Isolation and characterization of murine vav2, a member of the vav family of proto-oncogenes.

We describe the isolation and characterization of a cDNA encoding murine vav2. vav2 shares 63% and 55% identity at the nucleic acid and amino acid levels, respectively, with vav, a proto-oncogene that plays an essential role in embryonic development and hematopoietic signal transduction. The 100 kDa Vav2 protein contains the characteristic array of structural motifs found in Vav. However, unlike vav, vav2 transcripts are widely distributed in both hematopoietic and non-hematopoietic tissues. In the adult, vav2 mRNA is found at high levels in the spleen, liver, testes and placenta. Northern blo

Spatiotemporal expression of three gap junction gene products involved in fetomaternal communication during rat pregnancy.

The expression of three different members of the gap junction multigene family, alpha 1 (Cx43), beta 1 (Cx32), and beta 2 (Cx26), was analysed in the rat implantation chamber (a structural unit containing fetal, extraembryonic and maternal components within the pregnant uterus) during mid- and late stages of gestation as well as in the delivering, post-partum and non-pregnant uterus. A differential, spatiotemporal and cell-type-specific regulation of gap junctional coexpression was observed for beta 1 and beta 2 in all epithelia examined (visceral, luminal and glandular), as well as for alpha

Cell lineage and cell migration in the neural crest.

The neural crest is a transient embryonic structure whose cells migrate extensively before giving rise to a variety of differentiated cell types. Both intrinsic cell lineage information and environmental cues are thought to play a role in determining the fate of these cells. Early in development, these cells can be divided into distinct populations based on their axial level of origin. Cranial neural crest cells differentiate into facial cartilage and cranial ganglia, whereas vagal crest cells give rise to the enteric ganglia. Trunk neural crest cells normally give rise to melanocytes, neurons

Cell-type specific and differential regulation of the human metallothionein genes. Correlation with DNA methylation and chromatin structure.

The expression of three human metallothionein genes, MT-IIA, MT-IF, and MT-IG was studied in the human hepatoblastoma (HepG2), the hepatocarcinoma (Hep3B2), the embryonic kidney (Hek 293), and the lymphoblastoid-derived (Wi-L2) cell lines. The pattern of expression of each specific MT gene in response to various heavy metals was different among the four cell lines studied indicating differential regulation of MT gene expression. The MT-IF or MT-IG and the MT-IIA genes were regulated in a cell-type specific manner in response to heavy metals and dexamethasone, respectively. DNA methylation was

Chromatin and nucleolar changes in Xeroderma pigmentosum cells resemble aging-related nuclear events.

Xeroderma pigmentosum (XP) is a hereditary disease characterized by a defect in the excision-repair mode of ultraviolet light damage and a high incidence of skin tumors. Cultured fibroblasts from normal and XP cells at low population doubling times were compared by induction of mild spreading of their nuclear constituents in a highly alkaline solution containing detergent and formaldehyde. In each XP culture a certain fraction (10-80%) of the nuclei were abnormal (50-80% in cell lines from children with XP-C disorders and 10-35% from embryonic and adult XP cells). Although their chromatin thre

Design and biological evaluation of a histidine-bearing cyclometalated Pd(II) complex derived from 2-phenylpyridine.

A rationally designed approach was employed to synthesize a new C^N-cyclometalated Pd(II) complex (C^N = 2-phenylpyridine) as a potential anticancer metallodrug. The incorporation of histidine in the coordination sphere of metal complex offers dual effect, balancing hydrophobic and hydrophilic properties while enhancing biocompatibility. The structure of histidine-bearing C^N-cyclometalated Pd(II) complex was characterized using elemental analysis, FT-IR, and NMR spectroscopy. In vitro cytotoxic activity was evaluated against HEP G2 human hepatoma and MCF7 human breast cancer cells revealing h

Somatic mosaicism in the brain: linking development, ageing and neurodegeneration.

Somatic mosaicism is increasingly recognized as a pervasive feature of the human brain and a potential contributor to neurological disease across the lifespan. Unlike germline variants, somatic variants arise post-zygotically and are unevenly distributed across regions, cell types and even individual neurons, enabling focal biological effects that can scale to network-level dysfunction. In this Review, we synthesize current evidence that developmental timing, clonal architecture and cell-type-specific selective pressures shape how somatic variants influence brain structure and function. Early

Metastatic Retroperitoneal Rhabdomyosarcoma in a Child: A Case Report Highlighting the Role of Imaging in Diagnosis, Staging, and Follow-Up.

Retroperitoneal masses in children encompass a heterogeneous group of benign and malignant tumors with a largely nonspecific clinical presentation. Rhabdomyosarcoma (RMS) is a soft tissue sarcoma that uncommonly arises primarily in the retroperitoneum, a site associated with a comparatively unfavorable prognosis related to delayed diagnosis, large tumor volume at presentation, and frequent invasion of adjacent vascular and digestive structures. We report the case of a five-year-eight-month-old boy who presented with a one-month history of progressive abdominal distension and left lower limb ed

Cross-Species Conservation and Function of ALDH6A1/Aldh6a1 Validate Zebrafish and Mouse as Complementary Experimental Systems for Methylmalonate Semialdehyde Dehydrogenase Deficiency.

Methylmalonate semialdehyde dehydrogenase deficiency (MMSDD; OMIM #614265) is an ultra-rare autosomal recessive metabolic disorder caused by mutations in the ALDH6A1 gene, characterized by multi-system involvement including skeletal abnormalities, hypotonia, and visual impairment. To elucidate its pathogenic mechanisms, we characterized ALDH6A1/Aldh6a1 conservation and function using complementary zebrafish and mouse experimental systems. Bioinformatic and phylogenetic analyses revealed strong conservation of Aldh6a1/aldh6a1 gene structure, protein sequence, functional domains, and mitochondri

Human iPSC-cardiomyocyte-aggregate cell therapy in non-human primates and correlation of heart recovery with contractile and electrophysiological cardiomyocyte properties.

This study demonstrates the successful production and injection of human induced pluripotent stem cell cardiomyocyte aggregates into infarcted cynomolgus monkey hearts, resulting in substantial, structured human grafts three months after cell transplantation. Transient graft-induced arrhythmias decreased over time. Both the arrhythmogenicity and the substantial heart function recovery in vivo notably seemed to correlate with induced pluripotent stem cell clone-dependent contractile and electrophysiological cardiomyocyte properties in vitro. Overexpression of a red fluorescent reporter protein

Revealing the neurodevelopmental toxicity of face mask-derived microplastics to humans based on neural organoids.

The massive use of face masks during and after the COVID-19 pandemic has raised global concerns about environmental issues. Microplastics released from face masks pose great threats to ecosystems and human health. However, the potentially hazardous effects of face mask-derived microplastics (FMMs) on humans remain poorly understood. Using neural organoid models aims to reveal the toxicity of FMMs to human early neural development. Retinal organoids derived from human embryonic stem cells were exposed to FMMs for 21 days during early retinogenesis. FMMs were internalized by retinal organoids. E

Heterochronic changes in gene expression underlie placental evolution in the fish family Poeciliidae.

Placentas evolved nine times in the fish family Poeciliidae. Each time, the egg follicle is the maternal contribution to the placenta. In non-placental species, the follicle fully provisions the egg before fertilization. In placental species, provisioning continues throughout development, and the follicle becomes a more elaborate, well-vascularized organ. We generated transcriptomes for follicles from yolking eggs and developing embryos from two pairs of closely related placental and non-placental species that represent independent origins of placentation plus one non-placental outgroup. We id

An Astrocyte Transplantation Method to Investigate Astrocyte Development and Diversity in the Central Nervous System (CNS).

Astrocytes have rich structural, molecular, and physiological properties and show remarkable diversity in form and function that supports circuit-specific physiology within the central nervous system (CNS). However, the developmental mechanisms and cellular interactions that help to locally and globally diversify astrocytes with the CNS remain to be better understood. Understanding these processes will help to understand how astrocytes establish specialized microenvironments in the healthy brain and how they may differentially respond to CNS injury and disease. Here, we describe a cell transpl

Successful pregnancy in a complex chromosomal rearrangement carrier using preimplantation genetic testing for structural rearrangements: A case report.

RATIONALE: Complex chromosomal rearrangements (CCRs) frequently lead to unfavorable reproductive consequences, such as recurrent miscarriage. Preimplantation genetic testing for structural rearrangements (PGT-SR) has become a successful method for embryo diagnosis in CCR families. PATIENT CONCERNS: A woman of childbearing age experienced repeated miscarriages during her attempts to conceive. DIAGNOSES: A woman was diagnosed with a combined Robertsonian translocation involving chromosomes 13 and 15 (robs[13;15]) and a reciprocal translocation involving chromosomes 10 and 12 (t[10;12]). Genetics of Skin, Hair, and Eye Color in Human Pigmentation Disorders.

Skin, hair, and eye (oculocutaneous) color is due to melanin, a pigment produced by melanocytes. This review considers processes required for pigmentation and the complex genetic network that regulates them. The first requisite is migration of neural crest-derived melanoblasts, which populate various embryonic sites, then differentiate into melanocytes or seed stem cell niches. Differentiation is marked by expression of genes essential for melanogenesis, which takes place in melanosomes and involves conversion of tyrosine into melanin. Melanosome biogenesis requires premelanosome maturation th

Developmental alterations of indirect-pathway medium spiny neurons in mouse models of Huntington's disease.

Huntington's disease (HD) is a complex neurodegenerative disorder with cognitive and motor symptoms that typically manifest in adulthood. However, embryonic brain development impairments leading to cortical defects in HD mutation carriers has been shown recently supporting a neurodevelopmental component in HD. Despite HD is primarily recognized as a striatal pathology, developmental alterations in this structure, particularly during the early postnatal period, remain poorly understood. To fill this gap, we examined striatal development in newborn R6/1 mice. We found that D2 receptor-expressing

A Modular Microfluidic Organoid Platform Using LEGO-Like Bricks.

The convergence of organoid and organ-on-a-chip (OoC) technologies is urgently needed to overcome limitations of current 3D in vitro models. However, integrating organoids in standard OoCs faces several technical challenges, as it is typically laborious, lacks flexibility, and often results in even more complex and less-efficient cell culture protocols. Therefore, specifically adapted and more flexible microfluidic platforms need to be developed to facilitate the incorporation of complex 3D in vitro models. Here, a modular, tubeless fluidic circuit board (FCB) coupled with reversibly sealed ce

Frameshift variants in C10orf71 cause dilated cardiomyopathy in human, mouse, and organoid models.

Research advances over the past 30 years have confirmed a critical role for genetics in the etiology of dilated cardiomyopathies (DCMs). However, full knowledge of the genetic architecture of DCM remains incomplete. We identified candidate DCM causal gene, C10orf71, in a large family with 8 patients with DCM by whole-exome sequencing. Four loss-of-function variants of C10orf71 were subsequently identified in an additional group of492 patients with sporadic DCM from 2 independent cohorts. C10orf71 was found to be an intrinsically disordered protein specifically expressed in cardiomyocytes. C10o

Dynamic evolutionary interplay between ontogenetic skull patterning and whole-head integration.

The arrangement and morphology of the vertebrate skull reflect functional and ecological demands, making it a highly adaptable structure. However, the fundamental developmental and macroevolutionary mechanisms leading to different vertebrate skull phenotypes remain unclear. Here we exploit the morphological diversity of squamate reptiles to assess the developmental and evolutionary patterns of skull variation and covariation in the whole head. Our geometric morphometric analysis of a complex squamate ontogenetic dataset (209 specimens, 169 embryos, 44 species), covering stages from craniofacia
