Strain typing of acetic acid bacteria responsible for vinegar production by the submerged elaboration method.

Strain typing of 103 acetic acid bacteria isolates from vinegars elaborated by the submerged method from ciders, wines and spirit ethanol, was carried on in this study. Two different molecular methods were utilised: pulsed field gel electrophoresis (PFGE) of total DNA digests with a number of restriction enzymes, and enterobacterial repetitive intergenic consensus (ERIC) - PCR analysis. The comparative study of both methods showed that restriction fragment PFGE of SpeI digests of total DNA was a suitable method for strain typing and for determining which strains were present in vinegar ferment…

Energy metabolism of a unique acetic acid bacterium, Asaia bogorensis, that lacks ethanol oxidation activity.

Acetic acid bacteria (AAB) are known as a vinegar producer on account of their ability to accumulate a high concentration of acetic acid due to oxidative fermentation linking the ethanol oxidation respiratory chain. Reactions in oxidative fermentation cause poor growth because a large amount of the carbon source is oxidized incompletely and the harmful oxidized products are accumulated almost stoichiometrically in the culture medium during growth, but a newly identified AAB, Asaia, has shown unusual properties, including scanty acetic acid production and rapid growth, as compared with known AA…

Effect of metal ions on decomposition of chlorinated organic substances by ozonation in acetic acid.

The objective of this study is to find metal ions that enhance the ozone decomposition of chlorinated organic substances in acetic acid. Although the pseudo-first order degradation rate constant for 2,4-DCP by ozone in acetic acid in addition of Ca2+, Mg2+, Al3+ and Fe2+ were almost the same as that with no metal ion, the degradation rate in addition of Mn2+ and Fe3+ were 2.4 and 4.5 times as high as that with no metal ion, respectively. The presence of Fe3+ enhanced the degradation of 2,4-DCP by ozone in acetic acid because Fe3+-phenolate complex which have high reactivity with ozone was prod…

NMR analysis of the iron ligand ethylenediaminedi(o-hydroxyphenyl)acetic acid (EDDHA) employed in fertilizers.

The exceptional efficiency of the iron chelate of ethylenediaminedi(o-hydroxyphenyl)acetic acid (o,o-EDDHA) in correcting iron chlorosis in plants and the medical applications of various metallic chelates of this compound have long been recognized. As commercial preparations of o,o-EDDHA usually contain impurities, a method for their detection is proposed. By using one- and two-dimensional nuclear magnetic resonance two impurities were identified. The structure of one of these compounds was assigned to an isomer of EDDHA containing at least one p-hydroxyphenyl moiety. The structure of the othe…

Mass spectrometric identification and quantification of endogenous 5-methoxyindole-3-acetic acid in quail tissues.

The endogenous levels of 5-methoxyindole-3-acetic acid (5-MIAA) in quail pineal gland, retina and serum were determined by capillary column gas chromatography/mass spectrometry/selected ion monitoring using a deuterated internal standard and the N-pentafluoropropionyl-O-pentafluorobenzyl ester derivative. Diurnal rhythms of pineal and serum 5-MIAA with high levels at mid-light and low levels at mid-dark suggest a pineal origin of 5-MIAA in the blood. In contrast, retina displayed an opposite diurnal rhythm of 5-MIAA with high levels at night, suggesting that the biosyntheses of 5-MIAA in the q…

Biosynthesis of indole-3-acetic acid via the indole-3-acetamide pathway in Streptomyces spp.

Various Streptomyces spp. including S. violaceus, S. scabies, S. griseus, S. exfoliatus, S. coelicolor and S. lividans secrete indole-3-acetic acid (IAA) when fed with L-tryptophan (Trp). Production of IAA was detected in Streptomyces strains causing potato scab as well as in non-pathogenic strains. The pathways for IAA synthesis from Trp were investigated in S. violaceus and S. exfoliatus. Indole-3-acetamide (IAM), indole-3-lactic acid (ILA), indole-3-ethanol (IEt) and IAA were identified by HPLC and GC-MS. Streptomyces cells were capable of catabolizing IAM, ILA, IEt and indole-3-acetaldehyd…

Acidic fibroblast growth factor accelerates the healing of acetic-acid-induced gastric ulcers in rats.

Acidic fibroblast growth factor (aFGF) was evaluated for the healing of acetic-acid-induced gastric ulcers in rats. The effect of aFGF on angiogenesis in the gastric ulcer bed was determined by the carmine dye infusion method, while its effect on gastric acid secretion was assessed in chronic gastric fistula rats. Oral treatment with aFGF, in the presence of heparin, reduced (ED50 value = 30.2 micrograms/kg/day) the acetic-acid-induced gastric ulcer area, when assessed 1 week later. aFGF was about 1,333-fold more potent than famotidine for healing such ulcers. At a dose of 200 micrograms/kg/da

Misoprostol attenuates acetic acid-induced increases in mucosal permeability and inflammation: role of blood flow.

The objectives of this study were 1) to quantify the effects of misoprostol (Miso; prostaglandin E1 analogue) on acetic acid-induced increases in mucosal permeability and inflammation; 2) to determine what effect acetic acid, Miso, or the combination of Miso plus acetic acid has on colonic blood flow; and 3) to assess whether the protective effect of Miso may be attributable to its vasodilatory properties. We found that intrarectal administration of acetic acid produced a 6.4-fold increase in colonic myeloperoxidase activity (an index of granulocyte infiltration), an 8.2-fold increase in mucos…

Effects of trimoprostil on healing and recurrence of acetic acid-induced gastric ulcer in rats.

Chronic gastric ulcers were produced by injection of 20% acetic acid (0.05 ml) into the submucosal layer of the rat stomach in order to determine the effects of the prostanoid trimoprostil on the healing and recurrence of ulcers. Local injection of acetic acid solution produced large demarcated ulcers in all animals on day 5, which rapidly decreased to reach low levels on days 40-80 and then became exacerbated on day 100. The exacerbation of the ulcer is probably recurrence. Trimoprostil was administered ad libitum in drinking water containing 0.1, 0.3 and 1.0 microgram/ml (average dose 12.4, …

A morphologic study of the behavior of biogenic amines in experimental acetic acid gastric ulcer.

Morphological observations were made of the behavior of biogenic amines in the gastric wall of rats with acetic acid-induced ulcer. In the normal rat stomach, abundant adrenergic fibers were seen in the adventitia of arteries and arterioles, with frequently distributed mast cells in their environs, in all gastric wall layers. Mast cells had a more frequent distribution in the antral region than in the corpus ventriculi while enterochromaffin-like cells (EC-like cells) were found with greater frequencies in the latter region of gastric wall. Adrenergic fibers were abundant around blood vessels …

[Dynamics of acid-base equilibrium of the blood in humans a long-term exposure to an atmosphere containing acetic acid vapors].

Two experimental series, each of 20 days in duration, were carried out using 8 healthy test subjects, aged 25 to 44 years. The purpose of the experiments was to identify the effect of two different concentrations of acetic acid vapors in an enclosed environment on blood acid-base equilibrium and gases. In both experimental series ambient temperature was increased to 33 degrees C during the last three days of exposure in order to better evaluate the acetic acid vapor effect. The most significant changes in acid-base equilibrium were detected when the vapor concentration amounted to 25 mg/m3.

Effect of Acetic Acid and Lactic Acid at Low pH in Growth and Azole Resistance of Candida albicans and Candida glabrata.

Successful colonization of the acidic vaginal niche by C. glabrata and C. albicans depends on their ability to cope with the presence of lactic and acetic acids produced by commensal microbiota. As such, the inhibitory effect of these acids at a low pH in growth of C. glabrata and C. albicans was investigated. The effect of the presence of these organic acids in tolerance of the two Candida species to azoles used in treatment of vaginal infections was also investigated including eventual synergistic effects. Under the different experimental conditions tested lactic acid exerted no significant …

Cell Death Triggered by the YUCCA-like Bs3 Protein Coincides with Accumulation of Salicylic Acid and Pipecolic Acid But Not of Indole-3-Acetic Acid.

The pepper (Capsicum annuum) resistance gene bacterial spot3 (Bs3) is transcriptionally activated by the matching Xanthomonas euvesicatoria transcription-activator-like effector (TALE) AvrBs3. AvrBs3-induced Bs3 expression triggers a rapid and local cell death reaction, the hypersensitive response (HR). Bs3 is most closely related to plant flavin monooxygenases of the YUCCA (YUC) family, which catalyze the final step in auxin biosynthesis. Targeted mutagenesis of predicted NADPH- and FAD-cofactor sites resulted in Bs3 derivatives that no longer trigger HR, thereby suggesting that the enzymatic
