Downregulation of SIRT1 signaling underlies hepatic autophagy impairment in glycogen storage disease type Ia.

A deficiency in glucose-6-phosphatase-α (G6Pase-α) in glycogen storage disease type Ia (GSD-Ia) leads to impaired glucose homeostasis and metabolic manifestations including hepatomegaly caused by increased glycogen and neutral fat accumulation. A recent report showed that G6Pase-α deficiency causes impairment in autophagy, a recycling process important for cellular metabolism. However, the molecular mechanism underlying defective autophagy is unclear. Here we show that in mice, liver-specific knockout of G6Pase-α (L-G6pc-/-) leads to downregulation of sirtuin 1 (SIRT1) signaling that activates…

Apelin-13-Loaded Macrophage Membrane-Encapsulated Nanoparticles for Targeted Ischemic Stroke Therapy via Inhibiting NLRP3 Inflammasome-Mediated Pyroptosis.

PURPOSE: Ischemic stroke is a refractory disease wherein the reperfusion injury caused by sudden restoration of blood supply is the main cause of increased mortality and disability. However, current therapeutic strategies for the inflammatory response induced by cerebral ischemia-reperfusion (I/R) injury are unsatisfactory. This study aimed to develop a functional nanoparticle (MM/ANPs) comprising apelin-13 (APNs) encapsulated in macrophage membranes (MM) modified with distearoyl phosphatidylethanolamine-polyethylene glycol-RVG29 (DSPE-PEG-RVG29) to achieve targeted therapy against ischemic st

Insights on attenuating autophagy cellular and molecular pathways versus methotrexate-induced toxicity via liposomal turmeric therapy.

BACKGROUND: Methotrexate (MX), a competitive inhibitor of dihydrofolate reductase, can inhibit DNA and RNA production and is a powerful anticancer agent widely utilized in clinical practice for treating nonneoplastic maladies, as psoriasis and rheumatoid arthritis; meanwhile, its probable prescription dose and interval of administration are strictly limited due to dose-related organ damage. Former studies verified that kidney, brain, liver, and lung harms are prospective obstacles of methotrexate administration. To understand the machinery of methotrexate-prompt toxicity, various mechanisms we

Lactylation-mediated activation of HSPA5 upregulates VGLL3 expression to inhibit mitophagy-dependent ferroptosis following intracerebral hemorrhage.

Intracerebral hemorrhage (ICH) is a devastating neurological disorder characterized by extensive neuronal death, in which ferroptosis has em … ferase EP300 (E1A binding protein p300) and removed by the NAD-dependent protein deacetylase SIRT1 (sirtuin 1). Collectively, our findings delineate a lactylation-driven HSPA5-VGLL3-EGLN3-HIF1A/HIF1-1α axis that represses BNIP3-BNIP3L-mediated, mitophagy-dependent ferroptosis. Targeting this pathway may provide a mechanistic rationale and therapeutic strate …

The macrophage senescence hypothesis: the role of poor heat shock response in pulmonary inflammation and endothelial dysfunction following chronic exposure to air pollution.

INTRODUCTION: Cardiovascular diseases (CVD) have been associated with high exposure to fine particulate air pollutants (PM2.5). Alveolar macrophages are the first defense against inhaled particles. As soon as they phagocytize the particles, they reach an inflammatory phenotype, which affects the surrounding cells and associates with CVD. Not coincidentally, CVD are marked by a depleted heat shock response (HSR), defined by a deficit in inducing 70-kDa heat shock protein (HSP70) expression during stressful conditions. HSP70 is a powerful anti-inflammatory chaperone, whose reduced levels trigger

Mechanistic insights into the neuroprotective effect of syringic acid on methotrexate-induced neurotoxicity in rats via Sirt1/AKT/HO-1 pathway.

Syringic acid (SA), a naturally occurring derivative of benzoic acid, is widely distributed in many plants and fruits. It has been believed to exhibit potent anti-inflammatory, antioxidant, antiapoptotic, anticarcinogenic, and immunomodulatory effects. This study was designed to explore the protective role of syringic acid versus methotrexate (MTX)-induced neurotoxicity. Rats were randomly assigned into four experimental groups: Group 1: Normal control group; Group 2: MTX group (20 mg/kg, i.p.) single dose on the sixth day; Group 3 & 4: Syringic acid groups (50 and 100 mg/kg, orally) for 10 co

Genistein suppresses renal fibrosis in chronic kidney disease through regulation of the CCAR2/SIRT1/p53 signaling axis.

Chronic kidney disease (CKD), a major global health burden, is characterized by progressive renal fibrosis, for which effective therapies are lacking. While the natural isoflavone genistein (Gen) has been documented to possess broad anti-fibrotic properties, its specific function and underlying mechanism in renal fibrosis remain largely undefined. In this study, we demonstrate that Gen administration significantly attenuates renal fibrosis in both UUO and adenine-induced murine CKD models. To elucidate the molecular mechanism, we employed a strategy using biotin-conjugated Gen as a bait for pu

SIRT1 agonist SRT1720 alleviates cerebral ischemia-reperfusion injury by inhibiting lactate-mediated SOD2 Lactylation and suppressing NCOA4-mediated ferritinophagy-ferroptosis.

Cerebral ischemia-reperfusion injury (CIRI) represents a critical pathological process underlying stroke-induced neuronal loss and long-term neurological deficits, characterized by profound oxidative stress and dysregulated iron homeostasis. Although metabolic disturbances such as lactate accumulation are hallmarks of ischemic brain injury, their effects on protein post-translational modifications, particularly lysine lactylation, remain largely unexplored. In this study, male C57BL/6 J mice were subjected to transient middle cerebral artery occlusion (tMCAO) to induce focal CIRI, and microgli

SIRT1/NRF2 axis regulates NLRP3-mediated macrophage inflammation and renal injury in hypertension with hyperuricemia.

BACKGROUND: Hypertension and hyperuricemia commonly coexist and are associated with persistent inflammation and renal injury. However, the mechanism linking these two conditions remains unclear. This study investigated whether the SIRT1/NRF2 axis regulates NLRP3-mediated macrophage inflammation in hypertension complicated by hyperuricemia. METHODS: Clinical data from 84 patients with hypertension and hyperuricemia and 84 patients with hypertension alone were analyzed. Circulating NLRP3 and inflammatory cytokines were measured, and the value of NLRP3 in identifying hyperuricemia occurrence and

Chlorogenic Acid Modulates PINK1-Parkin-Mediated Mitochondrial Quality Control in T-2 Toxin-Induced Thymic Immunotoxicity.

T-2 toxin (T-2), a potent immunotoxic trichothecene, induces severe thymic injury and immune dysfunction. Building on our identification of the ROS-NF-κB-NLRP3 axis, mitochondrial involvement remains poorly understood. Using murine and thymic epithelial cell models, we show that T-2 induces mitochondrial damage, activating PINK1-Parkin-mediated mitophagy that is adaptive but insufficient to maintain mitochondrial homeostasis. Inhibition of mitophagy by 3-methyladenine or Parkin silencing further compromised mitochondrial membrane potential by ∼34% and ∼22% versus T-2, respectively, accompanied

Inhibition of GPR75 Alleviates Lipid Metabolism by Activating the AMPK-SIRT1 Signaling Pathway In Vitro and In Vivo.

The pathogenesis of non-alcoholic fatty liver disease (NAFLD) is primarily driven by excessive lipid accumulation and metabolic dysregulation, necessitating a comprehensive investigation into the underlying mechanisms. This study employed an in vitro model, wherein Huh7 cells were induced with a palmitic acid/oleic acid mixture, and an in vivo model involving the provision of a high-fat diet to SD rats for six weeks. Employing techniques such as oil red O staining, immunofluorescence, and Western blotting, we examined lipid synthesis, metabolism, and the associated molecular pathways. The find

Synthesis and anti-inflammatory properties of glycosylated cinnamaldehyde derivatives in mice models of colitis and gout.

Cinnamaldehyde, a natural compound with diverse biological activities, has limited biomedical application due to its potential cytotoxicity. In this study, we synthesized a series of novel glycosylated cinnamaldehyde derivatives by varying both the sugar moiety and its attachment position on the aromatic ring. Preliminary screening for cytotoxicity and anti-inflammatory activity revealed that these structural modifications critically influenced bioactivity. Among the derivatives, compound 1a, bearing a β-D-galactosyl group at the para-position of the cinnamaldehyde phenyl ring, exhibited the m

Ginsenoside Rh2 targets SIRT1-mediated deacetylation to modulate ERα/AR balance and overcome endocrine therapy resistance in prostate cancer using 3D organoid models.

Resistance to endocrine therapy remains a major challenge in treating prostate cancer (PCa), highlighting the need for alternative therapeutic approaches. In this study, we investigated the potential of Ginsenoside Rh2 to counteract such resistance by influencing the SIRT1-dependent deacetylation pathway, thereby modulating the equilibrium between estrogen receptor α (ERα) and androgen receptor (AR). We proposed that Rh2 may suppress therapy-resistant PCa progression by adjusting ERα/AR transcriptional dynamics. Through network pharmacology analysis, key anti-PCa targets of Rh2 were identified
