidation continuously benefits both dewatering and biological risk control remains unclear. Here, Fe2+-activated peracetic acid (PAA) was applied to WAS to evaluate the time-dependent responses of sludge dewaterability, ARGs, potential pathogens, extracellular polymeric substances, iron speciation and microbial functions. The Fe2+/PAA process exhibited di

Cefiderocol as an alternative antibiotic therapy for treating severe Stenotrophomonas maltophilia infections.

Stenotrophomonas maltophilia has emerged as an opportunistic pathogen that causes life-threatening hospital-acquired infections. This microorganism possesses a diverse array of chromosome-encoded antimicrobial resistance genes, which render it inherently multidrug-resistant (MDR). Its ability to acquire additional antimicrobial resistance via mutations and the horizontal transfer of resistome elements from neighboring microbial communities has further contributed to the development of extensively drug-resistant (XDR) and even pandrug-resistant (PDR) strains. These strains are resistant to rout

Nature AND nurture: enabling formate-dependent growth in Methanosarcina acetivorans.

Methanosarcinales are versatile methanogens, capable of regulating most types of methanogenic pathways. Despite the versatile metabolic flexibility of Methanosarcinales, no member of this order has been shown to use formate for methanogenesis. In the present study, we identified a cytosolic formate dehydrogenase (FdhAB) present in several Methanosarcinales, likely acquired by independent horizontal gene transfers after an early evolutionary loss, encouraging re-evaluation of our understanding of formate utilization in Methanosarcinales. To explore whether formate-dependent (methyl-reducing or

Novel Substitutes of Phthalate Esters (PAEs) Promote the Propagation of Antibiotic Resistance Genes via Ferroptosis: Implication for the Environmental Safety Evaluation of PAE Substitutes.

The horizontal transfer of antibiotic resistance genes (ARGs) has become a major threat to global public health. Recent studies have found that ferroptosis, an iron-dependent form of regulated cell death characterized by lipid peroxidation and glutathione depletion, may play a critical role in the dissemination of ARGs among environmental microbes. Here, we demonstrated for the first time that phthalate esters (PAEs) and their substitutes significantly enhanced plasmid conjugation by triggering ferroptosis-related pathways. Classical ferroptosis-associated responses, including the hyperpolariz

Early experience with tedizolid: clinical efficacy, pharmacodynamics, and resistance.

Antimicrobial resistance among gram-positive organisms such as methicillin-resistant Staphylococcus aureus (MRSA) and vancomycin-resistant enterococci (VRE) continues to limit therapeutic options. The oxazolidinones are a synthetic class of agents now commonly relied on for the treatment of serious MRSA and VRE infections. With increasing utilization of linezolid, resistant pathogens have once again begun to emerge. Tedizolid, a next-generation oxazolidinone, possesses a spectrum of activity including MRSA and VRE, with significantly enhanced potency also against linezolid-resistant strains. P

Epidemiology of meningitis and bacteraemia due to Streptococcus pneumoniae in The Netherlands.

In The Netherlands, accurate data on the epidemiology of pneumococcal meningitis are available through a clinical microbiology laboratory-based national surveillance of cerebrospinal fluid isolates. The Netherlands Reference Laboratory for Bacterial Meningitis receives isolates of about 80% of all meningitis cases and about 40% of bacteraemic cases. The incidence of pneumococcal meningitis has increased slowly from 1.0/100,000 in 1990 to 1.5/100,000 since 1996. The highest age-specific incidence of meningitis was observed in children < 5 y of age (8.2/100,000 in 1999). Of all isolates, 35% wer

Genetic characteristics and integration specificity of Salmonella enterica temperate phages.

INTRODUCTION: Temperate phages can engage in the horizontal transfer of functional genes to their bacterial hosts. Thus, their genetic material becomes an intimate part of bacterial genomes and plays essential roles in bacterial mutation and evolution. Specifically, temperate phages can naturally transmit genes by integrating their genomes into the bacterial host genomes via integrases. Our previous study showed that Salmonella enterica contains the largest number of temperate phages among all publicly available bacterial species. S. enterica is an important pathogen that can cause serious sys

Independent Microevolution Mediated by Mobile Genetic Elements of Individual Clostridium difficile Isolates from Clade 4 Revealed by Whole-Genome Sequencing.

Horizontal gene transfer of mobile genetic elements (MGEs) accounts for the mosaic genome of Clostridium difficile, leading to acquisition of new phenotypes, including drug resistance and reconstruction of the genomes. MGEs were analyzed according to the whole-genome sequences of 37 C. difficile isolates with a variety of sequence types (STs) within clade 4 from China. Great diversity was found in each transposon even within isolates with the same ST. Two novel transposons were identified in isolates ZR9 and ZR18, of which approximately one third to half of the genes showed heterogenous origin

From insect endosymbiont to phloem colonizer: comparative genomics unveils the lifestyle transition of phytopathogenic Arsenophonus strains.

UNLABELLED: Bacteria infecting the plant phloem represent a growing threat worldwide. While these organisms often resist in vitro culture, they multiply both in plant sieve elements and hemipteran vectors. Such cross-kingdom parasitic lifestyle has emerged in diverse taxa via distinct ecological routes. In the genus Arsenophonus, the phloem pathogens "Candidatus Arsenophonus phytopathogenicus" (Ap) and "Ca. Phlomobacter fragariae" (Pf) have evolved from insect endosymbionts, but the genetic mechanisms underlying this transition have not been explored. To fill this gap, we obtained the genomes

Contrasting Roles of Mobile Genetic Elements and Metal Resistance Genes in Shaping the Gut Resistome of Wild Fish from the Qiantang River.

The dissemination of antibiotic resistance genes (ARGs) in riverine ecosystems poses a pressing public health threat, while the mechanisms governing the assembly of the gut resistome in wild fish remain poorly elucidated. This study aimed to elucidate the distributional patterns of ARGs across multiple environmental compartments and to identify factors associated with their variation, particularly the contributions of mobile genetic elements (MGEs) and metal resistance genes (MRGs) to gut resistome variation. Metagenomic sequencing was conducted on 60 samples, comprising water, sediment, and g
