Rapid genetic analysis of oculocutaneous albinism (OCA1) using denaturing high performance liquid chromatography (DHPLC) system.

OBJECTIVES: To present the prenatal genetic diagnoses and counseling for two cases of oculocutaneous albinism (OCA) type I family by detection of mutations in the OCA1 gene by denaturing high performance liquid chromatography (DHPLC) system and a review of the literature. METHODS: All DNA samples were extracted from peripheral whole blood and amniocentesis-derived cells. Mutation analysis was performed for all five coding exons of the TYR gene, which were amplified by PCR. DHPLC was used for heteroduplex detection and sequence analysis was performed to demonstrate the mutation loci. RESULTS:

Novel NHLRC1 mutations and genotype-phenotype correlations in patients with Lafora's progressive myoclonic epilepsy.

BACKGROUND: Lafora's progressive myoclonic epilepsy (Lafora's disease) is an autosomal recessive neurodegenerative disorder characterised by the presence of polyglucosan intracellular inclusions called Lafora bodies. Mutations in two genes, EPM2A and NHLRC1, have been shown to cause the disease. A previous study showed mutations in the EPM2A gene in 14 Lafora's disease families and excluded the involvement of this gene in five other families who were biopsy proven to have the disease. OBJECTIVE: To relate the genetic findings to the clinical course of the disease. METHODS: As part of an ongo

Genetic risk factors for inhibitors to factors VIII and IX.

The formation of alloantibodies against factor VIII (FVIII) or factor IX (FIX) is the most severe complication of replacement therapy in patients with haemophilia. In the last decade, genetic factors have been shown to constitute a decisive risk determinant for the development of inhibitors. In severe haemophilia A and B, mutations that result in an absent or truncated FVIII/FIX protein are associated with a 20-80% risk of inhibitor formation. In mild to moderate haemophilia, missense mutations represent the main mutation type, with an inhibitor prevalence of 5%. These patients synthesize some

Association of ENPP1 gene polymorphisms with hand osteoarthritis in a Chuvasha population.

Periarticular calcification is a common attendant symptom of generalized arterial calcification of infancy, a rare Mendelian disorder caused by mutations of the gene coding for ectonucleotide pyrophosphatase/phosphodiesterase 1 (ENPP1). This prompted us to perform a family-based association study to test the hypothesis that genetic variation at the ENPP1 locus is involved in the etiology of osteoarthritis of the hand. The study population comprised 126 nuclear families with 574 adult individuals living in small villages in the Chuvasha and Bashkirostan autonomies of the Russian Federation. The

[Genetics of endocrine tumours].

Major advances have been made in the understanding of the genetic mechanisms underlying endocrine tumorigenesis, through the study of several syndromes of genetic predisposition and the identification of the genes involved. The syndrome of type 1 multiple endocrine neoplasia (MEN-1) is one of the best known; this autosomal dominant hereditary syndrome predisposes to the development of endocrine tumors of the pituitary, the parathyroids, the foregut and the adrenals. The responsible gene, known as MEN-1, encodes an original protein, menin, involved in several major cellular functions, such as t

Molecular diagnosis of Huntington disease in Portugal: implications for genetic counselling and clinical practice.

Huntington disease (HD) is a neurodegenerative, autosomal dominant disorder of late-onset, caused by the expansion of a CAG repeat in the coding region of the gene. Ours is the reference laboratory for genetic testing in HD, in Portugal, since 1998; 90.1% of all 158 families known were identified for the first time, including patients with unusual presentation or without family history. A total of 338 genetic tests were performed: 234 for diagnosis, 96 for presymptomatic and four for prenatal testing (four were done for family studies). Most referring physicians were neurologists (90.6%); 82.8

Genetics of the hydrophilic surfactant proteins A and D.

The use of candidate genes has increased the ability to identify genetic factors involved in diseases with complex and multifactorial etiology. The surfactant proteins (SP) A and D are involved in host defense and inflammatory processes of the lung, which are often components of pulmonary disease. Therefore, the SP-A and SP-D genes make particularly good candidates to study factors contributing to pulmonary disease etiopathogenesis. Moreover, SP-A also plays a role in the surface tension lowering abilities of pulmonary surfactant, which is essential for normal lung function. Although genetic v

The genetic basis of xeroderma pigmentosum.

The chemical integrity and proper functioning of DNA is threatened by numerous chemical and physical agents that cause a wide spectrum of DNA lesions. When unrepaired, DNA injury interferes with vital, cellular functions such as DNA replication and transcription and give rise to mutations leading to genetic defects, carcinogenesis and cell death. The contribution of DNA repair systems in preventing cancer is apparent from the high rate of tumorigenesis found in many repair syndromes. A classical example is the excision repair disorder xeroderma pigmentosum (XP) in which patients exhibit hypers

Targeted ablation of alpha-crystallin-synthesizing cells produces lens-deficient eyes in transgenic mice.

Genetic ablation techniques were used to study the role of the lens in mammalian eye development. Ablation was accomplished by microinjecting murine eggs with chimeric DNA constructs in which the alpha A-crystallin gene regulatory sequence (-366 to +46) was fused to the highly cytotoxic diphtheria toxin gene coding sequence. For genetic ablation to be successful the promoter regulating expression should be specific and completely silent in cells necessary for normal mouse development. In this report, we describe the generation and analysis of transgenic mice with this readily discernible pheno

Genomic spectrum of congenital heart disease combined with kidney and urinary tract anomalies uncovered by exome sequencing and array-CGH.

Congenital heart disease (CHD) combined with anomalies of the kidney and urinary tract (CAKUT) belongs to severe clinical conditions that represent isolated cases or can be part of complex inherited syndromes. To date, knowledge of the genetic basis of cardio-renal birth defects is very limited. Using chromosomal microarray analysis and exome sequencing, we investigated the spectrum of copy number variations (CNVs) and point genetic variants in a pediatric cohort of 30 patients presenting with combined CHD and CAKUT. Two patients possessed a pathogenic deletion in the 22q11.2 genomic region, w

MTHFR C677T polymorphism and T2DM risk in Iraqi Kurds: a cross-sectional study.

BACKGROUND/OBJECTIVES: Type 2 diabetes mellitus (T2DM) represents a major global health problem, and the methylenetetrahydrofolate reductase (MTHFR) C677T polymorphism has been repeatedly linked to T2DM susceptibility. However, no prior study has investigated this association in the Iraqi Kurdish population. This study aimed to examine the relationship between the MTHFR C677T polymorphism and T2DM in a sample of Iraqi Kurdish patients, and to evaluate its effects on metabolic parameters, lipid profiles, and the incidence of diabetic complications. Phylogenetic analysis blaTEM gene of Escherichia coli isolated from cave bats in West Nusa Tenggara Province, Indonesia.

BACKGROUND: The blaTEM, blaCTX-M, and blaSHV are the result of mutations in the extended-spectrum beta-lactamases (ESBL) enzyme, which can be seen genotypically. Wild bats have never received antibiotic therapy, and the presence of antimicrobial-resistance (AMR) in bats indicates the level of AMR in the environment, the circulation of ESBL Escherichia coli between humans or animals in Indonesia and wild animals is still not widely known. Whole genome sequencing is used to determine the circulation of ESBL Escherichia coli in detail. AIM: Our research aims to determine the genetic level of rel

Detection and Genetic Diversity of Bovine Kobuvirus in Diarrheic Calves in Türkiye: Genomic Characterization and Comparative B-Cell Epitope Analysis.

Kobuviruses infect a wide range of hosts and exhibit high genetic diversity, highlighting the need for further studies to better understand their evolution and impact on animal health. In the present study, bovine kobuvirus (BKoV) was detected in 40 of 154 fecal samples from calves using the primers targeting the 3D gene region, corresponding to an overall positivity rate of 25.97%. All positive samples were obtained from diarrheic calves younger than 30 days of age, (40/133; 30.08%), consistent with previous studies. To enable more reliable genotyping, the complete VP1 gene sequences of seven

An intersectional expression platform for gene complementation using RNA-fragment end joining (REJ).

UNLABELLED: Gene complementation is a powerful tool for genetic selection and protein functional studies but typically requires extensive screening for complementary components. Here we engineered a platform for gene complementation based on RNA end-joining (REJ) that precisely and efficiently splices separate RNA units into functional coding mRNAs. REJ is mediated by short structured modular RNA segments of ∼250bp that: (a) promote RNA::RNA interaction, (b) coopt the intrinsic cell splicing machinery to facilitate RNA trans-splicing, (c) minimizes translation of protein fragments from un-spli

TET2 promotes monocyte inflammatory activation in asthma via ALKBH5-m6A regulation and PI3K signaling: evidence from m6A-SNP and single-cell analyses.

Asthma is a complex inflammatory airway disease with strong genetic determinants, yet the functional relevance of most asthma-associated non-coding variants remains unclear. Emerging evidence suggests that N6-methyladenosine (m6A) modification may serve as a critical epitranscriptomic link between genetic variation and immune regulation. In this study, we aimed to systematically identify functionally relevant m6A-regulated genes in asthma by integrating large-scale GWAS data, m6A-SNP annotations, and single-cell transcriptomic analyses, and to investigate their roles in monocyte-driven airway

Genome-wide assessment of rare protein-coding variants identifies associations with non-syndromic cleft lip/palate.

Nonsyndromic cleft lip and palate (NSCLP) is a complex, multifactorial condition for which only about 25% of the genetic contribution has been identified. Although more than 60 genetic loci have been associated with NSCLP, a large portion of its heritability remains unexplained. Rare variants-often with stronger functional effects-are believed to account for some of this missing genetic risk. In this study, short-read whole-genome sequencing data from 786 NSCLP cases and 2149 controls were analyzed to identify genes enriched for rare protein-coding variants. Gene-based association testing, met

Biomarkers for Treatment Response in Orthodontics: Molecular Mechanisms, Clinical Utility, and Future Directions.

Orthodontic tooth movement (OTM) is a biologically driven process resulting from the mechanically induced remodeling of the periodontal ligament (PDL) and alveolar bone. A marked inter-individual variability exists in the rate of tooth movement, susceptibility to adverse outcomes such as external apical root resorption (EARR), and overall treatment response. This narrative review synthesizes current evidence on molecular, genetic, and epigenetic biomarkers that underline these differences. We summarize established local biomarkers derived from gingival crevicular fluid and saliva, including in

Factors contributing to the underdiagnosis of hereditary transthyretin amyloidosis (hATTR) in Black patients.

Hereditary transthyretin amyloidosis (hATTR) is a progressive, multisystemic, and life-threatening disease that disproportionately affects individuals of African descent, with an estimated prevalence of 3.4% of Black Americans. hATTR is often mis- or underdiagnosed, partially because many of its symptoms overlap with other cardiac conditions. This study highlights additional factors that may be contributing to the underdiagnosis of hATTR in Black patients. Participants were ascertained from the Emory University Amyloidosis Clinic with purposive sampling. A total of 11 interviews were conducted

Genomic analyses identify 15 risk loci and reveal HDAC2, SOX2-OT, and IGF2BP2 in a naturally occurring canine model of gastric cancer.

Gastric cancer ranks as the fifth most common human cancer worldwide and has a poor survival rate and limited treatment options. Despite the high prevalence and mortality rate, the genetic etiology is largely unknown. In dogs, a clinically and histologically similar disease disproportionately affects two breeds, the Belgian Tervuren and Belgian Sheepdog, which develop the intestinal and diffuse tumor subtypes observed in humans. We performed a Bayesian genome-wide association study and selection analyses in this naturally occurring canine model to elucidate underlying genetic risk factors for

A versatile genetic toolkit for engineering Wickerhamomyces ciferrii for tetraacetyl phytosphingosine production.

Wickerhamomyces ciferrii: a non-conventional yeast with significant industrial potential for tetraacetyl phytosphingosine (TAPS), remains underutilized due to the lack of a comprehensive genetic toolbox. In this study, we developed a modular genetic system tailored for Wickerhamomyces ciferrii to enable strain engineering and metabolic pathway optimization. This toolkit includes episomal plasmids incorporating multiple selectable markers, replication origins, and fluorescent reporters. Systematic evaluation of four antibiotic resistance markers demonstrated that nourseothricin, geneticin, and
