Immunological features and efficacy of the recombinant subunit vaccine LTB-EMY162 against Echinococcus multilocularis metacestode.

Alveolar echinococcosis is a zoonotic disease caused by the infection of the larval stage Echinococcus multilocularis with worldwide distribution especially in the northwest China. It is important to develop a well-tolerated immunoprophylaxis against E. multilocularis for alveolar echinococcosis control. In this study, a prokaryotic expression system for recombinant immunogen LTB-EMY162 was established, and the immunological features, sensitized lymphocyte, IL-4/IFN-γ secreted, prophylactic effect, and therapeutic effect were also evaluated. Arctic Express (DE3) system, Ni2+-charged and molecu

Inhibition of EBV-mediated membrane fusion by anti-gHgL antibodies.

Herpesvirus entry into cells requires the coordinated action of multiple virus envelope glycoproteins, including gH, gL, and gB. For EBV, the gp42 protein assembles into complexes with gHgL heterodimers and binds HLA class II to activate gB-mediated membrane fusion with B cells. EBV tropism is dictated by gp42 levels in the virion, as it inhibits entry into epithelial cells while promoting entry into B cells. The gHgL and gB proteins are targets of neutralizing antibodies and potential candidates for subunit vaccine development, but our understanding of their neutralizing epitopes and the mech

Identification and characterization of an interspersed repeat antigen of Babesia microti (BmIRA).

In this report, a novel gene encoding an interspersed repeat antigen from Babesia microti (BmIRA) was identified and described. The full-length cDNA containing an open reading frame of 1,947 bp was obtained by immunoscreening a B. microti cDNA expression library. The full-length of BmIRA gene was expressed as a GST fusion recombinant BmIRA (rBmIRA) in Escherichia coli. Sera of mice immunized with the rBmIRA detected a native parasite protein with a molecular mass of 76 kDa on Western blot analysis. The same protein was detected in the parasites by immunofluorescent antibody test (IFAT). An enz

Developing subunit immunogens using B and T cell epitopes and their constructs derived from the F1 antigen of Yersinia pestis using novel delivery vehicles.

Yersinia pestis is the etiological agent of pneumonic and bubonic plague. As the currently licensed vaccines for plague have their own limitations, there is a need for a rational and more effective form of a subunit vaccine to combat both forms of the disease. Newer methods of antigen delivery coupled with adjuvant offer an alternative approach toward a plague vaccine. In order to develop a new generation vaccine against plague, we chose an immunodominant, outer membrane capsular protein, F1 of Y. pestis. The immunogenicity of the peptide sequences, predicted to possess B (three sequences, B1,

MlaA, a conserved outer membrane protein of Neisseria gonorrhoeae, elicits potent functional immunity and protects against vaginal infection as a mucosal vaccine candidate.

The global spread of multidrug-resistant Neisseria gonorrhoeae strains presents a major challenge in the treatment of gonorrhoea. Vaccination represents a promising long-term strategy for disease control. However, no licensed vaccine is currently available. In this study, we investigated MlaA as a potential vaccine candidate antigen by assessing its endogenous expression, immunogenicity, and the functional activity of induced antibodies. Our findings show that the gene encoding MlaA is highly conserved, and expressed across all tested strains of N. gonorrhoeae. Intranasal immunization of mice

Exploring the structural basis to develop efficient multi-epitope vaccines displaying interaction with HLA and TAP and TLR3 molecules to prevent NIPAH infection, a global threat to human health.

Nipah virus (NiV) is an emerging zoonotic virus that caused several serious outbreaks in the south asian region with high mortality rates ranging from 40 to 90% since 2001. NiV infection causes lethal encephalitis and respiratory disease with the symptom of endothelial cell-cell fusion. No specific and effective vaccine has yet been reported against NiV. To address the urgent need for a specific and effective vaccine against NiV infection, in the present study, we have designed two Multi-Epitope Vaccines (MEVs) composed of 33 Cytotoxic T lymphocyte (CTL) epitopes and 38 Helper T lymphocyte (HT

Oral yeast-displayed SARS-CoV-2 spike protein vaccine enhanced by trimerization and flagellin peptide.

The global COVID-19 pandemic has underscored the need for innovative vaccine strategies, including oral vaccines, to improve accessibility and enhance immunity. In this study, we explored a novel oral vaccine approach by using yeast surface display to express the receptor-binding domain (RBD) of the SARS-CoV-2 spike protein on Pichia pastoris using SED1 as a carrier protein. The constructs included a fibritin foldon domain from bacteriophage T4 for trimerization and truncated flagellin from Salmonella typhimurium (FliC) as an adjuvant to boost immunogenicity. Three recombinant yeast constructs

N-terminal pyroglutamylation of an HLA-A24-restricted immunodominant epitope enhances SARS-CoV-2-specific T-cell responses.

T-cell responses play a critical role in the control of SARS-CoV-2 infections. Here, we examined the T-cell response to a conserved, immunodominant HLA-A*24:02-restricted SARS-CoV-2 spike protein epitope, QI9/A24 (QYIKWPWYI; residues 1208-1216). TCR repertoire analysis revealed that QI9/A24-specific T cells are highly diverse, with 219 clonotypes isolated and 169 αβ TCR pairs identified in six donors. To further characterize this response, we evaluated T-cell recognition of alanine-substituted and homologous coronavirus-derived peptides, using a TCR reconstitution system, and found that QI9/A2

Multi-epitope vaccine design against Monkeypox virus: An immunoinformatics approach.

BACKGROUND: The recent outbreak of the Monkeypox virus (MPXV) across multiple regions has highlighted the need for effective vaccine. This study employed an in-silico immunoinformatics approach to design a multi-epitope vaccine against monkeypox virus. METHODS: The IMV heparin binding surface protein (H3L) of MPV was selected as vaccine target. B and T cell epitopes were predicted and evaluated for their toxicity, allergenicity, antigenicity and global population coverage. A multi-epitope construct was assembled using adjuvant and appropriate linkers. The 3D vaccine structure was modelled, en

Computational design of a novel chimeric multi-epitope vaccine against chlamydia trachomatis serovar a to prevent trachoma.

Chlamydia trachomatis serovar A is a re-emerging worldwide health problem that causes the ocular trachoma, the leading cause of avoidable blindness. The disease continues to spread due to reinfection and failure to develop long-lasting immunity. This study aimed to develop an in silico novel multi-epitope that can induce long-term immune protection against C. trachomatis serovar A. Researchers harvested the protein sequences of serovar A and queried for virulence-related factors. The predictions of epitopes for CTLs, HTLs, and B cells using immunoinformatics tools, and tested for its ability t

Promiscuous class II-binding SARS-CoV-2-nuc derived vaccine-peptide induced extensive conventional, innate and unconventional T cell responses.

We describe the T-cell response of two healthy SARS-CoV-2-unexposed volunteers to a SARS-CoV-2 nucleoprotein-derived vaccine peptide predicted to promiscuously bind multiple HLA-DR allotypes. NGS-based bulk TCR-repertoire analysis of peptide-specific T-cell responses 4 (D2) and 27 (D1) weeks after vaccination identified CDR3 regions of TCRα, -β, -γ and -δ chains in T cells responding ex-vivo to the vaccine peptide LLLLDRLLNQLESKMS with IFNγ+-secretion. Adaptive repertoires were unique. Donors shared 15 TCRα and 9 TCRβ clonotypes, all public, showing no conserved motifs but TdT-independent "neo

Oral Administration of Bacillus subtilis Subunit Vaccine Significantly Enhances the Immune Protection of Grass Carp against GCRV-II Infection.

Grass carp reovirus (GCRV) is a severe virus that causes great losses to grass carp culture every year, and GCRV-II is the current popular and fatal strain. VP56, fibrin on the outer surface of GCRV-II, mediates cell attachment. In this study, we firstly divided the VP56 gene into four fragments to screen the optimal antigen by enzyme-linked immunosorbent assay and neutralizing antibody methods. The second fragment VP56-2 demonstrates the optimal efficiency and was employed as an antigen in the following experiments. Bacillus subtilis were used as a carrier, and VP56-2 was expressed on the sur

Comprehensive mutanome analysis of Lewis lung cancer reveals immunogenic neoantigens for therapeutic vaccines.

Personalized neoantigen vaccines are capable of eliciting vigorous T-cell responses and have been demonstrated to achieve striking therapeutic effects against cancer. Here we performed comprehensive mutanome analysis of the mouse Lewis lung cancer cells to identify tumor neoantigens followed by prediction of their MHC affinity and immunogenicity. We adopted a strategy that enables us to select neoantigens that were predicted to have high affinity to both MHC I and MHC II. Ten neoantigens were selected to synthesize peptide vaccines and tested in vivo for immunogenicity. Four neoantigen peptide

A nanovaccine induces durable and functional MUC4-targeted T cell responses against pancreatic cancer.

Vaccination against cancer for prophylaxis or treatment is becoming a viable approach owing to new developments in the identification of novel biomarkers and advances in our understanding of cancer immunology. Furthermore, nanotechnology has resulted in multifunctional delivery platforms with versatile chemistries and tunable release properties. However, despite this impressive progress, immunotherapy with the goal of inducing antigen-specific T cell immunity against pancreatic cancer (PC) remains challenging. Herein, we report the characterization of the long-lived humoral and cellular immuni

A human cytomegalovirus prefusion-like glycoprotein B subunit vaccine elicits humoral immunity similar to that of postfusion gB in mice.

Human cytomegalovirus (HCMV) is the leading infectious cause of birth defects. Despite the global disease burden, there is no Food and Drug Administration (FDA)-approved HCMV vaccine. The most efficacious HCMV vaccine candidates to date have used glycoprotein B (gB), a class III viral fusion protein, in its postfusion form. While some viral fusion proteins have been shown to elicit stronger neutralizing responses in their prefusion conformation, HCMV prefusion-like and postfusion gB were recently shown to elicit antibodies with similar fibroblast neutralization titers in mice. We aimed to defi

Expression of immunogenic poliovirus Sabin type 1 VP proteins in transgenic tobacco.

One of the milestones of vaccinology is the depletion of the global impact of Poliomyelitis. The current vaccines to deal with Polio comprise the Sabin and Salk formulations. The main limitation of the former is the use of attenuated viruses that can revert into pathogenic forms, whereas the latter is more expensive and induces no protection in the intestinal tract; the site of virus replication. Genetically engineered plants cope with such limitations. In addition, they offer a low-cost alternative for production, storage and delivery of vaccines. This technology has been narrowly applied in
