The effect of intestinal trichinellosis on oral bioavailability of albendazole in mice.

The effect of Trichinella spiralis infection on the pharmacokinetic profile of orally administered albendazol has been investigated in mice during the intestinal phase of the disease. Swiss CD-1 mice were orally infected with 300 +/- 50 muscle larvae of T. spiralis and then treated with albendazole (ABZ) formulated in hydroxy-propyl-beta-cyclodextrins at the dose of 10 mg/kg given orally on days 0, 5, 10 and 22 post-infection (p.i.). Blood samples were taken at 0.25, 0.5, 0.75, 1, 5, 6 and 24 h post-treatment (p.t.). Adult worm establishment as well as the histopathological alterations induced…

Preoperative albendazole and scolices viability in patients with hepatic echinococcosis.

To determine the plasmatic and intracystal concentrations of albendazole sulfoxide (AS) and correlate them with the viability of the scolices in patients surgically treated for hepatic hydatid cysts (HHC) that received albendazole preoperatively, as an indirect way of evaluating the scolicide efficacy of the drug. A non-consecutive series of patients with uncomplicated HHC, underwent operation at the Department of Surgery, Regional Hospital of Temuco, Chile, between 2001 and 2002. The patients were given 10 mg/kg/day of albendazole for 4 days prior to the surgery. Intraoperative samples of ven…

Placental and fetal toxicity of albendazole sulphoxide in Wistar rats.

This work characterized the effects of albendazole sulphoxide (ABZSO) on placental and fetal parameters in Wistar rats on day 20 of gestation. ABZSO was fed in laboratory chow at 0, 2.5, 5, 10, 20 or 30 mg/kg/d from day 6 to 15 of gestation to pregnant rats. Data of resorptions, placental and fetal characteristics and fetal skeletal malformations were recorded. Resorption percentages in the 20 and 30 mg/kg/d groups were significantly higher compared to the control group. Placentas of ABZSO-treated rats had lower weight and smaller size than untreated rats. The fetal weight and size were lower …

The pharmacokinetics, safety and tolerability of the co-administration of diethylcarbamazine and albendazole.

The pharmacokinetics, safety and tolerability of single, oral doses of diethylcarbamazine (DEC) and albendazole, given alone or in combination, were investigated in a double-blind, randomized and placebo-controlled trial involving 42 amicrofilaraemic subjects living in an area of India where lymphatic filariasis is endemic. The subjects (34 males and eight females, aged 18-52 years and weighing 46-66.5 kg) were randomly allocated to one of the three drug groups. Fourteen were given just DEC (6 mg/kg), another 14 were given just albendazole (400 mg) and the remaining 14 were given both DEC (6 m…

Quantitative determination of albendazole and its main metabolites in plasma.

Three different and complementary chromatographic methods are described for quantitative determination of albendazole (ABZ) and its two main metabolites: albendazole sulphoxide (SOABZ) and albendazole sulphone (SO2ABZ). ABZ, SOABZ and SO2ABZ can be quantified by two RP-HPLC methods with an ODS2 column and two different mobile phases. One of methanol-water (60:40) for ABZ and a second one of phosphoric acid in water-acetonitrile (80:20) for SOABZ and SO2ABZ. SOABZ bears an asymmetric sulphur centre. Quantitative assay of (+) SOABZ and (-) SOABZ can be performed by HPLC. A chiral AGP column and …

Discrepancy between in vitro and in vivo antifungal activity of albendazole.

Albendazole has in vitro activity against Cryptococcus neoformans and reduced in vitro activity for albendazole when compared with Candida albicans. The major metabolite of albendazole, albendazole sulphoxide showed no in vitro activity against isolates of either fungus. Immunocompetent mice infected intravenously (i.v.) with C. albicans were treated with albendazole doses of 20-600 mg kg-1 per day in noble agar or sesame oil for per oral (PO) administration, or 80 mg kg-1 per day in DMSO for intraperitoneal (i.p.) and i.v. administration for 10 days, and were observed for survival. Mice infec…

Albendazole versus thiabendazole as therapy for trichinosis: a retrospective study.

An outbreak of trichinosis caused by ingestion of horse meat occurred in December 1993 in France; more than 500 people were affected. We compared the immediate and midterm efficacy and tolerability of thiabendazole and albendazole as therapy for the 46 patients seen in our department. Forty-four patients (96%) were treated. The first 26 patients received thiabendazole therapy; the next 18 received albendazole therapy. All the patients were tested with prednisone. Eight relapses occurred (seven in the thiabendazole group and one in the albendazole group [not significant]). Side effects of treat…

[Computerized tomography of liver in alveolar echinococcosis treated with albendazole].

Eleven cases of alveolar echinococcosis were examined by using computerized tomographic (CT) scanning of liver 2-7 years after long-term continuous albendazole therapy. In seven cases the hepatic lesion was almost completely calcified and cured in three cases incomplete calcification of the peripheral margin of the hepatic lesion was observed. CT scanning of the remaining in effective case showed active lesion with heterogenous hypodense areas in the liver without calcification on the peripheral margin. Long-term continuous albendazole therapy can be lethal to the parasite.

Effects of albendazole on Entamoeba histolytica and Giardia lamblia trophozoites.

Albendazole, a benzimidazole carbamate commonly used for the treatment and control of intestinal helminthic infections, is also useful for the treatment of giardiasis. Therefore, it is of interest to determine whether the drug has activity against other intestinal protozoa, such as E. histolytica. The present results demonstrate that albendazole inhibits the growth of E. histolytica trophozoites in axenic cultures and induces fine structural changes such as polyribosome aggregation and loss of cytoplasmic vacuoles at concentrations up to 10 micrograms/ml. The viability of E. histolytica tropho…

Effect of albendazole on human hydatid cysts: an ultrastructural study.

Five patients with hepatic (3), pelvic (1) or spinal (1) hydatid cysts received 10 mg/kg/d albendazole for 1-3 months prior to surgery. Daughter cysts were present in the spinal hydatid and in one patient with hepatic disease. Electron microscope examination of the cyst tissue of the pelvic and the 2 hepatic cysts lacking daughter cysts showed no evidence of germinal layer, and the protoscoleces were dead. The primary cyst of the hepatic hydatid with daughter cysts (1 month therapy) was also judged dead but some pieces of the daughter cyst germinal layer appeared normal and had unaffected prot…
