Knockdown of BATF3 Inhibits Gastric Cancer Cell Growth and Radioresistance via S1PR1/STAT3 Pathway.

OBJECTIVE: Gastric cancer is one of the most common and deadly cancers worldwide. Basic leucine zipper transcription factor ATF-like 3 (BATF3) plays a key role in tumor immunity. However, the function of BATF3 in gastric cancer remains unclear. Here, we demonstrated BATF3 positively regulated proliferation and radioresistance of gastric cancer cells by regulating S1PR1/STAT3 pathway. METHODS: The RNA-seq analyzed the gene expression by UALCAN web portal and Tumor Immune Estimation Resource. RT-qPCR and western blot was performed to verify BATF3 expression in gastric cancer cells. The assays o

ROR1-targeted delivery of OSU-2S, a nonimmunosuppressive FTY720 derivative, exerts potent cytotoxicity in mantle-cell lymphoma in vitro and in vivo.

Mantle-cell lymphoma (MCL) remains incurable despite numerous therapeutic advances. OSU-2S, a novel nonimmunosuppressive FTY720 (Fingolimod) derivative, exhibits potent cytotoxicity in MCL cell lines and primary cells. OSU-2S increased the surface expression of CD74, a therapeutic antibody target in MCL cells. OSU-2S, in combination with anti-CD74 antibody milatuzumab, enhanced cytotoxicity in MCL. Moreover, MCL tumor antigen receptor tyrosine kinase-like orphan receptor 1 (ROR1) targeted immunonanoparticle-carrying OSU-2S (2A2-OSU-2S-ILP)-mediated selective cytotoxicity of MCL in vitro, as we

The protein kinase Sch9 is a key regulator of sphingolipid metabolism in Saccharomyces cerevisiae.

The Saccharomyces cerevisiae protein kinase Sch9 is an in vitro and in vivo effector of sphingolipid signaling. This study examines the link between Sch9 and sphingolipid metabolism in S. cerevisiae in vivo based on the observation that the sch9Δ mutant displays altered sensitivity to different inhibitors of sphingolipid metabolism, namely myriocin and aureobasidin A. Sphingolipid profiling indicates that sch9Δ cells have increased levels of long-chain bases and long-chain base-1 phosphates, decreased levels of several species of (phyto)ceramides, and altered ratios of complex sphingolipids. Sphingosylphosphorylcholine-induced ERK activation inhibits melanin synthesis in human melanocytes.

Sphingosylphosphorylcholine (SPC) is emerging as a potent signaling-lipid mediator. In this study, we investigated the effects of SPC on melanogenesis using cultured human melanocytes. Our results show that SPC significantly inhibits melanin synthesis in a concentration-dependent manner, and further that it reduces the activity of tyrosinase, the rate-limiting melanogenic enzyme. SPC treatment was also found to induce short-thick dendrites in human melanocytes, but not to reduce tyrosinase activity in a cell-free system, whereas kojic acid directly inhibited tyrosinase. These results suggest t

C(2)-ceramide-induced circular smooth muscle cell contraction involves PKC-epsilon and p44/p42 MAPK activation in cat oesophagus. Mitogen-activated protein kinase.

We investigated the mechanism of C(2)-ceramide (C(2))-induced circular smooth muscle cell contraction in cat oesophagus. C(2) produced contraction of smooth muscle cells isolated by enzymatic digestion, peaked at 30 s and was sustained at a plateau at 5 min. The response to C(2) was concentration-dependent. H-7 or chelerythrine inhibited C(2)-induced contraction, while the diacylglycerol (DAG) kinase inhibitor, R59949, had no effect, suggesting that the contraction is protein kinase C (PKC) pathway-dependent. To test if PKC-mediated contraction may be isozyme-specific, we examined the effects

FTY720 immunosuppression impairs effector T cell peripheral homing without affecting induction, expansion, and memory.

FTY720 (2-amino-2-(2-[4-octylphenyl]ethyl)-1,3-propanediol hydrochloride) prolongs survival of solid organ allografts in animal models. Mechanisms of FTY720 immunomodulation were studied in mice infected with lymphocytic choriomeningitis virus (LCMV) to assess T cell responses or with vesicular stomatitis virus to evaluate Ab responses. Oral FTY720 (0.3 mg/kg/day) did not affect LCMV replication and specific CTL and B cells were induced and expanded normally. Moreover, the anti-viral humoral immune responses were normal. However, FTY720 treatment showed first a shift of overall distribution of

Intranasally administered muse cells attenuate neurodegeneration in Parkinson's disease.

BACKGROUND: Parkinson's disease is a neurodegenerative disorder primarily caused by the degeneration and death of dopaminergic neurons in the substantia nigra. Multilineage differentiating stress enduring (Muse) cells are a novel type of stem cells discovered in recent years, exhibiting superior tissue regenerative capabilities compared to regular mesenchymal stem cells, including multi-lineage differentiation potential, stress tolerance, homing ability, in situ differentiation capacity, and non-tumorigenic properties. Here we investigated the effect and mechanism of muse cell in crossing bloo

Investigating the Role of Coenzyme A Restriction in the Pathophysiology of Preeclampsia: Protocol for a Combined Patient Screening and Laboratory Study.

BACKGROUND: Preeclampsia is 1 of the 3 leading causes of maternal mortality worldwide. Unfortunately, its exact pathogenesis is still unclear. Published metabolomic and gene expression analyses point to coenzyme A (CoA) restriction in the placenta as a factor underpinning the observed complications of preeclampsia, but this hypothesis has never been tested. OBJECTIVE: This pilot study aims to discover evidence supporting the CoA-restriction hypothesis through 2 avenues. The first of these involves developing a procedure for the quantitative determination of metabolites to discover if harmful

Opaganib Protects Against High-Fat Diet-Induced Hypercholesterolemia and Liver Steatosis in Mice.

INTRODUCTION: Sphingolipid metabolism is involved in many diseases including cancer, pathologic inflammation, viral infection, neuropathology and metabolic disorders, including obesity and diabetes. Opaganib inhibits the key enzymes sphingosine kinase-2 and dihydroceramide desaturase and has anticancer, anti-inflammatory and antiviral activities in multiple experimental models. Opaganib has been studied in clinical trials with nearly 500 patients having cancer or severe Covid-19 and demonstrated an excellent safety profile. We recently showed that opaganib protects against high-fat diet (HFD)-

Danlong oral liquid alleviates airway remodeling in asthma by targeting the S1PR2/ROCK1/YAP signaling pathway in airway smooth muscle cells.

ETHNOPHARMACOLOGICAL RELEVANCE: Danlong Oral Liquid (DLOL) is a proprietary Traditional Chinese Medicine (TCM) with documented clinical efficacy against asthma, yet its underlying mechanism of action remains incompletely understood. The bioactive sphingolipid mediator sphingosine-1-phosphate (S1P) and its receptor S1PR2 signaling axis are critically implicated in asthma pathogenesis, particularly in driving airway smooth muscle cell (ASMC) remodeling, a key pathological feature of asthma. Despite this, the mechanistic involvement of DLOL in this specific pathway has not been explored. AIM OF

Seeing beyond Gaucher disease: Early detection and treatment of ocular complications.

BACKGROUND: Gaucher disease is a rare genetic disorder caused by a deficiency in the enzyme glucocerebrosidase, which impairs the body's ability to break down certain fats. This leads to the accumulation of glucosyl sphingosine and glucosyl ceramide in the liver, spleen, and bone marrow. Gaucher disease has two major types: nonneuropathic (Type 1) and neuropathic (Type 2 and Type 3). Gaucher disease can have various ophthalmologic manifestations, particularly in Type 3, including posterior segment abnormalities, such as vitreous opacities, condensations, and/or preretinal white dots. Novel mechanism by which geniposide alleviates rheumatoid arthritis: Targeted inhibition of SphK1 negatively regulates S1P signaling, promotes HIF-1α acetylation-mediated degradation and blocks the VEGF-induced synovial angiogenesis.

BACKGROUND: Rheumatoid arthritis (RA) poses a significant health concern and profoundly affects patients' quality of life. Synovial angiogenesis, a key link in maintaining the formation of the RA pannus, is the fundamental reason for its prolonged treatment. Geniposide (GE) is an iridoid glycoside derived from Gardenia jasminoides Ellis (GJ) that shows promise as a treatment for RA, but its potential mechanism of regulating synovial angiogenesis remains unclear. PURPOSE: This study aimed to elucidate the therapeutic effect of GE on RA angiogenesis and its potential underlying mechanism and sp

A functional role for CCR6 on proallergic T cells in the gastrointestinal tract.

BACKGROUND & AIMS: CCL20 is a chemokine that regulates the homeostatic and inflammatory trafficking of leukocytes to the small intestine and regulates the development of the gastrointestinal lymphoid architecture. T cells expressing T helper cell (Th) 2 cytokines are critical for experimental food allergy, and we hypothesized that CCL20 is involved in the localization of these cells to the gut. METHODS: We evaluated the role of CCR6 in allergic diarrhea induced by sensitization and oral challenge with ovalbumin (OVA) using CCR6(+/+) and CCR6(-/-) mice. RESULTS: CCR6(-/-) mice were protected

First-line disease-modifying therapies in paediatric multiple sclerosis: a comprehensive overview.

Paediatric multiple sclerosis (MS) is defined as the onset of MS before the age of 18 years. Immunomodulatory disease-modifying therapies (i.e. the interferons [IFNs] and glatiramer acetate) are considered first-line treatments in adult patients with MS, but they are largely understudied in the paediatric population. IFNβ is a type 1 IFN produced by fibroblasts. The therapeutic effect achieved by IFNβ in MS is believed to be the result of a variety of mechanisms, including the inhibition of T-cell proliferation and a shift in cytokine production. There are currently two forms of recombinant IF
