BACKGROUND: Preclinical evidence suggests that diazepam enhances hippocampal γ-aminobutyric acid (GABA) signalling and normalises a psychosis-relevant cortico-limbic-striatal circuit. Hippocampal network dysconnectivity, particularly from the CA1 subfield, is evident in people at clinical high-risk for psychosis (CHR-P), representing a potential treatment target. This study aimed to forward-translate this preclinical evidence.
METHODS: In this randomised, double-blind, placebo-controlled study, 18 CHR-P individuals underwent resting-state functional magnetic resonance imaging twice, once foll

Dopaminergic neurons are located in several brain areas including the olfactory bulb (OB) and involved in many physiological and pathophysiological processes. In the OB, dopamine (DA) is released exclusively by a population of interneurons termed short axon cells (SACs) in the glomerular layer, the initial synaptic integration site of the whole olfactory system. SACs corelease GABA and extend their processes to many glomeruli forming the interglomerular circuit. Two major groups of DA receptors D1-like (D1LRs) and D2-like (D2LRs) types are differentially distributed in the OB, i.e., D1LRs are

OBJECTIVE: Approximately 30% of patients with epilepsy are refractory to existing antiseizure drugs (ASDs). Given that the properties of the central nervous systems of these patients are likely to be altered due to their epilepsy, tissues from rodents that have undergone epileptogenesis might provide a therapeutically relevant disease substrate for identifying compounds capable of attenuating pharmacoresistant seizures. To facilitate the development of such a model, this study describes the effects of classical glutamate receptor antagonists and 20 ASDs on recurrent epileptiform discharges (RE

The temporal fidelity of synaptic transmission is constrained by the reproducibility of time delays such as axonal conduction delay and synaptic delay, but very little is known about the modulation of these distinct components. In particular, synaptic delay is not generally considered to be modifiable under physiological conditions. Using simultaneous paired patch-clamp recordings from pre- and postsynaptic elements of the calyx of Held synapse, in juvenile mouse auditory brainstem slices, we show here that synaptic activity (20-200 Hz) leads to activity-dependent increases in synaptic delay a

The effects of muscarinic agonists on GABAergic synaptic transmission were examined using whole-cell patch-clamp recording in chick brain slices containing the lateral spiriform nucleus. Bath application of muscarine (10 microM) both increased the frequency of spontaneous GABAergic postsynaptic currents and reduced the amplitude of evoked GABAergic polysynaptic postsynaptic currents elicited by focal afferent fiber electrical stimulation. Both of these muscarinic actions were reversible and dose-dependent. Two M(1) antagonists, telenzepine and pirenzipine, and to a lesser extent the M(2) antag

Respiratory activity was studied in rats during light halothane anesthesia. Thyrotropin releasing hormone (TRH) and two TRH analogues: the desamidated form (TRH-OH) and gamma-butyrolactone-gamma-carbonyl-L-histidyl-L-prolinamide citrate (DN 1417) were administered intracerebroventricularly. TRH 0.5-5 micrograms induced a marked tachypnoea with a rapid onset and a duration of at least 20 min. DN 1417, a potent analogue of TRH with a very low TSH (thyroid stimulating hormone) releasing activity was more effective in stimulating respiratory frequency, while TRH-OH, regarded to have neither TSH re

Acorus tatarinowii, a traditional Chinese medicinal herb, is used clinically to treat neurological and respiratory disorders. Our previous work showed that α-asaronol (α-AOL), a low-toxicity natural constituent of A. tatarinowii, exerts robust antiepileptic effects through γ-aminobutyric acid (GABA)-ergic modulation. Given the critical role of the peripheral GABAergic system in asthma pathogenesis and inflammation regulation, the therapeutic potential of α-AOL for asthma warrants investigation. This study evaluated the efficacy, safety, and pulmonary targeting of both nebulized and oral α-AOL.

Essential oils are multicomponent natural products with potential neurobehavioral activity, but integrated comparative studies remain limited. This study compared the essential oils of Satureja brevicalyx, Peperomia dolabriformis, and Rosmarinus officinalis in relation to their chemical profiles, predicted target interactions, preliminary acute oral safety, anxiolytic-like and antidepressant-like effects, antagonist-sensitive behavioral patterns, and exploratory serum biomarkers. Oils were characterized by GC-MS, and their constituents were screened by molecular docking against anxiety-, depre

The theory of visually guided ocular growth is well supported in explaining myopia, but how the retina senses focus versus defocus and converts the signaling into growth-modulating genetic signals remains unresolved. Using whole-cell recordings and single-cell RNA-seq in the mouse retina, we show that lateral inhibitory networks-horizontal cells in the outer retina, but not AII amacrine cells in the inner retina, respond to optical defocus. Dopaminergic amacrine cells (DACs) are maximally excited by focused images and increasingly inhibited by high blur, consistent with dopamine's anti-myopiag

Endotoxemia-induced brain dysfunction is a complex neurological condition, with growing evidence underscoring the critical roles of neuroinflammation and oxidative stress in its pathogenesis. In this context, the current study was undertaken to evaluate the neuroprotective efficacy of the neuropeptide neurotensin analog PD149163 against endotoxemia-induced brain dysfunction and behavioural impairments in mice. Swiss-albino mice (female/08-weeks/25 ± 2.5 g) were divided into 6 groups: Group-I/control; Groups II and III were treated with 50 μg/kg BW and 100 μg/kg BW of PD149163, respectively, fo

Background/Objectives: It is widely recognized that malfunctions in the GABAergic system can be one of the underlying mechanisms in chronic pain. However, the use of GABAergic drugs to improve pain perception has strong and unwanted side effects, particularly in terms of sedation. Therefore, the present exploratory single-case study tested an alternative treatment using balance training to upregulate the GABAergic system in a 62-year-old patient with widespread chronic pain. Previously, balance training was shown to increase short-interval intracortical inhibition (SICI), a neurophysiological

BACKGROUND/OBJECTIVES: It remains unclear whether sleep deprivation induces constipation. This study aimed to evaluate the intervention effects of Bifidobacterium longum 151-1 and BN-BB01 on constipation induced by sleep deprivation in mice, and to explore the underlying mechanism from the perspective of the gut-brain axis, so as to provide a theoretical basis for probiotic intervention in sleep-related gastrointestinal dysfunction.
METHODS: A mouse model of sleep deprivation was established via the multiple platform water environment method. Through the detection and analysis of relevant ind

BACKGROUND: The occurrence of ischemic stroke is closely associated with the inflammatory infiltration of peripheral monocytes/macrophages, and the inflammatory activation of macrophages is often affected by metabolic reprogramming. The Slc25a1 regulates mitochondrial citrate transport and has been shown to affect cell proliferation and migration in cancer. Astragaloside IV exhibits significant anti-inflammatory activity and improves ischemic stroke, but its regulatory effect on macrophage metabolic reprogramming has not been elucidated.
OBJECTIVES: This study aims to explore the effect of as

BACKGROUND: The incidence of sleep disorders in children with autism spectrum disorder (ASD) is very high. Sleep disorders can exacerbate the development of ASD and impose a heavy burden on families and society. The pathological mechanism of sleep disorders in autism is complex, but gene mutations and neural abnormalities may be involved.
METHODS: In this review, we examined literature addressing the genetic and neural mechanisms of sleep disorders in children with ASD. The databases PubMed and Scopus were searched for eligible studies published between 2013 and 2023.
RESULTS: Prolonged awak

Opioid use disorder is characterized by excessive use of opioids, inability to control its use, a withdrawal syndrome upon discontinuation of opioids, and long-term likelihood of relapse. The behavioral stages of opioid addiction correspond with affective experiences that characterize the opponent process view of motivation. In this framework, active involvement is accompanied by positive affective experiences which gives rise to "reward craving," whereas the opponent process, abstinence, is associated with the negative affective experiences that produce "relief craving." Relief craving develo

Untargeted metabolomics is a global molecular profiling technology that can be used to screen for inborn errors of metabolism (IEMs). Metabolite perturbations are evaluated based on current knowledge of specific metabolic pathway deficiencies, a manual diagnostic process that is qualitative, has limited scalability, and is not equipped to learn from accumulating clinical data. Our purpose was to improve upon manual diagnosis of IEMs in the clinic by developing novel computational methods for analyzing untargeted metabolomics data. We employed CTD, an automated computational diagnostic method t

Pyridoxine (vitamin B6) toxicity is a well-known model for peripheral neuropathy. GABA and glutamate are two neurotransmitters in neural pathways involved in the peripheral neuropathy. Cichorium intybus (Chicory) contains glycosides and triterpenoids, which inhibit glutamatergic transmission and enhance GABAergic transmission. The present study was aimed at studying the effect of chicory extract (CE) on the pyridoxine-induced peripheral neuropathy with a particular focus on glutamatergic and GABAergic systems. In this experimental study, a high dose of pyridoxine (800 mg/kg, i.p.) was injected

Nicotinic acetylcholine receptors (nAChRs) regulate information transfer across the main olfactory bulb by instituting a high-pass intensity filter allowing for the filtering out of weak inputs. Excitation-driven inhibition of the glomerular microcircuit via GABA release from periglomerular cells appears to underlie this effect of nAChR activation. The multiplicity of nAChR subtypes and cellular locations raises questions about their respective roles in mediating their effects on the glomerular output. In this study, we address this issue by targeting heteromeric nAChRs using receptor knockout
